Abstract:Objective: to evaluate the therapeutic effect of targeted endostatin-loaded microbubbles, combined with improved, focused, directional ultrasound radiation for inhibition of subcutaneous translocation in situ colon tumor angiogenesis in colon cancer. Methods: 65 BALB/c nude mice with subcutaneous translocation in situ colon tumors were randomly divided into five groups. Group A was the control group, without any treatments. In Group B, the mouse was treated with ultrasonic radiation. In Group C, the mouse was treated with ultrasonic radiation combined with empty SonoVue microbubbles. In Group D, the mouse was treated with ultrasonic radiation combined with empty Targestar-SA microbubbles. In Group E, the mouse was treated with ultrasonic radiation combined with endostatin microbubbles. The tumor size was measured before and 1, 14, and 28 days after irradiation. The peak intensity (PI), regional blood volume (RBV) and regional blood flow (RBF) were recorded using contrast-enhanced ultrasound. The tumor tissue was removed for pathological examination; the tumor necrosis area and microvascular density (MVD) were evaluated by immunohistochemistry. Results: Tumors in Groups C, D and E were significantly smaller than in Groups A and B at 28 days after irradiation, with Group E the smallest. PI, RBF and RBV of Groups C, D, and E were significantly decreased 28 days after radiation with Group E the lowest, and significantly lower than Groups A and B (all P < 0.05). The tumor tissue necrosis area of Group E was clearly greater while MVD was obviously lower than the other groups (all P < 0.01) at 28 days after treatment. Conclusion: The targeted endostatin microbubbles, combined with focused, directional ultrasound radiation can damage tumor microvasculature of subcutaneous colon translocation in situ colon cancer, as well as inhibit the tumor angiogenesis.

Abstract:The purpose of this study was to investigate the effect of the angiogenesis inhibitor Endostar on carotid plaque neovascularization in patients with non-small cell lung cancer (NSCLC) using contrast-enhanced ultrasound (CEUS). Ninety-one patients who had NSCLC with soft carotid plaques were selected for treatment either with the NP regimen (vinorelbine + cisplatin) (43 patients) or with the ENP regimen (Endostar + NP) (48 patients). Plaque thickness and neovascularization of the plaque were assessed before and at 1 month after treatment using CEUS. Enhanced intensity (EI) of CEUS was used for quantification of plaque neovascularization. There was no significant changes in any group in thickness of plaque between recruitment and 1 month after treatment (P > 0.05 for all). There was no significant change in the EI of plaque in the controls or NP groups at 1 month after treatment (P > 0.05), while EI in the ENP group was significantly reduced at 1 month after treatment (P < 0.01) and significantly lower than that in the controls or NP group at 1 month after treatment (P < 0.001 both). This study indicates that carotid soft plaque neovascularization in patients with NSCLC can be reduced by anti-angiogenesis treatment.

Abstract:Objective To analyze the inhibitory effect of recombinant human Endostatin on tumor growth and the proliferation of endothelia cells.Methods Xenotransplanted nude mice model with human cancer and experimental implanted tumor mice model were used to evaluate the anti-tumor activity of rh-Endostatin.The cyto-toxicity drugs were used at the same time to compare with rh-Endostatin.MTT assay was applied to analyze the anti-proliferation activity of rh-Endostatin on human embryo umbilical cord vascular endothelial cell ECV and connective tissue fibroblast cell L929.Result rh-Endostatin was shown to have obvious anti-tumor effect on Xenotransplanted nude mice model with human hepatic cancer QGY,lung cancer A549 and breast cancer Bcap37 and on experimental implanted tumor mice model: Lewis lung cancer.MTT assay showed the inhibition of rh-Endostatin on the proliferation of ECV and L929.Conclusion rh-Endostatin was shown to have anti-tumor function on Xenotransplanted nude mice model with human cancer and implanted tumor mice model,and the mechanism may lie in the inhibition of forming new blood vessel in tumors.

Abstract:Purpose To study the effects of human recombinant human endostatin(rh-endostatin) on the microenvironment of human lung adenocarcinoma.Methods BALB/c nude mice were subcutaneously implanted with human adenocarcinoma cells and given rh-endostatin 5mg/kg s.c.daily for 14 days when tumor grew to about 100mm~3,with normal saline was administered as control.Seven days,14 days,and 21 days after the administration of rh-endostatin,the mice were sacrificed 30 minutes after intravenous injection of TRITC-Dextran(155kD) 50mg/kg.Tumor samples were excised for the examina- tion by fluorescence microscopy.The ratios of areas in the solid tumor where there was little TRITC - Dextran diffused were calculated.Results Fourteen days after administration of saline,the blood diffusion in the solid tumor was significantly lower than that on day 7 and day 21(10.01%±4.50%versus 1.49%±1.14%and 2.90%±2.02%).Rh-endostatin suppressed the blood diffusion at the tumor at the beginning of the treatment(day 7,the ratio of areas where there was little fluorescein distributed was 16.8%±4.02%).However,the blood diffusion increased by rh-endostatin 14 days after the treatment(P= 0.000) and there was no difference of blood diffusion between the two groups on day 21.Conclusion Rh-endostatin can transiently inhibit the blood diffusion in human adenocarcinoma.Furthermore,it can modulate the microenvironment by increasing the blood diffusion in a short period.

Abstract:It has been widely accepted that antiangiogenesis therapy could deprive tumor cells of nutrients and oxygen and suppress tumor growth. However, in the present study, Lewis lung carcinomas and A549 adenocarcinomas established in male C57BL/6 and BALB/c nude mice, respectively, were treated with recombinant human endostatin (rh-endostatin). Earlier studies document discrepancies in the antiangiogenic and antitumor outcomes of rh-endostatin treatment, at doses equivalent to clinical usage. Although there was no significant regression of tumor growth, tumor vasculature was widely disrupted within the first few days of treatment with rh-endostatin, as indicated by reduced blood perfusion (visualized by dynamic-contrast-enhanced magnetic resonance imaging) and reduced microvascular density. Interestingly, when rh-endostatin treatment was discontinued, there was an elevation in the diffusion of oxygen and tetramethylrhodamine isothiocyanate-dextran in both tumor classes, which was detected by hypoxyprobe (pimonidazole) and fluorescence microscopy. We conclude that the paradoxic outcomes in the antiangiogenic and antitumor properties of rh-endostatin might derive from the tumors' tolerance to antiangiogenesis inhibitors. Additionally, rh-endostatin might have the ability to transiently normalize tumor vasculature.

Abstract:Purpose Endostatin can normalize the tumor vasculature to some extent. However, exact length of its time window and corresponding markers for tumor vascular normalization are needed to be explored. Methods The A549 lung adenocarcinoma xenograft murine model was treated with recombinant human endostatin (rh-endostatin) for 14 days. Cisplatin was combined in different schedules. The effects of rh-endostatin on circulating endothelial cells (CECs) by flow cytometry, tumor vasculature and angiogenesis-related factors by confocal microscope and immunohistochemistry, and anti-tumor efficacy of cytotoxic drugs were observed. Results The activated CECs (aCECs) were increased on day 7 and decreased on day 10, and apoptotic CECs were increased on day 10. Tumor vasculature was transiently normalized with increased collagen coverage, decreased vessel permeability, intratumoral hypoxia, and microvascular density from day 7 to 10 after rh-endostatin administration. Extracellular matrix metalloproteinase inducer, vascular endothelial growth factor, matrix metalloproteinase (MMP)-2, and MMP-9 were transiently decreased by rh-endostatin from day 4 to 10, whereas the opposite effects were observed with tissue inhibitors of matrix metalloproteinase (TIMP)-1 and TIMP-2. The maximal anti-tumor effects of cisplatin were observed on administration from day 5 to 9 after rh-endostatin initial administration. Conclusions Rh-endostatin could transiently normalize tumor vasculature, probably via regulation of both pro- and anti-angiogenesis factors. The synergistic efficacy of anti-angiogenesis and chemotherapy was found during “the normalization window”. CEC could be a feasible blood biomarker for defining “vascular normalization window” and providing the evidence to make an optimizing combination therapeutic schedule in human tumor.

Abstract:OBJECTIVE:To assess the effect of endostatin on growth and neoplastic angiogenesis in transplanted human lung adenocarcinoma Calu-6 tumor in nude mice.METHODS:To treat Calu-6 tumor-bearing mice with endostatin at different doses, and to record the changes of the tumor size. The expressions of survivin, VEGF, COX-2 and MVD in tumor tissue were examined by immunohistochemistry staining, circulating endothelial cells (CECs) by flow cytometry and mRNA of CD146 and CD105 by RT-PCR and real-time PCR.RESULTS:After endostatin treatment, the tumor size was conspicuously shrunk, and the expressions of survivin, COX-2 and VEGF protein and MVD in tumor tissue decreased concomitantly with the significant difference between each of trial groups and control group (all P < 0.05). Both CECs and mRNA of CD146 and CD105 diminished remarkably. A positive correlation between both exhibition and change of amount of activated CECs and survivin, VEGF expression and MVD count in tumor tissue was found.CONCLUSION:Endostatin can decrease the expression of survivin, COX-2, VEGF and MVD, and to inhibit the growth of transplanted tumor. Activated CECs may probably serve as an ideal marker to predict the efficacy and prognosis of anti-angiogenesis therapy.

Abstract:Endostatin is an endogenous inhibitor of angiogenesis and has been shown to exhibit potent inhibitory activity in certain mice tumor models. In this study, a treatment strategy of combining recombinant human endostatin (rhEndostatin) and chemotherapeutics was implemented to evaluate the therapeutic efficacy of rhEndostatin against solid tumors. The antitumor effect of rhEndostatin in combination with several chemotherapeutic drugs, e.g., 5-fluorouracil, cyclophosphamide, methotrexate, and mitomycin C, on human QGY liver tumor and mice H22 liver tumor was compared with that of rhEndostatin treatment alone. The results showed that the combination of rhEndostatin and chemotherapeutic drugs resulted in a more potent inhibition of tumor growth. The potential advantages of rhEndostatin plus tumor chemotherapy provide a basis for further clinical trials of rhEndostatin.

Abstract:Objective:To evaluate antiangiogenic effect of endostar or endostar combined with avastin in a xenograft model of human lung cancer using dynamic contrast-enhanced magnetic resonance imaging(DCE-MRI).Methods:18 nude mice,which were inoculated subcutaneously with human lung adenocarcinoma cells A549,were divided into three groups,control group(n=6),endostar group(n=6) and combination of endostar and avastin(n=6).Tumor angiogenesis and vascular permeability were assessed with DCE-MRI and pathology methods.Results:7 days after antiangiogenic treatment,tumor volume among the control group,the endostar group and the combination treatment group were(0.85±0.25)cm3,(0.37±0.17)cm3 and(0.16±0.03)cm3,respectively.Ktrans,Kep,Ve and iAUC among three groups were(0.11±0.03)min-1,(0.67±0.54)min-1,(0.35±0.12) and(13.28±4.25),(0.06±0.01)min-1,(0.40±0.25)min-1,(0.31±0.09) and(7.33±3.93),(0.04±0.02)min-1,(0.27±0.12)min-1,(0.36±0.19) and(4.29±3.53).Significant decreases were reached in Ktrans,Kep and iAUC after antiangiogenic treatment,(F=15.257,P=0.000 and F=8.197,P=0.004 respectively).Microvessel density(MVD) among the control group,the endostar group and the combination treatment group were(14.24±3.38)item/hpf,(8.88±1.86)item/hpf and(6.47±1.78)item/hpf,respectively.A significant decrease in MVD was noted after antiangiogenic treatment(F=41.305,P=0.000).There were statistically significant differences in VEGF expression among three groups.A significant decrease in VEGF was reached after antiangiogenic treatment.Conclusion:DCE-MRI is a non-invasive functional imaging technique that permits indirect measurement of tumor angiogenesis.It may therefore be suitable for monitoring the antiangiogenic effects of endostar combined with avastin.

Abstract:Purpose Normalization of the tumor vasculature and microenvironment by several angiogenesis inhibitors has been reported. Given that recombinant human endostatin (rh-endostatin) is also an endogenous angiogenesis inhibitor, a comprehensive evaluation of the effects of rh-endostatin on tumor vasculature and microenvironment and chemotherapy sensitivity would be favorable. Methods Multiple treatment schedules of the combination of rh-endostatin and paclitaxel were tested in Lewis lung carcinoma. Further, we monitored microvascular density, tumor hypoxic fraction, and collagen covered tumor vessels at three different time points following the treatment of rh-endostatin, as well as the transcription of angiogenesis related factors (vascular endothelial growth factor-A and thrombospondin-1) and vasculature markers (regulator of G-protein signaling 5 and platelet/endothelial cell adhesion molecule-1). Results The anti-tumor efficacy of paclitaxel was significantly improved 7 days after the treatment of rh-endostatin. Tumor microvascular density was decreased by rh-endostatin, although it became even higher 7 days after termination of rh-endostatin. Non-necrotic hypoxic fraction was significantly reduced 7 days after treatment of rh-endostatin, accompanied with increased collagen covered tumor vessels and coverage of pericytes around endothelial cells. Rh-endostatin could transiently upregulate the transcription of thrombospondin-1 and modulate the imbalance between vascular endothelial growth factor-A and thrombospondin-1. Conclusion Rh-endostatin could normalize the tumor vasculature and microenvironment in Lewis lung carcinoma probably via modulation of the balance between vascular endothelial growth factor-A and thrombospondin-1. During the time of vascular normalization, paclitaxel treatment was found to have maximal effect on tumor growth delay.
Bioactivity, Pharmacokinetics, and Immunogenicity Assays in Preclinical and Clinical Trials for Recombinant Human Endostatin

Abstract:Aim: To determine the in vitro and in vivo bioactivity of recombinant human endostatin (rhEndostatin) and to analyze its pharmacokinetics and immunogenicity in rhesus monkeys and patients. Methods: The physical chemical characteristics of rhEndostatin were detected according to Pharmacopoeia of the People's Republic of China (2005 edition, part III). Its in vitro and in vivo bioactivities were assayed via proliferation-inhibition on human umbilical vein endothelial cells and their inhibitory effect on tumor-bearing mice models. Serum concentrations of rhEndostatin in monkeys and patients were determined by an enzyme immunoassay method. Results: The corresponding specific in vitro activities of rhEndostatin obtained from the cell counting method, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, and lactate dehydrogenase assay, respectively, were 6.4x10(7), 6.7x10(7), and 3.8x10(8) U/mg, and the in vivo antitumoral potency was 4.04x10(7) U/mg. In rhesus monkeys, there were no gender differences in all pharmacokinetic parameters. Serum anti-rhEndostatin immunoglobulin (Ig)G antibodies were generated quickly after intravenous (iv) administration and decreased rapidly when therapy was stopped. In phase I clinical trials, linearity in the pharmacokinetics of rhEndostatin was indicated by dose-proportionate increases in the area under the curve and the maximum serum concentration. Serum rhEndostatin reached a steady-state level after 7 d of successive administration with the average concentration at a steady state of 272.44 +/- 91.98 ng/mL. Neither IgG nor IgM antibodies against rhEndostatin were observed in patients. Conclusion: RhEndostatin exhibited a definite proliferation-inhibition effect on HUVEC, and significant antitumoral activity in mice. The immunoreactivity of rhesus monkeys to rhEndostatin is common, and rhEndostatin showed no immunogenicity in patients in this trial. The results provide a basis for further clinical trials.
Endostatin Inhibits Hypertrophic Scarring in A Rabbit Ear Model

Abstract:ObjectiveThe present study was designed to use an in vivo rabbit ear scar model to investigate the efficacy of systemic administration of endostatin in inhibiting scar formation.MethodsEight male New Zealand white rabbits were randomly assigned to two groups. Scar model was established by making six full skin defect wounds in each ear. For the intervention group, intraperitoneal injection of endostatin was performed each day after the wound healed (about 15 d post wounding). For the control group, equal volume of saline was injected. Thickness of scars in each group was measured by sliding caliper and the scar microcirculatory perfusion was assessed by laser Doppler flowmetry on Days 15, 21, 28, and 35 post wounding. Rabbits were euthanatized and their scars were harvested for histological and proteomic analyses on Day 35 post wounding.ResultsMacroscopically, scars of the control group were thicker than those of the intervention group. Significant differences between the two groups were observed on Days 21 and 35 (p<0.05). Scar thickness, measured by scar elevation index (SEI) at Day 35 post wounding, was significantly reduced in the intervention group (1.09±0.19) compared with the controls (1.36±0.28). Microvessel density (MVD) observed in the intervention group (1.73±0.94) was significantly lower than that of the control group (5.63±1.78) on Day 35. The distribution of collagen fibers in scars treated with endostatin was relatively regular, while collagen fibers in untreated controls were thicker and showed disordered alignment. Western blot analysis showed that the expressions of type I collagen and Bcl-2 were depressed by injection of endostatin.ConclusionsOur results from the rabbit ear hypertrophic scar model indicate that systemic application of endostatin could inhibit local hypertrophic scar formation, possibly through reducing scar vascularization and angiogenesis. Our results indicated that endostatin may promote the apoptosis of endothelial cells and block their release of platelet-derived growth factor (PDGF) and fibroblast growth factor (FGF), thereby controlling collagen production by fibroblasts. Blood vessel-targeted treatment may be a promising strategy for scar therapy.

Abstract:BACKGROUND & OBJECTIVE:It was reported that endostatin could inhibit tumor angiogenesis, then inhibit the growth and metastasis of tumor. However, there was few report about the treatment usage of endostatin. This study was designed to explore the effect of endostatin mediated with recombinant adeno-associated virus(rAAV) on tumor growth and metastasis.METHODS:To obtain the endostatin gene complete cDNA by RT-PCR, and clone it into the plasmid pSNAV and package the recombinant rAAV-SS-endostatin; to analyze its anti-tumor effect through animal experiments.RESULTS:In the B16F10 melanoma tumor model, C57BL/6 mice were inoculated with 10(11)TU rAAV-SS-endostatin i.m. injection, which the inhibition rate was 57.1%; In the lung metastatic model, the inhibition rate of metastases was 70.7%.CONCLUSIONS:rAAV-SS-endostatin can effectively inhibit tumor growth and metastasis.

Abstract:Endostatin is a C-terminal fragment of collagen XⅧ with M, of 20 000. Endostatin can inhibit endothelial proliferation and migration in vitro and also can inhibit angiogenesis and tumor growth in vivo by inducing apoptosis in endothelial cells. Endostatin is nontoxic and does not induce acquired drug resistance and has therefore become a potent new therapy strategy for solid tumor.This article will summarize the latest progress in endostatin studies.

Abstract:Recombinant human endostatin (rh-endostatin), a potential antiangiogenic agent, is used in non-small cell lung carcinoma treatment and represses vascular endothelial cell growth factor (VEGF) levels in tumor cell. However, precise affection of rh-endostatin on the proangiogenic VEGF isoforms (VEGF 165 ) or antiangiogenic VEGF isoforms (VEGF 165 b) is not clear. We therefore tested the hypothesis that rh-endostatin could alter expression of these isoforms to regulate tumor growth. A549 cells were exposed to rh-endostatin, and the expression of VEGF 165 and VEGF 165 b was detected. The role of SP1 as a regulator of isoform expression was investigated. We then examined the anticancer and antiangiogenic efficacy of rh-endostatin in combination with exogenous VEGF 165 b against A549 cells, EA.HY 926 cells and xenograft model of human lung cancer. rh-Endostatin reduced VEGF 165 and induced VEGF 165 b as well as inhibited SP1 in A549 cells. SP1 inhibitor (betulinic acid) also developed those changes. VEGF 165 b–rh-endostatin combination was highly synergistic and inhibited growth, survival, and migration of A549 cells, VEGF-mediated VEGFR2 phosphorylation in EA.HY 926 cells, and tumor growth in xenograft model of human lung cancer. rh-Endostatin downregulates proangiogenic vascular endothelial growth factor A (VEGFA) isoform and upregulates antiangiogenic VEGFA isoform, possibly through inhibition of SP1. Furthermore, VEGF 165 b sensitizes A549 to rh-endostatin treatment and enhances the anticancer effect of rh-endostatin.
Effects of Recombinant Human Endostatin and Docetaxel on MMP and Its Following Anti-neoplastic Effect under Different Administration Sequences

Abstract:BACKGROUND AND OBJECTIVE:The aim of this study is to observe the changes of MMP-2 and its regulators, and to investigate the mechanism of the two administration sequences of recombinant human endostatin (rh-endostatin) and docetaxel.METHODS:The experiment was performed as 2 stages. Firstly, nude mice with xenograft tumor were randomized into 2 groups as rh-endostatin-treated group with rh-endostatin 400 microg x d(-1), d1-d14 and docetaxel-traeted group with docetaxel 10 mg x kg(-1) x 3d(-1), d1-d14. Secondly, nude mice with xenograft tumor were randomized into 3 groups as concurrent administration group (rh-endostatin 400 microg x d(-1), d1-d35, docetaxel 10 mg x kg(-1) x 3d(-1), d1-d19), endo-first group (rh-endostatin 400 microg x d(-1), d1-d35, docetaxel 10 mg x kg(-1) x 3(d-1), d16-d34) and model group (positive control, mice burdened tumor without treatment). The volume of tumor was measured during treatment. Detection of the expressions of MMP-2, TIMP-2, EMMPRIN and the count of microvessel density (MVD) by immunohistochemistry stain examination were carried out at the end of experiment.RESULTS:Compared with the docetaxel-treated group, more obvious down-regulation of expression of MMP-2, EMMPRIN (P = 0.024, P = 0.081) were observed in rh-endostatin-treated group. No significant difference was found in TIMP-2 expression between the 2 groups. In combined treatment groups, at the endpoint tumor volumes of concurrent administration group and the endo-first group were remarkably smaller than that in model group (P < 0.001, P = 0.003). According to the administration procedure, concurrent administration inhibited tumor growth stronger than endo-first treatment did. Both of the combined groups down-regulated the expression of MMP-2 and decreased microvessel density (P < 0.05). Compared with model group, the expression of TIMP-2 was upregulated (P = 0.001) as well as EMMPRIN down-regulated (P = 0.018) in concurrent adminis- tration group. Oppositely, the same results were not observed in the endo-first group.CONCLUSION:The schedule of the concurrent administration group could inhibit the tumor growth better, and it down-regulated MMP-2 expression through TIMP-2 and EMMPRIN, and thus slow down the tumor growth superiorly to another schedule of treatment.

Abstract:Background Recombinant human endostatin (rh-endostatin, Endostar) has been proved to be an inhibitor of angiogenesis. Docetaxel has been also considered as a common chemotherapeutic agent with inhibition of angiogenesis of malignancies. However, their function has been seldom compared and a best synergism protocol is not determined. This study aimed to compare the effects of two drugs, investigate their combined impact on human umbilical vein endothelial cells (HUVECs), a molecular basis and find ideal protocols to inhibit endothelial cell proliferation. Methods HUVECs on confluent growth or activated by vascular endothelial growth factor (VEGF) were treated by rh-endostatin or/and docetaxel at respective gradient concentration in following operations as cell proliferation determined by MU assay, cell cycle distribution, apoptosis and markers of CD146, CD62E and CD105 detected by flow cytometery, the structure of the channel formed by HUVECs measured by tube formation count. Results Rh-endostatin exhibited time dependent inhibition of proliferation while docetaxel showed both time and dose dependent inhibition. HUVECs accumulated in G(0)-G(1) with decreased numbers of cells in G(2) after a single treatment of rh-endostatin or that followed by docetaxel treatment. Cells accumulated in G(2) after both a single docetaxel and simultaneous administration. Both the number of cells in G(0)-G(1) and apoptotic cells were increased by docetaxel followed by rh-endostatin treatment. The number of non-apoptotic cells at G(0)-G(1) was increased by first administering rh-endostatin then docetaxel. Sequential treatment of docetaxel followed by rh-endostatin resulted in the greatest increase in apoptosis (34.7%) and the second highest apoptosis was seen with simultaneous administration (18.2%). Expression of CD146 and CD105 on confluent HUVECs was reduced at certain doses of rh-endostatin and/or docetaxel. However, rh-endostatin reduced CD105 without any apparent impact on either CD146 or CD62E expression, whereas theses. markers were down-regulated by docetaxel after pre-activation by VEGF. Rh-endostatin treatment maintained tube-like structures for a limited time. In contrast, docetaxel swiftly reduced tube formation. Simultaneous treatment, or docetaxel followed by rh-endostatin, exhibited a stronger inhibition on tube formation than either agent alone. Conclusions Both rh-endostatin and docetaxel can inhibit HUVEC proliferation while the high apoptotic rate after combined administration was probably owing to different sequent administration by docetaxel followed by rh-endostatin or simultaneous treatment. Both proliferation and adhesion molecules on HUVECs of confluent growth are down-regulated by the two drugs. The rh-endostatin decreased proliferation markers, but only slightly modified adhesion molecules, while both markers were down-regulated by docetaxel on HUVECs activated by VEGF. Rh-endostatin could maintain adhesion of HUVECs at first then induce cells apoptosis to damage tube formation. We hypothesize that it could lead to vascular normalization in short time. In contrast, docetaxel can suppress HUVEC proliferation, adhesion, and reduced tube formation swiftly due to its cytotoxicity. Inhibitory effect of recombinant human endostatin on angiogenesis in atherosclerotic plaque of rats by regulating Dll4/Notch pathway

Abstract:AIM: To observe the inhibitory effect of recombinant human endostatin (rhES) on plaque angio-genesis, and to explore the regulatory mechanism of Dll4 /Notch pathway in the anti-angiogenic effect of rhES.METH-ODS: Male Wistar rats were randomized into 3 groups: normal control group (N group), atherosclerotic model group (AS group), and rhES treated group (AS +rhES group).The rats in N group were fed a normal diet, while the remaining 2 groups were established to atherosclerotic rat model via high-cholesterol diet, intraperitoneal injection of vitamin D3 and aor-tic balloon injury.The rats in AS +rhES group received intraperitoneal injection of rhES.The blood total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), C-reactive protein (CRP), interleukin-1 (IL-1) and troponin I (TnI) were measured.The atherosclerotic abdominal aortas were taken for pathological observation.Immu-nohistochemical staining was used to measure the density of neovessels in the plaques, which were marked by CD31.The protein levels of Dll4 and Notch1 in the aortas were analyzed by Western blot.RESULTS: The levels of blood TC, TG, LDL-C, CRP and IL-1 in AS group and AS +rhES group were much higher than those in N group (P <0.05), and no sta-tistical difference between AS group and AS +rhES group was observed.The expression of CD31 in AS group was the high-est among all groups.Compared with AS group, the density of neovessels in the plaques of AS +rhES group decreased sig-nificantly (P <0.05).The protein expression of Dll4 and Notch1 in AS group was lower than that in N group (P <0.05). Compared with AS group, the protein expression of Dll4 and Notch1 increased significantly (P <0.05).CONCLUSION:rhES has the ability to inhibit plaque angiogenesis in rats.The activation of Dll4 /Notch pathway may be the mechanism of rhES in inhibiting plaque angiogenesis.
