Abstract:Soy protein isolate (SPI) was modified by ultrasound pretreatment (200 W, 400 W, 600 W) and controlled papain hydrolysis, and the emulsifying properties of SPIH (SPI hydrolysates) and USPIH (ultrasound pretreated SPIH) were investigated. Analysis of mean droplet sizes and creaming indices of emulsions formed by SPIH and USPIH showed that some USPIH had markedly improved emulsifying capability and emulsion stabilization against creaming during quiescent storage. Compared with control SPI and SPIH-0.58% degree of hydrolysis (DH), USPIH-400W-1.25% (USPIH pretreated under 400W sonication and hydrolyzed to 1.25% DH) was capable of forming a stable fine emulsion (d43=1.79 μm) at a lower concentration (3.0% w/v). A variety of physicochemical and interfacial properties of USPIH-400W products have been investigated in relation to DH and emulsifying properties. SDS-PAGE showed that ultrasound pretreatment could significantly improve the accessibility of some subunits (α-7S and A-11S) in soy proteins to papain hydrolysis, resulting in changes in DH, protein solubility (PS), surface hydrophobicity (H0), and secondary structure for USPIH-400W. Compared with control SPI and SPIH-0.58%, USPIH-400W-1.25% had a higher protein adsorption fraction (Fads) and a lower saturation surface load (Γsat), which is mainly due to its higher PS and random coil content, and may explain its markedly improved emulsifying capability. This study demonstrated that combined ultrasound pretreatment and controlled enzymatic hydrolysis could be an effective method for the functionality modification of globular proteins.

Abstract:This research explored microwave treatment impact on the structuro-functional aspects of oxidized soy protein aggregates (OSPI). Data showed that oxidative treatment promoted the formation of high molecular weight aggregates through hydrophobic interactions, thereby disrupting the structure of natural soy protein isolates (SPI). Microwave treatment for an appropriate time (≤30 s) caused the molecular structure of OSPI to open up and reduction in molecular weight and disulfide bond content, while absolute zeta potential increased. These modifications increased emulsifying capacity of OSPI, as well as the interfacial adsorption of protein. Longer microwave treatment times (>30 s) caused OSPI to exhibit a tendency to aggregate in TEM and CLSM images. It indicated the appropriate microwave electromagnetic field effect and microwave heating effect could coordinatively regulate soy protein functional properties by modifying their aggregation behavior. The results provided new ideas for reducing resource waste, and further expanding soy protein application in the food industry.

Abstract:The optimal condition for extracting SSPS (soluable soybean polysaccharides) with microwave has been studied by the orthogonal experiment. The optimal extracting parameters were that the power and time of microwave and the ratio of raw material to liquid was 700 W, 120 s and 1:2 ,respectively. And the extracting rate was 1.806 4 %. It is confirmed that the optimal conditions were 4 times of heating up with microwave and 4 times of filtration during the process extracting of SSPS. And the extracting rate could be 10.544 7 %.

Abstract:The effects of power ultrasound and high-pressure on solubility and foaming ability of soybean protein iso- late (SPI) were studied. The value of solubility and foaming of SPI at various treatment times (5, 10, 15, 20, 30 and 50 min) by power ultrasound and at various treatment times (0, 5, 10, 15, 20 and 25 min at 300 MPa) by high-pressure were determined respectively. The results indicated that power ultrasound and high-pressure can im- prove the solubility and foaming ability of SPI. The improvement effects by the ultrasonic probe system were better than that by the bath system.

Abstract:This paper was to study the effects on NSI, emulsibility and foaming ability of SPC (Soy Protein Concentrate) in different pH. The structures of SPC were analysed by electrophoresis and infra-red spectrum so as to study the principle of increasing of NSI and improvement of emulsibility and foaming ability of SPC. The results showed that with the increasing of the pH, the NSI of SPC increases quickly, the NSI at pH 12.0 is much higher than that of at pH 7.0. Emulsibility and foaming ability are improved with the increasing of the NSI. The electrophoresis showed that the subunit of SPC does not dissociate during modification; but the structures of SPC are changed by infra-red spectrum.
Effects of low-frequency magnetic field on solubility, structural and functional properties of soy 11S globulin

Abstract:Background: Soy 11S globulin has high thermal stability, limiting its application in the production of low-temperature gel foods. In this study, the low-frequency magnetic field (LF-MF, 5 mT) treatment (time, 30, 60, 90, and 120 min) was used to improve the solubility, conformation, physicochemical properties, surface characteristics, and gel properties of soy 11S globulin. Results: Compared with the native soy 11S globulin, the sulfhydryl content, emulsifying capacity, gel strength, water-holding capacity, and absolute zeta potential values significantly increased (P < 0.05) after LF-MF treatment. The LF-MF treatment induced the unfolding of the protein structure and the fracture of disulfide bonds. The variations in solubility, foaming properties, viscosity, surface hydrophobicity, and rheological properties were closely related to the conformational changes of soy 11S globulin, with the optimum LF-MF modification time being 90 min. Conclusion: LF-MF treatment is an effective method to improve various functional properties of native soy 11S globulin, and this study provides a reference for the development of plant-based proteins in the food industry. © 2024 Society of Chemical Industry.

Abstract:<p>The effect of ultrasound on the conformational and physicochemical properties of soy protein isolate hydrolysates (SPHs) was investigated. SPHs were prepared at hydrolysis times of 20 min, 60 min, and 180 min, then treated with ultrasound for 10 min, 20 min, and 30 min at a frequency of 20 kHz and output powers of 150 W and 450 W. The structural properties and antioxidant capacities of the aqueous layer of SPHs (ASPHs) after sonication were evaluated by Fourier-transform infrared spectroscopy (FTIR), intrinsic fluorescence, DPPH radical scavenging activity assays, and microscopy observations. Results obtained showed that ultrasound treatment significantly disrupted the peptide aggregates formed during protein hydrolysis. The protein solubility was significantly increased after sonication (by up to 18.33%), as did the percentage of proteins with MW <1 kDa in ASPHs. The antioxidant capacity of ASPHs also increased, as measured by DPPH assay. FTIR analysis of ASPHs indicated that the protein secondary structures were different, with an increase in β-sheet and a decrease in α-helix and β-turn. Furthermore, the changes in fluorescence spectra of ASPHs showed the transition of protein tertiary structure with a greater exposure of Trp residues in the side chains. Scanning electron microscope (SEM) and atomic force microscope (AFM) observations of the morphological structure of ASPHs further confirmed the significant effect of sonication on disrupting peptide aggregates. In conclusion, ultrasound can be used as an efficient treatment to promote the solubility of protein hydrolysates.</p>

Abstract:This research explored the influences of ultrasonic and thermal treatments on the structure, functional properties, and beany flavor of soy protein isolate (SPI). In comparison with traditional thermal treatment, ultrasonic treatment effectively induced protein structural unfolding and exposure of hydrophobic groups, which reduced relative content of α-helix, increased relative content of β-turn, β-sheet and random coil, and improved the solubility, emulsifying and foaming properties of SPI. Both treatments significantly decreased the species and contents of flavor compounds, such as hexanal, (E)-2-nonenal, (Z)-2-heptenal and (E)-2-hexenal in SPI. The relative content of hexanal in the major beany flavor compound decreased from 11.69% to 6.13% and 5.99% at 350 W ultrasonic power and 150 s thermal treatment procedure, respectively. After ultrasonic treatment, structural changes in SPI were significantly correlated with functional properties but showed a weak correlation with flavor. Conversely, the opposite trend was observed for thermal treatment. Thus, using ultrasonic treatment to induce and stabilise the denatured state of proteins is feasible to improve the functional properties and beany flavor of SPI.

Abstract:Abstract Phosphatidylcholine-soybean protein isolate (PC-SPI) nanoemulsions were prepared by ultrasonication. The effects of preparation conditions (SPI and PC addition, ultrasonic power and time) on the structural properties of the nanoemulsions and their storage stability were investigated. The results showed that the most optimal adsorption capacity and adsorption tightness at the oil–water interface under optimal conditions (1.5% SPI, 0.20% PC, 500 W ultrasonic power and 9 min ultrasonic time) were exhibited by the SPI-PC conjugate, which demonstrated that this nanoemulsions can be categorized as a high-quality emulsion suitable for research. To test its stability, and the high-quality nanoemulsion of β-carotene was stored. After degradation of the nanoemulsions during storage, β-carotene was released. The β-carotene retention rate of the high-quality emulsion was maintained above 86% at different temperatures in the absence of light for up to 30 days. This study provides new information for the development of transport and stability systems for nanoemulsions.

Abstract:Soybean soluble polysaccharide(SSPS)is an acid polysaccharide extracted from the residue by-products produced during the making of Tofu and soybean protein isolate,Having many functional properties such as stabilization behaviour,emulsifying properties.The extraction of the soybean soluble polysaccharide and the effects on the functional properties were researched.The results showed that the optimum extraction technology of SSPS are as follows:temperature is 125℃,extraction time is 1.5h,pH5.0.At this condition SSPS have high yield and good effect on the stability of protein.

Abstract:Modifications in physicochemical, conformational and emulsifying properties of unheated and preheated soy protein isolates (SPIs) by microfluidization at a specific pressure level (120MPa) were investigated. The heat pretreatment was carried out at temperatures of 75, 85 and 95°C, respectively. The tested properties included protein solubility (PS), surface hydrophobicity (Ho), free sulfhydryl group (SH) and disulfide bond (SS) contents, conformational characteristics, as well as emulsifying properties (e.g. emulsifying ability index (EAI), particle size of oil droplets in the emulsions, creaming index). The results indicated that the microfluidization treatment generally increased PS, Ho, SS content, as well as EAI of both unheated and preheated SPIs, in a preheating temperature dependent way; the treatment also resulted in partial unfolding and denaturation, and even structural re-arrangement of the proteins; the treatment improved the emulsifying efficiency and stability against creaming, but accelerated the bridging flocculation of oil droplets in the preheated-SPI-stabilized emulsions, with higher extent of flocculation observed at higher temperatures of the pretreatment. Size exclusion chromatography and SDS-PAGE analyses confirmed that the microfluidization led to transformation of insoluble aggregates into soluble ones, in which SH/SS interchanges might be involved. The findings suggest that the microfluidization treatment, or in combination with a thermal pretreatment, would be an effective technique to improve surface properties of SPI.

Abstract:The effects of tempetrature and concentration on the solubility of soy protein isolates (SPI) were studied, and then the subunit compositions and form of aggregation of soluble fractions obtained from treated SPI were analyzed by SDS-PAGE. The results showed that the totally denatured SPI after thermal treatment at low concentration have good solubility due to formation of heat-induced soluble protein aggregates.

Abstract:Currently, the predominant process for soy protein concentrate (SPC) production is aqueous ethanol washing of hexane-extracted soy meal. However, the use of hexane is less desired, which explains the increased interest in cold pressing for oil removal. In this study, cold-pressed soy meal was used as the starting material, and a range of water/ethanol ratios was applied for the washing process to produce SPCs. Washing enriched the protein content for the SPCs, regardless of the solvent used. However, we conclude that washing with water (0% ethanol) or solvents with a high water/ethanol ratio (60% and above) can be more advantageous. Washing with a high water/ethanol ratio resulted in the highest yield, and SPCs with the highest protein solubility and water holding capacity. The water-only washed SPC showed the highest viscosity, and formed gels with the highest gel strength and hardness among all the SPCs at a similar protein concentration. The variations in the functionality among the SPCs were attributed to protein changes, although the effects of non-protein constituents such as sugar and oil might also be important. Overall, the aqueous ethanol washing process combined with cold-pressed soy meal created SPCs comparable to commercial SPC in terms of composition, but with varied functionalities that are relevant for novel soy-food developments.

Abstract:Soy protein isolate is the most refined form of soy protein widely used in food processing.It has higher nutritional value and better functional properties.The differences of functional properties between different extracting method were investigated.Three different methods including ultrasonic wave assisted(AOT)/isooctane reverse micelle system consisting bis-(2-ethylhexyl)sulfosuccinate,isooctane reverse micelle system and alkali-solution extraction followed by a acids precipitation.Experimental results showed that ultrasound-assisted reverse micelle had the higher value in protein content,water holding capacity,foaming capacity and foaming stability,emulsification and emulsion stability were also better than the other two methods,while solubility was slightly inferior to the reverse micelles extraction of soy protein.It was found that the micro-structure of soybean protein varied among three methods and were all characterized by electron microscope scanner.The results indicated that the reverse micelles in the micro-environment and ultrasound changed the micro-structure of protein,affected the functional properties of the protein.

Abstract:Soybean protein concentrates(SPC) combine the advantages of the lower cost and higher protein content,compared with soybean protein isolates(SPI) and soybean protein flours(SPF) respectively,and are utilized extensively in many food systems. At present,SPC are mostly produced by alcohol-leaching,their process conditions,including leaching temperature,time,the ratios of low temperature soybean flake and ethanol solution,concentration of ethanol solution,were studied in details in the paper. The optimum technological parameters of leaching temperature 60℃,30min,ratios 1∶5,and concentration of ethanol solution 60% were obtained by orthogonal test.
Structure and functional properties of soy protein isolate-lentinan conjugates obtained in Maillard reaction by slit divergent ultrasonic assisted wet heating and the stability of oil-in-water emulsions

Abstract:Effects of a novel slit divergent ultrasound (SDU) treatment on soybean protein isolate (SPI)-lentinan conjugates via Maillard reaction was investigated. Besides, the stability of emulsions prepared by SPI and SPI-lentinan conjugates (ultrasound and un-ultrasound) as emulsifiers was compared. The results showed that ultrasonic treatment (40 min) markedly increased the degree of grafting (26.48%) by 1.91 times comparing with traditional heating method (2 h, 13.89%). In addition, structural analysis showed that the conjugates obtained by SDU treatment changed the secondary structure and had higher surface hydrophobicity and fluorescence intensity than those obtained by traditional heating method. Apart from this, SDU treatment could significantly improve the functional properties (solubility, foaming, emulsifying capacity, thermal stability, and viscosity) of conjugates. Furthermore, the emulsions prepared by the SPI-lentinan conjugates (ultrasound) as emulsifiers possessed the highest stability against environmental stresses. Taken together, SDU-assisted heating could be an excellent method to improve the functional properties of conjugates.

Abstract:Soybean protein isolate-soluble soybean polysaccharide (SPI-SSPS) complexes and mixtures with varying SPI/SSPS concentration ratios (1: 1, 2:1, 4:1, 8:1) were prepared by Maillard reaction and blending, respectively, and their physicochemical, structure, and functional properties were compared studied. The physical stability of SPI-SSPS complex, which consisted of CN and CS bonds, was better than that of the SPI/SSPS mixture with electrostatic interactions and hydrogen bonds, and both were superior SPI alone. The complex with SPI/SSPS concentration ratio of 8:1 had the highest grafting degree (33.25 %) and a more ordered structure, making its solubility and emulsifying property lower than the SPI/SSPS mixture; however, the physical and thermal stability of the SPI-SSPS complex was higher than that of the SPI and SPI/SSPS mixture. In particular, the SPI-SSPS complex with a high grafting degree showed a higher thermal denaturation temperature (194.06 °C). This study aimed to provide effective modification methods to utilize soybean processing by-products by modifying soybean protein isolate with soluble soybean polysaccharide.

Abstract:The effects of low-frequency and high-intensity ultrasonic treatments and curdlan gels on the thermal gelling and structural properties of soy protein isolate (SPI) were investigated. Low-frequency (20 kHz), high-intensity ultrasonic treatment improved the solubility and rheological properties of the SPI and promoted gelling, especially with 400 W treatment for 10 min. The addition of thermo-reversible curdlan gel (TRC) or thermo-irreversible curdlan gel (TIRC) further improved gel strength and reduced cooking loss from SPI gels pretreated with ultrasound. Fourier transform infrared spectroscopy (FTIR) of the SPI composite gel showed that the peak at 3268 cm(-1) for the O-H vibration exhibited a blue shift compared to the control SPI and a red shift compared to pure TRC or TIRC, which indicated formation of hydrogen bonding interactions between SPI and TRC or TIRC. Circular dichroism spectroscopy (CD) showed that the addition of TRC or TIRC significantly decreased the a-helix content but increased the p-sheet contents of SPI gels due to denaturation and unfolding of SPI. Scanning electron microscopy (SEM) showed that the addition of TRC or TIRC led SPI gel to form a more regular and homogeneous network. These results revealed that the ultrasonic treatment and curdlan gels had obvious synergistic effects that improved the quality of SPI gels. Moreover, compared with TIRC, TRC addition promoted expansion and interactions of SPI molecules and formed a more stable network. Therefore, ultrasonic treatment combined with curdlan addition significantly improved the thermal gelation and structural properties of SPI.

Abstract:The effects of radio frequency (RF) heating treatments with different final temperatures (70, 80, and 90 °C) and electrode gaps (120, 160, and 200 mm) on the structural characteristics of soy protein isolate (SPI) dispersion were investigated. The results showed that RF heating significantly influenced free sulfhydryl groups and surface hydrophobicity of SPI. Free sulfhydryl groups increased with the increase of final temperature. The hydrophobicity of the RF-heated sample was higher than the original SPI without RF treatment. The highest hydrophobicity of the RF-heated SPI was found with electrode gap of 200 mm at 90 °C. RF heating treatment resulted in the reduction of ultraviolet absorption of SPI indicating the change of three-dimensional positions of soy protein but did not modify the protein primary structure of SPI. The Fourier transform infrared spectroscopy showed that hydration of SPI was decreased by RF heating. The self-reassembly from random coil structure to β-sheet structure suggested that RF heating treatment can change the secondary structure of soy protein to be more orderly.Graphical Abstractᅟ
