Abstract:Investigation of the bark and/or wood of seventeen African species of Diospyros has revealed the presence of a number of triterpenes and naphthoquinones. The triterpenes lupeol, betulin and betulinic acid were detected in all samples but three other triterpenes were each found in only one species; betulinaldehyde in D. canaliculata , cerin in D. iturensis , and glutinol in D. zenkeri . Naphthoquinones were detected in fourteen of the seventeen species and the common dimeric compounds diospyrin, isodiospyrin and diosindigo-A were isolated. D. canaliculata differed from all other species in producing derivatives of plumbagin including its reduced form, isoshinanolone (1 R ,2 R ) and a novel stereoisomer which was given the trivial name epiisoshinanolone (1 S ,2 R ).

Abstract:Three new triterpenoid derivatives, named (15 alpha)-15-hydroxysoyasapogenol B (1), (7 beta,15 alpha)-7,15-dihydroxysoyasapogenol B (2), and (7 beta)-7,29-dihydroxysoyasapogenol B (3), were isolated from cultures of the plant endophytic fungus Pestalotiopsis clavispora. Their structures and relative configurations were elucidated by extensive spectroscopic analysis and X-ray crystallography.

Abstract:A new naphthoquinone, 6-hydroxy-alpha-dunnione (1) and a new binaphthoquinone, methyl 1,1',4,4'-tetrahydro-3-hydroxy-1,1',4,4'-tetraoxo[2,2'-binaphthalene]-3'-carboxylate (2), along with ten known compounds, including naphthoquinones, anthraquinones, and phenylethanoid glucosides, were isolated from the roots of Didymocarpus hedyotideus CHUN. Their structures were identified by spectroscopic analyses, particularly 1D- and 2D-NMR spectroscopy. The cytotoxic activities of the two new naphthoquinones were also evaluated.

Abstract:Eleven undescribed guaiane-type sesquiterpenoids, named daphnegenols A-K (1–11) and a known analog (12) were targeted for isolation from the root of Daphne genkwa by LC-MS/MS-based molecular networking. Their diverse structures were elucidated through extensive spectroscopic methods, quantum chemical calculation and X-ray single crystal diffraction, including 5/7 bicycle, 5/6/7 tricycles and 5/6/5 tricycles guaiane-type sesquiterpenoid. Daphnegenol A (1) possessed a rare six membered oxyheterocyclic ring which was constructed by oxidation and cyclization at C-1 and C-11. The anti-acetylcholinesterase and anti-neuroinflammatory activities of compounds 1–12 were measured.

Abstract:The investigation of the constituents in the rhizomes of Dioscorea nipponica Makino subsp. rosthornii (Prain et Burkill) C. T. Ting afforded one new phenanthrene 2,2',7,7'-tetramethoxy-[1,1'-biphenanthrene]-4,4',6,6'-tetrol (7) and one new isocoumarin diorosthornoumarin (8), together with 16 known compounds (1-6 and 9-18). Their structures were established on the basis of extensive spectroscopic evidences (IR, HR-ESI-MS, NMR and optical rotation), as well as comparison with literature values. All the compounds 1-18 were firstly isolated from Dioscorea nipponica Makino subsp. Rosthornii (Prain et Burkill) C. T. Ting, and compound 9 was firstly obtained as a natural product from plants, while the compounds 11 and 14 were obtained from both the genus Dioscorea and the family Dioscoreaceae for the first time. Moreover, the antitumor activities of the compounds were tested against lung carcinoma NCI-H460 cell line. Compound 12, 13, 15 and 16 showed significant cytotoxic activities, whereas 7 displayed moderate cytotoxicity.

Abstract:A new benzoquinone, 2-hydroxy-3-methoxy-5-methyl-p-benzoquinone ( 1 ), along with 11 known compounds including three benzoquinones, 2-methoxy-6-methyl-p-benzoquinone ( 2 ), 2,3-dimethoxy-5-methyl-pbenzoquinone ( 3 ), and 2-hydroxy-5-methoxy-3-methyl-p-benzoquinone ( 4 ); three lignans, 4-hydroxysesamin ( 5 ), (+)-sesamin ( 6 ), and aptosimon ( 7 ); five benzenoids, 2,4-dimethoxy-6-methylbenzene-1,3-diol ( 8 ), benzocamphorin A ( 9 ), benzocamphorin D ( 10 ), 2,2′,5,5′-tetramethoxy-3,4,3′,4′-bi-methylenedioxy-6,6′-dimethylbiphenyl ( 11 ), and 4,7-dimethoxy-5-methyl-1,3-benzodioxole ( 12 ), were isolated from the fruiting body of Antrodia camphorata (Polyporaceae). These compounds were characterized and identified by physical and spectral evidences.

Abstract:Chemical investigation of an alcoholic extract from the stem of Daphne papyracea ("Xuehuagou") led to the isolation of the tetracyclic sesquiterpenoid daphnepapytone A (1), containing a unique caged skeleton with a cyclobutane ring having three tetrasubstituted chirality centers. Also isolated were new guaiane sesquiterpenoids, namely, daphnepapytones B-H (2-8), and one 1,5-diphenylpentanone 2-hydroxy-5-oxo-daphneone (9), together with 26 known compounds. The cyclic metabolites share a 5-isoprenyl-hexahydroazulene-2(1H)-one skeleton with different substitution patterns and a bridged cyclobutane, oxetane, or tetrahydrofuran ring. The planar structures and relative configuration of the new compounds were elucidated on the basis of spectroscopic analysis aided by DFT 13C NMR calculations. The absolute configurations of 1-7 were determined by X-ray single-crystal diffraction or TDDFT-ECD calculations. Daphnepapytones A and C (1 and 3), 2-hydroxy-5-oxodaphneone (9), daphnenone (10), daphneone (11), and 3-methyldaphneolone (12) showed α-glycosidase inhibitory activity, with IC50 values of 159.0, 102.3, 139.3, 43.3, 145.0, and 126.1 μM, respectively.

[Studies on Chemical Constituents in Bark of Dictamnus Dasycarpus].

Abstract:OBJECTIVE To study the chemical constituents in bark of Dictamnus dasycarpus. METHOD Isolation and purification were carried out on silica gel column chromatography, prepared thin layer chromatography and sephadex LH - 20, et al. The structures were identified by spectral analysis. RESULT Twelve compounds were obtained from bark of D. dasycarpus and the structures were determined as dictamnine (I), fraxinellone (II), skimmianine (III), gamma-fagarine (IV ), beta-sitosterol (V), obacunone (VI), limonin disophenol (VII), fraxinellonone (VIII), wogonin (IX), rutevin (X), kihadinin B (XI), dasycarine (XII). CONCLUSION Compounds IX and XI were isolated from genus Dictamnus for the first time, and compound VIII was isolated from the species for the first time.

Abstract:Four new lanostane-type triterpenes, 24-ethyl-3beta-methoxylanost-9(11)-en-25-ol (1), 3beta-methoxy-24-methylenelanost-9(11)-en-25-ol (2), 3beta-methoxy-25-methyl-24-methylenelanost-9(11)-en-21-ol (3) and 3beta-methoxy-24-methyllanosta-9(11),25-dien-24-ol (4) together with three known triterpenes, betulinaldehyde, betulinic acid methyl ester, and ursaldehyde have been isolated from the methanol extract of the twigs of Diospyros discolor. The structures of those new compounds were elucidated by spectroscopic methods.

Abstract:A bioactivity-guided study on the leaves of Picrasma javanica led to the isolation of 19 quassinoids, including 13 new compounds. The structures of the new compounds were elucidated by a combination of spectroscopic data analysis, X-ray crystallography studies, and electronic circular dichroism (ECD) data interpretation. Compounds 1–7 are rare examples of quassinoids with a keto carbonyl group at C-12. The biological activities of 11 of the more abundant isolates were evaluated against five phytopathogenic fungi in vitro, and several of them including 6 and 15 showed moderate inhibitory effects that were comparative to those of the positive control, carbendazim. Abstract:Nine undescribed carotane sesquiterpenes and rare rearranged guaiane sesquiterpenoids ( 1‒9 ), along with two known analogues ( 10‒11 ), were obtained from the aerial parts of Daphne penicillata . Their structures and absolute configurations were determined by extensive spectroscopic analyses, NMR/ECD calculations, and single-crystal X-ray diffraction analysis. All compounds were evaluated for anti-inflammatory and cytotoxic activities. The inhibitory effect of several active sesquiterpenoids against the cyclooxygenase-2 (COX-2) enzyme was also examined by an in vitro enzyme inhibition assay. Furthermore, molecular docking, molecular dynamics (MD) simulation and binding free energy calculation were applied to further analyze the interactions, stability and affinity, among which compounds 3 and 4 exhibited high binding affinities and stable binding to the active site of COX-2.

Abstract:Phytochemical characterization of the stem and root bark of Daphne giraldii Nitsche. led to the isolation of eight flavones (1–8), six flavans (9–14) and six coumarins (15–20). The structures of these compounds were examined with spectroscopic methods, and also by comparing to previously reported spectroscopic data. Among them, ten compounds (1–3, 9 and 15–20) are reported for the first time for this species, and all compounds with prenyl substitutions have not been identified in other species of genus Daphne, or even in the family Thymelaeaceae. We also describe the chemotaxonomic significance of these compounds.

Abstract:One (formosumone A, 1) new and fifteen (2-16) known phenolic compounds were isolated from the leaves of Cratoxylum formosum ssp. pruniflorumm, a substitute for the popular bitter nail tea ("Kuding Tea") generally used in Southeast Asia. Their structures were determined by extensive spectroscopic analysis and by comparison with literature data. Compound 1 possesses a rare scaffold of a flavanone coupled with a phloroglucinol moiety, representing the first example of such a scaffold from the Clusiaceae family. Among the isolates, toxyloxanthone B (11) and vismione D (12) were found to show remarkable anti-neuroinflammatory effects by inhibiting nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine BV-2 microglial cells. Additionally, toxyloxanthone B (11) exhibited significant neuroprotective effect against β-amyloid(25-35) (Aβ(25-35))-induced cell viability decrease in SH-SY5Y neuroblastoma cells.

Abstract:Abstract Three new phenolic derivatives, picraquanines A–C (1–3), along with 6 known ones 4–9 were obtained from the stems of Picrasma quassioides (D. Don) Benn. The new structures were determined by extensive spectroscopic data analysis, including IR, HRESIMS, 1H-NMR, 13C-NMR, HSQC, HMBC, 1H-1H COSY experiments. The absolute configuration of 1 was determined by comparison of its experimental and calculated ECD spectra. Furthermore, all the compounds were tested for their nitric oxide (NO) inhibitory effects against LPS-stimulated RAW 264.7 cells, however, none of them exhibited inhibitory effects (IC50 >100 μM).

Abstract:A new phenanthraquinone, 7,9-dihydroxy-2-methoxy-1,4-phenanthraquinone ( 1 ), and nine known compounds were isolated and identified from the whole plants of Dendrobium lindleyi . The structure of the new compound was determined on the basis of NMR (1D and 2D) and mass spectroscopic analysis. The new compound exhibited potent antimicrobial activity against Bacillus subtilis and Escherichia coli , and its activity was comparable with that of the well-known antibacterial drug berberine.
