Abstract:The chitosan derivatives were improved in preparation method and characterized by FT-IR spectrum analysis.The bacteriostasis,stability,dissolubility,pH and other properties of the chitosan derivatives were investigated.The experimental results indicated that the dissolubility and stability of the chitosan derivatives prepared by the improved method were prominently enhanced.Two kinds of the chitosan derivatives at the concentration of 0.1%(w/v) showed obvious bacteriostatic activities on Esherichia coli and Staphylococcus aureus.Their growths were terminated when the concentration reached 0.2%(w/v).The bacteriostatic activity of high molecular weight(Mw) chitosan derivative is higher on gram positive microbes while that of low Mw chitosan derivative is higher on gram negative bacteria.
Preparation and Characterization of N-succinyl-chitosan

Abstract:Poisonous solvents were widely used in the preparation of N-succinyl-chitosan,which were easy to cause environmental pollution and difficult to remove completely from the products.These disadvantages limited their uses in biomedical materials.In the present work,a novel method was proposed to successfully synthesize N-succinyl-chitosan,which was safe and environmentally friendly.A series of N-succinyl-chitosan with different degrees of substitution were prepared in absolute ethanol by controlling the reaction time.The different degrees of substitution,molecular weight and polydispersity index of the products were measured.The products were characterized by Fourier transform infrared(FTIR) spectroscopy,X-ray diffractometry(XRD) and thermogravimetry(TG).Furthermore,the water-solubility and biocompatibility were studied.When the reaction time is 15 h,the degree of substitution and molecular weight of the product are highest.Moreover,polydispersity index of the product is smallest among all the products(10 h,12 h,15 h,18 h,20 h).In addition,the results indicate N-succinyl-chitosan has been synthesized with decreased degree of crystallization and thermostability.Water-solubility of modified chitosan is improved due to the decrease of crystallization.N-succinyl-chitosan prepared in absolute ethanol has good biocompatibility,which has great potential in biomedical fields.

Abstract:In this study, nine chitosan derivatives containing aromatic five-membered heterocycles were prepared and the effects of different grafting methods on the biological activities of chitosan derivatives were investigated. The structures of all the compounds were characterized by Fourier Transform Infrared (FT-IR) spectroscopy and Nuclear Magnetic Resonance (NMR) spectroscopy, while the antioxidant, antifungal and antibacterial activities of the chitosan derivatives were tested. The experimental data suggested that the chitosan derivatives had outstanding inhibitory ability against Fusarium graminearum, Fusarium oxysporum f.sp.cucumbrum, Staphylococcus aureus and Escherichia coli. At the same time, some of the compounds showed strong scavenging ability against DPPH radical and superoxide radical. Cytotoxicity experiments have demonstrated that some chitosan derivatives are non-toxic to L929 cells. More importantly, compared to chitosan, these chitosan derivatives have good water solubility and can be used as potential polymers for antifungal and antibacterial biomaterials in agriculture.

Abstract:Hydroxypropyltrimethyl ammonium chitosan halogenated acetates were successfully synthesized from six different haloacetic acids and hydroxypropyltrimethyl ammonium chloride chitosan (HACC) with high substitution degree, which are hydroxypropyltrimethyl ammonium chitosan bromacetate (HACBA), hydroxypropyltrimethyl ammonium chitosan chloroacetate (HACCA), hydroxypropyltrimethyl ammonium chitosan dichloroacetate (HACDCA), hydroxypropyltrimethyl ammonium chitosan trichloroacetate (HACTCA), hydroxypropyltrimethyl ammonium chitosan difluoroacetate (HACDFA), and hydroxypropyltrimethyl ammonium chitosan trifluoroacetate (HACTFA). These chitosan derivatives were synthesized by two steps: first, the hydroxypropyltrimethyl ammonium chloride chitosan was synthesized by chitosan and 3-chloro-2-hydroxypropyltrimethyl ammonium chloride. Then, hydroxypropyltrimethyl ammonium chitosan halogenated acetates were synthesized via ion exchange. The structures of chitosan derivatives were characterized by Fourier transform infrared spectroscopy (FTIR), ¹H Nuclear magnetic resonance spectrometer (¹H NMR), 13C Nuclear magnetic resonance spectrometer (13C NMR), and elemental analysis. Their antifungal activities against Colletotrichum lagenarium, Fusarium graminearum, Botrytis cinerea, and Phomopsis asparagi were investigated by hypha measurement in vitro. The results revealed that hydroxypropyltrimethyl ammonium chitosan halogenated acetates had better antifungal activities than chitosan and HACC. In particular, the inhibitory activity decreased in the order: HACTFA > HACDFA > HACTCA > HACDCA > HACCA > HACBA > HACC > chitosan, which was consistent with the electron-withdrawing property of different halogenated acetates. This experiment provides a potential idea for the preparation of new antifungal drugs by chitosan.

Abstract:Chitosan exhibits good biocompatibility and some antibacterial activity, making it a popular choice in biomedicine, personal care products, and food packaging. Despite its advantages, the limited antibacterial effectiveness of chitosan hinders its widespread use. Introducing a six-membered heterocyclic structure through chemical modification can significantly enhance its antimicrobial properties and broaden its potential applications. In order to explore the effect of six-membered heterocyclic structure on the antibacterial and antibiofilm activities of chitosan. In this study, seven chitosan derivatives containing six-membered heterocyclics were prepared. They were characterized using Fourier transform infrared (FT-IR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy, and elemental analysis. Cell viability assay showed that they were non-toxic. The antibacterial and antibiofilm activities against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) were evaluated. Our research findings demonstrate that increasing the hydrophobicity, alkalinity and charge density of the substitute groups improved the antibacterial and antibiofilm activities of chitosan. This study also offers valuable insights into the quantitative structure-activity relationships of chitosan derivatives in terms of antibacterial and antibiofilm activities.

Abstract:Chitosan was prepared by alkali treatment. Its yield was 5.28%, degree of deacetylation 81.7%, intrinsic viscosity was 10.550 and molecular weight 54.42×104. Chitosan degradated by H2O2 oxidation time to form respectively different molecular weights 32.3×104, 6.7×104 and 0.496×104. Its antimicrobial activity for St.aureus, Bacillus pumilus strengthens with the increase of its MW(molecular weight) and concentration.

Abstract:Chitosan, a natural and abundant polysaccharide, offers several advantageous properties, such as excellent biocompatibility, biodegradability, and antimicrobial activity, making it a highly versatile material. However, its extensive application is hindered by its insolubility in water and most organic solvents, due to numerous intramolecular/intermolecular hydrogen bonds. To overcome this limitation and optimize its benefits, chitosan is often chemically modified to enhances its utility in various industries. Biomedical materials, environmental management, biological papermaking, food preservation, and the daily chemical industry are a few examples. Modified chitosan retains the inherent properties of chitosan and acquires new physicochemical characteristics as a result of introduced functional groups. Hydrophilic modifications typically include carboxylation, phosphorylation, and quaternary ammonium modification reactions. The specific properties and applications of chitosan derivatives depend on the grafted groups. This review summarizes the reaction conditions, properties, and applications of hydrophilic chitosan derivatives in drug delivery and food preservation, serving as a reference for the development and use of chitosan-based biomaterials. Abstract:Sulfanilamide derivatives of chitosan (2-(4-acetamido-2-sulfanimide)-chitosan (HSACS, LSACS), 2-(4-acetamido-2-sulfanimide)-6-sulfo-chitosan (HSACSS, LSACSS) and 2-(4-acetamido-2-sulfanimide)-6-carboxymethyl-chitosan (HSACMCS, LSACMCS)) were prepared using different molecular weights of chitosan (CS), carboxymethyl chitosan (CMCS) and chitosan sulfates (CSS) reacted with 4-acetamidobenzene sulfonyl chloride in dimethylsulfoxide solution. The structures of the derivatives were characterized by FT-IR spectroscopy and elemental analysis, which showed that the substitution degree of sulfanilamide group of HSACS, HSACSS, HSACMCS, LSACS, LSACSS and LSACMCS were 0.623, 0.492, 0.515, 0.576, 0.463 and 0.477, respectively. The solubility of the derivatives (pH<7.5) was higher than that of chitosan (pH<6.5). The antifungal activities of the derivatives against Aiternaria solani and Phomopsis asparagi were evaluated based on the method of Jasso et al. in the experiment. The results indicated that all the prepared sulfanilamide derivatives had a significant inhibiting effect on the investigated fungi in the polymer concentration range from 50 to 500 microg mL(-1). The antifungal activities of the derivatives increased with increasing the molecular weight, concentration or the substitution degree. The sulfanilamide derivatives of CS, CMCS and CSS show stronger antifungal activities than CS, CMCS and CSS.

Abstract:Chitosan is derived from chitin polysaccharide, the main component of crustacean shells. Chitosan is a biocompatible, nontoxic, and biodegradable polymer soluble in acidic solutions. It is widely used in the medical and pharmaceutical fields. Antimicrobial activities of chitosan against different bacterial, fungal, and viral pathogens have been considered one of its attractive properties, making chitosan valuable for biological applications, including textile, food, tissue engineering, agriculture, and environmental protection. Additionally, chitosan has beneficial effects on livestock, poultry, fish, and crustaceans, which can enhance immunity, improve feed conversion, and promote growth. However, the water solubility of chitosan influences antimicrobial capabilities, limiting its application. In the present work, we reviewed the preparation, factors affecting antimicrobial activity, morphological structure, antimicrobial mechanism, and application of chitosan derivatives, and the problems and prospects were pointed out. Collectively, this review provided an update on the application of chitosan derivatives and their potential for further advanced applications in the antimicrobial field.
Synthesis of Quaternary Ammonium Salts of Chitosan Bearing Halogenated Acetate for Antifungal and Antibacterial Activities

Abstract:In this paper, quaternary ammonium salts of chitosan bearing halogenated acetate, including N,N,N-trimethyl chitosan chloroacetate (TMCSC), N,N,N-trimethyl chitosan dichloroacetate (TMCSDC), N,N,N-trimethyl chitosan trichloroacetate (TMCSTC), and N,N,N-trimethyl chitosan trifluoroacetate (TMCSTF), were prepared via N,N,N-trimethyl chitosan iodide (TMCSI). The obtained chitosan derivatives were characterized by FT-IR, ¹H NMR spectra, 13C NMR spectra, and elemental analysis. Their antifungal property against Fusarium oxysporum f. sp. cucumebrium Owen (F. oxysporum f. sp. cucumebrium Owen), Botrytis cinerea (B. cinerea), and Phomopsis asparagi (P. asparagi) were evaluated by hyphal measurement method at concentrations ranging from 0.08 mg/mL to 0.8 mg/mL. Meanwhile, two common bacteria, Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), were selected as the model Gram-negative and Gram-positive bacteria to evaluate the antibacterial property of the chitosan derivatives by agar well diffusion method. The results showed that TMCSC, TMCSDC, TMCSTC, and TMCSTF had better antifungal and antibacterial activities than chitosan and TMCSI. In particular, a rule showed that the inhibitory activity decreased in the order: TMCSTF > TMCSTC > TMCSDC > TMCSC > TMCSI > chitosan, which was consistent with the electron-withdrawing property of different halogenated acetate. Apparently, the quaternary ammonium salts of chitosan with stronger electron withdrawing ability possessed relatively greater antifungal and antibacterial activities. This experiment provides a potentially feasible method for the further utilization of chitosan in fields of antifungal and antibacterial biomaterials.

Abstract:In this study, three types of dodecyl chitosan quaternary ammonium salts, each with different spacer groups were synthesized. These chitosan derivatives are N′ , N′ -dimethyl- N′- dodecyl-ammonium chloride- N -amino-acetyl chitosan (DMDAC), N′- dodecyl- N -isonicotinyl chitosan chloride (DINCC) and N′ , N′ -dimethyl- N′- dodecyl-ammonium chloride- N -benzoyl chitosan (DMDBC). The synthesized products were characterized using Fourier transform infrared spectrometers, nuclear magnetic resonance, thermogravimetric analysis, and elemental analysis. The antibacterial and antibiofilm activities against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) were investigated. The experimental results indicated that the introduction of hydrophobic groups of spacer groups could enhance the antibacterial and antibiofilm activities of the chitosan derivatives. The antibacterial rates of the chitosan derivatives were over 90 % for both E. coli and S. aureus at a concentration of 0.5 mg/mL. The chitosan derivatives removed >50 % of the mature biofilm of E. coli and over 90 % of the mature biofilm of S. aureus at a concentration of 2.5 mg/mL. Further, the synthesized chitosan derivatives were determined to be non-toxic to L929 cells. Among them, DMDBC exhibited the most promising overall performance and show potential for wide-ranging applications in food preservation, disinfectants, medical, and other fields.

Abstract:Three heterocyclic compounds namely: (Z)-4-benzylidene-2-phenyl-1,3-oxazol-5-one (A), (Z)-4-(4-methoxybenzylidene)-2-phenyl-1,3-oxazol-5-one (B), (Z)-4-(2-thienyl methylene)-2-phenyl-1,3-oxazol-5-one (C) were prepared in one step reaction between hippuric acid and benzaldehyde, p-anisaldehyde and thiophene-2-carboxaldehyde, respectively. The prepared compounds were characterized using FTIR and 1H NMR spectroscopy. Furthermore, chitosan was prepared from alkaline treatment of shrimp shells. These compounds were used to chemically modify chitosan biopolymer. The obtained modified chitosan biopolymers were characterized using FTIR spectroscopy. The swelling properties of the modified chitosan biopolymers were studied at different immersion times, temperature of 25–70 °C and at a wide range of pH of 2–9. The increase of immersion time and temperature increased the swelling efficiency, as well as, increasing the acidity and alkalinity of the aqueous solution increased the swelling than the moderate pH values at 5–6. The antimicrobial efficacies of the modified chitosan biopolymers were determined using agar diffusion technique against different strains of bacteria and fungi. The antimicrobial assay showed excellent activity of the modified biopolymers. The high antimicrobial activities were correlated and rationalized to the chemical structure of the heterocyclic moieties used in the chemical modification of chitosan.

Abstract:Chitosan derivatives, including O- carboxymethyl chitosan (CMC), N -(2-hydroxypropyltrimonium chloride)- O -carboxymethyl chitosan (QCMC), and N -(2-hydroxypropyltrimonium chloride)- O -dodecylylpyridine chitosan quaternary ammonium salt (DQCMC), were synthesized and characterized using Fourier transform infrared (FTIR) spectroscopy, 1 H nuclear magnetic resonance ( 1 H NMR) spectroscopy, Ultraviolet-visible (UV-vis) spectroscopy, and element analysis (EA). The antibacterial activities of chitosan and chitosan derivatives against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) were evaluated through the minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and antibacterial rate assays. Results demonstrated that DQCMC exhibited significantly higher antibacterial efficacy compared to chitosan, CMC, and QCMC. The MIC of DQCMC against E. coli and S. aureus were 31 μg/mL and 7 μg/mL, respectively, with a 100% antibacterial rate at a concentration of 0.5 mg/mL. Furthermore, assessment of mouse fibroblast (L929) cell viability using cell counting kit-8 (CCK-8) methods revealed no toxicity associated with the material.

Abstract:Chitosan was prepared by alkaline N-deacetylation of β-chitin and hydroxypropyl derivatives with different degrees of substitution (DS) were synthesized. It was characterized by Fourier transform infrared (FT-IR) and elemental analysis. The DS of hydroxypropyl chitosan (HPCS) calculated by an element analyzer were 0.42, 0.75, 1.20, 1.82 and 2.25. HPCS showed better foam capacity and stability than that of chitosan, and the effectiveness correlated well with the DS of HPCS. The highest bile acid-binding capacity of all five HPCS reached 56.02 mg/g, which was 4.0-fold higher than that of chitosan. The scavenging ability of HPCS against hydroxyl and ABTS radicals improved with increasing concentration. The correlation between the hydroxypropyl content (DS) of HPCS and scavenging ABTS radical ability was positive. The hydroxyl radicals scavenging activity of HPCS correlated well with its increasing concentration, and EC50 values were below 12.5 mg/mL. These results indicated that hydroxypropylation is a possible approach to obtain chitosan derivatives with desirable physiochemical properties.
Preparation of Chitosan Derivative by Schiff Base and Its Antibacterial Activity

Abstract:The Schiff base of chitosan was obtained by the reaction of chitosan with aromatic aldehydes, then the obtained Schiff base of chitosan reacts with chloroacetic acid, and finally, the amino protection group of the product was removed to get the chitosan derivative. The obtained chitosan derivative was characterized by FT-IR and 1H-NMR, and the results show that the obtained chitosan derivative is the target product, the o-carboxymethyl-chitosan. The analysis results of TG and DTG show that the thermal stability of the obtained chitosan derivative is obviously different from that of chitosan. The antimicrobial properties of the chitosan and obtained chitosan derivative were analyzed by their antimicropial activities against Staphylococcus aureus and Aspergillus niger. The results indicate that both the antimicrobial activities of the chitosan derivative and chitosan increase with the increase of their concentrations, and the antimicrobial activities of the chitosan derivative are stronger than that of chitosan. The minimum inhibitory concentration (MIC) of the chitosan derivative against Staphylococcus aureus and Aspergillus niger was found to be 0.5%. Since the solubility of the chitosan derivative is good in organic solvent, it could be expected to have a big advantage of to be used in the fields of food and medicine.

Abstract:Two anionic soluble monomers, mono(2-methacryloyl oxyethyl)acid phosphate and vinylsulfonic acid sodium salt, were grafted onto chitosan to obtain copolymers with zwitterionic property. Graft reaction improved the antimicrobial activities of chitosan. Antimicrobial activities of chitosan and graft copolymers depended largely on the amount and type of grafted chains as well as changes of pH, against Candida albicans, Trichophyton rubrum, and Trichophyton violaceum. The most excellent antimicrobial activity among tested samples was shown at pH 5.75 with demonstrating strain selectivity against Candida albicans and Trichophyton violaceum due to the difference in affinity between cell wall of fungi and chitosan or its derivatives. Abstract:Water-soluble hydroxybutyl chitosan(HBCS) was prepared by the reaction of chitosan(CS) with 1,2-butane oxidation in an alkali solution.The structure of HBCS was characterized by FT-IR and solid state 13C NMR,and the results prove that the hydroxybutyl has been successfully connected to chitosan molecular chain.The degree of hydroxybutyl substitution estimated by means of elemental ananlysis is 1.2.Besides,HBCS exhibited good water-solubility and could dissolve in aqueous solutions in a broader pH range.In comparison with CS,HBCS shows a poorer thermostability.This is because the modified reaction destroys the crystalline region of chitosan.Antibacterial activities of CS and the modified derivative were explored by turbidity method.The results show that HBCS exhibits antibacterial activity against E.coli and S.aureus,and with the increase of concentration,the antibacterial effect enhances obviously.It was found that the antimicrobial activity of HBCS is weaker than that of CS under the same mass concentrations.

Abstract:A novel and green method for the preparation of chitosan derivatives bearing organic acids was reported in this paper. In order to improve the antioxidant activity of chitosan, eight different hydroxypropyltrimethyl ammonium chitosan derivatives were successfully designed and synthesized via introducing of organic acids onto chitosan by mild and non-toxic ion exchange. The data of Fourier Transform Infrared (FTIR), 13C Nuclear Magnetic Resonance (NMR), 1H NMR, and elemental analysis for chitosan derivatives indicated the successful conjugation of organic acid salt with hydroxypropyltrimethyl ammonium chloride chitosan (HACC). Meanwhile, the antioxidant activity of the chitosan derivatives was evaluated in vitro. The results indicated that the chitosan derivatives possessed dramatic enhancements in DPPH-radical scavenging activity, superoxide-radical scavenging activity, hydroxyl radical scavenging ability, and reducing power. Furthermore, the cytotoxicity of the synthesized compounds was investigated in vitro on L929 cells and showed low cytotoxicity. Thus, the enhanced antioxidant property of all novel chitosan products might be a great advantage, while applied in a wide range of applications in the form of antioxidant in biomedical, food, and cosmetic industry.

Abstract:The structure modification of chitosan has great application potential. 6-Oxychitosan was prepared by specially oxidizing the C6-OH of chitosan, then 6-oxychitosan was reacted with three kinds of aldehydes to prepare N-quaternized 6-oxychitosan derivatives in this paper. The derivatives were characterized by FT-IR, NMR, and elemental analysis. The antimicrobial activity of these derivatives was tested against two common plant-threatening fungi and three plant disease bacteria. The results showed that N-quaternized 6-oxychitosan derivatives had good water-solubility and excellent antimicrobial activity. Moreover, derivative 3 which connected 8-hydroxyquinolines had the highest antimicrobial activity than the other derivatives. The inhibitory indices of derivative 3 against V. albo-atrum and P. hibernalis are 89.1% and 72.8% at 0.4 mg/ml. The MICs of 3 against X. oryzae, P. syringae, and E. rhapontici were 625, 625, and 156 mg/l, respectively. All the results indicate that derivative 3 has the potential of becoming an alternative to harmful agricultural chemicals.
