Wnt Signaling Pathway and Preadipocytes

Abstract:Wnt proteins are a family of secreted growth factors, which act through paracrine and autocrine to regulate cell fate and development in many species. Current studies have shown that Wnt signaling is a critical pathway in the biogenesis and differentiation of preadipocytes. Wnt ligands bind to their transmembrane receptors of the Frizzled family (Fz1/2/5) members, and the coreceptor, low density lipoprotein receptor-related protein (LRP5/6) to activate canonical or noncanonical signaling pathways and influence the phosphorylation of downstream targeted genes products. Thereby subsequently to suppress adipogenesis and maintain preadipocytes in an undifferentiated state through inhibition of the adipogenic transcription factors such as CCAAT/enhancer binding protein (C/EBPα) and peroxisome proliferators activated receptor (PPARγ). When Wnt signaling in preadipocytes is prevented by overexpression of axin or dominant-negative TCF4, these cells differentiate into adipocytes. Disruption of Wnt signaling also causes transdifferentiation of myoblasts into adipocytes in vitro. In summary, Wnt signaling functions as an adipogenic switch. When it is on, adipogenesis is repressed; when it is off, adipogenesis is initiated.
