Abstract:We have realized a cyclization reaction of 2,3-allenoic acids 1 in the presence of simple alkylor aryl-substituted allenes 3. In this reaction, the cyclic oxypallaclation of 2,3-allenoic acid with Pd(II) would afford the furanonyl palladium intermediate 2, which could be trapped by the simple allene to afford a pi-allylic intermediate ant-9. This intermediate ant-9 could be nucleophilically attacked by Br- to yield 4-(1'-b romoalk-2'(2)-en-2'-yl)furan-2(5H)-one derivatives Z-5 and Pd(0). The in-situ formed Pd(0) was efficiently converted to the catalytically active Pd(II) species by benzoquinone in HOAc. The functional groups, such as malonate, acetoxyl, and phthalic amide in allene 3, are tolerable under the current conditions. High efficiency of chirality transfer was observed when optically active 2,3-allenoic acids were used, which reveals that the formation of the intermediates 2 was a highly stereoselective anti-oxypalladation process. The highly selective formation of Z-isomer may be explained by face-selective coordination of allene 3 with the palladium atom in intermediate 2: the palladium atom coordinates to the terminal C = C double bond of allene 3 from the face opposite to the substituent group to avoid the steric congestion. The products Z-5 could be further elaborated via the SN2 nucleophilic substitution with amine or sodium benzenesulfinate, the reduction of the C-Br bond by NaBH4, and the CuBr-SMe2-catalyzed SN2'-substitution with CH3MgBr.

Abstract:A tandem coupling-ketooxygenation reaction of readily accessible enediyne-carboxylic compounds with inner alkynes has been developed that utilizes the PdCl2/CuBr2 catalytic system under an O2 atmosphere and assembles a class of isoindolinones and o-acylbenzoic acids. The two oxygen atoms are regioselectively incorporated into enediyne units at the 1- and 6-positions from atmospheric molecular oxygen and H2O, respectively, during the present process. This study uncovered a formal [4C + 2C] benzannulation-diketonization of enediynes and alkynes via a coupling and decoupling strategy.

Abstract:Here, we report a strategy enabling triple switchable chemo-, regio-, and stereodivergence in newly developed palladium-catalyzed cycloadditions of allenes. An asymmetric pseudo-stereodivergent cycloaddition of allenes bearing a primary leaving group at the α-position, where a dynamic kinetic asymmetric hydroalkoxylation of racemic unactivated allenes was the enantio-determining step, is realized, providing four stereoisomers [(Z,R), (Z,S), (E,S), and (E,R)] containing a di-substituted alkene scaffold and a stereogenic center. By tuning reaction conditions, a mechanistically distinctive cycloaddition is uncovered selectively with the same set of substrates. By switching the position of the leaving group of allenes, a cycloaddition involving an intermolecular O-attack is disclosed. Diverse mechanisms of the cycloaddition reactions of allenes enable rapid access to structurally and stereochemically diverse 3,4-dihydro-2H-1,4-benzoxazines in high efficiency and selectivity.

Abstract:Two sets of reaction conditions were established to enable the palladium(0)-catalyzed addition of organoboronic acids with 1.2-allenic sulfones, sulfoxides, or alkyl- or aryl-substituted allenes in the presence of acetic acid. This reaction provides a new way for the stereoselective synthesis of tri- or tetrasubstituted E-alkenes. With arylboronic acids, the reactions of 1,2-allenic sulfones, sulfoxides, and alkyl-substituted allenes gave only 1-12% of specific regoisomers; the reactions of aryl-substituted allenes afforded only E-alkenes in very high regioselectivity.

Abstract:An effective method for the synthesis of beta-allyl polysubstituted butenolides from the easily available allylic halides and 2,3-allenoic acids is described. By using this method optically active butenolides can be obtained. According to the results presented in this paper, the reaction may proceed via three consecutive steps: cyclic oxypalladation of the allene, insertion of the C=C bond in allylic halides, and beta-dehalopalladation.

Abstract:CONSPECTUS: Modern synthetic chemists have looked for rapid and efficient ways to construct complex molecules while minimizing synthetic manipulation and maximizing atom-economy. Over the last few decades, researchers have made considerable progress toward these goals by taking full advantage of transition metal catalysis and the diverse reactivities of allenes, functional groups which include two cumulative carbon carbon double bonds.This Account describes our efforts toward the development of Pd-catalyzed cyclization reactions of allenes in the presence of compounds that contain unsaturated carbon carbon bonds such as alkenyl halides, simple alkenes, allenes, electron-deficient alkynes, or propargylic carbonates. First, we discuss the coupling-cyclization reactions of allenes bearing a nucleophilic functionality in the presence of alkenyl halides, simple alkenes, functionalized and nonfunctionalized allenes, or electron-deficient alkynes. These processes generally involve a Pd(II)-catalyzed sequence: cyclic nucleopalladation, insertion or nucleopalladation, and beta-elimination, reductive elimination, cyclic allylation or protonation. We then focus on Pd(0)-catalyzed cyclization reactions of allenes in the presence of propargylic carbonates. In these transformations, oxidative addition of propargylic carbonates with Pd(0) affords allenylpalladium-(II) species, which then react with allenes via insertion or nucleopalladation. These transformations provide easy access to a variety of synthetically versatile monocyclic, dumbbell-type bicyclic, and fused multicyclic compounds. We have also prepared a series of highly enantioenriched products using an axial-to-central chirality transfer strategy.A range of allenes are now readily available, including optically active ones with central and/or axial chirality. Expansion of these reactions to include other types of functionalized allenes, such as allenyl thiols, allenyl hydroxyl amines, and other structures with differing steric and electronic character, could allow access to cyclic skeletons that previously were difficult to prepare. We anticipate that other studies will continue to explore this promising area of synthetic organic chemistry.

Abstract:In this paper, a bimolecular-cyclization reaction between two different bis(allene)s with at least one heteroatom as the tether under the catalysis of trans-[RhCl(CO)(PPh3)(2)] is described. This protocol provides an efficient entry to different heterocyclic 18.19-norsteroid-like scaffolds. The tri-cyclic product was formed highly selectively from the cyclization reaction of bis (2,3-butadienyl) sulfide with dimeth- yl 2-bis(2',3'-butadienyl)malonate, which sheds light on the mechanism involving the metalla-[4.3.0]-bicyclic intermediate formed by the cyclometallation of the terminal and the internal C=C bonds of each of the two allene moieties in 2-bis(2',3'-butadienyl)malonate.

Abstract:Herein we report the first example of a Pd-catalyzed highly selective three-component reaction of alkynyl-1,4-diol dicarbonates, organoboronic acids, and malonate anions for the efficient synthesis of trisubstituted 2,3-allenyl malonates not readily available by the known protocols. The reaction demonstrates an excellent regio- and chemo-selectivity for both the oxidative addition referring to the two C-O bonds and the subsequent coupling with the nucleophile with a remarkable functional group compatibility. A series of control experiments confirm a unique mechanism involving beta-O elimination forming alka-1,2,3-triene and the subsequent insertion of its terminal C = C bond into the Ar-Pd bond.

Abstract:Herein, we report a visible-light-mediated palladium-catalyzed three-component radical-polar crossover carboamination of 1,3-dienes or allenes with diazo esters and amines, affording unsaturated γ- and ε-amino acid derivatives with diverse structures. In this methodology, the diazo compound readily transforms into a hybrid α-ester alkylpalladium radical with the release of dinitrogen. The radical intermediate selectively adds to the double bond of a 1,3-diene or allene, followed by the allylpalladium radical-polar crossover path and selective allylic substitution with the amine substrate, thereby leading to a single unsaturated γ- or ε-amino acid derivative. This approach proceeds under mild and simple reaction conditions and shows high functional group tolerance, especially in the incorporation of various bioactive molecules. The studies on scale-up reactions and diverse derivatizations highlight the practical utility of this multicomponent reaction protocol.

Abstract:A palladium-catalyzed highly E-selective triple cyclization of 2,7-alkadiynylic carbonates with 2-butyne-1,4-diol or 2-butyne-1,4-disulfonamides to construct different tricycles such as 6,8-dihydrobenzo[1,2-c:3,4-c ']difuran-1(3H)-one and 1,3,6,7-tetrahydro-8H-furo[3,4-e]isoindol-8-one derivatives has been reported. The alkene group produced and the nucleophilic unit kept in the product provide further opportunity to construct more complicated polycycles. The control experiments showed that the reaction mainly go through an intermolecular insertion of the alkynes before the lactonization or lactamization.

Abstract:A straightforward protocol for the synthesis of tetra-substituted 2,3-allenoic acids via a catalytic carboxylation of the readily available propargylic alcohols in the presence of CO using palladium and a Bronsted acid as the cocatalysts has been established. Abstract:The development of new strategy for construction of chiral cyclic sulfide-containing multiple stereogenic centers is highly desirable. Herein, by combination of base-promoted retro-sulfa-Michael addition and palladium-catalyzed asymmetric allenylic alkylation, the streamlined synthesis of chiral thiochromanones containing two central chiralities (including a quaternary stereogenic center) and an axial chirality (allene unit) was successfully realized with up to 98% yield, 49.0:1 dr and >99% ee.
