Preparation of Acetylcholinesterase by Crucian Carp Brain and Its Sensitivity to Several Organophosphorus Pesticides

Abstract:Acetylcholinesterase ( AChE) is one kind of crucial enzyme in nerve conduction. For the serine residue at the active site of AChE could be phosphorylated by organophosphorous pesticides ( OPs) , AChE could be used as the target enzyme for OPs nerve positions. Here described the purification and characterization of AChE from carp brain. Firstly, the crude AChE from the crucial carp brain was extracted, and then the enzyme properties including enzyme activity, sensitivity and detection limit were analyzed in the optimized conditions. After purification of crude enzyme by chromatography, the activity of enzyme reached 458.43μmol/min/mg while assay conditions were as 200mmol/L substrate, pH 8.0 and bathing for 30min at 30℃. Enzyme activity could be affected by substrate concentration, pH, temperature, time and dose of enzyme in the descending order. Results also indicated dichlorvos showed the most susceptible to OPs, and then trichlorfon, acephate, tiazophos, poxim and methoate.
