Protective effects of antithrombin Ⅲ on experimentally secondary spinal cord injury in rats

Abstract:Objective To investigate the protective effects of antithrombin Ⅲ(ATⅢ) on experimentally secondary injury of spinal cord in rats and its mechanism.Methods Thirty-six Sprague-Dawleys rats were randomly divided into three groups:sham group(n=12),spinal cord injury(SCI) group(n=12),ATⅢ treatment group(n=12).Only vertebral lamina excision was performed without SCI in sham group.In the other two groups,the SCI rat models were established according to Allen's method.Fifteen minutes after SCI,rats in treatment group were intraperitoneally injected with ATⅢ(250 IU/kg),while those in control group were given normal sodium.The myeloperoxidase(MPO) and superoxide dismutase(SOD) activities and malondialdehyde(MDA) level in spinal tissues were measured at 24 h after operation.Function of nerves was assessed by inclined-plane test and BBB(Basso,Beattie and Bresnahan) scales at 24 h and at 1,2,3 week after operation respectively.Results Compared with sham group,the MPO activity and the level of MDA in control group significantly increased(P<0.01),and the SOD activity reduced(P<0.01).Compared with control group,MPO activity and the level of MDA in treatment group decreased and SOD activity increased significantly(P<0.01).Behavioral score in treatment group was significantly higher than that in control group(P<0.05).Conclusion ATⅢ has neuroprotective effects on secondary spinal cord injury,which might be associated with the inhibition of neutrophils-mediated immune inflammatory reaction.
