Effects of deep brain stimulation of nucleus accumbens on heroin-seeking behavior in self-administration rats

Abstract:Objective To observe effects of deep brain stimulation(DBS) of nucleus accumbens on heroin-seeking behavior in self-administration rats.Methods Self-administration rat models of relapsing in heroin was established with fixed ration program,and model rats were randomly divided into 3 groups: a control group,a sham stimulation group and a DBS group;the rats of sham stimulation group and DBS group were implanted bilaterally with concentric bipolar microelectrodes.The the rats in DBS group were given a 1-hour-long high frequency electrical stimulation(Parameters:square pulse,130Hz frequency,150μA current intensity,100μs pulse width) for 7 days during the extinction phase.Heroin-seeking behavior was elicited by conditioned cues and small dose of heroin.Results After treatments,the active nose-poke responses of rats in DBS group(8.00±5.33) was significantly decreased as compared with the control group(36.50±9.16) and sham stimulation group(34.00±7.96) in the reinstatement induced by conditioned cues(P<0.01);and the nose-poke responses of rats in DBS group(11.17±7.78) was significantly decreased as compared with the control group(29.67±5.24) and sham stimulation group(28.00±11.92) in the reinstatement induced by small dose of heroin(P<0.01);there were no significant differences in the total and phases of locomotor activity(P>0.01).Conclusion High frequency stimulation of nucleus accumbens can reduce the heroin-seeking behaviors in self-administration rats,and has no effect on the locomotor activity.
