Decreased mature adrenomedullin level in umbilical plasma in preecalmpsia and its role in enhancing invasion of trophoblast

Abstract:Objective To determine the mature Adm level in umbilical plasma of both normal and preeclamptic pregnancies, and to explore the effect of mature Adm on the invasive ability of trophoblast from preeclampsia in vitro. Methods Umbilical fresh plasma was taken and trophoblasts were isolated and cultured from 10 preeclamptic(≥35 wk)and 7 normal(≥39 wk)pregnancies. Plasma mature Adm levels were measured using a radioimmunoassay and the difference between normal and preeclamptic pregnancy was compared with non-paired t-test; After treating the cultured trophoblasts with 10-8M Adm for 48hs, the number of cells penetrating the filter pores, either in treated group or control group, was counted and compared.Results The mature Adm value in umbilical plasma of preeclamptic pregnancy(2.97±1.31 fmol/ml,n=10)was significantly lower than that of normal pregnancy (3.95±1.86 fmol/ml,n=7)(P<0.05); The number of the cells penetrating the filter pores after treated with Adm was significantly higher than of control (n=6,P<0.01). Conclusion The defective implantation and placentation of preeclamsia might be due to the low level of mature Adm in fetal circulation and increasing the content of the functional form Adm might become a new choice for preeclamptic pregnancy.
