THE ANTIFATIGUE EFFECTS AND MECHANISM OF GINSENG STEM AND LEAF SAPONIN ON WEIGHT-BEARING SWIMMING MICE

Abstract:Objective To explore the antifatigue effects and the mechanism of Ginseng stem and leaf saponin(GSLS).Methods Twenty male and 20 female Kunming clean mice with the body mass of(21±3)g were randomly divided into four groups: control group(NC group),GSLS 10 mg/(kg·d)(LD group),GSLS 40 mg/(kg·d)(MD group),and GSLS 160 mg/(kg·d)(HD group).In each group,it was subdivided into 7-day and 28-day subgroup.The mice in NC group were given intragastric administration of normal saline for seven or 28 days.Those in LD,MD and HD group were respectively given intragastric administration of corresponding dose of GSLS for seven or 28 days.The time of exhausting swimming,the levels of muscle glycogen,serum lactic dehydrogenase(LDH) and blood urea nitrogen(BUN) were measured.Results After 28-day GSLS administration,the levels of muscle glycogen in the mice in MD and HD group markedly elevated(F=6.62;q=6.565,5.102;P0.01),and postexercise serum LDH activity in MD raised(F=6.91;q=9.021;P0.01),BUN lowered,and BUN clearance rate accelerated(F=5.25;q=4.329;P0.05).Conclusion GSLS plays an anti-fatigue action through regulating glycogen storage,increasing LDH activity,and adjusting BUN level.The optimized dosage of GSLS was 40 mg/(kg·d).The dosage of 160 mg/(kg·d) for 28 days might cause a certain toxic reaction.
