Abstract:One novel amperometric immunosensor for α-fetop- rotein was fabricated which was modified by antibody immobilized on nano ZrO2-Au-polyLysine composite membrane. Firstly, the deoxyribonucleic acid – phenoxyacetic acid isoniazid- Schiff base Co(II) complex (DNA-CoL) mixture film was casted on surface of multi carbon nanotubes(MWCNTs) modified glassy carbon electrode(GCE/CNTs/DNA-CoL). Then the composite nanoparticles (ZrO2-Au-pLL) was employed to immobilize the antibody of α-fetoprotein(AFP) to produce the probes(ZrO2-Au-pLL-anti AFP) for AFP. Finally the probes were modified on GCE/CNTs/DNA-CoL through the specific connection between DNA and ZrO2 to form a novel nanocomposite membrane immunosensor for AFP(GCE/CNTs/ DNA-CoL/ZrO2-Au–pLL-anti AFP). The function of immunosensor was investigated by scanning electron microscopy (SEM), cyclic voltammetry(CV), differential pulse voltammety (DPV) and electrochemical impedance spectroscopy experiment (EIS). The results indicated that combining the advantages of the MWNTs-DNA material and ZrO2-Au-pLL-anti AFP nano probes, the immunosensor with excellent sensitivity and selectivity to AFP was prepared successfully. The modified electrode was sensitive to AFP with a linear relationship between 0.05 and 10 ng · mL−1 and a correl- ation coefficient of 0.9905. The detection limit at a signal to noise ratio of 3 was 0.01 ng·mL−1 under the optimal conditions. The described immunosensor preparation and immunoassay methods offer promise for simple and cost-effective analysis of AFP in serum samples.

Abstract:A high performance electrochemical immunosensor was constructed for sensitive label-free detection for alpha-fetoprotein (alpha-FP) based on Au nanoparticles/poly(3,4-ethylenedioxythiophene) modified multi-walled carbon nanotubes (AuNPs/PEDOT-MWCNTs), which acted as signal amplification matrix. PEDOT functionalized MWCNTs were prepared via interfacial polymerization method. PEDOT was uniformly distributed on MWCNTs, which can provide more binding sites for high mass loading of metal nanoparticles. Subsequently, AuNPs were attached on MWCNTs by introducing conductive PEDOT to bridge the AuNPs and MWCNTs walls. The AuNPs/PEDOT-MWCNTs nanocomposites displayed good electron transfer ability, which enabled good analytical properties for alpha-FP detection.

Abstract:A sensitive label-free amperometric immunosensor was developed based on the amine-functionalized graphene (GR-NH2) and gold nanoparticles (AuNPs) composite modified carbon ionic liquid electrode (CILE). CILE was fabricated by using an ionic liquid of 1-octyl-3-methylimidazolium hexafluorophosphate as binder. The nanocomposite AuNPs/GR-NH2 had prominent biocompatibility, good electron transfer ability, large specific surface area, and primarily excellent adsorption. The negatively charged AuNPs could be adsorbed on the positively charged GR-NH2 modified CILE surface by electrostatic adsorption, and then to immobilize α-fetoprotein antibody (anti-AFP) for the assay of α-fetoprotein (AFP). The fabricated procedures and electrochemical behaviors of the immunosensor were characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The anti-AFP/AuNPs/GR-NH2 modified CILE was sensitive to AFP in linear relation between 1 and 250ngmL−1 with the correlation coefficient of 0.995, and the detection limit (S/N=3) was 0.1ngmL−1 under the optimal conditions. In addition, the proposed immunosensor exhibited good sensitivity, selectivity, stability and long-term maintenance of bioactivity and it may be used to immobilize other biomoleculars to develop biosensor for the detection of other antigens or biocompounds.

Abstract:The authors describe an electrochemical immunoassay for α-fetoprotein (α-FP) using a glassy carbon electrode (GCE) modified with a nanocomposite made from gold nanoparticles, graphene oxide and multi-walled carbon nanotubes (AuNPs/GO-MWCNTs) and acting as a signal amplification matrix. The nanocomposite was synthesized in a one-pot redox reaction between GO and HAuCl4 without using an additional reductant. The stepwise assembly of the immunoelectrode was characterized by means of cyclic voltammetry and electrochemical impedance spectroscopy. The interaction of antigen and antibody on the surface of the electrode creates a barrier for electrons and causes retarded electron transfer, this resulting in decreased signals in differential pulse voltammetry of hexacyanoferrate which is added as an electrochemical probe. Using this strategy and by working at a potential of 0.2 V (vs. SCE), a wide analytical range (0.01 - 100 ng∙mL‾1) is covered. The correlation coefficient is 0.9929, and the limit of detection is as low as 3 pg∙mL‾1 at a signal-to-noise ratio of 3. This electrochemical immunoassay combines the specificity of an immunological detection scheme with the sensitivity of an electrode modified with AuNPs and GO-MWCNTs.

Abstract:The authors describe a voltammetric immunosensor with antibody immobilized on a glassy carbon electrode (GCE) modified with N-doped graphene (N-GS), electrodeposited gold nanoparticles (AuNPs) and chitosan (Chit). The preparation is simple and the thickness of the electrodeposited films can be well controlled. Due to the specific advantages of N-GS, AuNPs and Chit, the electrode has a large specific surface, improved conductivity, high stability. A new label-free immunosensor for the model antigen (alpha fetoprotein, AFP) detection was then designed by employing N-GS-AuNP-Chit as the antibody immobilization and signal amplification platform. Differential pulse voltammetry and electrochemical impedance spectroscopy were used for the characterization of the stepwise assembly process. Under the optimized conditions, at a typical working potential of +0.20 V (vs. SCE), and by using hexacyanoferrate as an electrochemical probe, the immunosensor has a detection limit as low as 1.6 pg mL−1 and a linear analytical range that extends from 5 pg mL−1 to 50 ng mL−1. AFP was quantified in spiked human serum samples with acceptable precision.

Abstract:The synthesis of Au nanocubes is used to label alpha-fetoprotein antibody (anti-AFP) and horseradish peroxidase (HRP) to form an immune complex for antibody detection. Graphene oxide-methylene blue-gold nanoparticles (GO-MB-AuNPs) nanocomposites were used as the immunosensing platform. This proposed sandwich-type immunoassay shows good performance. This method establishes a feasible amperometric immunoassay method for sensitive analysis of AFP in serum samples. Under the optimal experimental conditions, the DPV current response of the immunosensor is proportional to the logarithmic value of the AFP concentration. The linear detection range can achieve to 0.005–20 ng/mL with a detection limit of 1.5 pg/mL. The proposed immunosensor has good precision, selectivity and stability, and can be used for AFP determination in clinical tests.

Abstract:A sensitive electrochemical immunosensor was developed for detection of alpha-fetoprotein (AFP) based on a three-dimensional nanostructure gold electrode using a facile, rapid, "green" square-wave oxidation-reduction cycle technique. The resulting three-dimensional gold nanocomposites were characterized by scanning electron microscopy and cyclic voltammetry. A "sandwich-type" detection strategy using an electrochemical immunosensor was employed. Under optimal conditions, a good linear relationship between the current response signal and the AFP concentrations was observed in the range of 10-50 ng/mL with a detection limit of 3 pg/mL. This new immunosensor showed a fast amperometric response and high sensitivity and selectivity. It was successfully used to determine AFP in a human serum sample with a relative standard deviation of <5% (n=5). The proposed immunosensor represents a significant step toward practical application in clinical diagnosis and monitoring of prognosis.

Abstract:An electrochemical immunosensor for quantitative detection of α-fetoprotein (AFP) in human serum was developed using graphene sheets (GS) and thionine (TH) as electrode materials and mesoporous silica nanoparticles (MSNs) loaded with ferroferric oxide (Fe3O4) nanoparticles and horseradish peroxidase (HRP) as labels for signal amplification. In this study, the compound of GS and TH (GS-TH) was used as a substrate for promoting electron transfer and immobilization of primary antibody of AFP (Ab1). MSNs were used as a carrier for immobilization of secondary antibody of AFP (Ab2), Fe3O4, and HRP. The synergistic effect occurred between Fe3O4 and HRP and greatly improved the sensitivity of the immunosensor. This method could detect AFP over a wide concentration range from 0.01 to 25 ng ml(-1) with a detection limit of 4 pg ml(-1). This strategy may find wide potential application in clinical analysis or detection of other tumor markers.

Abstract:Herein, a stable and sensitive label-free electrochemical immunosensor for detecting α-fetoprotein (AFP) was proposed based on gold nanoparticles-poly(3,4-ethylenedioxythiophene)/Prussian blue-reduced graphene oxide (AuNPs-PEDOT/PB-rGO) nanocomposite as sensing substrate. PB was used as the electron transfer mediator to provide the electrochemical signal. By utilizing rGO as the supporting material for PB, the stability and intensity of the redox signal were greatly improved. The incorporation of highly conductive AuNPs-PEDOT can further enhance the electrochemical signal and its stability. Electrochemical test showed the anodic current of AuNPs-PEDOT/PB-rGO modified electrode could remain up to 89.45% after 100 cyclic voltammetries. The assembly of PB-rGO and AuNPs-PEDOT exhibited 3D nanostructure and showed high specific surface area, which could effectively capture the antibody of AFP. With the advantages of AuNPs-PEDOT/PB-rGO nanocomposite, the prepared immunosensor showed a sensitive response to AFP in a linear range from 0.01 to 50ngmL−1 with a low detection limit of 3.3pgmL−1 (S/N=3). The proposed immunosensor also has been employed to determine AFP in human serum with satisfactory results. These excellent properties make the AuNPs-PEDOT/PB-rGO nanocomposite an ideal interface for efficient biomarkers detection.

Abstract:A functional electrochemical immunosensor was successfully designed for simultaneous and high sensitivity detection of carcinoembryonic antigen (CEA) and α-fetoprotein (AFP). This electrochemical immunosensor was mainly designed by surface-initiated poly(glycidyl methacrylate) (PGMA) from multi-walled carbon nanotubes (MWCNTs) with the methodology of mussel-inspired chemistry (polydopamine, PDA) and metal-free photoinduced electron transfer-atom transfer radical polymerization (PET-ATRP). Controlled PGMA was initiated from MWCNTs, in which Br-functional PDA-modified MWCNTs and eosin Y (EY) were used as initiator and photocatalyst, respectively. The epoxy group of PGMA is reacted with ethylenediamine (EDA) by a ring-opening reaction, the amino and hydroxyl groups in the polymer chain provide numerous effective binding sites for carboxyl groups of signal molecules and capture antibodies, realizing highly sensitive electrochemical detection. Under optimized conditions, the obtained hybrid nanoprobes (Aq/PGMA-g-MWCNTs and Fc/PGMA-g-MWCNTs) were used as efficient electrochemical immunosensor to simultaneously detect CEA and AFP. It is worth noting that the electrochemical immunosensor performed a wide detection range and a low detection limit of CEA (163 fg/mL ~ 163 ng/mL, 56.1 fg/mL) and AFP (100 fg/mL ~ 100 ng/mL, 32.8 fg/mL), respectively. This high sensitivity and selectivity detection platform has a potential application in the simultaneous detection of more tumor markers.

Abstract:This paper presents a novel nano-material composite membrane for detecting aflatoxin B1 (AFB1). The membrane is based on carboxyl-functionalized multi-walled carbon nanotubes (MWCNTs-COOH) @ antimony-doped tin oxide (ATO)-chitosan (CS). To prepare the immunosensor, MWCNTs-COOH were dissolved in the CS solution, but some MWCNTs-COOH formed aggregates due to the intertwining of carbon nanotubes, blocking some pores. ATO was added to the solution containing MWCNTs-COOH, and the gaps were filled by adsorbing hydroxide radicals to form a more uniform film. This greatly increased the specific surface area of the formed film, resulting in a nano-composite film that was modified on screen-printed electrodes (SPCEs). The immunosensor was then constructed by immobilizing anti-AFB1 antibodies (Ab) and bovine serum albumin (BSA) on an SPCE successively. The assembly process and effect of the immunosensor were characterized using scanning electron microscopy (SEM), differential pulse voltammetry (DPV), and cyclic voltammetry (CV). Under optimized conditions, the prepared immunosensor exhibited a low detection limit of 0.033 ng/mL with a linear range of 1 × 10−3–1 × 103 ng/mL. The immunosensor demonstrated good selectivity, reproducibility, and stability. In summary, the results suggest that the MWCNTs-COOH@ATO-CS composite membrane can be used as an effective immunosensor for detecting AFB1.

Abstract:An ultrasensitive multiplexed electrochemiluminescence immunoassay method was developed for the detection of tumor markers by combining a functionalized graphene nanosheets and gold-coated magnetic Fe3O4 nanoparticles (GMPs) labeled alpha-fetoprotein (AFP) antibody (GMP similar to Ab1). The functionalized graphene nanosheets (PDDA-G) with poly (diallyldimethylammonium chloride) (PDDA) were synthesized and used to combine with luminol-capped gold nanoparticles (Lu-Au NPs). The resulting PDDA-G@Lu-Au composite displayed an enhanced capability for the immobilization of horseradish peroxidase (HRP) and signal antibody (Ab2) to realize its electrochemiluminescence immunoassay. Great signal amplification was achieved since PDDA-G could adsorb large amount of reporter molecules (Lu-Au). Besides gold nanoparticles were not only used as carriers of Ab2 and HRP but also catalyzed the ECL reaction of luminol, which further amplified the ECL signal of luminol in the presence of H2O2. GMPs as supporting material, not only been performed the rapid separation and purification of signal antibody on magnetic field, but also enhanced the fixed capacity of Ab1 to improve detection range. In addition, the magnetic probes were readily immobilized on the working electrode of screen-printed carbon electrode (SPCEs) by magnets. Under the optimized conditions, the ECL method shows a linear range of alpha-fetoprotein (AFP) from 0.002 to 20 ng mL(-1) and an extremely low detection limit of 0.2 pg mL(-1) (3 sigma). Moreover, the proposed method is also simple, stable, specific, and time-saving, avoiding the complicated process modification on the electrodes' surface, which may open a new door to ultrasensitive detection of tumor markers in clinical analysis.
Label-Free Amperometric Immunosensor Based on Versatile Carbon Nanofibers Network Coupled with Au Nanoparticles for Aflatoxin B1 Detection

Abstract:Facile detection methods for mycotoxins with high sensitivity are of great significance to prevent potential harm to humans. Herein, a label-free amperometric immunosensor based on a 3-D interconnected carbon nanofibers (CNFs) network coupled with well-dispersed Au nanoparticles (AuNPs) is proposed for the quantitative determination of aflatoxin B1 (AFB1) in wheat samples. In comparison to common carbon nanotubes (CNTs), the CNFs network derived from bacterial cellulose biomass possesses a unique hierarchically porous structure for fast electrolyte diffusion and a larger electrochemical active area, which increases the peak current of differential pulse voltammetry curves for an immunosensor. Combined with AuNPs that are incorporated into CNFs by using linear polyethyleneimine (PEI) as a soft template, the developed Au@PEI@CNFs-based immunosensor showed a good linear response to AFB1 concentrations in a wide range from 0.05 to 25 ng mL-1. The limit of detection was 0.027 ng mL-1 (S/N = 3), more than three-fold lower than that of an Au@PEI@CNTs-based sensor. The reproducibility, storage stability and selectivity of the immunosensor were proved to be satisfactory. The developed immunosensor with appropriate sensitivity and reliable accuracy can be used for the analysis of wheat samples.

Abstract:In this work, a label-free electrochemical immunosensor was structured for detecting alpha fetal protein (AFP) effectively. The proposed electrochemical immunosensor was structured by gold silver platinum nanodendrites loaded on amino-functionalized MoO 2 nanosheets (AuAgPt NDs/NH 2 -MoO 2 NSs). The AuAgPt NDs with rich dendrite structure provided abundant specific surface area for effective capture antibody. MoO 2 possessed the satisfactory catalytic ability in terms of H 2 O 2 reduction and charge transfer capacity. AuAgPt NDs possessed favourable catalytic performance to H 2 O 2 reduction. Therefore, the prepared AuAgPt NDs/NH 2 -MoO 2 NSs was better than AuAgPt NDs and NH 2 -MoO 2 NSs in H 2 O 2 reduction on account of a synergistic effect. The constructed electrochemical immunosensor possessed a satisfying detection limit of 3.3 fg ml −1 and a broad detection range which was from 10 fg ml −1 to 100 ng ml −1 (S/N = 3) for the AFP detection under optimal conditions. The constructed electrochemical immunosensor possessed favourable detection performance in sensitivity, stability, selectivity and reproducibility, which indicated that it possessed a practical application in the aspect of clinical detection.

Abstract:A renewable and magnetic electrochemiluminescence (ECL) immunosensor for alpha-fetoprotein (AFP) was fabricated by using RuL-MWNTs (Tris(2,2'-bipyridine) ruthenium(II) complex immobilized on multiwall carbon nanotubes through pi-pi stacking function between the carbon aromatic systems) doped Au (RuL-MWNTs@Au) composite as labels. Polyelectrolyte poly(dimethyldiallylammonium chloride) (PDDA) was employed to covered on the surfaces of RuL-MWNTs which made it have positive electric charge and can absorb negative colloid Au nanoparticles to fabricate RuL-MWNTs@Au through electric attraction. The primary antibody, anti-AFP was first immobilized on the Fe3O4(core)/gold(shell) nanoparticles (Fe3O4@Au, GMP) as probes. Then the antigen and the RuL-MWNTs@Au composite nanoparticles labeled Ru(bpy)(3)(2+)-conjuged secondary antibody (RuL-MWNTs@Au similar to RuLAb2) were combined successively to form RuL-MWNTs@Au similar to RuLAb2/AFP/anti-AFP-GMP sandwich-type immunocomplexes through the specific interaction. The resulting magnetic sandwich immune complexes can be absorbed on the planar screen printed carbon electrode (SPCE) due to the external magnetic field added on the bottom of it. The magnetic sandwich complexes can be easily washed away from the surface of SPCE when the magnetic field was removed which made the immunosensor reusable. The prepared RuL-MWNTs@Au composite nanoparticles own large surface area, good biocompatibility and highly effective ECL properties. The morphologies of the RuL-MWNTs@Au and its antibody labeled composite nanoparticles were characterized by transmission electronic microscope (TEM). The sensor performed high sensitivity and wide liner for detection AFP in the range of 0.01-50 ng.mL(-1) and the limit detection was 3 pg.mL(-1) (defined as S/N=3). Furthermore the sandwich immunocomplexes can be performed the rapid separation from the unbound secondary antibody quickly by magnetic field. So the immunosensor can simultaneously realize separation, enrichment and determination, with high sensitivity, which would be valuable for clinical immunoassay for AFP in human serum.

Abstract:Nanogold functionalized mesoporous carbon foam (Au/MCF) coupling with a signal amplification by C–Au synergistic silver enhancement was designed for sensitive electrochemical immunosensing of biomarker. The Au/MCF was prepared by in situ growth of nanogold on carboxylated MCF and used as a tracing tag to label signal antibody via the inherent interaction between protein and nanogold. The immunosensor was prepared by covalently immobilizing capture antibody on an electrochemically reduced graphene oxide/chitosan film modified glassy carbon electrode. Through a sandwich-type immunoreaction, Au/MCF tags were captured on the immunoconjugates to induce a silver deposition process. The electrochemical stripping signal of the deposited silver was used to monitor the immunoreaction. The Au/MCF-mediated silver enhancement along with the graphene-promoted electron transfer led to high detection sensitivity of carcinoembryonic antigen. Under optimal conditions, the proposed immunoassay method showed wide linear range from 0.05pgmL−1 to 1ngmL−1 and a detection limit down to 0.024pgmL−1. The newly designed amplification strategy holds great potential for ultrasensitive electrochemical biosensing of other analytes.
Colorimetric Immunosensor Based on Au@g-C3N4-Doped Spongelike 3D Network Cellulose Hydrogels for Detecting α-Fetoprotein

Abstract:A colorimetric immunoassay is a powerful tool for detecting tumor markers, with outstanding advantages of visualization and convenience. This study designed a colorimetric immunoassay using the antibody/antigen to control the catalytic activity to be "switched on/off". This system, where Au NPs (18.5 ± 3.9 nm) were loaded on the g-C3N4 nanosheets that were fixed in a three-dimensional porous cellulose hydrogel, was used as a binding site for the antibody/antigen. After being incubated with an antibody of a cancer marker, the turned-off catalytic sites on Au NPs in Au@g-C3N4/microcrystalline cellulose hydrogels would not be "turned on" until the corresponding antigen was added. The number of the recovered Au active sites was related to the amount of the antigen added. The Fourier transform infrared and X-ray photoelectron spectroscopy measurements did not detect the existence of Au-S bonds. Catalyzed by the turned-on Au NPs, 4-nitrophenol was reduced to 4-aminophenol accompanied by a color fading. The color and the absorption spectrum changes in the process were used as the colorimetric quantitative basis for immunoassays. The colorimetric immunoassay showed a linear relationship with the liver cancer marker (α-fetoprotein, AFP) in the range of 0.1-10 000 ng/mL with the detection limit of 0.46 ng/mL. In addition, 4-nitrophenol had a significant color fading when the AFP concentration exceeded the healthy human threshold. The clinical patient's serum test results obtained from the developed colorimetric immunosensor were consistent with those obtained from the commercial enzyme-linked immunosorbent assay. Furthermore, the immunosensor exhibited a good selectivity, repeatability, and stability, which demonstrated its potential for practical diagnostic application.

Abstract:Aflatoxin in food is a significant concern that threatens human health and life and causes a heavy burden on international trade and healthcare. In this work, we developed a new type of label-free electrochemical immunosensor for the high-sensitivity determination of aflatoxin B1 (AFB1) based on Au nanoparticles decorated urchin-like Bi 2 S 3 (Au/Bi 2 S 3 ) anchored on electrochemically reduced graphene oxide (ERGO) modified carbon fiber (CF) microelectrode (Au/Bi 2 S 3 /ERGO/CF). The morphology and characterization of the modified microelectrode were investigated using scanning electron microscopy (SEM), cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and differential pulse voltammetry (DPV). Under the optimum conditions, the label-free electrochemical immunosensor detected AFB1 within the linear range of 10 pg mL -1 to 20 ng mL -1 , with a low detection limit of 8 pg mL -1 and high sensitivity (0.48 μA ng -1 mL). The immunosensor showed high selectivity, good reproducibility, and acceptable stability. The immunosensor was applied to detecting AFB1 in cornflour samples, offering excellent reliability and accuracy compared with typical detection methods. Finally, this platform provides a superior prospect for the development of a nanohybrid immunosensor.

Abstract:Metal oxide-based sensors have the benefit of inexpensive, quick response, and high sensitivity in detecting specific biological species. In this article, a simple electrochemical immunosensor was fabricated using antibody-chitosan coated silver/cerium oxide (Ab-CS@Ag/CeO2) nanocomposites on a gold electrode for sensitive alpha-fetoprotein (AFP) diagnosis in human serum samples. Successfully synthesis of AFP antibody-CS@Ag/CeO2 conjugates was confirmed through FTIR spectra of the product. The amine coupling bond chemistry was then used to immobilize the resultant conjugate on a gold electrode surface. It was observed that the interaction of the synthesized Ab-CS@Ag/CeO2 nanocomposites with AFP prevented an electron transfer and reduced the voltammetric Fe(CN)63-/4- peak current, which was proportional to the amount of AFP. The linear ranges of AFP concentration were found from 10-12-10-6 gmL-1. The limit of detection was calculated using the calibration curve and came out to be 0.57pgmL-1. The designed label-free immunosensor successfully detected AFP in human serum samples. As a result, the resulting immunosensor is a promising sensor plate form for AFP detection and could be used in clinical bioanalysis.
