Pharmacokinetics of Aconitine and Glycyrrhetic Acid in Separation and Combination with Aconiti Lateralis Radix Praeparata and Glycyrrhizae Radix Et Rhizoma in Rats

Abstract:AIM To investigate the plasma pharmacokinetics change of aconitine and glycyrrhetic acid in rats compared among Shengfuzi Decoction(Aconiti lateralis Radix praeparata),Gancao Decoction(Glycyrrhizae Radix et Rhizoma) and Fuzi Gancao Decoction(Aconiti lateralis Radix praeparata,Glycyrrhizae Radix et Rhizoma).METHODS The plasma pharmacokinetics of aconitine and glycyrrhetic acid were studied in 156 male Sprague-Dawley rats with body weight 237-292 g.The rats were randomly divided into three decoction groups.The decoction was single intragastricly administered to each rat at a volume of 25 mL/kg,including Shengfuzi Decoction,Gancao Decoction and Fuzi Gancao Decoction in which aconitine and glycyrrhetic acid contents were detected respectively.Blood samples(2.0 mL)were collected at thirteen time intervals after dosing.Four rats were used for each set time point.LC-MS/MS was used for determination of aconitine and LC-MS was used for determination of glycyrrhetic acid in rat plasma.The main pharmacokinetic parameters were calculated by DAS2.1 pharmacokinetic soft.RESULTS There was no significant difference in the dissolution of aconitine and glycyrrhetinic acid after compatibility of licorice and aconite.The main pharmacokinetic parameters of aconitine in Shengfuzi Decoction and Fuzi Gancao Decoction were as follows:AUC0-t were 730.66 pg·h/L and 538.97 pg·h/L respectively.Cmax were 91.43 pg/L and 42.47 pg/L,respectively.Tmax were 1.0 h and 7.25 h,respectively.t1/2 were 14.69 h and 7.81 h,respectively.The main pharmacokinetic parameters of glycyrrhetic acid in Gancao Decoction and Fuzi Gancao Decoction were as follows:AUC0-t were 8 159.83 ng·h/L and 7 035.89 ng·h/L,respectively.Cmax were 526.5 ng/L and 458.25 ng/L,respectively.Tmax were 8 h and 12 h,respectively.t1/2 were 11.99 h and 9.57 h,respectively.CONCLUSION Compared with Fuzi Gancao Decoction,both the peak time of aconitine in Shengfuzi Decoction and glycyrrhetic acid in Gancao Decoction are delayed,the maximal concentration and its absorption decrease,but the elimination speeds up.
