Preparation and identification of monoclonal antibodies against human prostate stem cell antigen

Abstract:Objective To establish a hybridoma cell line secreting monoclonal antibody (McAb) against human PSCA. Methods BALB/c mouse was immunized by purified PSCA,A murine McAb designated D3 was generated and the specificity was tested by immunohistochemistry and Western bloting. Results A positive hybridome cell lines which contained McAb against PSCA were obtained.It belong to IgG1 according to the examination of immunoglobulin classification. Western-blot test showed that the McAb reacted specifically with PSCA without other crossing reactions.The antibody detected PSCA expression on the cell surface of normal and malignant prostate cells,PSCA protein expression was identified in 38/40(95%)primary prostate cancers.There was no statistical difference in PSCA expression level between normal prostate tissues and BPH,but the PSCA expression level of prostate cancer was of significant statistical difference. Conclusions It was showed the antibody could be used in diagnosis and immunotherapy of prostate cancer.
