The influence of mitogen activated protein kinases inhibitor on the Toxoplasma gondii tachyzoites invading to host cells

Abstract:Objective To observe the effect of the mitogen activated protein kinases(MAPK) inhibitor on Toxoplasma gondii tachyzoites invading to host cells,and to explore the effect of MAPK inhibitor on the Toxoplasma gondii tachyzoites invading and proliferation in the host cells. Methods The MAPK inhibitor was added in the tachyzoite-NIH3T3 cell and HL-60 cell culture system in different dosage,then the infect cells were counted in different time(3-9h) by light microscope with the Giemsa staining, and by flow cytometry(FCM). Results The microscopic counting showed that compared to the control the infective rate of host cells with U0126 1μM,10μM and 100μM was decreased to 20.22%,(P<0.01), 52.91%(P<0.01)and 75.27%(P<0.01)at 3 hours respectively.After 6 and 9 hours,Compared to the control the infective rate of host cells with U0126 1μM,10μM and 100μM was decreased to 32.88%(P<0.01),57.24%(P<0.01),79.21 %(P<0.01)and 41.26%(P<0.01), 54.73%(P<0.01),80.80%(P<0.01), respectively. Conclusion The infective rates of host cells with U0126 inhibitor were remarkably lower than that of the control group ,the cell invading process of Toxoplasma gondii tachyzoites was inhibited by affecting the MAPK cellular signal pathway.
