Abstract:Osteopontin (OPN), a secreted phosphoprotein, is a member of the small integrin-binding ligand N-linked glycoprotein (SIBLING) family of cell matrix proteins and participates in many biological activities. Studies have shown that OPN plays a role in bone metabolism and homeostasis. OPN not only is an important factor in neuron-mediated and endocrine-regulated bone mass, but also is involved in biological activities such as proliferation, migration, and adhesion of several bone-related cells, including bone marrow mesenchymal stem cells, hematopoietic stem cells, osteoclasts, and osteoblasts. OPN has been demonstrated to be closely related to the occurrence and development of many bone-related diseases, such as osteoporosis, rheumatoid arthritis, and osteosarcoma. As expected, the functions of OPN in the bone have become a research hotspot. In this article, we try to decipher the mechanism of OPN-regulated bone metabolism and bone diseases.

Abstract:As a whole,the calcium in diet of national people is not sufficient.The calcium mainly exists in bone,and the adequate calcium content is very important for healthy bone.So,calcium should be supplied sufficiently,in which,the absorbsion of calcium is the most important thing.Accordingly,it is worthy of developing the additive of calcium supplying and fixing.It was shown in some specialists' research that osteoblasts milk protein(OMP)has good and especial effects on increasing the bone density and promoting the growth of bone.In this paper,the relationship among the calcium supplement,healthy bone and OMP,consequently,the theory base of bioactivity of OMP was analyzed.

Abstract:Matrix protein is a kind of secretory protein that regulates the biomineralization of the bivalve shell. In this study, a water-soluble matrix protein (WSMP) from Pacific oysters (Crassostrea gigs) shell was isolated, and its structure was analyzed in detail, in addition to its anti-osteoporosis activity in vitro and in vivo. Results showed that WSMP was an acidic protein with an apparent molecular mass of 47 and 79 kDa and contained a glycoprotein structure. In vitro, the reduction of Tartrate-resistant acid phosphatase (TRAP) and deoxypyridinoline (DPD) indicated that osteoclast activity was inhibited compared with the model group. Moreover, the increased osteocalcin (OCN) and BMD levels suggested that the high osteoblast activity and bone mineralization was improved. SEM analysis of the femur showed that there were fewer bone pits in experimental groups, which was consistent with the above results. In vivo, WSMP promoted the expression of alkaline phosphatase (ALP) and osteogenic differentiation factor BMP-2 in osteoblasts. In addition, the activity of osteoclasts was inhibited by regulating the process of osteoclast differentiation induced by RANKL. Both in vitro and in vivo studies showed that WSMP could promote osteogenesis and inhibit osteoclast absorption, thus demonstrating their potential applications in osteoporosis.

Abstract:Osteoporosis is a systemic bone disease characterized by decreased bone mass, decreased bone mass, and bone microstructure degradation. The cause is that the activity of osteoclasts is greater than that of osteoblasts, resulting in bone resorption greater than bone formation. The current method of treating osteoporosis is mainly through calcium supplementation or drug treatment, but it would be accompanied by disadvantages such as poor absorption, insignificant effect, and large side effects. Plant natural active ingredients have the characteristics of wide sources, abundant structure and less adverse reactions. They could be used as a new method to prevent and treat osteoporosis. This article summarizes the anti-osteoporosis effects of osteoporosis and plant active ingredients. Through the analysis of related signal pathways, transcription factors, protein expression and other aspects, the anti-osteoporosis mechanism of plant active ingredients and related clinical research are described. On this basis, the anti-osteoporosis products of medicinal and edible plants are introduced, aiming to provide theoretical support for the development of new drugs and functional foods.

Abstract:Milk protein, consists of caseins, whey proteins, milk fat globule membrane proteins ( MFGMPs) and other ingredients, is one of the most important components of the milk and is a major index for evaluating milk quality. MFGMPs is a special sub-unit of milk protein, which has the functions of anti-cancer, anti-virus infection, increasing immunity, etc. Additionally, MFGMPs has a close relationship with the occurrence of multiple sclerosis, autism, coronary heart disease. This paper mainly reviewed the components of MFGMPs in sheep and goat milk and its differential components among different breeds, different lactation stages, different species, and between acute phase and normal phase, which could facilitate us to understand the components, functions and secretion mechanism of MFGMPs better, and thus provide theoretic references for making reason-able weaning strategy for lambs and developing optimal formula of lambs' milk replacer.

Abstract:Introduction Milk is a good source of bioavailable calcium compared with other foods. Recent in vitro and in vivo studies have shown that milk whey protein, especially its basic protein fraction (milk basic protein, MBP), contains several components capable of promoting bone formation and inhibiting bone resorption. The objective of this study was to examine the effects of MBP on bone mineral density (BMD) and bone metabolism of healthy young women.Methods Eighty-four healthy young women were randomly assigned to three groups: control group, whole milk group or MBP group treated with milk containing 40 mg MBP for 8 months. The bone mineral density of total body, the lumbar vertebrae L2-L4 and the left forearm of each subject were measured by dual-energy X-ray absorptiometry (DEXA) at 0 and 8 months of treatment. Serum indexes of bone metabolism were measured at 0, 3, 6 and 8 months. Eighty-one subjects who completed the study in accordance with the protocol were included in the analysis.Results Total BMD in all groups significantly increased compared with baseline values. However, no significant difference on the mean rate of gain of total BMD was observed among the MBP group (2.19%), the whole milk group (2.63%) and the control group (1.61%). Serum crosslinked N-teleopeptides of type-I collagen (NTx) in MBP group at 8 months and in whole milk group at 6 months were significantly decreased from baseline. There were no significant differences between whole milk group and MBP group; however, after combining the milk groups, NTx had significantly decreased from baseline. No significant increase was observed in serum bone-specific alkaline phosphatase (BAP) in both whole milk group and MBP group.Conclusion No significant effect of MBP on bone mineral density and bone metabolism was observed, but milk supplementation was effective in suppressing bone resorption.

Effects of Chinese Medicine of Nourishing Yin and Tonifying Yang on OPG and RANKL Expression in Osteoblast

Abstract:Objective: Effects of Gule granules of nourishing Yin and tonifying Yang on bioactive factor OPG and RANKL expression in osteoblast and investigate the mechanism for the treatment of osteoporosis. Methods: A large number of cells were identified into osteoblast by enzyme digestion and alizarin red staining. MTT assay and Western blot methods were used to determine the proliferation activity in vitro and the contents of RANKL and OPG in osteoblast. Results: The cells were mononuclear,polygon and spindle observed under the inverted microscope,and local density cell became cell aggregate. Calcified nodules were stained orange with alizarin red stain. The high and medium doses of Gule granules significantly promoted the proliferation of osteoblasts,but low doses of Gule granules had no effect on osteoblasts. The high and medium doses of Gule granules significantly increased the expression of OPG and inhibited the secretion of RANKL,but the low dose of Gule granules had no effect on OPG and RANKL. Conclusion: Gule granules of nourishing Yin and tonifying Yang can promote the proliferation of osteoblast,increase OPG expression and inhibit RANKL secretion.

Abstract:INTRODUCTION:The function of promoting bone regeneration of Moutan Cortex (MC), a traditional Chinese medicine, has been widely known but, the effective components of MC in promoting osteoblast-mediated bone regeneration were still unclear.OBJECTIVE:The method of osteoblast membrane bio-specific extraction conjugated with HPLC analysis was established to screen bone regeneration active components from MC.METHODS:The fingerprints, washing eluate and desorption eluate of MC extract were analyzed by the established HPLC-DAD method. The established MC3T3-E1 cells membrane chromatography method was used for the bio-specific extraction of MC. The isolated compounds were identified by MS spectrometry. The effects and possible mechanisms of the isolated compounds were evaluated by molecular docking, ALP activity, cell viability by MTT Assay and proteins expression by Western Blot Analysis.RESULTS:The active compound responsible for bone regeneration from MC was isolated using the established method of osteoblast membrane bio-specific extraction conjugated with HPLC analysis, and it was identified as 1,2,3,4,6-penta-O-β-galloyl-D-glucose (PGG) by MS spectrometry. It was further demonstrated through molecular docking that PGG could fit well into the functional ALP, BMP2, and Samd1 binding pocket. The proliferation of osteoblasts was promoted, the level of ALP was increased, and the protein expression of BMP2 and Smad1 was increased as shown by further pharmacological verification.CONCLUSION:It was concluded that PGG, the bone regeneration active compound from MC, could stimulate the proliferation of osteoblasts to promote osteoblast differentiation, and its mechanism might be related to the BMP/Smad1 pathway.

Abstract:BACKGROUND:It is confirmed that traditional Chinese medicine can improve osteoblast proliferation and differentiation.Cell molecular research on the pharmacological mechanism behind this has made great progress.OBJECTIVE:To summarize the research progress on the mechanisms of osteoblasts proliferation and differentiation induced by traditional Chinese medicine at home and abroad.METHODS:An online search of CNKI,PubMed and ScienceDirect databases was performed for articles on the mechanism of osteoblast proliferation and differentiation induced by traditional Chinese medicine,published between January 2000 and December 2010,using keywords of "traditional Chinese herbs,osteoblasts,proliferation,differentiation and mechanism".A total of 176 articles were collected after initial search,and 30 of them were retained.RESULTS AND CONCLUSION:Traditional Chinese medicine can improve osteoblast proliferation and differentiation by regulating the genes,signal pathways,cytokines,OPG/RANK/RANKL system of osteoblasts and estrogen-like biological activities.The mechanism is clear.It has a good future prospect for the application of traditional Chinese medicine in bone tissue engineering.

Abstract:Osteoporosis stands out as a prevalent metabolic disorder, bearing significant repercussions on human well-being and overall quality of life. It remains an urgent concern within the global public health framework due to its widespread occurrence. Osteoporosis arises from an abnormal metabolism in osteoblasts and osteoclasts, resulting in a disruption of the delicate equilibrium between bone formation and bone resorption. Within this context, deer antler peptides emerge as natural active compounds, wielding a pivotal role in governing the differentiation, proliferation, and mineralization of osteoblasts, as well as influencing the activity of osteoclasts. This article aims to consolidate our comprehension of the mechanisms underpinning the dynamic balance between bone formation and resorption, meticulously orchestrated by osteoblasts and osteoclasts in osteoporosis. Furthermore, it offers a comprehensive overview of how deer antler peptides, through their modulation of relevant signaling pathways, contribute to the enhancement of bone homeostasis. These insights deepen our understanding of the pathological processes through which deer antler peptides ameliorate bone homeostasis, while also presenting novel strategies for osteoporosis management.

Abstract:This study aimed to investigate the effects of osteoprotegerin on the activity of osteoclasts.Primary mice osteoclasts were cultured in vitro,and various concentrations of OPG were added in the different groups.TRAP staining,F-actin staining,bone resorption activity analysis were employed in the present study.The results showed that OPG could influence the osteoclasts directly,lessened the number,disrupted the F-actin rings and weakened the bone resorption activity of osteoclasts,in particular the high concentrations of OPG.It was concluded that OPG could decrease the number of osteoclasts and decline their bone resorption activity,in a concentration-dependent manner.

Abstract:Osteoporosis is a systemic metabolic bone disease characterized by a reduction in bone mass, bone quality, and microarchitectural deterioration. An imbalance in bone remodeling that is caused by more osteoclast-mediated bone resorption than osteoblast-mediated bone formation results in such pathologic bone disorder. Traditional Chinese medicines (TCM) have long been used to prevent and treat osteoporosis and have received extensive attentions and researches at home and abroad, because they have fewer adverse reactions and are more suitable for long-term use compared with chemically synthesized medicines. Here, we put the emphasis on osteoblasts, summarized the detailed research progress on the active compounds derived from TCM with potential anti-osteoporosis effects and their molecular mechanisms on promoting osteoblast-mediated bone formation. It could be concluded that TCM with kidney-tonifying, spleen-tonifying, and stasis-removing effects all have the potential effects on treating osteoporosis. The active ingredients derived from TCM that possess effects on promoting osteoblasts proliferation and differentiation include flavonoids, glycosides, coumarins, terpenoids (sesquiterpenoids, monoterpenoids, diterpenoids), phenolic acids, phenols and others (tetrameric stilbene, anthraquinones, diarylheptanoids). And it was confirmed that the bone formation effect induced by the above natural products was regulated by the expressions of bone specific matrix proteins (ALP, BSP, OCN, OPN, COL I), transcription factor (Runx2, Cbfa1, Osx), signal pathways (MAPK, BMP), local factors (ROS, NO), OPG/RANKL system of osteoblasts and estrogen-like biological activities. All the studies provided theoretical basis for clinical application, as well as new drug research and development on treating osteoporosis.

Abstract:Tremendous experiments and studies have shown that matrix metalloproteinase (MMP) particiates in the process of bone remodeling and construction. Matrix metalloproteinase is associated with bone resorption activity, mainly distributed in bone matrix. It has large family ranging from MMP-1 to MMP-26. From the previous research, MMP-2、-9、-13 have been identified to have an effect on pathogenesis of primary osteoporosis. Based on the basic knowledge about MMP family from research, we understand it deeply. Therefore, in present study, we reviewed the effects of MMP on pathogenesis of primay osteoporosis and osteoporosis fracture.

Abstract:With aging of the society, the incidence of postmenopausal osteoporosis（PMOP） has become a common clinical disease in postmenopausal women. Its pathogenesis is complex and has not yet been fully elucidated. A large body of study has shown that the gut microbiota（GM） is closely related to PMOP, which provides a new perspective to explore the reconstruction of bone homeostasis. Traditional Chinese medicine（TCM） has great potential to prevent and treat PMOP through GM, which is a new effective treatment approach. Therefore, the review aims to summarize studies from domestic and abroad to further illustrate the theoretical research, clinical research, and mechanism research of GM on regulating PMOP. Additionally, we also summarize studies that explore the effect of herbal compounds, related herbs（single components）, Chinese patent medicines, and other TCM treatments on improving bone health through GM. In conclusion, regulating GM may broaden perspective of PMOP research, which is significant for clinical prevention and treatment.
