EFFECT OF ACUTE ALCOHOL ADMINISTRATION ON MEMORY-IMPAIRMENT AND PHOSPHORYLATION LEVEL OF c-AMP RESPONSIVE ELEMENT BINDING PROTEIN OF HIPPOCAMPUS IN RATS

Abstract:[Objective] To observe the effect of acute alcohol administration on memory-impairment and phosphorylation level of cAMP-responsive element binding protein(CREB) of hippocampus in rats.[Methods] Alcohol was administrated to rats by intragastric lavage with doses of 1,2 and 3g /kg.2 hours later,Morris Water Maze performance was applied to evaluate memory function,and the phosphorylation level of cAMP-responsive element binding protein in rat hippocampus was determined by Western Blotting.Also,memory function and p-CREB level were determined at 0.5,1,2 and 4 hour after administration with alcohol 3g /kg.[Results] The Morris Water Maze performances demonstrated that when the alcohol doses increasing,la-tency and traveling distance prolonged,with significant differences between 2g /kg,3g /kg dose groups and the control(P﹤ 0.05);The latency and traveling distance extended 0.5,1 and 2 h after administration with alcohol 3g /kg with significant dif-ference in contrast to the control group(P ﹤ 0.05).The Western Blotting results indicated that the phosphorylation level of CREB elevated gradually after administration,peaking at 2h and returning to normal content at 4h.[Conclusion] The increase of phosphorylation level of CREB in rat hippocampus is in accordance with the instance of memory-impairment after acute lavage with alcohol,which implies that alcohol may induce memory-impairment by enhancing the phosphorylation level of CREB.
