Abstract:Objective To analyse the relationship between glucose transporter 1 (GluT1) gene polymorphism and prognosis of patients with diabetic nephropathy(DN). Methods The GluT1 gene polymorphism was determined by PCR-RFLP in 102 DN patients. Clinical features of the patients with different genotype were compared. Among them, 61 DN subjects were followed up for 1~7 years. Results (1) The frequencies of XbaI(+) and XbaI(-) allele were 79% and 21% respectively in normal subjects, and were 72% and 28% respectively in DN subjects: the frequence of XbaI(-) allele was significantly higher in DN subjects than in normal subjects(P<0.05); (2) The average renal survival time in DN patients with XbaI(+/+), XbaI(+/-) and XbaI(-/-)genotypes was 67 months, 67 months and 44 months respectively. The renal survival rate was significantly lower in patients with XbaI(-/-)genotype than in the patients with XbaI(+/-)and XbaI(+/+)genotypes(P<0.05). There was no significant difference in renal survival rate between the patients with XbaI(+/-)and XbaI(+/+)genotypes. Conclusions The renal survival rate is significantly lower in patients with XbaI(-/-)genotype than in patients with XbaI(+/-)and XbaI(+/+)genotypes.
AGTR1 A1166C Polymorphism is Associated with Risk of Diabetic Nephropathy

Abstract:OBJECTIVE:To investigate whether the angiotensin II type I receptor gene (AGTR1) A1166C polymorphism is associated with a high risk of diabetic nephropathy.METHODS:The allele frequency and the genotype distribution of the AGTR1 A1166C polymorphism were studied in normal controls (157 cases), simple diabetes (141 cases, duration of diabetes >10 years), and diabetic nephropathy (152 cases) by means of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).RESULTS:Patients with diabetic nephropathy had a higher frequency of C allele of the AGTR1 A1166C polymorphism than that of normal controls and simple diabetes (P<0.05); but there was no significant difference in frequency of C allele between the normal controls and patients with simple diabetes.CONCLUSION:The diabetic patients with AGTR1 C allele may be more susceptible to diabetic nephropathy than diabetic patients with A allele.
Glucose transporter 9 regulates hepatic steatosis through modulating intrahepatic uric acid contents in mice

Abstract:Abstract Background: Nonalcoholic fatty liver disease (NAFLD) is the most common chronic liver disease worldwide. The important role of urid acid (UA) in NAFLD has been widely investigated. Our previous studies unveiled the elevation of serum UA levels independently predicts an increased risk of incident NAFLD. However, the role of intrahepatic UA in NALFD has not been investigated yet. Glucose transporter 9 (GLUT9) is a key transporter that mediates the uptake of UA in hepatocytes. Methods: In this study, we first explored the clinical association between GLUT9 polymorphism and NAFLD. Blood samples of 247 male Chinese (127 were NAFLD patients) were collected and tested for the blood UA levels and genotype of the single nucleotide polymorphism (SNP) of GLUT9 (rs1014290). Next, Glut9 hepatic-specific knockout mice (Glut9Hep-ko) were generated to investigate the role of hepatic GLUT9 in NAFLD in male mice. Results: We found that the GA/AA genotypes (rs1014290) were associated with elevated serum UA levels in NAFLD patients. Meanwhile, we found that Glut9Hep-ko mice displayed lower intrahepatic UA levels, down-regulated lipogenesis genes expressions, and attenuated NAFLD symptoms after 12 weeks of high fat diet feeding, compared with Glut9Fl/Fl littermates. However, Glut9Hep-ko mice and wild type littermates showed no significant difference on hepatic fatty acid oxidation or inflammation. Conclusions: our results suggested that GLUT9 polymorphism was significantly associated with NAFLD, and hepatic-specific knockout of Glut9 significantly decreased intrahepatic contents and ameliorated diet-induced NAFLD in mice.
Uncoupling Protein 2 and Peroxisome Proliferator-Activated Receptor γ Gene Polymorphisms in Association with Diabetes Susceptibility in Chinese Han Population with Variant Glucose Tolerance

Abstract:Objective . To investigate the association of polymorphisms in uncoupling protein 2 (UCP2) and peroxisome proliferator-activated receptor (PPAR γ ) with glucolipid metabolism in Chinese Han population. Methods . Five hundred eighty-nine subjects were divided into normal glucose tolerance (NGT) group ( n = 198 ) and abnormal glucose tolerance group ( n = 358 ). HbA1c, blood lipid profile, plasma glucose, and insulin were determined. Insulin sensitivity (HOMA-IR and Matsuda index (ISI M )) and insulin secretion indexes (HOMA- β , early and total phase disposition index) were evaluated. Eight potential functional SNPs in UCP2 and 7 in PPAR γ were selected. SNPs were genotyped on Sequenom MassARRAY platform. Results . The GG genotype of rs2920502 in PPAR γ was associated with decreased risk of impaired glucose tolerance (G allele: OR: 0.818, 95%CI: 0.526–0.969, P = 0.042 ; GG: OR: 0.715, 95%CI: 0.527–0.97, P = 0.031 ). The TT genotype of rs3856806 in PPAR γ was associated with increased risk of impaired glucose tolerance (T allele: OR: 1.46, 95%CI: 1.055–2.017, P = 0.022 ; TT: OR: 1.58, 95%CI: 1.104–2.761, P = 0.032 ). The GG genotype of rs2920502 in PPAR γ had better blood glucose and increased insulin secretion and had lower HOMA-IR than GC/CC genotypes. Conclusion . It probably could prevent insulin resistance in early stage by classifying the genotype of rs649446 and rs7109266 in UCP2. The GG genotype of rs2920502 in PPAR γ had a decreased risk for diabetes. The TT genotype of rs3856806 in PPAR γ had an increased risk for diabetes.

Abstract:Context: Several genome-wide association studies identified a strong association of SLC30A8 with type 2 diabetes in individuals of European ancestry. The effect of the association of rs13266634 with type 2 diabetes or related glycemic traits has not been fully extended to non-European populations, and a comprehensive examination of common variants in the gene has not yet been carried out in Han Chinese. Objective: The objective of the study was to investigate the association of SLC30A8 with type 2 diabetes in Chinese. Design: A comprehensive gene-based association study was performed using 14 tagging single-nucleotide polymorphism (SNPs) of SLC30A8 in Han Chinese subjects with normal glucose tolerance (NGT; n = 721), impaired glucose regulation (IGR; n = 375), and type 2 diabetes (n = 521). Results: A significant association for SNP rs13266634 was observed between patients with type 2 diabetes and NGT controls (P = 0.016). The association was also observed between combined type 2 diabetes/IGR and NGT subjects (P = 0.002). The adjusted odds ratios for homozygote CC vs. TT at this locus were 1.71 for type 2 diabetes (95% confidence interval 1.19-2.45, P = 0.002) and 1.77 for type 2 diabetes and IGR (95% confidence interval 1.29-2.42, P = 0.0001). We further studied the genotype-phenotype correlation in 70 Han Chinese using iv glucose tolerance test and found an association between SNP rs13266634 and acute insulin response to glucose and disposition index (adjusted P = 0.012 and 0.004, respectively). Conclusions: Our results provide evidence that SLC30A8 is a susceptible locus for type 2 diabetes in Chinese population, and its variant can influence insulin secretion.

Abstract:Diabetes mellitus (DM) is a significant public health problem. Diabetic kidney disease (DKD) is the most common complication of DM, and its incidence has been increasing with the increasing prevalence of DM. Given the association between DKD and mortality in patients with DM, DKD is a significant burden on public health resources. Despite its significance in DM progression, the pathogenesis of DKD remains unclear. Aberrant glucose uptake by cells is an important pathophysiological mechanism underlying DKD renal injury. Glucose is transported across the bilayer cell membrane by a glucose transporter (GLUT) located on the cell membrane. Multiple GLUT proteins have been identified in the kidney, and GLUT1 is one of the most abundantly expressed isoforms. GLUT1 is a crucial regulator of intracellular glucose metabolism and plays a key pathological role in the phenotypic changes in DKD mesangial cells. In an attempt to understand the pathogenesis of DKD better, we here present a review of studies on the role of GLUT1 in the development and progression of DKD.
Sanger Sequencing Reveals Novel Variants in GLO-1, ACE, and CBR1 Genes in Patients of Early and Severe Diabetic Nephropathy

Abstract:Background and Objectives: Diabetes is a global health issue, with approximately 50% of patients developing diabetic nephropathy (DN) and 25% experiencing early and severe forms of the disease. The genetic factors contributing to rapid disease progression in a subset of these patients are unclear. This study investigates genetic variations in the GLO-1, CBR-1, and ACE genes associated with early and severe DN. Materials and Methods: Sanger DNA sequencing of the exons of CBR1, GLO1, and ACE genes was conducted in 113 patients with early and severe DN (defined as occurring within 10 years of the diagnosis of diabetes and with eGFR < 45 mL/min/1.73 m2) and 100 controls. The impact of identified genetic variations was analyzed using computational protein models created in silico with SWISS-Model and SWISS-Dock for ligand binding interactions. Results: In GLO1, two heterozygous missense mutations, c.102G>T and c.147C>G, and one heterozygous nonsense mutation, c.148G>T, were identified in patients. The SNP rs1049346 (G>A) at location 6:38703061 (GRCh38) was clinically significant. The c.147C>G mutation (C19S) was associated with ligand binding disruption in the GLO1 protein, while the nonsense mutation resulted in a truncated, non-functional protein. In CBR1, two heterozygous variations, one missense c.358G>A, and one silent mutation c.311G>C were observed, with the former (D120N) affecting the active site. No significant changes were noted in ACE gene variants concerning protein structure or function. Conclusions: The study identifies four novel and five recurrent mutations/polymorphisms in GLO1, ACE, and CBR1 genes associated with severe DN in Pakistani patients. Notably, a nonsense mutation in GLO1 led to a truncated, non-functional protein, while missense mutations in GLO1 and CBR1 potentially disrupt enzyme function, possibly accelerating DN progression.
SNPs in PRKCA-HIF1A-GLUT1 are associated with diabetic kidney disease in a Chinese Han population with type 2 diabetes.

Abstract:ObjectiveTo explore the relationship between SNPs in PRKCA-HIF1A-GLUT1 and diabetic kidney disease in Chinese Han people. Materials and methodsA total of 2552 participants from Shanghai Diabetes Institute Inpatient Database of Shanghai Jiao Tong University Affiliated Sixth People's Hospital were involved in the stage 1 cross-sectional population. A total of 6015 subjects from the Hong Kong Diabetes Register were included for validation. Genotyping of participants was conducted by the MassARRAY Compact Analyzer (Agena Bioscience). The data were analysed by plink, SAS, Haploview. ResultsWe identified variants associated with diabetic kidney disease in stage 1. Rs1681851 (P = .0105, OR = 1.331) in GLUT1 as well as rs2301108 (P = .0085, OR = 1.289) and rs79865957 (P = .0204, OR = 1.263) in HIF1A were significantly associated with diabetic kidney disease. Regarding DKD-related traits, rs1681851 was associated with plasma creatinine levels (P = .0169, beta = 4.822) and eGFR (P = .0457, beta = -6.956). Moreover, the results showed the interactions between PRKCA-GLUT1 in the occurrence of DKD. We further sought validation of the 17 SNPs in a prospective cohort and found that rs900836 and rs844501 were associated with the percentage change in eGFR slope. We performed a meta-analysis of case-control analysis data from the Hong Kong samples together with the stage 1 data from Shanghai. Rs9894851 showed significant correlation with the serum creatinine level as well as eGFR and no SNP showed association with DKD after meta-analysis. ConclusionsOur results suggest potential association between SNPs in PRKCA-HIF1A-GLUT1 and diabetic kidney disease in Chinese Han people.
Low-Frequency Genetic Variant in the Hepatic Glucokinase Gene is Associated with Type 2 Diabetes and Insulin Resistance in Chinese Population

Abstract:Glucokinase (GCK) regulates insulin secretion and hepatic glucose metabolism, its inactivating variants could cause diabetes. We aimed to evaluate the association of a low-frequency variant of GCK (rs13306393) with type 2 diabetes (T2DM), prediabetes or both (IGR) in Chinese population. An association study was firstly conducted in a random cluster sampling population (Samples 1, 537 T2DM, 768 prediabetes and 1912 controls), then another independent Samples 2 (3896 T2DM, 2301 prediabetes and 868 controls) was used to confirm the findings in Samples 1. A-allele of rs13306393 was associated with T2DM [OR 3.08 (95%CI 1.77-5.36), p=0.00007] in Samples 1. Rs13306393 was also associated with prediabetes [OR 1.67 (95%CI 1.05-2.65), p=0.03] in Samples 2. In pooled analysis of two samples, A-allele increased the risk of T2DM [OR1.57 (95%CI 1.15-2.15), p=0.005], prediabetes [OR 1.83 (95%CI 1.33-2.54), p=0.0003) or IGR [OR 1.68 (95% CI 1.26-2.25), p=0.0004], insulin resistance estimated by homeostasis model assessment (beta=0.043, p=0.001), HbA1c (beta=0.029, P=0.029) and urinary albumin excretion (beta=0.033, p=0.025) irrespective of age, gender and BMI. Thus, the Chinese specific low-frequency variant increased the risk of T2DM through reducing insulin sensitivity rather than islet beta cell function, which should be considered in clinical use of GCK activators in the future.
Clinical and Genetic Features of Patients With Type 2 Diabetes and Renal Glycosuria.

Abstract:Context: A sodium glucose cotransporter 2 (SGLT2) inhibitor, which increases urinary glucose excretion, was reported to decrease blood glucose levels and deaths among patients with type 2 diabetes mellitus (T2DM) and established cardiovascular disease. SLC5A2 and HNF1A mutations are associated with renal glycosuria, but their contributions to renal glycosuria in patients with T2DM are not well understood. Objective: To assess the clinical features of patients with T2DM and renal glycosuria and those with T2DM and low urinary glucose excretion (LUGE) and identify variants in the exons of SLC5A2 and HNF1A in patients with renal glycosuria and T2DM. Design: A total of 2044 Chinese patients with T2DM, including 64 patients with renal glycosuria and 58 patients with LUGE, were tested for their plasma and urine glucose concentrations after fasting. SLC5A2 and HNF1A exons were sequenced. Results: Compared with patients with LUGE, those with renal glycosuria were younger (P = 0.008), had lower body mass index (BMI) (P = 0.002) and Homeostatic Model Assessment of Insulin Resistance (HOMA-IR) values (P < 0.0001), and were less likely to have hypertension (P = 0.006). HOMA-IR and BMI were negatively associated with renal glycosuria after adjusting for age, sex, hypertension, and insulin therapy. One novel mutation (V359G) of SLC5A2 in 32 patients with renal glycosuria and one known mutation (R131W) of HNF1A in 28 nonobese patients with renal glycosuria were identified. Conclusions: These findings suggest that there are subtypes of T2DM characterized by different urinary glucose excretion and cardiovascular risk factors. SLC5A2 and HNF1A mutations partially explain renal glycosuria in patients with T2DM.
Polymorphisms in the Glucagon-Like Peptide 1 Receptor (GLP-1R) Gene Are Associated with the Risk of Coronary Artery Disease in Chinese Han Patients with Type 2 Diabetes Mellitus: A Case-Control Study

Abstract:Background. Glucagon-like peptide 1 (GLP-1) bestows protective effects upon the cardiovascular system through direct cardiovascular interactions or by improvements to metabolic function. Both these effects are thought to be at least partly mediated by the GLP-1 receptor (GLP-1R). This case-controlled study investigated whether polymorphisms in the GLP-1R gene affect the risk of cardiovascular disease in type 2 diabetic patients in the Chinese Han population. Methods. Eleven haplotype-tagging single nucleotide polymorphisms (SNPs), distributed across 22 kb of the 39 kb GLP-1R gene, were selected and genotyped in diabetic patients from a Chinese Han population. Patients were classified based on the severity of coronary artery stenosis. Coronary artery stenosis was ≥50% in 394 patients (coronary artery disease- (CAD-) positive group), and coronary artery stenosis was <50% in 217 patients (control group). Allele and genotype frequencies were compared between the two groups at all 11 SNPs. Results. When considered in recessive inheritance mode, patients with the GG genotype at rs4714210 had a lower CAD risk than patients with other genotypes (OR = 0.442, 95% CI = 0.258–0.757, p=0.002), even when other known CAD risk factors were taken into account (ORa = 0.440, 95% CIa = 0.225–0.863, pa=0.017). In additive inheritance mode, GG genotype carriers at rs4714210 exhibited a lower risk of CAD than AA carriers (ORa = 0.475, CIa = 0.232–0.970, pa=0.041). Conclusion. In type 2 diabetic patients from a Han Chinese population, some variations in the GLP-1R gene were associated with a lower risk of developing CAD.
THE RELATIONSHIP BETWEEN GLUCOKINASE GENE POLYMORPHISMAND NON-INSULIN-DEPENDENT DIABETES MELLITUS IN CHINESE

Abstract:Objective: To evaluate the relationship between glucokinase(GCK)gene and non-insulin-dependent diabetes mellitus(NIDDM) in Chinese. Methods:Utilizing the microsatellite polymorphism of GCK gene3' as a genetic marker,we detected the differences of frequences of alleles at this locus between patients with NIDDM and normal control subjects by polymerase chain reaction(PCR). Results:In the population studied, three alleles were found, which were named alleles A, B, and C, and whose sizes were 195 bp, 197 bp, and 199 bp, respectively; six genotypes(AA,BB,CC,AB,AC, and BC) were found. The frequences of alleles A, B, and C were 53.4%, 25.2%, and 21.4% in control group, and 60.0%, 17.1%, and 22.9% in NIDDM group, respectively. The frequency of genotypes AA, BB, CC, AB, AC, and BC were 28.3%, 6.2 %, 2.1%, 24.8%, 25.5% , and 13.1% in normal control group,and 36.2 %, 1.9%, 3.8%, 20.0%, 27.6% and 10.5% in NIDDM group, respectively. In addition, the differences of frequency of other alleles and each genotype between NIDDM and normal control group were not detected. The NIDDM group had a lower B allels distribution frequency compared with normal control group(17.1% vs 25.2%,P=0.03). The NIDDM patients without B allele (B-) had similar bodymass indexes(BMI) and the numbers of patients with family history of NIDDM to the NIDDM patients with B allele (B+). Patients of Bgroup were diagnosed as NIDDM at earlier age (51 vs 56 years old,P0.05) compared with the patients of B+ group. Conclusion:There is a positive association of GCK gene or nearby gene with NIDDM in Chinese,and the allele B may be a protective factor for NIDDM in Chinese.
Glucose and Lipid Metabolism in Relation to Novel Polymorphisms in the 5′-Amp-activated Protein Kinase Γ2 Gene in Chinese

Abstract:The 5'-AMP-activated protein kinase (AMPK) behaves as a fuel sensor in glucose and lipid metabolism. We sequenced exon 1 and flanking regions of the gene encoding for the gamma2 subunit of AMPK (AMPKgamma2) and identified two novel common polymorphisms at position -26 and IVS1+43. We then studied these two polymorphisms in relation to plasma glucose, insulin resistance, beta-cell function, and serum lipids in 290 Han Chinese undergoing an oral glucose tolerance test and a frequently sampled intravenous glucose tolerance test. The -26C/T and IVS1+43C/T polymorphisms were in tight linkage disequilibrium (P=0.0002). In adjusted categorical analyses, the -26TT genotype tended to be associated with a higher risk of type 2 diabetes (odds ratio 4.52, P=0.07). The adjusted continuous analyses were confirmatory. -26TT subjects, compared with -26C allele carriers, had higher concentrations of plasma glucose, both fasting (7.3 vs. 6.1 mmol/L, P=0.02) and after oral glucose loading (area under the curve for glucose, 1984 vs. 1596 minmmol/L, P=0.002), and had lower acute insulin response to glucose (143 vs. 404, P=0.0005) and disposition index (151 vs. 459, P=0.008). In further adjusted analyses, we observed that IVS1+43TT subjects, compared with IVS1+43C allele carriers, had significantly higher serum concentrations of triglycerides (4.20 vs. 2.00 mmol/L, P<0.0001) and total cholesterol (5.88 vs. 4.99 mmol/L, P=0.01). In conclusion, in Chinese, the AMPKgamma2 polymorphisms might be associated with glucose and lipid metabolism.
Arg913Gln of SLC12A3 Gene Promotes Development and Progression of End-Stage Renal Disease in Chinese Type 2 Diabetes Mellitus

Abstract:Whether the Arg913Gln variation (rs11643718, G/A) of SLC12A3 contributes to diabetic nephropathy (DN) remains controversial. We undertook a case–control study to evaluate the association of the SLC12A3-Arg913Gln variation with the risk of end-stage renal disease (ESRD) in Chinese type 2 diabetes mellitus (T2DM) patients undergoing hemodialysis, and analyzed the genotype–phenotype interaction. Unrelated Chinese T2DM patients (n = 372) with diabetic retinopathy were classified into the non-DN (control) group (n = 151; duration of T2DM >15 years, no signs of renal involvement) and the DN–ESRD group (n = 221; ESRD due to T2DM, receiving hemodialysis). Polymerase chain reaction-direct sequencing was used to genotype the SLC12A3-Arg913Gln variation for all participants. The frequency of the GA+AA genotype in the DN–ESRD group was significantly higher than that of the non-DN group (23.1 vs. 9.9%; adjusted OR 2.2 (95% CI 1.3–4.5), P = 0.019). In the non-DN group, GA+AA carriers had a significantly higher urinary albumin excretion rate (UAER) and diastolic blood pressure compared with GG carriers (both P < 0.05). The SLC12A3-Arg913Gln variation may be associated with increased blood pressure and UAER and, therefore, could be used to predict the development and progression of DN–ESRD in Chinese T2DM patients undergoing hemodialysis.
Association of Single Nucleotide Polymorphism in ATP-Binding Cassette Transporter 1 Gene with Type 2 Diabetes Mellitus and Obesity

Abstract:Objective To study the association of ATP-binding cassette transporter 1(ABCA1) gene polymorphism with type 2 diabetes and obesity in Chinese Han population living in Shanghai. Methods Four hundred and forty patients with type 2 diabetes mellitus and 315 normal control subjects were enrolled in the study.Diabetic patients were further divided into normal weight patients(n=188,BMI25 kg/m~(2)) and obese patients(n=252,BMI25 kg/m~(2)).A single nucleotide polymorphism(SNP) rs2020927 was chosen and allele-specific real-time PCR method was employed to identify the loci genotype in all the subjects. Results The distribution of genotype frequency of the loci rs2020927 of ABAC1 was significantly different between the groups of type 2 diabetes and normal controls(P=0.001).The frequency of AA,AG,GG genotype was 43.8%,35.9% and 20.3%,respectively in the control group,and 33%,53.2% and 13.8%,respectively in the normal-weight diabetic group,and 40.9%,47.2% and 11.9%, respectively in the obese diabetic group.The frequency of genotype GG was significantly diffe-(rent) between the control group and obese diabetic group(P=0.009),while no significant difference was found between the control group and normal-weight diabetic group(P0.05). Conclusion The SNP rs2020927 of ABCA1 gene may be associated with type 2 diabetes mellitus and obesity in Chinese Han population(living in Shanghai.)
Sodium-glucose cotransporter 1/2 inhibition and risk of neurodegenerative disorders: A Mendelian randomization study

Abstract:Introduction: This study aims to evaluate the effects of sodium-glucose cotransporter 1 inhibitors (SGLT1i) and sodium-glucose cotransporter 2 inhibitors (SGLT2i) on neurodegenerative disorders and to investigate the role of hemoglobin A1c (HbA1c) levels. Methods: Utilizing drug target Mendelian randomization, we employed single nucleotide polymorphisms (SNPs) proximal to the SLC5A1 and SLC5A2 genes to analyze the influence of SGLT1i and SGLT2i on Alzheimer's disease (AD), Parkinson's disease (PD), multiple sclerosis (MS), frontotemporal dementia (FTD), Lewy body dementia (LBD), and amyotrophic lateral sclerosis (ALS), with type 2 diabetes (T2D) as a positive control. An additional analysis examined the impact of HbA1c levels on the same disorders. Results: SGLT1i exhibited a significant association with decreased risk for ALS and MS. Conversely, SGLT2i were linked to an increased risk of AD, PD, and MS. Elevated HbA1c levels, independent of SGLT1 and SGLT2 effects, were associated with an increased risk of PD. Sensitivity analyses supported the robustness of these findings. Conclusion: Our study suggests that SGLT1i may confer protection against ALS and MS, whereas SGLT2i could elevate the risk of AD, PD, and MS. Additionally, elevated HbA1c levels emerged as a risk factor for PD. These findings underscore the importance of personalized approaches in the utilization of SGLT inhibitors, considering their varying impacts on the risks of neurodegenerative diseases.

Abstract:Abstract Background: Serum amyloid A (SAA) was reported to be associated with insulin resistance and type-2 diabetes. The present study aimed to investigate the association of SAA genetic polymorphisms with plasma glucose levels in non-diabetic subjects. Methods: All participants (n=1220) were selected from subjects participating in the Cardiovascular Risk Survey (CRS) study. Five single-nucleotide polymorphisms (SNPs) (rs2229338, rs12218, rs4638289, rs7131332 and rs11603089) of SAA gene were genotyped using polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) analysis. Results: In the present study, we found rs2229338, rs4638289 and rs12218 were significantly associated with plasma glucose levels in a dominate model, recessive model or additive model before and after multivariate adjustment (all p<0.05). These associations were not found in rs7131332 and rs11603089 before and after adjustment of key co-variants. Conclusions: The genetic polymorphisms of SAA1 were associated with plasma glucose levels in non-diabetic subjects.

Abstract:Aims: We aimed to identify whether the risk G-allele was associated with fasting glucose level and other pre-diabetic and obesity-related phenotypes in Chinese children and adolescents. Methods: The rs10830963 polymorphism in MTNR1B was genotyped in 2,030 Chinese children and adolescents of two independent studies. Association with fasting glucose levels and risk of impaired fasting glucose (IFG) were initially tested. Subsequently we analyzed the association with fasting insulin, homeostasis model assessment for insulin resistance (HOMA-IR) and for beta cell function (HOMA-B), the quantitative insulin sensitivity check index (QUICK) and obesity-related phenotypes (BMI standard deviation score, waist circumference etc.). Results: The G-allele of rs10830963 was associated with increased fasting glucose level in Chinese children and adolescents (increase of 0.072 mmol/l per G-allele, 95% CI 0.034-0.111, p = 2.46 10(-4)). The G-allele was also associated with an increased risk of IFG (OR = 1.21, 95% CI 1.00-1.46, nominal p = 0.048). We found the glucose-raising G-allele was nominally associated with reduced HOMA-B. No association to other pre-diabetic or obesity-related phenotypes was detected. Conclusions: The rs10830963 polymorphism in MTNR1B was associated with increased fasting glucose and risk of IFG in Chinese children and adolescents. The effect may result from reduced pancreatic beta cell function, but the mechanism awaits further studies.

Abstract:AimTo elucidate the effects of rs2289669, an intron variant of the SLC47A1 gene, on glucose response to metformin in Chinese people with newly diagnosed Type 2 diabetes. MethodsRs2289669 was genotyped, using Sequenom, in 291 participants receiving 48 weeks of metformin monotherapy. The changes in HbA(1c) were compared among rs2289669 genotypes, and associations with rs2289669 were evaluated using linear regression analysis. ResultsWe found that, compared with participants with a homozygous G allele, those carrying the minor A allele had significantly greater HbA(1c) reduction and greater increases in basal glucagon-like peptide-1 concentration. Regression analysis showed that there was a significant association between rs2289669 and the glucose response to metformin after adjusting for confounding factors, except for changes in basal glucagon-like peptide-1, for which an association was not observed. ConclusionsOur findings suggest that rs2289669 might help predict the glycaemic response to metformin in Chinese people newly diagnosed with Type 2 diabetes, and that differential increases in basal glucagon-like peptide-1 concentration among rs2289669 genotypes might be associated with inter-individual response to metformin.

Abstract:Background Galactose-deficient IgA1 plays a key role in the pathogenesis of IgA nephropathy, the most common primary GN worldwide. Although serum levels of galactose-deficient IgA1 have a strong genetic component, the genetic link between this molecule and IgA nephropathy has not yet been clearly established. Methods To identify novel loci associated with galactose-deficient IgA1, we performed a quantitative genome-wide association study for serum galactose-deficient IgA1 levels, on the basis of two different genome-wide association study panels conducted in 1127 patients with IgA nephropathy. To test genetic associations with susceptibility to IgA nephropathy, we also enrolled 2352 patients with biopsy-diagnosed IgA nephropathy and 2632 healthy controls. Peripheral blood samples from 59 patients and 27 healthy controls were also collected for gene expression analysis. Results We discovered two loci, in C1GALT1 and GALNT12, that achieved genome-wide significance, explaining about 3.7% and 3.4% of variance in serum galactose-deficient IgA1 levels, respectively. We confirmed the previously reported association of C1GALT1 with serum galactose-deficient IgA1 levels, but with a different lead single-nucleotide polymorphism (rs10238682; beta=0.26, P=1.20x10(-9)); the locus we identified at GALNT12 (rs7856182; beta=0.73, P=2.38x10(-9)) was novel. Of more interest, we found that GALNT12 exhibits genetic interactions with C1GALT1 in both galactose-deficient IgA1 levels (P=1.40x10(-2)) and disease risk (P=6.55x10(-3)). GALNT12 mRNA expression in patients with IgA nephropathy was significantly lower compared with healthy controls. Conclusions Our data identify GALNT12 as a novel gene associated with galactose-deficient IgA1 and suggest novel genetic interactions. These findings support a key role of genetically conferred dysregulation of galactose-deficient IgA1 in the development of IgA nephropathy.
