Abstract:2-(3-Thienyl)malonic acid,an intermediate for antibiotic penicillin,was synthesized from 3-methylthiophene by bromonation and cyano substitution to give 3-thienylacetonitrile,which was subjected to alcoholysis,condensation with diethyl carbonate and then hydrolysis with an overall yield of 36%.

Abstract:The process of preparing 4-hydroxy-3-methoxyphenylacetonitrile,as one of the important intermediates for synthesis was studied.Beginning from the raw material—vanillin,the title compound was prepared,via protection of hydroxyl,reduction,chloridization and nitrilation.The influences of various reaction conditions on the yield were investigated,and the optimal reaction conditions(reagent,temperature,time and yield) were as follows:acetic anhydride as acetylation reagent,reacting at room temperature for 3 h and yield was 99%;Raney Ni as catalyst,reaction time was 8 h at room temperature,and yield was 96.8%;dropping thionyldichloride at 0℃,then reacting at room temperature for 2 h,and the yield was 78%;refluxing with sodium cyanide for 9 h,and the yield was 78.5%.The products were characterized with MS,1H NMR and elemental analysis.The results showed that the process proposed overcame the defects of traditional synthesis route.

Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:As an intermediate for COX-2 inhibiters,4-(methylthio)benzeneacetic acid(Ⅳ) was prepared via Friedel-Crafts and Willgerodt-Kindler reaction from thioanisole(Ⅰ).When the molar ratio of Ⅰ to acetyl chloride and AlCl_3 was 1∶1.1∶1.3 and CS_2 was used as solvent,after reacting at(0°C) for 6 h the yield of 4-(methylthio) acetophenone(Ⅱ) was 75.9%.Ⅱ was refluxed 5 h with morpholine and sulfur to give thioacetmorpholide(Ⅲ),then Ⅲ was refluxed in 10% NaOH for 10 h to give Ⅳ with an overall yield of 63.5% and purity of 99.0%(by HPLC).The structure was confirmed by IR and()~1H NMR.

Abstract:2-Bromo-5-hydroxy-4-methoxybenzaldehyde(I)and 5-hydroxy-4-methoxy-2-nitrobenzaldehyde(II)were synthesized.The intermediates 2-bromo-4,5-dimethoxybenzaldehyde and 4,5-dimethoxy-2-nitrobenzaldehyde were synthesized firstly from veratraldehyde through bromination and nitration respectively.In the presence of molar quantity methionine,the 5th-position methoxys of the intermediates were in turn hydrolysed to hydoxy by concentrated sulfuric acid to give I and II.The chemical structures of the titled compounds were confirmed by 1H-NMR,IR and MS spectrum.The yields of compounds I and II are 85.0% and 65.0% respectively.The orientations of the bromination and nitration steps were suggested and discussed.The mechanism of methionine in the hydrolysis step may involve in the formation of the multiple hydrogen bonded association complex,and the calculated single point energy(E=-1532.5 hartree)of the multiple hydrogen bonded association complex seems to show it is possibly existed at the RHF/6-31G level.

Abstract:Synthesis process of 2,6-dichloro-3-methylaniline(Ⅶ) was investigated. 3-methylani-line was first N-acetylated with acetic anhydride, then treated with chlorosulphonic acid and ammonia yielding the 2-methyl-4-( N-acetyl )-benzenesulfonic amide, which was in turn deacetylated, chlorinated and hydrolyzed to the title compound. Hydrochloric acid was used instead of sodium hydroxide in deacetylation and sodium chlorate instead of hydrogen peroxide as oxidant in chlorination. Separation for 2-methyl-4-amino sulfanilamide intermediate was omitted. Then, high total molar yield was obtained, and based on 3-methylaniline, total molar yield of the product(Ⅶ) was 60. 8%. Steam distillation operation was employed to purify product (Ⅶ), the purity was more than 99. 5 percent.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:The target compounds——3-aminopyrrolidine dihydrochloride and its acetate are the key intermediates of tosufloxacin and other quinolone antibiotics. One of the synthetic routes of the target compounds is from benzylamine and ethylacrylate, which lead to 3-benzylaminopropionate by additional reaction, then affords 3-[N-(methoxycarbonylmethene)benzylamino]propionate through N-alkylation with methyl chloroacetate, further by Dieckmann cyclization, decarboxylation to obtain 1-benzyl-3-pyrrolidine, then oximate and hydrogenate it to gain the intermediate 3-amino-1-benzy1-pyrrolidone, which can be changed to target compounds easily. This route is much of industrial value because of the cheap and easily gained materials. But the original method presents many weakness such as low overall yield and troublesome operate process. We carried out our research work to improve the above method and got an obvious increase of N-alkylation yield from 62.4% to more than 95%, and also some improvement in some reaction process. Thus we succeed in getting an industrial route characterized by cheap and easily gained materials, convenient operate process and low cost.

Abstract:5-[2-(ethylthio)propyl]-1,3-cyclohexanedione, the title compound of this study, is an important intermediate for preparation of 3-hydroxy-cyclohex-2-enones herbicides, such as clethodim, sethoxydim, cloproxydim, etc.. The optimization of the synthesis route and the preparation method of the title compound were studied and the results were described concisely as follows. Firstly, 3-ethylthiobutanal was prepared with high yield by reacting crotonaldehyde with ethanethiol in triethylamine. Secondly, methyl acetoacetate was converted to 6-ethylthiohept-3-en-2-one via three steps, hydrolysis, condensation with 3-ethylthiobutanal and dehydration, then the 6-ethylthiohept-3-en-2-one was cyclocondensed with di-Me malonate by Michael addition and Clasisen condensation, and further via saponification and decarboxylation , the title compound was obtained. The products of each steps and the final title compound obtained were compared with each its own standerd samples and were confirmed by LC/MS.

Abstract:Reaction of thiophene with N,N-dimethylformamide in presence of phosphorus oxychloride gave 2-formylthiophene(Ⅰ)in 62% yield.Reaction of chloroacetic acid with isopropanol gave isopropyl chloroacetate(Ⅱ) in 55% yield.2-Thienylacetaldehyde(Ⅲ) was obtained by Darzens reaction with ⅠandⅡ,and Ⅲ reacted with hydroxylamine hydrochloride to give 2-thienylacetoxime (Ⅳ) in 72% yield.Reduction of Ⅳgave 2-(2-thienyl)ethylamine in 61% yield.Structure of the product was identified by methods of MS and TLC.

Abstract:A new strategy for enantioselective assembly of the trisubstituted tetrahydrofuran ring has been established for synthesis of the C1-C12 acid fragment of amphidinolide T series marine macrolides. The key steps involve the SmI2-mediated highly enantioselective reductive coupling of an aldehyde with the (1S,2R)-N-methylephedrine-derived crotonate to form the cis-3,4- disubstituted gamma-butyrolactone and the subsequent BF3-mediated 1,3-anti-selective allylation of the five-membered-ring oxocarbenium ion with allyltrimethylsilane. The desired C1-C12 acid fragment was obtained in >25% overall yield via a 9-step sequence. (C) 2009 Elsevier Ltd. All rights reserved.

Abstract:2-(5-Amino-1,2,4-thiadiazol-3-yl)-2-(Z)-methoxyiminoacetic acid 2-benzothiazolyl thioester,an important intermediate of the fourth generation cephalosporins,was efficiently synthesized by reacting 2-(5amino-1,2,4-thiadiazol-3-yl)-2-(Z)-methoxyiminoacetic acid(Ⅰ) with 2,2′-dibenzothiazole disulfide(Ⅱ) in the presence of triphenylphosphine.An improved procedure suitable for industrial production was established.Using 1,2dichloroethane as solvent,triphenylphosphine as reducer and triethylamine as catalyst,n(Ⅰ)∶n(Ⅱ)∶n(triphenylphosphine)=1.0∶1.0∶1.0,the product was obtained at room temperature with a yield of 98.1%.The purity of the product without further purification is 98.7% by HPLC method.This procedure was a suitable alternative to the traditional processes due to easy handling,high yield and low cost.

Abstract:4,4-Diethylthio/benzylthio-but-3-en-2-ones 2 were prepared in excellent yields,by the acid-catalyzed deacetylation of 3-(diethylthio/benzylthio) methylene-pentane-2,4-diones (1). With ketene diethylthio/benzylthioacetals (2) as the odorless thiol equivalents, in the presence of acetyl chloride, a series of diethylthio-/dibenzylthioacetals were prepared via the reaction of compound 2 with aldehydes/ketones in methanol under mild conditions.

Abstract:(E)-2-(6-((E)-2-carboxyvinyl)-2,3-dihydroxyphenyl)-3-(3,4-dihydroxyphenyl) acrylic acid, designated SMND-309, was synthesized starting from 2-hydroxy-3-methoxybenzaldehyde in 12 steps and with an overall yield of 44%. The synthetic key features were the conversion of aryl aldehyde into aryl acetic acid for one-carbon extended, the Perkin condensation, and the Heck coupling for construction of α,β-unsaturated ester.

Abstract:Telithromycin belongs to novel class of macrolide antibiotics. A general synthesis route was described which started from clarithromycin via protection of the hydroxyl in 2' position, hydrolysis with acid in 3 position, oxidation with the improved Pfitzner-Maffat method in 3 position to obtain ketolides, elimination of 11 and 12 positions with lithium hydride, cyclization with 4-[4-(3-pyridyl)-1H-imidazole]-1-butyl-amine, esterification exchange with methanol, the total yield is 18.8%. Comparing with the current technology, this described technology has the merits of the easily obtained raw material, the easy purification of intermediates and product, and it is more convenient to be used for industrial production.

Abstract:A new method for the solid phase synthesis of benzothiazoles and 2-arylamino-3-carboxyl-4-hydroxy-5-arylthiophenes is described. Resin bound cyclic malonic acid ester was reacted with aryl isothiocyanates and then with bromine or α-bromoketones, followed by treatment with perchloric acid or sodium methoxide to afford substituted 2-methylbenzothiazoles or 2-arylamino-3-carboxyl-4-hydroxy-5-arylthiophenes, respectively, in high yields and purities.

Abstract:Three polymerizable vinyl thiamphenicol esters with different acyl donor carbon chain length (C4, C6, C10) were regioselectivly synthesized by Lipozyme(R) (immobilized from mucor miehei) in acetone at 50 degreesC for 12 h to give 73%, 81%, 63% yield, respectively. The products were valuable monomers for preparation of macromolecular antibiotic.
