Establishment and evaluation of a mouse model of type Ⅱ diabetes mellitus complicated by tuberculosis

Abstract:Objective To establish a mouse model of type II diabetes mellitus complicated by tuberculosis and evaluate the pathogenetic process in the lung. Methods One hundred SPF 3-week old C57BL /6J mice were used in this study,40 mice as normal and 60 as diabetic group. Mice were fed with high-fat and high-sugar diet,and were intraperitoneally injected with streptozotocin( STZ) to generate mouse models of type II diabetes mellitus. Then,the mice were infected with Mycobacterium tuberculosis H37Rv by nasal drip to establish a mouse model of type II diabetes mellitus complicated by tuberculosis. The mice were sacrificed at 1st to 8th,10th and 12th weeks after infection,respectively,and the lungs were taken for gross and histopathological observation,and bacterial colony counting using HE and acid-fast staining. Results The gross inspection revealed changes in the lungs,and the lesions were gradually increased within the first 8 weeks,up to 80% of the whole organ. Since the 10th week the lesions became reduced. Typical tuberculosis granulomas were observed at 8th week and reparative changes began to occur in some areas of the lung at 10th week. Acid-fast staining showed the presence of Mycobacterium tuberculosis in the lung tissues. Conclusions An experimental mouse model of type II diabetes mellitus complicated by tuberculosis is successfully established. Compared with the mouse models of tuberculosis infection alone,the pathological changes in the mouse models of type II diabetes mellitus complicated by tuberculosis are characterized by early onset,rapid disease progression,and more similar to the clinical patients.
