Abstract:Aim: To analyze the effects of extract ginkgobiloba (EGb) on the blood circulation of arteriole in dorsal trapezius muscle and the contractility of gastrocnemius muscle in rats as well as the motor capacity in mice. Methods: The experiment was conducted in the College of Physical Education of Guangxi Normal University in February 2004. The EGb was manufactured by Guilin Site High-Tech Corporation of Natural Plants, including 24% of EGb and 6% of bilobalide, which was in accordance with the international criteria. 1 Observation on blood circulation of arteriole in dorsal trapezius muscle of rats: Forty SD rats were selected and randomly divided into blank control group and 2.0, 3.0, 4.0 g/L EGb groups with 10 rats in each group. Rats in each group were made into specimens of free muscle with specially made perfusion kit fixed on the free surface. Ringer's solution was used for perfusion, and the transversal arteriole with the caliber of 15-20 μm was observed for 30 minutes. 50 μL of norepinephrine (0.1 g/L) was dripped in the observing sites respectively, and 1mL of corresponding EGb solution was dripped in 2.0, 3.0, 4.0 g/L EGb groups respectively when the blood vessels contracted. Rats in the blank control group were dripped with normal saline at the same volume. The caliber of arteriole and the changes of fluid state were measured. 2 Observation on contractility of gastrocnemius muscle in rats: The general status and grouping of enrolled rats as well as were the same as those in experiment of blood circulation of arteriole in dorsal trapezius muscle. There were totally 36 rats with 9 in each group. The gastrocnemius muscle was cut off at the Achilles tendon under anesthetization, and the suspending electrodes was inserted into the gastrocnemius muscle by the pointed end to stimulate the specimens of each group once per second at 0.8 V until the muscle-contraction, which was considered as the time of muscular fatigue. Specimens in 2.0, 3.0, 4.0 EGb groups were injected with 1 mL of EGb solution at corresponding concentration, while rats in the blank control group were injected with normal saline at the same volume, 5 minutes after that the stimulus was continued, and the muscular contraction as well as the time of fatigue muscles recovered to normal were recorded. 3 Motor experiment: Sixty healthy Kunming male mice were selected and randomly divided into quiet control group, quiet administration group, simple exercise group and administration and exercise group with 15 mice in each group. Mice in the quiet administration group as well as administration and exercise group were administrated by gastric perfusion twice respectively in the morning and at night at the dose of 2.4 mL/kg. Mice in the quiet control group and simple exercise group were given gastric perfusion of plain boiled water at the same volume. Mice in the simple exercise group and administration exercise group were required to swim in a 100 cm ×50 cm ×80 cm transparent glass pool with tail loading once a day and 30 minutes each time for 6 continuous days. The exercise was conducted for 6 weeks in all with an interval of 1 day between each week. Mice in the quiet control group and quiet administration group swam without loading for 15 minutes per day. Climbing experiment was conducted on mice of each group at the 5th week and the static endurance was determined. Exhaustive swimming experiment was performed at the 6th week. Results: Forty SD rats enrolled in the experiment of blood circulation of arteriole in dorsale trapezius muscle, 36 SD rats enrolled in the experiment of contractility of gastrocnemius muscle and 60 kunming mice enrolled in the motor capacity were involved in the analysis of results, and 3 experiments were independent of each other. 1 Effects of EGb on the caliber of arterial blood vessels: Compared with the blank control group, the caliber in all EGb groups were obviously enlarged at 5 minutes after administration (P < 0.05 or 0.01). 2 Effects of EGb on the fluid state of arterial blood vessels: Compared with the blank control group, grade III evaluation on the fluid state of each Ebb group and the time of blood-stopping as well as the numbers of vessels recovered to normal were significantly different (P < 0.05 or 0.01). 3 Effects of EGb on the contraction of gastrocnemius muscle: Compared with the blank control group, the time of muscular fatigue in each EGb group were not remarkably changed (P>0.05), while the time was significant decreased (P < 0.05 or 0.01). 4 Comparison in capacities of climbing and weighted-swimming among each group: the static time of climbing was obviously longer for mice in the quiet administration group than that in the quiet control group (P < 0.05), and the swimming time with loading was remarkably longer in the administration swimming group than that in simple swimming group (P < 0.01). Conclusion: EGb can promote the blood circulation, enhance the metabolic rate, reduce the time of muscular fatigue and so on, based on which exercise training on rats can remarkably enhance the motor capacity and postpone the fatigue.
