Abstract:Objective To study the chemical constituents from the fruits of Lycium barbarum.Methods The compounds were isolated and purified by column chromatography and RP-HPLC.Their structures were elucidated by means of physicochemical properties and spectroscopic analyses(UV,MS,1H-NMR,and13C-NMR).Results Twelve compounds were separated and identified,including six phenolic acids: protocatechuic acid(1),dihydroisoferulic acid(2),caffeic acid(3),Z-p-hydroxy-cinnamic acid(4),E-p-hydroxy-cinnamic acid(5),and trans-cinnamic acid(6);Three coumarins: scopoletin(7),isoscopoletin(8),esculetin(9);Two pyrrole derivatives: 4-[formyl-5-(hydroxymethyl)-1H-pyrrol-1-yl] butanoic acid(10) and 4-[formyl-5-(methoxymethyl)-1H-pyrrol-1yl] butanoic acid(11);and 4′-hydroxyacetophenone(12).Conclusion Except compounds 3,5,and 7,others are isolated from this plant for the first time.Compounds 2,4,8,9,and 12 are firstly isolated from the plants in genus of Lycium L.
[A New Method for Simultaneous Assay of Six Organic Acids in Lonicerae Japonicae Flos].

Abstract:OBJECTIVE:To establish a method of quantitative analysis of multi-components by single marker(QAMS) for quality control of Lonicerae Japonicae Flos.METHODS:The contents of chlorogenic acid(CA) and caffeic acid(CfA) were determined, and the relative correction factors(RCF) of other organic acids were calculated, which were used for the indirect determination.RESULTS:The RCFs for the neochlorogenic acid(NCA), 3, 4-O-dicaffeoylquinic acid(3, 4-DCA), 3, 5-O-dicaffeoylquinic acid(3, 5-DCA), and 4, 5-O-dicaffeoylquinic acid(4, 5-DCA) were 5.462, 5.689, 2.313, 2.382(to CA) and 3.941, 4.103, 1.669, 1.718(to CfA), respectively. The established method was validated in different laboratories with different high performance liquid chromatography(HPLC) instruments and different chromatographic columns; the result indicated that the reproducibility was satisfactory. There was no significant difference between the established QAMS method and external reference method (P>0.05).CONCLUSION:The established QAMS method can be used for simultaneous determination of 6 organic acids as quality control of Lonicerae Japonicae Flos with easy available standard substances.

Abstract:OBJECTIVE To research chemical constituents from Caulis Lonicerae Japonicae.METHODS The compounds were separated and purified by column chromatography on macroreticular resin and silica gel.Their structures were identified by MS,~(1)H-NMR and ~(13)C-NMR spectral analysis. RESULTS Two iridoid glycosides were obtained and identified as loganin(I) and sweroside(II).CONCLUSION Compound I and II are isolated from the Caulis Lonicerae Japonicae for the first time.

Abstract:Nine (1–9) and seven (1–6, 10) compounds were isolated from the fruits of Sonneratia caseolaris and Sonneratia ovata, respectively. Their structures were identified by comparing their MS and NMR data as well as the physical properties with the literature. All the isolated compounds were screened against a rat glioma C-6 cell line using the MTT assay method; only compounds (-)-(R)-nyasol (1), (-)-(R)-4′-O-methylnyasol (2) and maslinic acid (6) were found to show moderate cytotoxic activity. Our findings from these two kinds of fruits can be used as a foundation for further chemotaxonomic studies on Sonneratia species. The nor-lignans (1, 2) and 6H-benzo[b,d]pyran-6-one derivatives (3, 4) were isolated from this genus for the first time, indicating that these two classes of compounds may tentatively be considered as taxonomic markers for Sonneratia genus.

Abstract:AIM:To investigate the active constituents in Flos Lonicera (Jinyinhua,in Chinese),and to estimate the quality of Flos Lonicera more roundly.METHOD:Many kinds of column chromatography were used to isolate the compounds from the EtOH ext. of Lonicera confusa DC. The chemical constituents of the plant were identified by means of IR,MS, 1HNMR, 13CNMR,respectively,in some cases by direct comparison with authentic samples.RESULT:Eight flavonoids were isolated from L.confusa DC. and identified as:luteolin (Ⅰ),quercetin (Ⅱ),tricin (Ⅲ),tricin-7-O-β-D-glucopyranoside (Ⅳ),luteolin-7-O-β-D-galactoside(Ⅴ),rutin(Ⅵ),chrysoeirol-7-O-neohesperidoside(Ⅶ),and tricin-7-O-neohesperidoside(Ⅷ).CONCLUSION:Compounds Ⅲ,Ⅳ,Ⅵ,Ⅶ and Ⅷ were firstly isolated from this genus,and others were firstly isolated from this plant.

Abstract:Lonicerae japonicae flos (LJF), Lonicerae japonicae caulis (LJC), Lonicerae folium (LF) and Lonicerae fructus (LFR) are derived from Lonicera japonica Thunb., which are formed due to different medicinal parts. The efficacy of the 4 medicinal materials has similarities and differences. However, little attention has been paid to illustrate the differences in efficacy from the perspective of phytochemistry. In this study, ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry (UPLC-Q-Exactive-Orbitrap-MS) was used to qualitatively analyze the ingredients in 4 herbs. A total of 86 compounds were plausibly or unambiguously identified, there were 54 common components among the 4 medicinal materials, and each kind of medicinal materials had its own unique components. On the basis of qualitative analysis, ultra-performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (UPLC-QQQ-MS/MS) was used to quantitatively analyze 31 components contained in 4 medicinal materials, and principal component analysis (PCA), orthogonal partial least squares discriminant analysis (OPLS-DA) and other multivariate statistical analysis were furtherly performed for comparing the component contents. The results showed that the samples from the same parts were clustered into one group, and the samples from different medicinal parts were significantly different. The analysis of variable importance projection (VIP) value of the OPLS-DA model showed that 10 components including chlorogenic acid, secologanic acid, isochlorogenic acid A, loganin, lonicerin, loganic acid, secoxyloganin, sweroside, luteolin and rhoifolin were the main difference components among the 4 medicinal materials. The study not only lays a solid foundation for the intrinsic quality control of 4 medicinal materials and the study of different effects of the 4 medicinal materials at the phytochemical level, but also provides a basis for more rational utilization of various parts of L. japonica and expansion of medicinal resources.

Abstract:Morphological characteristics, ITS sequences, and active compounds have been used to differentiate between species of Lonicera used in the traditional Chinese medicines Flos Lonicerae Japonicae (FLJ) and Flos Lonicerae (FL). FLJ includes L. japonica whereas FJ includes Lonicera macranthoides, Lonicera hypoglauca, Lonicera confusa and Lonicera fulvotomentosa. FLJ could be distinguished from FL using four quantitative and 10 qualitative characters, ITS sequences, chlorogenic acid and luteoloside contents. Analyses revealed that L. japonica was very different from the other species. The results have implications for the identification and quality control of species of Lonicera used medicinally, suggesting that species should not be interchanged in medicinal preparations.

Abstract:Lonicerae japonicae flos and Lonicerae flos are increasingly widely used in food and traditional medicine products around the world. Due to their high demand and similar appearance, they are often used in a confused or adulterated way; therefore, a rapid and comprehensive analytical method is highly required. In this case, the comparative analysis of a total of 100 samples with different species, growth modes, and processing methods was carried out by nuclear magnetic resonance (1H-NMR) spectroscopy and chemical pattern recognition analysis. The obtained 1H-NMR spectrums were employed by principal component analysis (PCA), partial least-squares discriminant analysis (PLS-DA), orthogonal partial least-squares discriminant analysis (OPLS-DA), and linear discriminant analysis (LDA). Specifically, after the dimensionality reduction of data, linear discriminant analysis (LDA) exhibited good classification abilities for the species, growth modes, and processing methods. It is worth noting that the sample prediction accuracy from the testing set and the cross-validation predictions of the LDA models were higher than 95.65% and 98.1%, respectively. In addition, the results showed that macranthoidin A, macranthoidin B, and dipsacoside B could be considered as the main differential components of Lonicerae japonicae flos and Lonicerae Flos, while secoxyloganin, secologanoside, and sweroside could be responsible for distinguishing cultivated and wild Lonicerae japonicae Flos. Accordingly, 1H-NMR spectroscopy combined with chemical pattern recognition gives a comprehensive overview and provides new insight into the quality control and evaluation of Lonicerae japonicae flos.

Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.
Accumulation Pattern of Flavonoids during Fruit Development of Lonicera maackii Determined by Metabolomics

Abstract:Lonicera maackii (Caprifoliaceae) is a large, upright shrub with fruits that contain many bioactive compounds. Flavonoids are common active substances in L. maackii. However, there is a dearth of information about the accumulation of these flavonoids and their possible medicinal value. We used targeted metabolomics analysis based on ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) to analyze five developmental stages of L. maackii fruit. A total of 438 metabolites were identified in the five developmental stages, including 81 flavonoids and derivatives. The 81 flavonoids included 25 flavones and derivatives, 35 flavonols and derivatives, two isoflavones, three cyanidins and derivatives, eight procyanidins, and eight flavanones. In addition, we outlined the putative flavonoid biosynthesis pathway and screened their upstream metabolites. More importantly, we analyzed the accumulation patterns of several typical flavones and flavonols. The results reported here improved our understanding of the dynamic changes in flavonoids during fruit development and contributed to making full use of the medicinal value of L. maackii fruit.

Abstract:OBJECTIVE Elucidation of the chemical constituents from fruits of Schisandra chinensis(Turcz) Baill.METHODS Chromatographic means and spectral analysis were applied to identify chemical constituents in the title plant. RESULTS Four compounds were isolated and characterized as ganwuweizic acid(Ⅰ),mono-ethyl fumarate(Ⅱ),tetracosanoic acid(Ⅲ) and meso-dihydroguaiaretic acid(Ⅳ) respectively.CONCLUSION Ⅰ～Ⅲ were isolated from S.chinensis for the first time and the ~(13)C NMR data of meso-dihydroguaiaretic acid(Ⅳ) was firstly offered and confirmed by 2D NMR analysis.

Abstract:Six new isolates including three new alkaloids (1–3), one new secoiridoid glycoside (4) and one new 11-delactonization-secoiridoid glycoside (5), and one new organic acid (6) were identified from the leaves of Lonicera japonica, among which 1 and 2 assigned as a pair of configurational isomers possessed two oxazolidin-2-one fragments connected through NN bond. The structures were elucidated by the NMR data and ICD analysis. All the compounds (1–6) were tested for their antioxidant and hepatoprotective activities using cell models of ABAP-induced HepG2 cell and APAP-induced HepG2 cell by the MTT method. Outstandingly, compound 2 exhibited remarkable antioxidative activity with inhibitory rate of 117.2%, and compounds 2, 4, 6 exhibited significant effects with inhibitory rates of 68.1%, 69.3%, 69.2%, respectively.

Abstract:The flowers of Lonicera japonica Thunb. were extensively used to treat many diseases. As the demands for L. japonica increased, some related Lonicera plants were often confused or misused. Caffeoylquinic acids were always regarded as chemical markers in the quality control of L. japonica, but they could be found in all Lonicera species. Thus, a simple and reliable method for the evaluation of different Lonicera flowers is necessary to be established. In this work a method based on single standard to determine multi-components (SSDMC) combined with principal component analysis (PCA) for control and distinguish of Lonicera species flowers have been developed. Six components including three caffeoylquinic acids and three iridoid glycosides were assayed simultaneously using chlorogenic acid as the reference standard. The credibility and feasibility of the SSDMC method were carefully validated and the results demonstrated that there were no remarkable differences compared with external standard method. Finally, a total of fifty-one batches covering five Lonicera species were analyzed and PCA was successfully applied to distinguish the Lonicera species. This strategy simplifies the processes in the quality control of multiple-componential herbal medicine which effectively adapted for improving the quality control of those herbs belonging to closely related species.

Determination of Six Chlorogenic Acids in Lonicerae Flos Extract by Quantitative Analysis Multi-Components with Single Marker

Abstract:This study is to determine six chlorogenic acids (chlorogenic acid, 5-O-caffeoylquinic acid, 4-O-caffeoylquinic acid, 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid and 4,5-di-O-caffeoylquinic acid) by quantitative analysis of multi-components with a single marker (QAMS). Chlorogeinc acid was used as internal reference to calculate the relative correction factors (RCF) of five compounds. Then the ruggedness of relative correction factors was tested on different instruments and columns. Meanwhile, a total of 4 batches of Lonicerae Flos and 20 batches of Lonicerae Flos extract with five different processing procedures were analyzed by external standard method (ESM) and QAMS, respectively. The ruggedness of relative correction factors was good. And the analytical results calculated by ESM and QAMS showed no difference. The quantitative method established was suitable for the quality evaluation of Lonicerae Flos extract.

Quality control of Lonicerae Japonicae Flos using near infrared spectroscopy and chemometrics

Abstract:A set of qualitative and quantitative methods based on near infrared (NIR) spectroscopy was established for the geographical origin identification, quantitative determination of active compounds, and fingerprint analysis of Lonicerae Japonicae Flos. The spectra of 140 samples from different origins were collected with two NIR instruments from different manufacturers (Thermo Scientific and Buchi). A Soft Independent Modeling of Class Analogy model was established for the identification of Lonicerae Japonicae Flos from the genuine producing area. Using the established discriminant analysis model, 22 samples from Henan province were predicted with 100% rate of accuracy, while the 68 samples from other producing areas were predicted with 9 samples incorrectly judged. Futhermore, partial least square regression method was used for developing the quantitative calibration models with the reference values determined with a validated HPLC-UV method. The RMSEP values of external validation are 0.169%, 0.048%, 0.172%, 0.007%, 0.203%, and 0.066% for NCA, CA, CfA, 3,4-DCA, 3,5-DCA, and 4,5-DCA, respectively, which indicated that the established models possess satisfactory predictive abilities. In addition, the models established on the primary instrument can also be transferred to the secondary instrument using direct standardization algorithm, which enlarged the application scope of the established models. Since the NIR spectra can reflect the comprehensive quality information of Lonicerae Japonicae Flos, a fingerprint analysis method was finally proposed for the quality consistency check of raw materials. The proposed methods are rapid and effective, and possess good portability, which is helpful to the quality control of Lonicerae Japonicae Flos.

Abstract:Amur honeysuckle (Lonicera maackii) is a widely used medicinal plant of the Caprifoliaceae family that produces chlorogenic acid. Research on this plant mainly focuses on its ornamental value and medicinal compounds, but a reference genome sequence and molecular resources for accelerated breeding are currently lacking. Herein, Nanopore sequencing and high-throughput chromosome conformation capture (Hi-C) allowed a chromosome-level genome assembly of Lonicera maackii (2n = 18). A global view of the gene regulatory network involved in the biosynthesis of chlorogenic acid and the dynamics of fruit coloration in L. maackii was established through metabolite profiling and transcriptome analyses. Moreover, we identified the genes encoding hydroxycinnamoyl-CoA quinate transferase (LmHQT) and hydroxycinnamoyl-CoA shikimic/quinate transferase (LmHCT), which localized to the cytosol and nucleus. Heterologous overexpression of these genes in Nicotiana benthamiana leaves resulted in elevated chlorogenic acid contents. Importantly, HPLC analyses revealed that LmHCT and LmHQTs recombinant proteins modulate the accumulation of chlorogenic acid (CGA) using quinic acid and caffeoyl CoA as substrates, highlighting the importance of LmHQT and LmHCT in chlorogenic acid biosynthesis. These results confirmed that LmHQTs and LmHCT catalyze the biosynthesis of CGA in vitro. The genomic data presented in this study will offer a valuable resource for the elucidation of CGA biosynthesis and facilitating selective molecular breeding.

Abstract:Aim: To investigate the active constituents in the buds of Lonicera macranthoides Hand.-Mazz., and to study the chemical constituents to estimate the quality of Flos Lonicerae. Methods: Many kinds of column chromatography were used to isolate the constituents from the extracts of the buds of L. macranthoides. The chemical constituents of the plant were elucidated by means of spectroscopic methods, respectively. Results: Eleven compounds were obtained and identified as hederagenin-28-O-[β-D-glucopyranosyl- (1→6)-β-D-glucopyranosyl]ester(I), 3-O-[α-L- rhamnopyranosyl]-(1→2)-α-L-arabinopyranosyl] hederagenin(II), 3-O-(β-D-glucopyranosyl-(1→-3)-α-L- rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl] hederagenin (III), 3-O-α-L-arabinopyranosyl hederagenin-28-O-[β-D- glucopyranosyl-(1 → 6)-β-D-glucopyranosyl ester (IV), 3-O-[β-D-glucopyranosyl-(1 → 4)-β-D- glucopyranosyl-(1→3)-α-L-rhamnopyranosyl- (1→2)-α-L-arabinopyranosyl] hederagenin (V), 3-O-[α- L-rhamnopyranosyl-(1→2)-α-L-araninopyranosyl] hederagenin-28-O-[β-D-glucopyranosyl-(1→6) -β-D-glucopyranosyl] ester(VI), 3-O-[β- D-glucopyranosyl-(1→ 3)-α-L-rhamnopyranosyl-(1 → 2)-α-L-arabinopyranosyl] hederagenin- 28-O-[β-D-glucopyranosyl-(1 → 6)-β-D-glucopyranosyl] ester (VII), 3-O-[β-D-glucopyranosyl- (1→4)-β-D-glucopyranosyl-(1→3)- α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl] hederagenin-28-O-[β-D-glucopyranosyl- (1→6)-β-D-glucopyranosyl] ester(VIII), kaempferol-3-O-β- D-glucopyranoside (IX), rutin(X), luteolin-7-O-β-D-glucopyranoside(XI). Conclusion: Saponins I - V, flavonoids IX and X were isolated from the plant for the first time.
