Abstract:A green and fully enzymatic procedure for the resolution of chiral alcohols through lipase/esterase-catalyzed acylation and subsequent lipase-catalyzed aminolysis using anhydrous ammonia was demonstrated. Both enantiomers can be obtained in high ee values (up to >99%) under ambient reaction conditions. The solvent and acyl donors can be recycled, and the enzyme can be reused for up to five times.
Preparation of (R)-1-(2-naphthyl)ethylamine by Enzymatic Kinetic Resolution with A New Acyl Donor

Abstract:Enantiomerically pure (R)-1-(2-naphthyl)ethylamine was firstly prepared by an efficient enzymatic kinetic resolution process. The effects of lipases, solvents, acyl donors, substrate concentration and temperature on the reaction were investigated. It was found that as the acyl donor, 4-chlorophenyl valerate could effectively inhibit the non-selective amidation reaction. The optimal solvent, amine concentration and temperature were toluene, 300 mmol/L and 40 degrees C, respectively. Under the optimal conditions mentioned above, an excellent ee(p), value (>99%) at high conversion (50%) in 8 h could be achieved on the kinetic resolution reaction catalyzed by Novozym 435 with the new acyl donor.

Abstract:The enzymatic resolution of 1-phenylethylamine by amidation reaction in organic medium was studied. Based upon the screening of acyl donors, enzymes and solvents, it was concluded that isopropyl acetate was the most suitable acyl donor and a small amount of NaHSO3 can reduce side reactions. Novozym 435 showed both the highest catalytic activity and the highest enantioselectivity. Toluene was the most suitable medium for the reaction. The effects of ester concentration, amine concentration, enzyme concentration, shaking rate and temperature on the reaction were examined. The optimum ratio of amine to ester, amine concentration, enzyme concentration, shaking rate and temperature were 1:0.6, 200 mmol·L-1, 4 mg·ml-1, 200 r·min-1, 30°C, respectively. Under the optimal conditions mentioned above, E value was 89. A product enantiomeric excess (eep) of 96% was achieved with conversion of 39% after 4 h of reaction and a substrate enantiomeric excess (ees) of 98% was achieved with conversion of 52.4% after 10 h of reaction.

Abstract:In contrast to alcohols and amines, racemic lactams and thiolactams cannot be resolved directly via enzymatic acylation or classical resolution. Asymmetric N-acylation promoted by amidine-based catalysts, particularly Cl-PIQ 2 and BTM 3, provides a convenient method for the kinetic resolution of these valuable compounds and often achieves excellent levels of enantioselectivity in this process. Density functional theory calculations indicate that the reaction occurs via N-acylation of the lactim tautomer and that cation-π interactions play a key role in the chiral recognition of lactam substrates.

Abstract:The lipase-catalyzed remote kinetic resolution of 4-[4-(dimethylamino)-1-(4-fluorophenyl)-1-hydroxy-1-butyl]-3-(hydroxymethyl)-benzonitrile (1), a useful intermediate of citalopram with a quaternary chiral center, was studied. Through screening, the preferable system for the asymmetric reaction was: Candida antarctica lipase B (Novozym435) as a catalyst, acetonitrile as a solvent and vinyl acetate as an acyl donor. The influence of various parameters on the enzymatic resolution was studied. The optimal condition was as follows: temperature of 30 degrees C, the molar ratio of diol 1 to vinyl acetate 1 to 5, 10 mg/mL lipase, and 200 r/min rotation speed. Reaction curves of 180 and 60 mmol/L diol 1 concentrations ensured the high reaction rate and enantioselectivity. Reusability study ensured the operational stability of lipase over repeated cycles.

Abstract:Regio- and enantioselective hydroarylamination, hydroalkylamination and hydroamidation of styrenes have been developed by NiH catalysis with a simple bioxazoline ligand under mild conditions. A wide range of enantioenriched benzylic arylamines, alkylamines and amides can be easily accessed by nitroarenes, hydroxylamines and dioxazolones, respectively as amination reagents. The chiral induction in these reactions is proposed to proceed through an enantiodifferentiating syn-hydronickellation step.

Abstract:An efficient approach for synthesizing chiral β-amino nitroalkanes has been developed via the Ni-catalyzed asymmetric hydrogenation of challenging β-amino nitroolefins under mild conditions, affording the desired products in excellent yields and with high enantioselectivities. This protocol had good compatibility with the wide substrate scope and a range of functional groups. The synthesis of chiral β-amino nitroalkanes on a gram scale has also been achieved. In addition, the reaction mechanism was elucidated using a combined experimental and computational study, and it involved acetate-assisted heterolytic H2 cleavage followed by 1,4-hydride addition and protonation to achieve the nitroalkanes.

Abstract:Direct synthesis of aliphatic amines from alkynes is highly desirable due to its atom economy and high stereoselectivity but still challenging, especially for the long-chain members. Here, a combination of Au-catalyzed alkyne hydration and amine dehydrogenase-catalyzed (AmDH) reductive amination was constructed, enabling sequential conversion of alkynes into chiral amines in aqueous solutions, particularly for the synthesis of long-chain aliphatic amines on a large scale. The production of chiral aliphatic amines with more than 6 carbons reached 36-60 g/L. A suitable biocatalyst [PtAmDH (A113G/T134G/V294A)], obtained by data mining and active site engineering, enabled the transformation of previously inactive long-chain ketones at high concentrations. Computational analysis revealed that the broader substrate scope and tolerance with the high substrate concentrations resulted from the additive effects of mutations introduced to the three gatekeeper residues 113, 134, and 294.

Abstract:Due to their importance in natural products, organic synthesis, life science, and pharmaceuticals, reliable methods for the synthesis of chiral amines are urgently required. Here in this contribution, the dual chiral catalytic method has been established for the enantioselective reaction of terminal alkynes, aldehydes, and amines affording optically active propargylic amines with a decent enantioselectivity and a broad substrate scope. These propargylic amines have been demonstrated as platform molecules for the syntheses of different types of chiral amines. By applying this protocol, the asymmetric total syntheses of two linear monoamine natural products, (S)-5 and (S)-11, which were isolated from marine cyanobacteria Microcoleus lyngbyaceus, have been realized. The envisioned chiral ion pair, which has boosted the enantioselectivity greatly, and other key intermediates have been successfully characterized by MS studies.

Abstract:An efficient and convenient strategy for synthesis of enantiomerically pure S -2-(1-hydroxy-3-butenyl)-5-methylfuran was for the first time described utilizing a lipase-mediated asymmetric acylation in organic solvents. Rhizopus arrhizus lipase was chosen as the biocatalyst, and different immobilization methods including sol–gel encapsulation and covalent attachment were adopted to improve its catalytic characteristics. Various influential factors of the reaction were also investigated. Finally, the results showed that the lipase covalently attached onto epoxy resin exhibited the highest enantioselectivity and operational stability. Under optimized reaction conditions, i.e., n -hexane as the solvent, 5/1 (mol/mol) of vinyl acetate to 2-(1-hydroxy-3-butenyl)-5-methylfuran and 30 °C, the ee value of S - 1 reached up to above 98% at 52% conversion with an E value of 99.

Abstract:Catalytic enantioselective α-C–H functionalization of widely available achiral alkyl amines could provide an ideal synthetic approach towards chiral amines. However, the inert nature of the α-C–H of alkyl amines renders their activation as carbanionic nucleophiles for catalytic asymmetric reactions an important yet unmet challenge. Here we describe how N -arylidene-protected alkyl amines could be activated as carbanions for asymmetric conjugate addition and the Mannich reaction. These results represent an intriguing and generally useful approach to the synthesis of chiral α,α-dialkyl amines. More importantly, they highlight the enormous potential of N -arylidene-protected amines as readily available and widely applicable synthons for the asymmetric synthesis of chiral amines.

Abstract:Enantioselective protonation, delivery of a proton to a carbanion intermediate, is the most straightforward and fundamental method for the preparation of a chiral tertiary carbon stereocenter. Recent efforts for this objective have been realized through enamine catalysis, which has now become a prominent catalytic strategy enabling a range of fascinating chiral transformations. This review will summarize recent advances in the field of enantioselective enamine protonation for the synthesis of optically active carbonyl compounds. Dynamic kinetic resolutions of alpha-substituted carbonyl compounds through enamine intermediates will be discussed as well.

Abstract:A chemo-enzymatic synthesis method of S-citalopram was developed to overcome the disadvantage of relatively low selectivity of enzyme towards tertiary alcohols. The combination of kinetic resolution, cyclic resolution and stereoinversion synthesis was successfully applied in the asymmetric synthesis of the S-citalopram. Using the kinetic model to predict the cyclic resolution, R-diol with high ee value was obtained by controlling the conversion rate. Subsequently, the unwanted R-diol was inverted to S-citalopram by stereoinversion of chiral quaternary center with 98.0% yield and ee value of 91.0%. Based on dynamic simulation and experiments, the kinetic resolution was scaled up from 10 mL to 1 L and 14 L, gradually. There was no significant scale-up effect and the dynamic simulation result fitted the experimental data well, with an error of 12.5 and 14.0%, respectively. This chemo-enzymatic synthesis route is a promising model system for the production of pharmaceuticals with the chiral tertiary alcohols intermediate.

Abstract:The first example of copper-catalyzed ring-opening, enantioselective arylation of cyclic ketoxime esters to access ω,ω-diaryl alkyl nitriles has been developed in high yield (up to 92% yield) with excellent enantioselectivity (up to 91% ee). Side-arm bis(oxazoline) ligand plays a significant role in this asymmetric catalytic transformation, which provides an efficient route to construct diverse chiral ω,ω-diaryl alkyl nitriles. Synthetic utility has also been demonstrated in the further derivatization of the ω,ω-diaryl alkyl nitrile to the corresponding amide.
