Abstract:Embryo Implantation is a sophisticated process by which the blastocyst makes the first physical and physiological contact with the uterine luminal epithelium. A successful implantation requires a receptive endometrium, a normal and functional embryo at the blastocyst developmental stage and a synchronized crosstalk between maternal and embryonic tissues. Angiogenesis is crucial to successful implantation and decidualization, however, as an important angiogenic growth factor, the effect of Angiopoietin-2 (Ang-2) in the process of implantation and decidualization is still unknown. The aim of this study was to investigate the expression of Ang-2 gene in different developmental stages of mouse preimplantation embryos and in the mouse uterus during early pregnancy, and pseudopregnancy, delayed implantation and artifical decidualization, and also its regulation by steroid hormones using in situ hybridization and reverse transcription polymerase chain reaction (RT-PCR). The results showed that there was no detectable Ang-2 mRNA signal on days 1-5 of pregnancy by in situ hybridization. On days 6-8, Ang-2 mRNA was mainly expressed in the primary decidua of mesometrial side, and the expression gradually increased. By RT-PCR, a significantly high level of Ang-2 expression was seen on day 8 of pregnancy although Ang-2 expression could be found through days 1-8. Similarly, the high level of Ang-2 mRNA was induced in decidualized cells under artificial decidualization. Ang-2 mRNA was not expressed in the uterus from days 1 to 5 of pseudopregnancy, delayed implantation uterus and activated implantation sites by in situ hybridization. However, Ang-2 expression was detected through days 1-5 of pseudopregnancy by RT-PCR and highly expressed on day 3. Simultaneously, Ang-2 mRNA was also highly expressed in activated implantation sites compared with the delayed implantation uterus. In the ovariectomized mouse uterus, Ang-2 expression gradually increased after estrogen injection, and with peak levels at 12h, while progesterone injection could cause a decline in uterine Ang-2 mRNA level which reaches a nadir at 12h. These results suggest that the Ang-2 may play a key role in the process of mouse decidualization. Estrogen can induce the expression of Ang-2 while progesterone can inhibit its expression in the ovariectomized mouse uterus.
