Abstract:Lipases are ubiquitous in animals, plants and microorganisms, but industrial lipases are mainly produced by microorganisms. The industrial application of lipases is constrained by its high cost and enzymatic characteristics such as temperature and pH tolerance, specific activity, substrate specificity and solvent resistance. Despite the abundance of information regarding microbial lipase genes and proteins, there are currently very few commercial lipase preparations that are suitable for application in the food, pharmaceutical and energy industries. Accordingly, tremendous research efforts have been made to mine novel alternative lipase resources. This article mainly presents a review of the approaches used to mine microbial lipase resources including environmental screening and metagenomics screening for mining new lipases with good properties, genetic improvement of lipase-producing strains, recombinant expression of lipases in genetically engineered strains, lipase modification by protein engineering, lipase immobilization and optimization of reaction conditions of immobilized lipase.

Abstract:Biodiesel is an environment‐friendly and renewable fuel produced by transesterification of various feedstocks. Although the lipase‐catalyzed biodiesel production has many advantages over the conventional alkali catalyzed process, its industrial applications have been limited by high‐cost and low‐stability of lipase enzymes. This review provides a general overview of the recent advances in lipase engineering, including both protein modification and production. Recent advances in biotechnology such as in protein engineering, recombinant methods and metabolic engineering have been employed but are yet to impact lipase engineering for cost‐effective production of biodiesel. Abstract:In recent years, biological ways for biodiesel production have drawn an increasing attention and compared to chemical approaches, lipase-mediated alcoholysis for biodiesel production has many advantages. Currently, there are extensive reports about enzyme-mediated alcoholysis for biodiesel production, and based on the application forms of biocatalyst, the related research can be classified into immobilized lipase, whole cell catalyst, and liquid lipase-mediated alcoholysis for biodiesel production, respectively. This mini-review is focusing on the study of the aforementioned three forms of biocatalyst for biodiesel production, as well as its impacts and prospects.

Abstract:Increasing environmental concerns and significant demand for industrial fuels necessitate the development of alternate and sustainable energy sources. Biodiesel is a renewable and environmentally friendly fuel produced by trans-esterifying a variety of feedstocks. Lipases are robust triacylglycerol ester hydrolases that catalyze hydrolysis, esterification, interesterification, transesterification, acylation, acidolysis, alcoholysis, aminolysis, and resolution of racemates. Although the lipase-assisted greener biosynthesis of biodiesel has numerous advantages over the traditional alkali-based process, low catalytic efficiency, marginal stability, and high cost of lipase enzymes limit its widespread industrial processability. Protein engineering methodologies such as directed evolution, semi-rational design, and rational design can effectively tailor or improve the biocatalytic characteristics of lipase enzymes used in biodiesel generation, such as thermostability, solvent tolerance, activity, and substrate specificity. These innovative techniques improved our ability to predict structure-function relationships, engineering qualities, computational tools for designing new biocatalysts, and functional screening to manipulate enzyme traits for application in prevalent industrial bioprocesses. Many recent studies have demonstrated improved lipase performance, such as activity, stability, and specificity via protein engineering. This review spotlights a current overview of lipase engineering at the molecular scale with robust biocatalytic properties for biodiesel synthesis.

Abstract:To valorize oleaginous Schizochytrium sp. biomass, a novel sequential process was addressed and developed to simultaneously produce high value DHA-enriched glycerides and low value bulky biodiesels. In this process, Schizochytrium sp. biomass was first treated by alkaline protease; then the resulting biomass was directly subjected to enzymatic ethanolysis catalyzed by liquid enzymes from C. antarctica (CALA), T. lanuginosus (TL), R. miehei (RM) and A. oryzae (PLA). For all four liquid lipases, the alkaline protease treatment on Schizochytrium sp. biomass significantly enhanced total lipid recovery, biodiesel yield and yield of DHA-enriched glycerides. Under optimized conditions, CALA and TL achieved the highest biodiesel yields 76.28-76.66 wt% based on microalgae TFAs while PLA had the lowest (55.25 wt%). Moreover, CALA attained the highest DHA (77.93 wt%) and LC PUFAs (93.08 wt%) contents in glyceride fraction, suggesting a superior performance to concentrate DHA and LC PUFAs in glycerides. C-13 NMR analysis illustrated that non-regiospecificity and unfavorability towards LC PUFAs of CALA made it possible to transesterify saturated fatty acids located in all 3 positions of microalgal oil; thus LC PUFAs were highly enriched in glycerides. Overall, the sequential process using enzymes and ethanol as green media was successfully established for the co-production of high-value microalgal bioproducts rich in LC PUFAs and sustainable biodiesels in a comprehensive manner.

Abstract:Recently,with the global shortage of fossil fuels,price hike of crude oil and increased environmental concerns have resulted in the rapid growth of biodiesel production.The central reaction in the biodiesel production is the transesterification reaction which can be catalyzed either chemically or biologically.Enzymatic transesterification has certain advantages over chemical catalysis of transesterification,as it is less energy intensive,allows easy recovery of glycerol as a by-product and the transesterification of glycerides with high free fatty acid contents.This article reviews the current status and latest development of biodiesel production with immobilized lipase,including various feedstocks,source of lipases,immobilization techniques,lipase inactivation caused by methanol(ethanol)and glycerol.Lastly,future perspective for biodiesel production catalyzed by immobilized lipase is discussed.

Abstract:Tert-butanol was adopted as a reaction medium for lipase-catalyzed methanolysis of oils for biodiesel production,in which both the negative effects caused by excessive methanol and by-product glycerol could be eliminated.The optimum conditions of methanolysis of rapeseed oil were as follow:tert-butanol/oil(volumetric ratio)1:1,methanol/oil(molar ratio)4:1,3% Lipozyme TLIM and 1% Novozym 435 based on oil weight,35℃,130 r/min and 12 h.The highest biodiesel yield was 95% and the lipases could be reused 200 cycles without obvious loss of their activity.The technology was further tested in a pilot plant and the product was up to the biodiesel standards of US(ASTM PS121-99)and Germany(DINE51606).Furthermore,soybean oil,tung oil,cottonseed oil,sapium fat and acidified waste oil were also explored for biodiesel production,and lipases also showed good stability in tert-butanol reaction medium.

Abstract:A kind of microzyme was used in lipid production,and the effects of operation conditions in 1 L fermentor culture were studied.The quantitative test lipid by Sudan Black B straining and confirmed the component of lipid by gas chromatograph and mass spectrum(GC-MS) were carried out.The results showed that the optimal operation conditions in 1 L fermentor culture were as follows:at 30°C,1.1 vvm of ventilation,pH=6,and 300 r/min of stirrer speed.The analysis indicated that the components of lipid produced by fermentation were mainly made of palmitic acid,steric acid,oleic acid and linoleic acid,which were 17.80%,10.27%,53.02%,and 5.29%,respectively,and the components and their portions are very like those in vegetable oil.Furthermore the lipid produced by fermentation was used as raw material to produce biodiesel with alkali as catalyst.

Abstract:With the industrialization and development of modern science, the application of enzymes as green and environmentally friendly biocatalysts in industry has been increased widely. Among them, lipase (EC. 3.1.1.3) is a very prominent biocatalyst, which has the ability to catalyze the hydrolysis and synthesis of ester compounds. Many lipases have been isolated from various sources, such as animals, plants and microorganisms, among which microbial lipase is the enzyme with the most diverse enzymatic properties and great industrial application potential. It therefore has promising applications in many industries, such as food and beverages, waste treatment, biofuels, leather, textiles, detergent formulations, ester synthesis, pharmaceuticals and medicine. Although many microbial lipases have been isolated and characterized, only some of them have been commercially exploited. In order to cope with the growing industrial demands and overcome these shortcomings to replace traditional chemical catalysts, the preparation of new lipases with thermal/acid-base stability, regioselectivity, organic solvent tolerance, high activity and yield, and reusability through excavation and modification has become a hot research topic.

Abstract:Lipase is one of the most widely studied and applied biocatalysts. Due to the high enzyme leakage rate of the immobilization method of physical adsorption, we propose a new lipase immobilization method, based on the combination of macroporous resin adsorption and organic polymer coating. The immobilized Candida antarctica lipase B (CALB@resin-CAB) was prepared by combining the macroporous resin adsorption with cellulose acetate butyrate coating, and its structure was characterized by various analytic methods. Immobilized lipase was applied for biodiesel production using acidified palm oil as the starting material, the conversion rate achieved as high as 98.5% in two steps. Furthermore, the immobilized lipase displayed satisfactory stability and reusability in biodiesel production. When the aforementioned reaction was carried out in a continuous flow packed bed system, the yield of biodiesel was 94.8% and space-time yield was 2.88 g/(mL∙h). The immobilized lipase CALB@resin-CAB showed high catalytic activity and stability, which has good potential for industrial application in the field of oil processing.

Abstract:Although Saitozyma podzolica Zwy2-3 can use the enzymatic hydrolysate of corn stalks treated with an ammonium carbonate-steam explosion (EHCS-ACSE) as a substrate for lipid accumulation, the inefficient conversion of sugars from EHCS-ACSE into lipids necessitates the further optimization of fermentation parameters. Response surface design was used to optimize the primary fermentation parameters. Under the optimized conditions of the reducing sugar concentration of 89.44 g/L, yeast extract concentration of 3.88 g/L, rotational speed of 219 rpm, and incubation time of 122 h, the maximum lipid production achieved 11.45 g/L, which was 2.28 times higher than the results of the previous study. In addition, lipid profiling showed the presence of four fatty acid methyl esters, with the highest percentage being 61.84% oleic acid, followed by 21.53% palmitic acid, 13.05% stearic acid, and 3.58% linoleic acid. It is noteworthy that the composition and relative abundance of microbial lipids remained constant under different culture conditions. The characteristics of Zwy2-3 biodiesel, such as the iodine value (62.09), cetane number (59.29), density (0.87 g/cm3), and oxidation stability (35.53), meet the international standards (ASTM D6751-02 and EN 14214) for biodiesel. The present study further demonstrated that S. podzolica Zwy2-3 can efficiently utilize EHCS-ACSE for microbial lipid accumulation, and its lipids have favorable qualities that make them suitable for biodiesel production.

Abstract:Enzymatic biodiesel production has attracted tremendous interest due to its well-recognized advantages. However, high enzyme costs limit the application of enzymatic processes in industrial production. In the past decade, great improvements have been achieved in the lab and the industrial scale, and the production cost of the enzymatic process has been reduced significantly, which has led to it being economically competitive compared to the chemical process. This paper summarizes the progress achieved in enzymatic biodiesel research and commercialization, including reducing enzyme cost, expanding low-quality raw materials, and novel reactor designs. The advantages and disadvantages of different enzymatic processes are also compared.

Abstract:The characteristic of Rhodotorula gutini As2.107 for producing lipid was studied. The biomass was 11 g/L and the lipid concentration reached 4.5 g/L with the optimal fermentation medium containing glucose 5%, (NH4)2SO40.25%, MgSO40.2%, CaCl20.01%, FeCl30.007%. The optimal fermentation temperature, rotation speed and inoculation size were 30°C, 180 rpm and 5% respectively. The acid value, saponification value and the average molecular weight were analyzed as 26.31 mgKOH/g oil, 231.86 mgKOH/g oil and 818.78. The components of the microbial lipid were palmitic acid 17.8%, steric acid 10.27%, oleic acid 53.02%, linoleic acid 5.29%, which was similar with that of vegetable oil. Based on the above results, biodiesel production with the microbial lipid of Rhodotorula gutini As2.107 was investigated. The results indicated that the maximum methyl esters (ME) yield reached 93% by using alkali as the catalyst, with the molar ratio of methanol to oil 20:1, 1.5% KOH and reaction temperature 60°C, after the high acid value of the microbial lipid was first lowed.
Comparative study on lipase-catalyzed transformation of soybean oil for biodiesel production with different acyl acceptors

Abstract:Methyl acetate, a novel acyl acceptor for biodiesel production has been developed, and a comparative study on Novozym 435-catalyzed transesterification of soybean oil for biodiesel production with different acyl acceptors was conducted and reported in this paper. Methanol has a serious negative effect on enzymatic activity. A molar ratio of methanol to oil of above 1:1 leads to serious inactivation of the enzyme. However, when methyl acetate was used as the acyl acceptor, a yield of 92% of methyl ester could be obtained with a molar ratio of methyl acetate to oil of 12:1, and methyl acetate showed no negative effect on enzymatic activity. Additionally, with crude soybean oil as the oil source and methanol as acyl acceptor, a much lower methyl ester yield was obtained than that with refined soybean oil, while with methyl acetate as acyl acceptor, an equally high yield of methyl ester (92%) was achieved for both soybean oils. Lipase loses its activity very rapidly during repeated experiments with methanol as the acyl acceptor, while there is almost no detected loss in lipase activity, even after being continuously used for 100 batches, when methyl acetate was used for biodiesel production. Moreover, the by-product triacetylglycerol is an important chemical with a higher value than glycerol, and this novel acyl acceptor seems very promising for lipase-catalyzed large-scale production of biodiesel.
Production of Biodiesel Catalyzed by a Novel Lipase LipB52 from Pseudomonas fluorescens

Abstract:A novel lipase gene UpB52 from Pseudomonas fluorescens was overexpressed in E. coli BL21 (DE3) and the lipase LipB52 was purified through a Ni-NTA chelated Sepharose column. Immobilized lipase LipB52 was used to catalyze the transesterification of soybean oil and methanol to produce biodiesel. The effects of temperature, substrate molar ratio, lipase dosage, organic solvent, different enzymes and oil type on transesterification were investigated and the optimal conditions of methanolysis were as follows: 20% LipB52 based on oil weight; oil/methanol molar ratio 1:4; n-heptane as solvent and 30°C. Under the optimal conditions lipase LipB52 exhibited high catalysis activity and methyl ester yield of transesterification was 92% at 40 h.

Abstract:Recently, lipase processing for biodiesel production has shown a global increase as it is considered a potential alternative clean-fuel source. The current study's objective is to investigate of lipolytic activity of lipase produced from different strains of Pseudomonas aeruginosa ( P. aeruginosa ) in biodiesel production using edible plant oils. The goal is to develop an efficient and cost-effective method for producing inexpensive and environmentally friendly biodiesel.

Abstract:Biodiesel as an alternative to fossil diesel has attracted great attention for its environmental friendliness. In China, the raw feedstrocks for biodiesel production are mainly low-quality oils which contain a certain amount of phospholipids in addition to triglycerides and free fatty acids. Studies have shown that phospholipids present complex effects on both chemical-mediated and lipase-mediated biodiesel production, which further affect the final yield and quality of biodiesel. This review summarizes species, structures and characteristics of phospholipids in oils, and their effects on biodiesel quality are further discussed. Finally, the effects of phospholipids on biodiesel production with chemical catalyst or lipase are reviewed, and possible research directions are prospected.
Lipase nanogel for biodiesel production

Abstract:The production of biodiesel from plant oil can be efficiently catalyzed by lipase in a water-containing system without an organic solvent. However, this process is hindered by methanol induced deactivation of lipase. Lipase nanogel was prepared and characterized. The half-life time of native lipase was 50 min at 50°C while almost no visible loss of lipase activity was observed in the 500 min for the nanogel. After incubation in 40% methanol aqueous solution, lipase nanogel preserved about 80% of its hydrolytic activity while native lipase reserved only 10%. All these are of great interest for further exploration of the potential application of lipase nanogel for the production of biodiesel.
