NF-κB mediates ketamine-induced loss of phenotype in parvalbumin-containing interneurons

Abstract:Objective The loss of phenotype of parvalbumin(PV)-positive interneurons play an important role in various psychiatric alterations.This study is to observe whether nuclear factor-kappa (NF-κB) is in involved in ketamine-induced loss of phenotype in PV-containing interreurons in primary neuronal cultures in vitro.Methods 0.5 μmol/L ketamine (ketamine group),0.5 μmol/L ketamine and 5 μmol/L NF-κB translocation inhibitor SN50 (SN50 group) or neuron culture media (control group) was applied to primary neuron cultures at development in vitro (DIV) 21 d.Twenty four hours later,NF-κB activity,reactive oxygen species (ROS) production,PV and glutamic acid decarboxylase-67 (GAD67) expression were measured.Results NF-κB activity,ROS production,PV and GAD67 expression in ketamine group were 1.5-folder,2.1-folder,67.6%,and 63.0% of those in control group,respectively (P<0.05).Compared with control group,SN50 group had no statistically significant changes in NF-κB activity,ROS production,PV and GAD67 expression (P>0.05).Conclusion In primary neuron cultures,activation of NF-κB mediates ketamine-induced decrease of PV and GAD67 in PV-containing interneurons.
