A New Route to 3-Amino Sugars. A Concise Synthesis of L-Daunosamine and D-Ristosamine Derivatives

Abstract:An asymmetric aldol strategy has been developed for the synthesis of L-daunosamine and D-ristosamine derivatives starting from noncarbohydrate precursors. Lithium and boron enolate mediated aldol reactions of 12 with O-TBS lactaldehyde gave non-Evans syn and Evans syn aldol products, respectively, with high selectivity. The chemical efficiency of the lithium enolate reactions were higher than the corresponding reactions with the boron enolates. Curtius rearrangement of lactone acids 23 and 26 gave the corresponding N-BOC amino lactones 30 and 32 in 64% and 62%, respectively, with complete retention of configuration. Lactone 30 was converted by a two-step sequence to N-benzoyldaunosamide 40. The overall yield for the amino sugar 40 was 18% over six steps. Similarly, lactone 32 was converted to N-benzoylristosamide 42 with an overall yield of 18% starting from 12.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:Our studies of the TIBAL-promoted Claisen rearrangement reaction and ring-closing metathesis (RCM) resulted in the development of an efficient synthetic route to polyfunctional seven-membered carbasugar synthons from d-arabinose. Moreover, the construction of 8-oxa-bicyclo[3.2.1]octane derivatives 10 and 13 was achieved by BCl3 or iodide-promoted intramolecular electrophilic addition reactions, which were regio- and stereoselective.

Abstract:OBJECTIVE To find a commercially useful route for the preparation of 1-(2′-bromo-2′-deoxy-3′,5′-di-O-propionyl-β-D-ribosyl)thymine. METHOD To uses thymine and tetra-O-acylribofuranose as starting materials. The process involved the following steps: silylanization, condensation, deacetylation and halogenation. RESULTS The procedure was simple and the yield of this route was higher than other synthetic routes by using cheaper reagents.CONCLUSION The improved synthetic process is practical.

Abstract:A concise synthesis of 3-deoxy-3-nucleobase-2,5-anhydro-d-mannitol 6(a–d) has been achieved, using the deamination of 2-amino-2-deoxy-d-glucose to construct in one step the sugar skeleton with the desired sense of chirality at each asymmetric center. The selective dibenzoylation of 2,5-anhydro-d-mannitol 9 was investigated, and the key epoxide intermediate 13 was obtained in good yield via an intramolecular Mitsunobu reaction. The process of opening of epoxide 13 by nucleobases appeared to be regioselective.

Abstract:An approach to the regio- and stereo-selective construction of 2-deoxy-C-glycosides via Lewis acid-mediated rearrangement of 2,3-anhydro-1-thiopyranosides is disclosed. Treatment of 2,3-anhydro-1-thiopyranosides with phenols in the presence of TMSOTf, the migration-O-glycosylation and Fries-like O to C rearrangement took place in succession, providing aryl 2-thio-2-deoxy-C-glycosides with the single thermodynamically favourable configuration (C-1(4) or C-4(1)) in good to excellent yields. The coupling reaction of 2,3-anhydro-1-thiopyranosides with trimethylsilylated or tributylstanylated nucleophiles in the presence of TMSOTf or Sc(OTf)(3), via the migration-C-glycosylation process, afforded 2-thio-2-deoxy-C-glycosides in a stereospecific manner in moderate to good yields and with the C-1 and C-2 substituents opposite. The 2-thio-functionality was further removed to produce the corresponding 2-deoxy-C-glycosides. This method may provide a convenient route for the preparation of 2-deoxy-C-glycosides.

Abstract:Chiral aryl glycines are biologically relevant compounds that received a great deal of attention over the last years. They can be prepared by aminohydroxylation (K. L. Reddy, K. B. Sharpless J. Am. Chem. Soc. 1998, 120, 1207-1217), amination of enolates (D. A. Evans, S. G. Nelson J. Am. Chem. Soc. 1997, 119, 6452-6453) or reductive amination (R. Kadyrov, T. H. Riermeier, U. Dingerdissen, V. Tararov, A. Börner J. Org. Chem. 2003, 68, 4067-4070) but in some cases low regioselectivity, high catalyst loading or poor reactivity is observed. The authors applied their recently developed TangPhos ligand (Angew. Chem. Int. Ed. 2002, 41, 1612-1614) to the enantioselective hydrogenation of a variety of α-imino esters.

Abstract:A versatile and concise synthesis of N-alkylated 1,4-dideoxy-1,4-imino-d-arabinitol and 1,4-dideoxy-1,4-imino-l-xylitol derivatives is described. These were prepared using pseudohemiketal lactams as key intermediates, which in turn were obtained from sucrose. The key intermediates were prepared by a diastereospecific tandem reaction which facilitated the introduction of various substituents on the nitrogen atom of the iminosugars.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:OBJECTIVE To study the synthetic method of 2-aminoxy-1- aminoethane dihydrochloride. METHOD The objective compound was synthesized by steps of ring-opening, condensation, hydrazinolysis and hydrolysis, starting from phthalic anhydride.RESULTS The chemical structure of synthetic product was confirmed by IR and 1HNMR, the total yield was amount to 39.4%. CONCLUSION This synthetic route is practical.

Abstract:2,5:3,4-Dianhydro-L-talofuranose dimethylacetal 7 was synthesized from D-glucose in 7 steps. A series of 4-deoxy-4-nucleobase-2,5-anhydro-L-mannitols 13-16 were synthesized regioselectively from 7 in good yields. 6-O-p-Tolylsulfonyl-2,5:3,4-dianhydro-L-talofuranose dimethylacetal 8 reacted with uracil or thymine to give the corresponding isonucleosides 17 and 19, but in the case of reaction of 8 with adenine, reformation of tetrahydrofuran ring took place, giving 4-(S)-adenyl-5-(R)-[1'-(R)-hydroxy-2',2'-dimethoxyl] ethyl-2,3-dihydrofuran 21 and 4-deoxy-4-adenyl-2,5:3,6-dianhydro-L-mannofuranose dimethylacetal 22. Abstract:In order to optimize the synthesis and separation of anomers of 1-(diethyl)-2,5-anhydro-D-ribofuranosyl phosphonate,they were synthesized by Wadsworth-Emmons reactions in which two different protected D-ribose derivatives were used as the starting materials.In method A,2,3-O-isopropylidene-5-O-pivaloyl-D-ribofuranose reacted with tetraethyl methylenediphosphonate(TEMDP) to give 1-(diethyl)-2,3-O-isopropylidene-5-O-pivaloyl-D-ribofuranosyl phosphonate.Sodium ethoxide was used to remove the pivaloyl group in 81% yield.The α and β anomers were isolated and then hydrolyzed under acidic condition to afford the desired products in 94% and 91% yields,respectively.In method B,the D-ribose 1-phosphonates were obtained through the reaction of 2,3,5-tri-O-benzyl-D-ribose with TEMDP in 70% yield.After separating the anomers,the desired products were obtained through deprotection of α and β anomers with Pd/C and H_2 in 93% and 91% yields,respectively.The products were characterized by ~1HNMR to verify the feasibility of our synthesis.

Abstract:2-Deoxy-4'-thio-D-sugars are potential thiosugar donors for glycosylation reactions with nucleobases to produce various D-deoxy-4'-thionucleosides. However, thio-inversion reactions that produce D-deoxy-4'-thio-D-sugars depend on using a starting material, L-deoxysugar, which is expensive. Herein, we report a unique synthetic methodology to produce 2-deoxy-4'-thio-D-sugars using a D-sugar as an inexpensive starting material. Following our synthetic procedures for carbonyl translocation, D-ribose was transformed to 2-deoxy-L-ribosyl dithioacetal as a key intermediate. Mesylation and triflation provide the corresponding dialkylthio acetals as key precursors. Under thio-inversion reaction with iodide reagents, D-2-deoxy-4'-thioxyloside was successfully produced for the first time.

Abstract:Reaction of tartaric acid with thiocarbohydrazide (2) and thiosemicarbazide (6) afforded 1,2-bis(4-amino-5-mercapto-4 H -1,2,4-triazol-3-yl)-ethane-1,2-diol (3) and 1,2-bis(5-mercapto-4 H -1,2,4-triazol-3-yl)-ethane-1,2-diol (7) . Reaction of compounds 3 and 7 with DMAD (dimethylacety lendi carboxylate) and DEAD (diethylacetylendicarboxylate) gave 1,2-bis(7-[( z )-methoxycarbonylmethylen]-5,6-dihydro-5 H -6-one-[1,2,4] riazolo[3,4-b] [1,3,4] thiadiazin-3-yl)-ethan-1,2-diol (4) , 1,2-bis(7-[( z )-ethoxycarbonylmethylen] -5,6-dihydro -5 H -6-one-[1,2,4]triazolo[3,4-b][1,3,4]thiadiazin-3-yl)-ethan-1,2- diol (5) and 1,2-bis(6-[( z )-methoxycarbonylmethylen]-5-oxo-[1,3]thiazolo[2,3-c] [1,2,4]triazol-3-yl)-ethan-1,2-diol (8) in good yields.

Abstract:A highly efficient, solid-phase synthesis of 2-arylamino-5-(4-hydroxy-phenyl)-1,3,4-thiadiazole derivatives under mild conditions has been developed. The 1,3,4-thiadiazole derivatives were synthesized from resin-bound acylhydrazines in several steps, which gave 78-88% overall yields and excellent purities of the products.

Abstract:A convenient,high-yield synthesis of 1-O-acetyl-3,5-di-o-(p-chlorobenzoyl)-2-deoxy-α,β-D-ribofuranoside from 2-deoxy-D-ribose was reported.The product was a very important medical intermediate.Its anomers were separated by column chromatography.The optimum conditions were as follows: methyl-3,5-di-o-(p-chlorobenzoyl)-2-deoxy-α,β-D-ribofuranoside:acetic anhydride,1∶4.4;2 mL conc.sulfuric acid;temperature of solution when adding conc.sulfuric acid,(-2°C);hydrolysis temperature,(6°C).The yield was above 84.14%.The separated α and β anomers were characterized by()~1H NMR and melting point.
