Abstract:OBJECTIVE: To evaluate the relationship between autoantibodies against oxidized low density lipoprotein(ox- LDL), immune complex and coronary artery disease(CAD). METHODS: The levels of autoantibodies against ox-LDL and immune complex were assayed by ELISA, the content of ox-LDL and lipid levels were also measured. The serum samples were taken from 61 patients with CAD, 116 patients with essential hypertension(EH) and 123 healthy individuals as control. RESULTS: The patients with CAD had significantly higher levels of anti-ox-LDL IgG[21.48(17.58 approximate, equals 29.01)U/L], anti-ox-LDL IgM4.71[3.88 approximate, equals 7.06)U/L] and ox-LDL[0.87(0.44 approximate, equals 1.08) mg/L] than EH group[15.93(11.12 approximate, equals 22.26) U/L 2.54(1.17 approximate, equals 5.05) U/L 0.32(0.16 approximate, equals 0.61) mg/L]and healthy control group[11.12(4.70 approximate, equals 16.57)U/L 1.61(0.60 approximate, equals 3.03)U/L 0.23(0.12 approximate, equals 0.36)mg/L], P<0.001. However, the serum ox-LDL immune complex[2.63(1.69 approximate, equals 5.90)U/L] was significantly lower in CAD than that in EH group[15.71(6.25 approximate, equals 28.74)U/L] and that in healthy control group[12.54(8.28 approximate, equals 23.90)U/L], P<0.001. There were discrepancies in the association between ox-LDL and autoantibodies against ox-LDL among different groups. CONCLUSION: The changes of autoantibodies against ox-LDL and immune complex in patients with CAD may be related to the role of ox-LDL and autoantibodies against ox-LDL in the process of coronary atherosclerosis.

Conclusion These results partially show that the relation of chronic inflammation, autoimmunity and CAD. oxLDL-Ab and hs-CRP respectively maybe play an important role in the development of atherosclerosis.

Abstract:Objective Dynamic observation of oxidized low density lipoprotein(Ox-LDL) and thrombus precursor protein(TpP) expressed in patients with acute cerebral infarction ,and explore its clinical significance .Methods To detect the Ox-LDL and the plasma TpP level of 44 patients with acute cerebral infarction (ACI)respectively in acute stage and recovery stage ,and the healthy 30 cases (control group)by using enzyme-linked immunosorbent assay (ELISA) method ,and all the ACI cases were scored by the national institutes of health stroke scale (NIHSS) .To analyze the expression of Ox-LDL and plasma TpP in patients with ACI ,and the relationship with the NIHSS score .Results Ox-LDL and plasma TpP in patients with ACI all increased compared with the con-trol group .Ox-LDL and TpP in acute stage were significantly higher than the corresponding recovery stage and the control group , the difference was statistically significant (P<0 .01) .The levels of plasma TpP were positively correlated with the NIHSS score (P<0 .01) ,and Ox-LDL levels were uncorrelated with the NIHSS score (P>0 .05) .The levels of Ox-LDL were positively correla-ted with TpP in ACI group (r=0 .521 ,P<0 .05) .Conclusion The expression of Ox-LDL and plasma TpP in ACI were increased , Ox-LDL and plasma TpP level in ACI varies with changes of clinical course ,and maybe involved in the coordination and the devel-opment process of ACI ;Plasma TpP may be relevant to the seriousness of cerebral infarction ,and may be arguably used as the measurement of ACI patient′s conditions as well as the prognosis estimation .

Abstract:OBJECTIVE:To assess the value of serum soluble lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) levels and LOX-1 mRNA expression in peripheral blood mononuclear cells in early diagnosis of acute coronary syndrome (ACS).METHODS:Enzyme-linked immunosorbent assay was used to detect the levels of plasma ox-LDL and LOX-1 in 95 patients with ACS, 60 with stable angina pectoris (SAP) and 40 normal control subjects. The expression of LOX-1 mRNA in peripheral blood mononuclear cells was detected by RT-PCR in the 3 groups.RESULTS:The levels of ox-LDL, LOX-1 and LOX-1 mRNA in the peripheral blood mononuclear cells were significantly higher in ACS patients than in SAP patients and normal control subjects (P<0.05). In ACS group, the level of plasma ox-LDL was significantly correlated to serum LOX-1 and LOX-1 mRNA expression in peripheral mononuclear cells.CONCLUSION:The level of plasma soluble LOX-1 and LOX-1 mRNA in peripheral mononuclear cells are significantly increased in ACS, and when combined, they provide a useful means for detecting ACS in the prophase.
The Correlation between Lipids Ratio and Degree of Coronary Artery Stenosis

Abstract:Objective: The aim of the study was to explore the relationship between lipids ratio and the degree of coronary artery stenosis. Methods: According to the Gensini score of inpatients in the Cardiology Department of the Second Affiliated Hospital, Zhejiang University College of Medicine, Hangzhou, China, from January 2008 to June 2010, a total of 207 patients (group 1: control group = 34 patients; group 2: 1–30 score group = 84 patients; group 3: 31–90 score group = 66 patients; and group 4: >90 score group = 23 patients) were enrolled, and the relationship between lipids and the degree of coronary artery stenosis was determined. Results: Our study showed that the severity of coronary lesions was increased with the elevation of low-density lipoprotein cholesterol (LDL-C)/high-density lipoprotein cholesterol (HDL-C), total cholesterol (TC), LDL-C, triglycerides (TG), TC/HDL-C, TG/HDL-C and reduction of HDL-C. Compared with group 1, LDL-C/HDL-C, TC/HDL-C were significantly increased in groups 2, 3 and 4 (p < 0.05), and compared with group 2, LDL-C/HDL-C, TC/HDL-C in groups 3 and 4 were significantly increased (p < 0.05). There were no differences in the value of TG, TG/HDL-C among groups 2, 3 and 4 (p > 0.05). Furthermore, the Pearson correlation analysis revealed that LDL-C/HDL-C (r = 0.54, p < 0.05) and TC/HDL-C (r = 0.50, p < 0.05) were significantly and positively correlated with the coronary artery lesions. Conclusions: The results suggested that the severity of coronary artery lesions were correlated with abnormal lipid metabolism, and LDL-C/HDL-C, TC/HDL-C are better indicators than other measures. However, the predictive value of TG/HDL-C needs further study.
Comparison of low-density lipoprotein cholesterol/high-density lipoprotein cholesterol and total cholesterol/high-density lipoprotein cholesterol for the prediction of thin-cap fibroatheroma determined by intravascular optical coherence tomography

Abstract:Background The correlation among the ratios of low-density lipoprotein cholesterol/high-density lipoprotein cholesterol (LDL-C/HDL-C), total cholesterol/high-density lipoprotein cholesterol (TC/HDL-C) and thin-cap fibroatheroma has not yet been established. Methods It was a single center, retrospective observational study. In total, we recruited 421 patients (82.4% men; mean age 65.73 +/- 10.44 years) with one culprit vessel which determined by intravascular optical coherence tomography (OCT). The thinnest-capped fibroatheroma (TCFA) group was defined as lipid contents in > 2 quadrants, with the thinnest fibrous cap measuring less than 65 mu m. Univariate and multivariate logistic regression were carried out to explore the relationship between lipoprotein ratios, TCFA and other characteristics of plaque. To compare different ratios, the area under curve (AUC) of receiver-operating characteristic (ROC) curve was assessed. Results OCT was performed in 421 patients (TCFA group (n = 109), non-TCFA group (n = 312)). LDL-C/HDL-C in the TCFA group was significantly higher than in the non-TCFA group (2.95 +/- 1.20 vs. 2.43 +/- 0.92, P < 0.05), as was TC/LDL in TCFA and non-TCFA group (4.57 +/- 1.58 vs. 4.04 +/- 1.13, P < 0.05). Both LDL-C/HDL-C (OR: 1.002 (1.002-1.003), P < 0.05) and TC/HDL-C (OR: 1.001 (1.001-1.004), P < 0.05) were considered independent factors for the prediction of TCFA according to the logistic regression. Based on the AUC comparison, LDL-C/ HDL-C and TC/HDL-C had no significant difference statistically (LDL-C/HDL-C AUC: 0.63; TC/HDL-C AUC: 0.61; P = 0.10) for the prediction of TCFA. Conclusions LDL-C/HDL-C and TC/HDL-C could be the independent factors for predicting the presence of TCFA, indicating coronary plaque vulnerability in CAD patients. Moreover, TC/HDL-C also showed a comparative performance for the prediction of TCFA as LDL-C/HDL-C.
Association Between Non-High-density Lipoprotein Cholesterol and Coronary Artery Disease in Patients with Acute Coronary Syndrome: a Single-Centre Retrospective Cross-Sectional Analysis

Abstract:Background A significant relationship exists between atherosclerotic cardiovascular disease (ASCVD) and non-high-density lipoprotein cholesterol (non-HDL-C). However, the relationship between non-HDL cholesterol levels and coronary artery disease in patients with acute coronary syndrome (ACS) remains under-researched and poorly understood. Therefore, this study aimed to explore the association between non-HDL-C and coronary artery disease in individuals with ACS. Methods This single-centre retrospective cross-sectional study included 1695 patients with ACS who underwent percutaneous coronary intervention at Zunyi Medical University's Affiliated Hospital from May 2019 to May 2023. The patients were categorised into three groups based on non-HDL cholesterol tertiles. The severity of coronary artery disease was assessed using the Gensini score, and patients were categorised into high and low Gensini score groups. Logistic regression models, including multifactorial adjustments, were used to analyse the association between non-HDL-C levels and high Gensini scores. Results After adjusting for potential risk factors, multifactorial logistic regression (odds ratio [OR] = 1.20; 95% confidence interval [CI] = 1.10–1.31; p < 0.001) revealed that non-HDL-C was an independent risk factor for high Gensini scores. After subgrouping based on age, sex, and glucose metabolic status, non-HDL-C remained an independent risk factor for high Gensini scores in these subgroups. Restricted cubic spline analysis revealed that a higher non-HDL-C level was associated with a greater the risk of having a high Gensini score. Receiver operating characteristic curve analysis revealed that non-HDL-C was incrementally predictive of high Gensini scores in patients with ACS. Conclusions Non-HDL-C is dose-dependently associated with the severity of coronary artery disease in patients with ACS. These findings suggest the potential of non-HDL-C in predicting its severity, supporting its use as a cost-effective and accessible marker for assessing coronary artery disease risk.
Relationship Between Oral Infections and Coronary Heart Disease

Abstract:Objective To investigate the correlation of oral infections and coronary heart disease(CAD). Methods Fifty nine patients with CAD and fifty one with no CAD were compared with their traditional cardiovascular risk factors, oral health status and the amount of Streptococcus Sanguis(SS) in saliva and subgingival plaque(SSs and SSp). The natural logarithm transformation of SSs and SSp [LN(SSs+1), LN(SSp+1)] were used to analyse due to their skew distribution. The related risk factors of CAD were included in a stepwise logistic regression analysis. Results Univariate analysis showed that there was significant difference in plasma total cholesterol(TC), lower density lipoprotein cholesterol(LDL-C), the number of tooth in gingivitis, the number of tooth in peridontitis, LN(SSp+1)between patients with CAD and those with no CAD(P0.05～0.01). Logistic regression analysis showed that LN(SSp+1) and LDL-C were strongly correlated with incidence of CAD. The relative risk were 1.438(95%CI was 1.127～1.834)and 2.271(95%CI was 1.249～4.129) respectively. Conclusion The amount of oral SS may be one of the predictors for CAD.
Limited Association between Antibodies to Oxidized Low-Density Lipoprotein and Vascular Affection in Patients with Established Systemic Lupus Erythematosus

Abstract:Patients with systemic lupus erythematosus (SLE) are at an increased risk of cardiovascular disease. We aimed to evaluate whether antibodies to oxidized low-density lipoprotein (anti-oxLDL) were associated with subclinical atherosclerosis in patients with different SLE phenotypes (lupus nephritis, antiphospholipid syndrome, and skin and joint involvement). Anti-oxLDL was measured by enzyme-linked immunosorbent assay in 60 patients with SLE, 60 healthy controls (HCs) and 30 subjects with anti-neutrophil cytoplasmic antibody-associated vasculitis (AAV). Intima-media thickness (IMT) assessment of vessel walls and plaque occurrence were recorded using high-frequency ultrasound. In the SLE cohort, anti-oxLDL was again assessed in 57 of the 60 individuals approximately 3 years later. The levels of anti-oxLDL in the SLE group (median 5829 U/mL) were not significantly different from those in the HCs group (median 4568 U/mL), while patients with AAV showed significantly higher levels (median 7817 U/mL). The levels did not differ between the SLE subgroups. A significant correlation was found with IMT in the common femoral artery in the SLE cohort, but no association with plaque occurrence was observed. The levels of anti-oxLDL antibodies in the SLE group were significantly higher at inclusion compared to 3 years later (median 5707 versus 1503 U/mL, p < 0.0001). Overall, we found no convincing support for strong associations between vascular affection and anti-oxLDL antibodies in SLE.

Effects of Oxidative Low-Density Lipoprotein on Number and Function of Endothelial Progenitor Cells from Peripheral Blood

Abstract:Objective To observe the effects of oxidative low density lipoprotein (ox LDL) on number and function of endothelial progenitor cells (EPCs) from peripheral blood. Methods Total mononuclear cells (MNC) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin coated culture dishes. After incubation for 7 days, attached cells were collected and stimulated with different cencentrations of ox LDL (25, 50, 100 and 200 μg/ml), native LDL (100 μg/ml) or vehicle control for different time (6, 12, 24 and 48 h). EPC were characterized and adherent cells which were double positive for DiLDL uptake and lectin binding by direct fluorescent staining were observed under a laser scanning confocal microscope. EPC were further identified by demonstrating the expression of CD34, VEGFR 2 and AC133 with flow cytometry. Proliferation, migration and in vitro vasculogenesis activity of EPC were assayed by MTT assay, modified Boyden chamber assay and in vitro vasculogenesis kit, respectively. EPC adhesion assay was performed by replating those on fibronectin coated dishes, then adherent cells were counted. Results Incubation of isolated human MNC with ox LDL decreased the number of EPC in a concentration dependent manner, being maximum at 200 μg/ml (approximately 70% reduction, P0.01). In time course experiments performed with a concentration of 100 μg/ml ox LDL, the decrease of EPC number became apparent at 12 h and the maximal decrease was at 24 h (approximately 50% reduction, P0.01). In addition, ox LDL impaired proliferative, migratory, adhesive and in vitro vasculogenesis capacities of EPC in dose and time dependent manners. Conclusion Incubation of EPC with ox LDL reduces the number of EPC and impaires their function.

Abstract:OBJECTIVES We investigated whether associations exist between immune reactions to oxidized lowdensity lipoproteins (OxLDLs), chronic infections, and carotid atherosclerosis as quantified by ultrasound. BACKGROUND Atherosclerosis is a chronic immuno-inflammatory disease wherein both oxidized lipids and infectious agents are incriminated as possible contributors. METHODS We measured immunoglobulin (Ig)G and IgM autoantibody titers to copper-oxidized-LDL and malondialdehyde-LDL (OxLDL-AB), IgG and IgM apolipoprotein B-100-immune complexes (ApoB-IC), and titers of antibodies to Escherichia coli and chlamydial lipopolysaccharide (LPS), mycobacterial heat shock protein 65 (mHSP65), Chlamydia pneumoniae, Helicobacter pylori, and cytomegalovirus and evaluated their relationship to cardiovascular risk factors, chronic infections, and incident/progressive carotid atherosclerosis in the Bruneck study. RESULTS The OxLDL-AB and ApoB-IC levels remained stable over time as indicated by strong correlations between 1995 and 2000 measurements (p 0.001 each). Significant associations existed between all OxLDL markers and antibody titers to pathogens, especially to E. coli-LPS and mHSP65. Both OxLDL-AB and ApoB-IC levels showed a rise with increasing pathogen burden. Notably, OxLDL-ABs were also elevated in subjects with chronic infection as defined by clinical criteria. Titers of IgG, but not IgM, OxLDL-AB, or ApoB-IC inversely correlated with total cholesterol, LDL cholesterol, and apoB concentrations. The IgG OxLDL markers were positively and IgM markers were inversely associated with incident and progressive carotid atherosclerosis in univariate analyses but were not independent predictors in multivariate analyses. CONCLUSIONS Our study provides evidence for an association between human oxLDL markers and chronic infections. Abstract:Objective To explore the association of antibody against oxidized low density lipoprotein with atherosclerotic coronary artery disease.Methods Serum anti Ox LDL was measured by enzyme linked immunosorbent assay in 76 patients who had divided into 3 groups,stable effort angina,unstable angina and control.Results The anti Ox LDL titer was higher in patients with unstable angina(491.19±307.01 mU/ml)than in patients with stable effort angina or in the control;However,no significant difference was shown between the stable effort angina group(242.18±95.91 mU/ml)and the control(304.57±170.96 mU/ml).The titer was higher in patients with multi vessel CAD(453.19±285.93 mU/ml)than in patients with single vessel CAD or in the control;However,no significant difference was shown between the single vessel CAD group(283.71±197.36 mU/ml)and the control(304.57±170.96 mU/ml).Conclusion Serum anti Ox LDL titer was not only can predict a presence of atherosclerotic CAD,but also may be a marker of plaque instability.

Abstract:Background: Oxidized lipoprotein(a) [ox-Lp(a)] and oxidized low density lipoprotein (ox-LDL) levels have been reported to be useful predictors of cardiovascular events. The study developed a chemiluminescence (CL) imaging immunoassay method for simultaneously detecting serum concentrations of ox-Lp(a) and ox-LDL. Methods: Ox-Lp(a) and ox-LDL levels were measured by CL imaging immunoassay using a disposable immunosensor array as the carrier and a charge-coupled device as the detector, and were studied in 46 acute coronary syndromes (ACS) patients, 58 stable coronary artery disease (CAD) and 61 control subjects. Results: This method showed good linear relations (R-2>0.99) in the concentration range of 2.00x10(-5)-2.00x10 (1) and 2.40x10 (4)-2.40 U/mL for ox-Lp(a) and ox-LDL, respectively. The detection limits for ox-Lp(a) and ox-LDL were 2.40x10(-6) and 3.00x10(-5) U/mL, respectively. The intra- and inter-assay coefficients of variation (CV) were 4.90%-6.76% and 7.11%-10.06% for ox-Lp(a), and 5.01%-6.04% and 5.47%-9.77% for ox-LDL, respectively. The mean recovery was 99.31% for ox-Lp(a) and 99.57% for ox-LDL, respectively. Significant correlations were observed between ox-Lp(a) levels detected by CL imaging immunoassay and ELISA, and between ox-LDL levels detected by the two methods, respectively. Furthermore, ox-Lp(a) and ox-LDL levels increased in stable CAD, and especially in ACS. Conclusions: The CL imaging immunoassay provided a simple, sensitive and reliable method for the simultaneous determination of serum ox-Lp(a) and ox-LDL. The clinical monitoring ox-Lp(a) and ox-LDL levels may possess distinctly clinical value for assessment of CAD risk.

Abstract:This study aimed to study the value of oxidized low-density lipoprotein (OX-LDL) and related ratio in the diagnosis of acute cerebral infarction and the classification of acute cerebral infarction.Of the 129 patients enrolled in this study, 94 patients with acute cerebral infarction were assigned to the case group, and 35 healthy subjects were enrolled as control group (n = 35). And then the case group were divided into large-artery atherosclerosis (LAA) group (n = 61) and small-artery occlusion (SAO) group (n = 33) according to the TOAST classification standard. Plasma OX-LDL levels were determined by enzyme-linked immunosorbent assay. OX-LDL/total cholesterol (OX-LDL/TC), OX-LDL/high-density lipoprotein (OX-LDL/HDL), OX-LDL/LDL were calculated.There were significant differences in OX-LDL, OX-LDL/TC, OX-LDL/HDL, and OX-LDL/LDL in patients with acute cerebral infarction and those in control group (P < .001). The area under the receiver-operating characteristic curve of OX-LDL and related ratio was >0.7 (P < .001). There was a slight positive correlation between OX-LDL/TC and National Institutes of Health Stroke Scale score at admission (r = 0.265, P = .039) in the LAA group.OX-LDL, OX-LDL/TC, OX-LDL/HDL, and OX-LDL/LDL were closely related to acute cerebral infarction, especially with large atherosclerotic cerebral infarction. OX-LDL/TC can reflect the severity of acute cerebral infarction for LAA, but it cannot predict the short-term prognosis of acute cerebral infarction.

Abstract:Toll-like receptor 4 (TLR4) may provide a potential pathophysiological link between lipids and infection/inflammation and atherosclerosis. Oxidized low-density lipoprotein (ox-LDL) makes it more atherogenic than its native form. The purpose of the study was to investigate the relationship between the effects of ox-LDL in human atherosclerosis and the expression of TLR4. We studied the relationship between TLR4 and ox-LDL, pro and con, using both real-time quantitative RT-PCR and RNA interference technology through in vitro cell culture. Nuclear factor kappa B (NF-κ B) activity and the concentrations of monocyte chemoattractant protein-1(MCP-1) and interleukin-8 (IL-8) were detected by ELISA. The results showed that the expression of TLR4 increased in response to ox-LDL. Simultaneously, NF-κ B relative activity and the concentrations of MCP-1 and IL-8 in cell supernatant were upregulated by ox-LDL in a dose-dependent manner. TLR4 expression was inhibited by small interference RNA(siRNA) plasmid expression vectors; NF-κ B activity and the secretions of MCP-1 and IL-8 in response to ox-LDL were significantly lower in the group whereinTLR4 expression has been inhibited than that in the group wherein TLR4 expression has not been inhibited. We suggest that the atherogenic effects of ox-LDL could be mediated in part via the TLR4 pathway. Furthermore, inhibition of TLR4 expression may downregulate the NF-κ B activity and secretions of MCP-1 and IL-8 in monocytes due to oxidized LDL, resulting in the alleviation of the progress of atherosclerosis.

Abstract:BACKGROUND:Oxidized Low-Density Lipoprotein (ox-LDL) is crucial in the recrudescence and prognosis of acute ischemic stroke (AIS). We aimed to probe into the influence of cumulative ox-LDL exposure on the 90-day prognosis of AIS.METHODS:Patients with AIS were recruited in this research. AIS severity at admission was estimated with infarct volumes and National Institute of Health Stroke Scale (NIHSS) scores. AIS prognosis was assessed using Modified Rankin Scale (mRS) scores at 90 days and the change in NIHSS scores from admission to discharge. Cumulative ox-LDL exposure was defined as ox-LDL level (pg/mL) multiplied by age(y). Multivariate logistic regression analysis was employed to reveal the correlation between exposure factors and the prognosis of AIS. The prognostic prediction ability of cumulative ox-LDL exposure was compared with cumulative LDL exposure by the receiver operating characteristic curve (ROC).RESULTS:Higher cumulative ox-LDL exposure was related to worse prognosis, including neurological worsening at discharge (NIHSS increasing more than 2 points) (OR = 3.02, 95% CI, 1.30-6.98, P = 0.01) and poor functional prognosis at 90 days (mRS ≥ 3) (OR = 21.21, 95% CI, 4.72-95.36, P < 0.001). As multivariate regression analysis showed, significantly increased cumulative ox-LDL exposure was relevant to poor functional prognosis at 90 days (OR = 9.92, 95% CI, 1.23-79.76, P = 0.031), but not with neurological worsening at discharge (P = 0.414). ROC curve revealed that cumulative ox-LDL exposure had a higher predictive value (AUC = 0.843, P < 0.001) for functional prognosis of AIS than cumulative LDL exposure (AUC = 0.629, P = 0.023).CONCLUSION:Cumulative ox-LDL exposure has a positive correlation with poor prognosis at 90 days of AIS, and has a more accurate predictive ability than cumulative LDL exposure.

Abstract:Objective To explore the changed mechanism of plasma oxidized low-density lipoprotein(ox-LDL)in the patients with coronary heart disease,and to evaluate the clinical value of detection for ox-LDL level.Methods Enzyme-linked immunosorbent assay(ELISA)was used to measure the plasma ox-LDL in 139 patients with coronary heart disease,among them 76 were acute coronary syndrome(ACS)and 63 were non-ACS patients,and in 100 healthy controls.The patients were divided by coronary angiography into multi-vessel disease group,double-vessel disease group and mono-vessel disease group.Results The ox-LDL levels of the patients in both ACS and non-ACS groups[(188.01±69.88)μg/ml and(137.88±59.74)μg/ml] were significantly higher than that in normal controls group[(82.68±29.06)μg/ml;P0.001],and the ox-LDL level of patients in ACS group was significantly higher than that in non-ACS group(P0.001).Different plasma levels of ox-LDL were found in the multi-vessel,double-vessel and mono-vessel disease group[(183.63±79.42)μg/ml,(164.15±63.79)μg/ml,and(146.97±58.40)μg/ml,respectively,P0.05].The ox-LDL levels in multi-vessel disease group were significantly higher than that in mono-vessel group(P0.01).Multiple linear regression analysis showed that ox-LDL levels significantly correlated with the severity of coronary artery disease(R2=0.100,β=0.316,P=0.000).Conclusion Elevated levels of ox-LDL presented in the patients with coronary heart disease and correlated with the severity of coronary artery lesion,especially in the patients with acute coronary syndrome.
[Clinical Significance of Oxidized Low-Density Lipoprotein Antibody in Antiphospholipid Syndrome].

Abstract:OBJECTIVE:To investigate the significance and distribution of oxidized low-density lipoprotein antibodies (ox-LDL-Ab) in patients with antiphospholipid syndrome (APS).METHODS:In this study, 334 patients who were hospitalized in the Department of Rheumatology and Immunology, Peking University People's Hospital were included. There were 162 APS patients, 122 patients with other autoimmune diseases without thrombosis or obstetric disease as disease control and 50 healthy controls. The clinical data and laboratory indicators were retrospectively collected. The ox-LDL-Ab, anticardiolipin (aCL) IgG/IgA/IgM, and anti-β2-glycoprotein Ⅰ (aβ2GPI) IgG/IgA/IgM were detected by enzyme-linked immunosorbent assay (ELISA). The relationship between ox-LDL-Ab and clinical and laboratory parameters were analyzed by SPSS 27.0.RESULTS:In APS group, 60.5% of patients had thrombosis, 48.1% had pregnancy morbidity, 34.0% had thrombocytopenia. The positive rates of aCL, aβ2GPI and lupus anticoagulant (LAC) were 17.9%, 34.6%, and 46.9%, respectively. The ox-LDL-Ab titers and positive rate in APS group were higher than that in healthy controls [titers: 40.8 (25.4-66.0) U/mL vs. 24.1 (12.3-36.5) U/mL, P=0.001; positive rate: 67.3% vs. 36.0%, P=0.001]. The diffe-rences in titers and positive rate of ox-LDL-Ab between APS patients and disease controls were not statistically significant [titers: 40.8 (25.4-66.0) U/mL vs. 35.9 (24.2-53.1) U/mL, P=0.118; positive rate: 67.3% vs. 61.5%, P=0.318]. The area under curve (AUC) for aβ2GPI, aCL, and ox-LDL-Ab were 0.745 (95%CI: 0.692-0.797), 0.666 (95%CI: 0.608-0.724), 0.609 (95%CI: 0.549-0.669), respectively. The Youden's index was 0.388, 0.269, and 0.132, respectively. The AUC for ox-LDL-Ab in seronegative APS patients was 0.562 (95%CI: 0.480-0.645). The sensitivity and specificity of ox-LDL-Ab in seronegative APS patients were 63.9% and 47.0%, respectively, and the Youden's index was 0.109. The ox-LDL-Ab positive group had higher positive rate of aβ2GPI (42.2% vs. 18.9%, P=0.003) and aCL (22.9% vs. 7.5%, P=0.017) than the ox-LDL-Ab negative group. There was no correlation between ox-LDL-Ab and thrombosis, coronary artery disease, pregnancy morbidity, hyperlipidemia, hypocomplementemia, and LAC positivity.CONCLUSION:Ox-LDL-Ab was correlated with aCL and aβ2GPI, and no association were observed between ox-LDL-Ab and thrombosis, coronary artery disease, and pregnancy morbidity.

Abstract:To investigate retinol-binding protein 4 (RBP4), small dense low-density lipoprotein cholesterol (sdLDL-C) and oxidized low-density lipoprotein (ox-LDL) levels and their associations in dyslipidemia subjects.We determined RBP4, sdLDL-C, ox-LDL levels in 150 various dyslipidemia subjects and 50 controls. The correlation analysis and multiple linear regression analysis were performed.The RBP4, sdLDL-C and ox-LDL levels were found increased in various dyslipidemia subjects. The sdLDL-C levels were positively correlated with RBP4 (r=0.273, P=0.001) and ox-LDL (r=0.273, P=0.001). RBP4 levels were also correlated with ox-LDL (r=0.167, P=0.043). The multiple regression analysis showed that only sdLDL-C was a significant independent predictor for RBP4 (β coefficient=0.219, P=0.009; adjusted R(2)=0.041) and ox-LDL (β coefficient=0.253, P=0.003; adjusted R(2)=0.057) levels, respectively.The independent associations of sdLDL-C with RBP4 and ox-LDL were observed in dyslipidemia subjects. RBP4 may play an important role in lipid metabolism of atherosclerosis, particularly in formation of sdLDL.
