Effect of Cigarette Smoking on Macrophage Cholesterol Efflux Mediated by Mice Serum and the Potential Mechanisms.

Abstract:Objective:To investigate the effect of cigarette smoking on cholesterol efflux mediated by mice serum and the possible mechanisms.Methods:14 C57BL/6 mice were randomly divided into control group and cigarette smoking group.After 8 weeks of passive smoking,the serum was collected for determining the concentrations of total cholesterol(TC),low density lipoprotein cholesterol (LDL-C),high density lipoprotein cholesterol(HDL-C),triglyceride(TG)and maleic dialdehyde(MDA).In addition,we determined the rate of cholesterol efflux mediated by serum from control group,serum from cigarette smoking group and serum from cigarette smoking group incubated with N-acetylcysteine(NAC,2mM)by liquid scintillation counting.Results:Compared with the control group,the level of serum HDL-C decreased significantly in the mice of smoking group,while the concentrations of serum TG and MDA increased,no significantly difference between TC and LDL-C;the rate of cholesterol efflux mediated by serum from cigarette smoking group was significantly decreased,which could be significantly increased by incubation with NAC.Conclusions:Cigarette smoking significantly decreased the rate of cholesterol efflux mediated by serum,which was possible related to the effects of cigarette smoking decreasing HDL-C concentration and increasing the lipid peroxidation of HDL.
