Abstract:N-phosphorylated amino acids without a side chain functional group can transfer themselves into peptides after prolonged standing in solvents at warm temperatures. Seven N-phosphorylpeptides and free peptides were isolated and their structures determined. The phosphoryl group participation is the key to the peptide formation. An intramolecular mixed carboxylic-phosphoric anhydride intermediate was proposed for this type of reaction which might provide a clue to the function of the phosphoryl group in the phosphorylated enzymes and in the prebiotic synthesis of protein.

Abstract:O-phosphorylated peptide and amino acid are important in living system. In this paper, Dialkyl-N,N-dialkyl phosphoramidites (DDPA) 3 were synthesized using two methods. The reaction of DDPA with the hydroxyl group of the corresponding amino acids in the presence of tetrazole, followed by oxidation, gave O-phosphoryl amino acid methyl esters in a good yield. A systematical study of the reactivity of DDPA was presented too.

Abstract:Frame-Guided Assembly (FGA) strategy has been recently reported to prepare vesicles with customized shapes and sizes. However, the effects of the interaction between leading hydrophobic groups (LHGs) and amphiphiles on the thermodynamic and kinetic control of the FGA process haven't been fully understood. In this work, we employed the pH low-insertion peptide (pHLIP) as the LHGs because its interaction with lipids could be finely tuned by pH and investigated the mechanism of FGA in detail. Our study demonstrated the peptide frames could successfully guide the assembly of lipids to form heteroliposomes below the pH transition point owing to the strong peptide and lipids interaction. The pHdependent kinetic controlled FGA process was proved and factors affecting the FGA process were also investigated systematically. We believe this pH-responsive FGA strategy improved our knowledge on the mechanism of the FGA and provide inspiration in understanding the sophisticated assembly behavior in life.

Abstract:Reactions of fresh pentacoordinate phosphoric-amino acid mixed anhydrides with amine and alcohol yielded peptide analogs, N-phosphoamino acid esters, phosphodiesters and some products of ester exchange, which might provid some evidence that the formation of pentacoordinate intermediates was crucial to some bioorganic reactions of N-Phosphoamino acids.

Abstract:The peptide formation of N -phosphoryl amino acids with amino acids proceeds in aqueous solution without any coupling reagents. After being separated in sephadex gel column, the phosphoryl dipeptides were analyzed by the electrospray ionization tandem mass spectrometry (ESIMS/ MS). The result demonstrates that phosphoryl dipeptides were detected in all the reaction systems. It is found that the formation of N -phosphoryl dipeptides is oriented: the N-terminal amino acid residues of the N -phosphoryl dipeptides are from N -phosphoryl amino acids, and the peptide elongation happened at the C-terminal. Only a-dipeptide, no β-dipeptide, is formed in the N -phosphoryl dipeptides, showing that a-carboxylic group is activated selectively by N -phosphorylation. Theoretical calculation shows that the peptide formation of N -phosphoryl amino acids might happen through a penta-coordinate carboxylic-phosphoric intermediate in solution. These results might give some clues to the study on the origin of proteins and protein biosynthesis.

Abstract:Coupling of N-phosphoamino acids 2 and amino acid methyl esters by dichlorotriphenylphosphorane 3 produced N-phosphopeptide methyl esters 5, which were sequentially hydrolyzed in triethylamine or dilute NaOH aqueous solution. The target products N-phosphodipeptides 6 were obtained in reasonable yields and high purity after isolation using extraction method. The convenient and efficient approach could be generally used for synthesis of polypeptides.

Abstract:L-alpha-Amino acids and D-alpha-amino acids, mediated by phosphorus pentachloride, could take place the phosphorylation and then assembled into polypeptides. However, beta-amino acids could not assemble into polypeptides, and the self-assembly of DL-alpha-amino acids into peptides were difficult. alpha-Amino acids could not assemble into peptides mediated by thionyl chloride. The self-assembly reactions were studied by positive ion electrospray ionization mass spectrometry, and the five-membered cyclic penta-coordinated phosphoryl amino acid intermediates were found in the course of self-assembly into peptides. The intermediates could be trapped by P-31 NMR spectroscopy after amino acids were trimethylsilylized to N, O-bis(trimethylsilyl)aniino acids.

Abstract:We reported that for a series of amines with different structures and functional groups, the deprotection of the dialkylphosphoryl group from the N-alkyl-N-dialkyl- phosphoramidates can be achieved under very mild conditions as compared to the corresponding sulfonyl derivatives. From the kinetic study, we find that the diisopropylphosphoramidates derivatives have substantial stability in the acid media. The O, O-diisopropyl-N-phenyl-phosphoramidates has a half life of 53 min in 6M HCl at 50℃. Also, the P-N bond persists during the Lewis acid catalyzed Freidel-Crafts acylation of N-dialkylphosphoryl-N-substituted phenethylglycine (4) to give the stable 2, 3, 4, 5-tetrahydro-7, 8-dialkoxyl-N-diisopropylphosphoryl- 3-benzazepine-1-one (5). 5 can be quantitatively transformed into the amine HCl salt under very mild condition. Thus. we have shown that diisopropyl phosphite can be used as an economical amino protection reagent, the steric hindrance of the diisopropyl group slows down the rate of acid hydrolysis, therefore, increases its stability in the acid media. This chemical property will be useful in organic synthesis.

Abstract:An iridium photoredox catalyst in combination with a phosphoric acid catalyzes the decarboxylative alpha-aminoalkylation of natural and unnatural alpha-amino acid-derived redox-active esters (N-hydroxyphthalimide esters) with a broad substrate scope of N-heteroarenes at room temperature under irradiation. Dipeptide- and tripeptide-derived redox-active esters are also amenable substrates to achieve decarboxylative insertion of a N-heterocycle at the C-terminal of peptides, yielding molecules that have potential medicinal applications. The key factors for the success of this reaction are the following: use of a photoredox catalyst of suitable redox potential to controllably generate alpha-aminoalkyl radicals, without overoxidation, and an acid cocatalyst to increase the electron deficiency of N-heteroarenes.

Abstract:Four phosphonodipeptides containing 1-aminoalkylphosphonate were synthesized by applying four different coupling methods. One of them was then converted into monoacid, ethyl hydrogen 1-(N-acetyl-prolyl)amino-2-phenylethyl phosphonate. Three phosphonodipeptides containing ethyl 1w-amino-2-phenylethylphosphonamidate were also prepared by converting phosphono monoacid into phosphonochloridate and reacting it with amino acid ester. Benzyl ethoxyphosphonyl-glycyl-glycine ethyl ester also was synthesized from ethyl glycylglycinate and ethyl benzylphosphonochloridate derived from direct chlorination of diethyl benzylphosphonate with phosphorus oxychloride.

Abstract:The amino acids' side chains act as the relay device to modulate the chemical reactivity of the N-phosphoryl amino acids. The N-dialkyl phosphoryl cysteine is stable, but the N-dialkyl phosphoryl serine or threoine was converted into many kinds of products at 40-degrees-C. The N-dialkyl phosphoryl gltamic acid is a stable compound, while the N-dialkyl phosphoryl aspartic acid was transferred into the peptides, esters and the phosphoryl ester-exchanged products under mild conditions. The N-dialkyl phosphoryl histidine has the similar reactivity through the co-participation of the side chain, carboxyl and phosphoryl groups. A hexacoordinate phosphorus was proposed to account for this differentiation and promotion effect.

It was demonstrated that the co-participation of the three functional groups of phosphoryl, imidazolyl, and carboxyl in the molecule is essential for these reactions to occur. Also, the neat phosphoryl transfer reaction occurs with N(alpha), N(Im)-Bis-phosphohistidine.

Abstract:The structural characteristics of N phosphorylated branched peptides 2 5 in solution have been studied by NMR. Diastereotopic phenomena, the interaction of hydrogen bonds and no regular secondary structures have been observed. It has been shown that participation of phosphoryl group only affects amino acid, which was bond on the phosphoryl group. Due to the formation of peptide planes and the increase of the effect of hydrogen bond between inter or intra molecules, the position of P=O group relating with C\-β=O group in compounds 2 5 was changed from trans into gauche conformation as compared with that in Dipp Ala of compound 1. The phosphoryl group was still lain on trans position with N C\-α group. It suggests that there is not any polar groups on the side chain of the amino acid. These structural characteristics will provide some evidence for their studies.

Abstract:in recent years beta-amino acids have increased their importance enormously in defining secondary structures of beta-peptides. Interest in beta-amino acids raises the question: Why and how did nature choose alpha-amino acids for the central role in life? In this article we present experimental results of MS and P-31 NMR methods on the chemical behavior of N-phosphorylated alpha-alanine, beta-alanine, and gamma-amino butyric acid in different solvents. N-Phosphoryl alpha-alanine can self-assemble to N-phosphopeptides either in water or in organic solvents, while no assembly was observed for beta- or gamma-amino acids. An intramolecular carboxylic-phosphoric mixed anhydride (IMCPA) is the key structure responsible for their chemical behaviors. Relative energies and solvent effects of three isomers of IMCPA derived from alpha-alanine (2a-c), with five-membered ring, and five isomers of IMCPA derived from beta-alanine (4a-e), with six-membered ring, were calculated with density functional theory at the B3LYP/6-31G** level. The lower relative energy (3.2 kcal/mol in water) of 2b and lower energy barrier for its formation (16.7 kcal/mol in water) are responsible for the peptide formation from N-phosphoryl alpha-alanine. Both experimental and theoretical studies indicate that the structural difference among alpha-, beta-, and gamma-amino acids can be recognized by formation of IMCPA after N-phosphorylation. Origins of Life and Evolution of Biospheres

Abstract:An intramolecular mutual activation between a phosphoryl group and carboxyl group results in the simultaneous formation of nucleotides and peptides by the reaction of nucleosides with N-(O,O-diisopropyl)phosphothreonine in anhydrous pyridine. These results suggest pathways for the simultaneous prebiotic synthesis of peptides and oligonucleotides.
