Abstract:Objective:To observe the synergistic effect of hyperthrnia and cisplatin on human lung adenocarcinoma cell line H1299.Methods:The cells treated 42 degree for 2 hours or not were detected cisplatin sensitivity with CCK-8 assay. After 42 degree for 2 hours, ciplatin 10μg/ml for 24 hours or the combination with both sides, the inhibition ratio was detected wtih CCK-8 assay and flow cytometry analyzed DNA contents in every phase of cell proliferation, and we observed their synergistic effect on cytotoxicity and cell cycel.Results:After hyperthermia, the IC_ 50 values to cisplatin decreased from 9.51μg/ml to 6.07μg/ml. CCK-8 assay showed hyperthermia and ciplatin had synergistic cell killing effect, and FCM also showed hyperthermia had significant effect on cell cycle with increase of G0/G1 phase population and reduction of S phase population.Conclusion:Hyperthermia can significantly increase the sensitivity of H1299 cells to cisplatin.
Enhancement of Osteosarcoma Cell Sensitivity to Cisplatin Using Paclitaxel in the Presence of Hyperthermia

Abstract:Purpose: This paper aimed to evaluate the effects of a combination of paclitaxel and cisplatin on osteosarcoma (OS) cell lines in the presence of hyperthermia and to investigate the related mechanism.Materials and methods: Two types of OS cell lines (OS732 and MG63) were treated with paclitaxel and cisplatin in the presence of hyperthermia. The survival rate was measured by MTT assay, and the clonogenic rate was measured by a clonogenic assay. The cellular changes were observed with an inverted phase contrast microscope and a fluorescence microscope. The apoptotic effect was analysed with flow cytometry (FCM). Fas expression by the OS cell lines was measured by western blot. Fas expression in OS tissue was measured by immunohistochemistry.Results: Our study indicated that 1 h after the application of a combination of 10 mu g/mL paclitaxel and 5 mu g/mL cisplatin to OS cells at 43 degrees C, the survival rate of the OS cells was 11.96%, which was significantly lower than when either 10 mu g/mL paclitaxel (45.02%) or 5 mu g/mL cisplatin (48.69%) was applied alone (p<0.01). Additionally, the clonogenic assay demonstrated that the clonogenic survival rate in the OS cells of the combination group was lower than that in the individual groups. Moreover, the cellular changes and apoptosis rates indicated that apoptosis in the combined application group was much greater than when either drug was applied individually. Fas expression by OS cell lines was increased by the combination of paclitaxel and cisplatin under hyperthermic conditions. More importantly, our study revealed low Fas expression in OS, which better explained the up-regulation of Fas achieved by the combination of paclitaxel and cisplatin in the presence of hyperthermia.Conclusions: The combination of paclitaxel and cisplatin increases the effects of thermochemotherapy on OS cell lines, primarily through the induction of apoptosis by the up-regulation of Fas expression.

Abstract:OBJECTIVE:To evaluate the effect of hypoxia improvement in Hep-2 cell on cisplatin-induced apoptosis.METHOD:Human laryngeal squamous cell carcinoma Hep-2 cells and HIF-1α-RNAi-Hep-2 cells were cultured in normoxic, hypoxic and reoxygenation condition. The inhibition of cisplatin on cell proliferation was evaluated by MTT assay. The influence of cisplatin on cell cycle and apoptosis were detected by flow cytometry.RESULT:The inhibition of cisplatin on cell proliferation was reduced by hypoxia. After HIF-1α gene was silenced, the inhibition of cisplatin on Hep-2 cell proliferation was increased apparently, but was still interfered partly by hypoxia. Hypoxia could induce cell apoptosis decreased and enhance chemotherapeutic resistance. After reoxygenation, cell apoptosis induced by cisplatin was increased significantly (P < 0.05). HIF-1α-RNAi-Hep-2 cells under hypoxia also showed certain resistance to apoptosis but the sensitivity to cisplatin was higher than that of Hep-2 cells. When cells were returned from hypoxic condition to normoxic condition for some time, the apoptosis induced by cisplatin was increased significantly (P < 0.05).CONCLUSION:The improvement of hypoxic microenvironment with HIF-1α gene knockout could increase the sensitivity of Hep-2 cells to chemotherapy.

Abstract:Abstract Cytotoxic effects of hyperthermia combined with DDP on MKN28 and MKN45 cells were studied by MTT assay according to a nested design. The results showed: hyperthermia alone above 43 °C for 30 mins was cytotoxic; hyperthermia at temperature lower than 43 °C for 30 mins could increase sensitivity of cancer cells to DDP. The cytotoxic effect of simultaneous use of hyperthermia and DDP was more marked than that of sequential use of the 2 treatments. Hyperthermia combined with DDP could inhibit growth of human gastric adenocarcinoma cells regardless of their degree of differentiation.
Effect of Alarelin Combined with Cisplatin on Subcutaneous Xenograft of Human Endometrial Carcinoma in Nude Mice

Abstract:Objective To investigate the effects of alarelin combined with cisplatin on subcutaneous xenograft of human endometrial carcinoma in nude mice and to explore its mechanism.Methods Human endometrial carcinoma model was established by subcutaneous inoculation in nude mice,and then they were divided into 4 groups:control group,alarelin group,cisplatin group,alarelin+cisplatin group.The anti-tumor efficacy was evaluated by determining tumor volume and tumor growth.After treatment,all mice were sacrificed.The inhibitory rates were calculated,and the pathological changes were observed.The expression levels of Caspase-3 were determined by immunocytochemistry.Results ①The inhibitive rate of the tumor was higher in alarelin+cisplatin group than in alarelin group and cisplatin group(63.32% vs 32.12%,22.14%,P0.05).②The immunohistochemical results showed that the expression of Caspase-3 increased obviously in treated groups compared with control group.Meanwhile the expression of Caspase-3 showed significant difference among the treated groups(P0.001).Conclusion Either alarelin or cisplatin can suppress tumor growth.Alarelin combined with cisplatin could have a synergistic effect on inhibiting the growth of human endometrial carcinoma in nude mice of subcutaneous xenograft,which may be related to the up-regulation of caspase-3 protein.

Abstract:Objective To observe the effects and mechanisms of hyperthermia therapy combined with chemotherapy on multidrug-resistant human lung adenocarcinoma cell line A549/DDP, it will provide theory evidence in thermochemotherapy for clinic. Methods A549 or A549/DDP treated with different temperature were detected of sensitivity to cisplatin with CCK-8 assay. After hyperthermia, A549/DDP cell cycle was analyzed with flow cytometry and ERCC1mRNA expression was detected using RT-PCR. Results Parent human lung adenocarcinoma cell A549 was more sensitive to cisplatin than drug-resistant cell line A549/DDP. Treated with different temperature, drug sensitivity of A549/DDP to cisplatin all increased. Hyperthermia had significant effect on cell cycle with increase of G2/M phase population and reduction of G0/G1 phase population, and thermochemotherapy was more notable(P<0.01). After hyperthermia, the expression of ERCC1 mRNA did not change. Conclusion Hyperthermia can significantly enhance the sensitivity to cisplatin in human multidrug-resistant lung cancer cell line A549/DDP. Thermochemotherapy has synergistic effects of inhibiting cell proliferation. The major synergistic mechanisms of thermochemotherapy are that they can induce cell cycle arrest?inhibit cell proliferation ,but have no correlation with gene ERCC1.

Abstract:Objectives This experiment aims to investigate whether concurrent exposure of human hepatocellular carcinoma cells SMMC-7721 to cisplatin combined with hyperthermia will increase anticancer effects.Methods In our study,the MTT assay has been developed for quantitative evaluation of the proliferation of SMMC-7721 and cytotoxicity of cisplatin with or without hyperthermia.Flow cytometric analyses were used for the assessment of apoptosis rates after treatment.Results The combination treatments induced significantly higher apoptosis and cytotoxicity than exposure to single 43 ℃ hyperthermia or cisplatin.Conclusion While either hyperthermia or cisplatin can induce apoptosis and anti-proliferation effect,they may offer synergistic benefits when used concurrently.
Synergistic Effect of Elemene Combined with Hyperthermia and Cisplatin on Human Lung Adenocarcinoma Cell Line A549

Abstract:Objective To observe the synergistic effect of elemene combined with hyperthermia and cisplatin on human lung adenocarcinoma cell line A549.Methods The sensitivity of the cells treated with gradient concentration cisplatin and elemene in 42℃ or 37℃ water bath for 1.5 hours were detected by MTT assay.One hour and a half after treatment with water bath at 42℃ and cisplatin 4 μg/mL combined with elemene 15 μg/mL or 60 μg/mL,the inhibition ratio of the cells was detected by MTT assay and appearance of the cells was detected by transmission electron microscope.Results After hyperthermia,the IC50 values to cisplatin decreased from 22.44 μg/mL to 11.74 μg/mL.The IC50 values of elemene decreased from 35.43 μg/mL to 28.68 μg/mL.MTT assay showed elemene combined with hyperthermia and cisplatin had synergistic cell killing effect,and the cells of A549 appeared apoptosis.Apoptotic body and cellular necrosis could be seen.Conclusion Low concentration elemene can significantly increase the sensitivity of A549 cells to cisplatin combined with hyperthermia.

Abstract:目的观察热疗联合放化疗对人肺腺癌耐药细胞株A549/DDP的协同杀伤作用,为临床上应用热放疗和热化放疗联合治疗非小细胞肺癌提供实验依据.方法采用CCK-8法和克隆形成实验检测热放疗或热放化疗对A549/DDP细胞抑制率和克隆形成率的影响;采用流式细胞仪检测热放疗或热放化疗联合作用后A549/DDP细胞的凋亡现象;采用光镜和透射电镜观察细胞形态学改变.结果热放疗尤其是热放化疗联合对A549/DDP细胞有明显抑制作用(P＜0.01);流式细胞仪显示热放疗或热放化疗有诱导A549/DDP细胞凋亡作用(P＜0.01);电子显微镜观察到细胞核固缩、变形,染色质浓聚、边集等凋亡现象.结论热放疗或热放化疗能诱导A549/DDP细胞凋亡。
Effect of Hyperthermia Combined with Anti-Cancer Drugs for Human Lung Adenocarcinoma Cell Lines

Abstract:Objective To observe the kill effect of hyperthermia combined with anti-cancer drugs(mitomycin,vindesine,cisplatin) on human lung adenocarcinoma cell lines(D6).Methods Experimental objects were divided into four groups:Control group,hyerthermia group,chemotherapy group and chemohyperthermia group.Dynamic change of cell proliferation was detect by cell growth curve and clone formation assay;Cell cycle and apoptosis were test by flow cytometry;Cell morphology was observed by electron microscope;Ag-NOr3 in nucleus was determined by multi-function image analysis system.Results After treatment,the dynamic change of cell proliferation all was obviously inhibited in chemotherapy group,hyperthermia group,particularly in the chemohyperthermia group.Of cell cycle,the blockage in S-phase was found at 24 hours after chemohyperthermia group,it reached 88.18%,small apoptotic peak was also presented.There found obvious cell damage and cell vacuolation after 24 hours of chemotherapy or chemohyperthermia treatment.Even as cell disruption to typic apoptosis was appeared in chemohyperthermia group.While the Ag-NOR3 in nucleus appeared black granules,nuclei got yellow under the electron microscope.Conclusion The study suggests that the hyperthermia combined with anti-cancer drugs has a kill effect for treating the human lung adenocarcinoma cells.
Synergistic suppression effect on tumor growth of hepatocellular carcinoma by combining oncolytic adenovirus carrying XAF1 with cisplatin

Abstract:Purpose The potent anticancer efficacy of oncolytic viruses has been verified in Clinic in recent years. Cisplatin (DDP) is one of most common chemotherapeutic drugs, but is accompanied by side effects and drug resistance. Our previous studies have shown the strategy of cancer -targeting gene-viro-therapy (CTGVT) mediated by the oncolytic virus ZD55 containing the XAF1 cDNA (ZD55-XAF1), which exhibited potent antitumor effects in various tumor cells and no apparent toxicities on normal cells. In the study, the CTGVT strategy is broadened by combining DDP with ZD55-XAF1 for growth inhibition of hepatocellular carcinoma (HCC) cells. Methods The transgenic expression was evaluated by both in vitro and in vivo experiments, and the enhanced inhibitory effect of ZD55-XAF1 combined with cisplatin was assessed in HCC cells. The cytotoxicity on normal liver cells was evaluated by MTT assay and apoptotic cell staining. Activation of caspase-9 and PARP for apoptosis was further detected by Western blot analysis. The in vivo antitumor efficacy of combination treatment with cisplatin and ZD55-XAF1 was estimated in an HCC xenograft mouse model. Results We found that the combination of ZD55-XAF1 and cisplatin showed enhanced inhibitory effects on the proliferation of HCC cells in vitro and tumor growth in mice. Furthermore, the combined treatment of ZD55-XAF1 and DDP decreases the chemotherapy dose needed to achieve the same inhibitory effect without overlapping toxicities on normal liver cells and induces tumor cell apoptosis via the activation of caspase-9/PARP pathway. Conclusion Thus, these data suggest that the chemo-gene-viro-therapeutic strategy by combining ZD55-XAF1 and DDP reveals a novel therapeutic strategy for hepatocellular carcinoma.

Abstract:OBJECTIVE:To determine the sensitivity of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) induced apoptosis in Hep-2 cells by means of systematically evaluating the cytotoxicity of TRAIL alone and TRAIL in combination with chemotherapeutic agents (cisplatin, paclitaxel) or radiation in Hep-2 cells in vitro, and whether the synergistic killing effects correlated with the expression level of TRAIL receptors and the activity of caspase-8 or caspase-9.METHODS:The cytotoxicities of TRAIL, cisplatin and paclitaxel were investigated by cell counting kit-8 (CCK-8) assay. The expression levels of four TRAIL receptors in Hep-2 cells after treated by TRAIL, chemotherapeutic agents or radiation alone and by combined treatments were measured by flow cytometry and Western blotting respectively. The growth inhibition effects of caspase-8 or caspase-9 inhibitor on Hep-2 cells were determined by CCK-8 assay, and the activities of caspase-8, caspase-9 and caspase-3 were measured by Western blotting.RESULTS:The half maximal inhibitory concentration (IC50) of TRAIL to Hep-2 cells on 24 h was 596.92 microg/L. Cisplatin, paclitaxel and radiation had synergistic inhibitory effects with TRAIL on the growth of Hep-2 cell line. After the activity of caspase-9 was inhibited by Z-LETD-FMK, the inhibition effects of TRAIL, cisplatin and paclitaxel on Hep-2 cells decreased significantly (all P<0.05). The expressions of caspase-8, caspase-9 and the death receptors (DR4 and DR5) increased significantly (all P<0.05) after combined administration.CONCLUSIONS:Hep-2 cells are resistant to the apoptosis induced by TRAIL and TRAIL can induce the apoptosis of Hep-2 cells through the endogenous apoptotic pathway. The increase of death receptors expression by chemotherapeutic agents or radiation could enhance the sensitivity of Hep-2 cells to TRAIL and the synergistic killing effects.

Comparison of Either Cisplatin or Oxaliplatin with Hyperthermia Against Human Gastric Cancer Cell Line BGC-823

Abstract:Objective:To compare the anticancer effect of cisplatin or oxaliplatin combined with hyperthermia against human undifferentiated gastric adenocarcinoma cell line BGC-823. Methods:The working concentrations of cisplatin and oxaliplatin against BGC-823 were determined by MTT assay that has been developed for quantitative evaluation of the proliferation of cells. Then the chemotherapy or hyperthermia, singly or concurrently, were used and the cell survival rates were obtained. The anticancer effect was evaluated and compared by Veleriote method at 24 h or 48 h. Results:The working concentrations were defined as the concentration causing 20%～30% inhibition rate. They turned out to be 1μg/ml and 5μg/ml of cisplatin and oxaliplatin, respectively. Exposing to 43℃ hyperthermia for 1 hour did not show obvious inhibition to BGC-823 at 24 h or 48 h(P0.05). The chemotherapies with cisplatin 1μg/ml or oxaliplatin 5μg/ml did not exhibit obvious inhibition to BGC-823 at 24 h (P0.05), but they did at 48 h(P 0.01). The combination of cisplatin or oxaliplatin with 43℃ hyperthermia for 1 hour showed obvious inhibition to BGC-823 at 24 h and 48 h.(P0.05). And the effect of chemohyperthermia was amplified as the time prolonged(P0.05). There was no difference between the two drugs at 24 h and 48 h when used singly. The inhibitory effect of cisplatin was much higher than that of the oxaliplatin when used with hyperthermia. Judged with the Veleriote method at 24 h and 48 h, there was obvious synergism of cisplatin 1μg/ml combined with hyperthermia. Oxaliplatin combined with hyperthermia exhibited subaddictive effect at 24 h and synergism at 48 h. Conclusion:The combination of cisplatin or oxaliplatin and 43℃ hyperthermia for 1 hour!showed subaddictive effect or synergism to the inhibition of human undifferentiated gastric adenocarcinoma cell line BGC-823,and cisplatin is much better than oxaliplatin when combined with hyperthermia.
Dihydroartemisinin breaks the immunosuppressive tumor niche during cisplatin treatment in Hepatocellular carcinoma

Abstract:Objective Hepatocellular carcinoma, characterized by high mortality rates, often exhibits limited responsiveness to conventional treatments such as surgery, radiotherapy, and chemotherapy. Therefore, identifying a sensitizer for cisplatin has become crucial. Dihydroartemisinin, known for its potent role of tumor treatment, arises as a prospective candidate for cisplatin sensitization in clinical settings. Methods A mouse model of liver tumor was established through chemical induction of DEN/TCPOBOP. Upon successful model establishment, ultrasound was employed to detect tumors, Hematoxylin and eosin staining was conducted for observation of liver tissue pathology, and ELISA was utilized to assess cytokine changes (IFN-γ, IL-2, IL-4, IL-10, TGF-β, IL-1β, CCL2, and CCL21) in peripheral blood, para-tumor tissues, and tumor tissues. The infiltration of CD8 + T cells and macrophages in tumor tissue sections was detected by immunofluorescence. Results Dihydroartemisinin combined with cisplatin obviously restrained the growth of liver tumors in mice and improved the weight and spleen loss caused by cisplatin. Cisplatin treatment of liver tumor mice increased the content of CCL2 and the number of macrophages in tumor tissues and promoted the formation of an immunosuppressive microenvironment. The combination therapy decreased the content of TGF-β in tumor tissues while increasing CCL2 levels in para-tumor tissues. Both combination therapy and cisplatin alone increased the number of CD8 + T cells in tumor tissue, but there was no difference between them. Conclusion Dihydroartemisinin combined with cisplatin obviously prevented the deterioration of liver tumor in hepatocellular carcinoma mice and improve the therapeutic effect of cisplatin by improving the immunosuppressive microenvironment induced by cisplatin. Our findings provide a theoretical basis for considering dihydroartemisinin as an adjuvant drug for cisplatin in the treatment of hepatocellular carcinoma in the future.

Abstract:Background Hyperthermia can play a synergistic role with chemotherapy in combination therapy. Although the association between caspase activation, apoptosis, and pyroptosis have been published for both cisplatin (CDDP) and hyperthermia therapies independently, the interactions between these molecular pathways in combination therapy are unknown. The present study aimed to investigate the possible interactions between caspase 8 activation, apoptosis, and pyroptosis in combination therapy.

Abstract:OBJECTIVE:To observe the cytotoxic effect of thermo-chemotherapy with cisplatin on osteosarcoma OS-732 cell line and to explore its mechanism.METHODS:The osteosarcoma OS-732 cell line was treated with different temperature, cisplatin alone and 43 degrees C +cisplatin for 1 h, respectively and the in vitro cytotoxic effect was observed with MTT assay. The cell cycle and the apoptotic rate were analyzed with flow cytometry (FCM); the cellular apoptosis was observed also with electron microscope.RESULT:The cytotoxicity index increase markedly as the temperature elevated or the concentration of cisplatin increased. While treated with 43 degrees C hyperthermia and cisplatin simultaneously, the cytotoxicity index increased to 72.37%;the cell cycle of the treated OS-732 cells line was changed with marked increase in S phase and decreasing in G(2)/M phase. The apoptotic rate increased markedly with the highest of 56.47%. Electron microscope showed the characteristic apoptotic alterations.CONCLUSION:Hyperthermia with cisplatin enhance cytotoxicity on osteosarcoma OS-732 cell line and the induced cell apoptosis may be one of the mechanisms of enhanced cytotoxicity by thermo-chemotherapy.

Abstract:Objectives To investigate whether concurrent exposure of human hepatocellular carcinoma cells SMMC-7721 to hydroxycamptothecine with hyperthermia will increase anticancer effects. Methods The MTT assay has been developed for quantitative evaluation of the proliferation of SMMC-7721 and cytotoxicity of hydroxycamptothecine with or without hyperthermia. Flow cytometric analyses were used for the assessment of apoptosis rates after treatment. Results The combined treatments significantly induced apoptosis and cytotoxicity higher than exposure to 43 ℃ hyperthermia alone or hydroxycamptothecin alone. Conclusion While both hyperthermia and hydroxycamptothecin can individually induce apoptosis and anti-proliferation effect, they may offer synergistic benefits when used concurrently.
Enhancing Intrapleural Hyperthermic Chemotherapy for Lung Cancer: Insights from 3D and PDX Models

Abstract:Background/Objectives: Malignant pleural effusion (MPE) in lung cancer indicates systemically disseminated advanced lung cancer and is associated with poor survival. Intrapleural hyperthermic chemotherapy (IPHC) is a promising treatment for MPE; however, its biological basis is not fully understood. IPHC can enhance anticancer drug efficacy, particularly in drug-resistant cancers. This study investigated the effects of hyperthermia on cisplatin cytotoxicity in lung cancer cell lines, patient-derived tumor cells, and a patient-derived xenograft (PDX) model. Methods: Lung cancer cell lines (A549 and H2170) and patient-derived tumor cells were cultured in 2D/3D systems and treated with cisplatin under varying temperatures (37 °C, 43 °C, and 45 °C) and exposure times (5, 15, and 30 min). Antiproliferative effects were evaluated using LDH and CCK-8 assays. Optimal conditions identified in cell culture experiments were validated using a PDX model; tumor growth inhibition, delay, and protein expression were analyzed post-treatment. Results: Hyperthermia significantly enhanced the antitumor efficacy of cisplatin at 43 °C and 45 °C, with comparable effects under 15 and 30 min exposure. In the PDX model, IPHC showed increased tumor inhibition and necrosis and delayed tumor regrowth, particularly at higher cisplatin doses. Protein expression analysis revealed that hyperthermia decreased EGFR expression and increased levels of apoptosis-related proteins, including cleaved PARP and caspase-3. Conclusions: IPHC with cisplatin demonstrated enhanced antitumor efficacy in vitro models, particularly in drug-resistant lung cancer, indicating its potential as a valuable adjunct to existing treatment regimens for lung cancer and for improving patient outcomes in advanced lung cancer with MPE or pleural metastasis.
