Abstract:This study was conducted to study the effects of γ-aminobutyric acid (GABA) on growth performance, serum biochemical indices and hormones of HPA (cortisol, corticosterone and adrenocorticotropic hormone), HPT (free triiodothyronine, free thyroxine, triiodothyronine and thyroxine) axis in growing-finishing swine. Ninety-six (Duroc× Landrace×Yorkshine) growing-finishing pigs with an average initial body weight of (43. 0 ± 3. 2) kg were randomly allocated to four treatments with three replicates in each group and eight pigs in each replicate (1/2 male and female). The treatments included control group without GABA, 10 mg/kg GABA group (group I), 20 mg/kg GABA group (group Ⅲ) and 40 mg/kg GABA group (group Ⅲ) .The experiment lasted for 48 days. At the end of the experiment, the growth performance were determined, and two pigs in every replicate in control group and optimum growth performance group were randomly selected to determine serum biochemical parameters and hormones of HPA and HPT axis. The results showed that different levels of GABA affected growth performance of growing-finishing pigs, and 10 mg/kg was the best dosage which significantly increased the average daily gain and feed/gain ratio by 13. 08% and 7. 81% , respectively (P0. 01). Ten mg/kg GABA increased the serum concentration of triiodothyronine and free triiodothyronine (P0. 05); glutathione peroxidase (GSH-Px) activity was significantly increased (P0. 01); but lactate dehydrogenase (LDH) activity and cortisol, corticosterone and adrenocorticotropic hormone (ACTH) concentrations were decreased(P0. 05). It was concluded that GABA could improve growth performance of growing-finishing pigs by increasing average daily gain and decreasing feed/gain ratio; 10 mg/kg GABA also could enhance antioxidation performance and thyroid function.

Abstract:This experiment was conducted to study the effects of gamma-aminobutyric acid(GABA) on antioxidative property and hormones of HPA,HPT axis in growth-finishing swine.48 crossbred(Duroc×Landrace×Large White) pigs with average weight 43kg were randomly assigned to 2 treatments with 3 replicates and 8 pigs with per pen.Two treatments were fed basal diets (the control group) and basal diets supplemented 10 mg/kg GABA(the experimental group).After the feeding trial,2 pigs (one male and one female) in each replicate were chosen for blood sampling,and related blood parameters were determined.The results showed that:10mg/kg GABA supplemented in basal diets could increase the activities of GSH-Px (P<0.01) and SOD,and decrease the concentration of MDA (P>0.05).The concentrations of cortisol, corticosterone and ACTH in serum of pigs in experimental groups were decreased (P<0.05),and the concentrations of T3 and FT3 (P<0.05) were increased,however,10mg/kg GABA supplemented in diets had no effect on the concentration of T4 and FT4(P>0.05).

Abstract:A study was conducted to determine the effects of feeding γ-aminobutyric acid(GABA) on the growth performance,antioxidation and hormones of HPA,HPT axis in growth-finishing swine in the hot summer.96(Duroc×Landrace×Yorkshine) growth-finishing pigs with an average initial body weight(BW) of(43.07±5.76) kg,were randomly allotted into 4 treatment groups with 3 replications of 8 pigs per replicate pen for each treatment in 48 days trial.The treatments were designed in a single factorial arrangement,and were as follows: basal diet without GABA,basal diet with 10 mg·kg-1 GABA,basal diet with 20 mg·kg-1 GABA,and basal diet with 40 mg·kg-1 GABA.The results showed that growth performance of growth-finishing swine was affected in all treatments.10 mg·kg-1 was the best dosage which significantly increased the average daily gain(ADG),feed/gain(F/G) by 13.08% and 7.81%,respectively(P<0.01).However,average daily feed intake(ADFI) was not affected in every treatment.In addition,10 mg·kg-1 GABA increased the activity of GSH-Px(P<0.01),the activity of SOD was increased and MDA concentration was decreased(P>0.05).The concentration of serum cortisol,corticosterone and ACTH were all decreased too(P<0.05).10 mg·kg-1 GABA obviously promoted the secretion of T3 and FT3(P<0.05) but had no obvious effect on the secretion of T4 and FT4(P>0.05).In conclusion,GABA can effectively improve the growth performance of growth-finishing swine in the hot summer when they were added into diet,10 mg·kg-1 was the optimal dosage under this condition.10 mg·kg-1 GABA also mitigated stress induced by high temperature in the summer and enhanced antioxidation performance and thyroid function.

Abstract:48 Duroc×(Landrace×Large White) finishing hogs that weighed about 43 kilograms were randomly divided into two groups, one was fed with basal diet, and the other was fed with basal diet added 10 mg/kg GABA. The results showed that in 10 mg/kg GABA group, serum calcium, phosphorus, kalium, chloride concentrations were increased respectively, all the differences were not significant(P>0.05); so were the serum total protein, albumin, urea nitrogen concentrations have the improvements uptrend, but the differences were not significant(P>0.05); glucose was significantly increased by 13.29%(P<0.05); GPT, GOT activity were both not significantly increased, while LDH and CPK were decreased by 16.93%(P<0.05), 38.22%(P<0.01); GSH-Px, SOD activity were increased, while MDA concentration was decreased.

Abstract:This experiment was conducted to study the effects of γ-aminobutyric acid(GABA) on the laying and reproductive performance of broiler breeders under heat stress,and to investigate the mechanism from antioxidation,immunologic function and liver-associated serum enzyme activities.A total of 360 Lingnan yellow broiler breeders(28-week-old) were randomly divided into 4 groups with 3 replicates in each group and 30 broiler breeders in each replicate.Diets of the four groups were supplemented with 0(control group),25,50 and 75 mg/kg GABA in a basal diet,respectively.The preliminary trial period lasted for 14 days;trial period lasted for 56 days.The results showed as follows: compared with the control group,1) supplemented 75 mg/kg GABA significantly increased laying rate,average egg weight,feed/egg ratio of broiler breeders(P0.05),and also significantly increased fertilization and hatchability of eggs(P0.05),birth weight(P0.05) and birth rate(P0.01) of chicks.Furthermore,supplemented 25 and 50 mg/kg GABA significantly increased hatchability and birth rate(P0.05),and supplemented 25 GABA significantly increased birth rate of chicks(P0.05).2) Supplemented 75 mg/kg GABA significantly increased the contents of serum immunoglobulin A(IgA),immunoglobulin M(IgM) and complement C4(P0.05);supplemented 25 mg/kg GABA significantly increased the serum IgM content(P0.05).3) Supplemented 75 mg/kg GABA significantly increased the activities of serum superoxide dismutase,glutathione peroxidase(P0.05) and total antioxidant capability(P0.01),and decreased the serum malondialdehyde concentration(P0.01);supplemented 25 mg/kg GABA also significantly decreased serum malondialdehyde concentration(P0.05).It is concluded that supplementation of GABA can improve the laying and reproductive performance of broiler breeders under heat stress.

Abstract:γ-Aminobutyric acid (GABA) is a non-protein amino acid which naturally and widely occurs in animals, plants, and microorganisms. As the chief inhibitory neurotransmitter in the central nervous system of mammals, it has become a popular dietary supplement and has promising application in food industry. The current article reviews the most recent literature regarding the physiological functions, preparation methods, enrichment methods, metabolic pathways, and applications of GABA. This review sheds light on developing GABA-enriched plant varieties and food products, and provides insights for efficient production of GABA through synthetic biology approaches.

Abstract:This study was conducted to investigate the effects of rumen-protected γ-aminobutyric acid (GABA) on feed intake, growth performance and expression of related genes in growing lambs. A total of 24 lambs weaned at age of 50 days were divided into four block of six based on their BW, six lambs within a block were allocated to three pairs, which were then assigned randomly to three treatments with addition of rumen-protected GABA at levels of 0, 70 or 140 mg/day for 6 weeks. Dry matter intake was recorded weekly in three consecutive days, and BW was recorded every two weeks. At the end of the trial, four lambs from each group were slaughtered, and duodenum and ileum mucosa were obtained for measurement of mRNA abundance of GABA receptor and cholecystokinin receptor. Dry matter intake was higher (P<0.01) in the lambs fed 140 mg/day GABA than that in the control or 70 mg GABA-fed lambs. Average daily gain and nutrients digestibility were not different (P>0.05) among treatments. Lambs fed 140 mg/day GABA had higher mRNA abundance of GABA-B receptor (P<0.01) and lower mRNA abundance of cholecystokinin-2 receptor (P<0.01) in duodenum mucosa. Serum CCK content was lower (P<0.01) in lambs fed 140 mg/day GABA than that in control. It is indicated that GABA may enhance feed intake by regulating GABA- and cholecystokinin-related genes.

Abstract:γ-aminobutyric acid (GABA)is a functional component which extensively exists in cereals,fruits and animal brains.It presents various functional activities including relieving anxiety,lowering blood pressure and regulating body weight,ect.The process of plant metabolism (germination) and microbial fermentation can effectively enrich the content of GABA in food though it is generally low in natural foodstuffs.In this paper,the technology process of GABA enrichment in cereals were reviewed.

Abstract:This study was conducted to evaluate the effect of γ-aminobutyric acid (GABA) on laying performance, egg quality, digestive enzyme activity, hormone level and immune activities in Roman hens under heat stress. Roman hens (320 days old) were fed with 0, 25, 50, 75 and 100 mg/kg GABA, respectively during a 60-day experiment. Compared with control, supplementation of 50 mg/kg GABA improved the laying performance and egg quality by significantly increasing egg production, average egg weight and shell strength (P < 0.05), while decreasing the feed-egg ratio and cholesterol level. Anti-oxidation activity was improved by significantly increasing the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), but decreasing malondialdehyde level in serum (P < 0.05), while significantly increasing the glucose and total protein (TP) level, follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E(2) ), insulin, triiodothyronine (T(3) ) and free triiodothyronine (FT(3) ) levels, and IgG, IgA and complement (C3)activity in serum (P < 0.05). The results indicated that oral GABA improved laying performance and physical condition mainly by modulating hormone secretion, enhancing anti-oxidation and immune activity, and maintaining electrolyte balance. Fifty mg/kg was the optimum level for laying hens under heat stress in the present study.

Abstract:In swine production, stress is a common encounter that leads to serious bacterial infection and adverse effects on growth performance. Though antibiotics have been frequently used to control pathogen spread, sustained negative impacts from antibiotics have been found to affect intestinal integrity and the immune system. Multiple nutritional strategies have shown potential to counteract stress and replace antibiotics, including functional amino acids, low protein diet, plant extracts, organic acids, prebiotics, probiotics, minerals and vitamins. These additives relieve the stress response in swine via different mechanisms and signal transduction pathways. Based on the overview of signaling pathways and stress models, this review highlights the potential of nutritional strategies in swine for preventing or treating stress-related health problems. For wider application in the pig industry, the dose ranges measured require for further validation in different physiological contexts and formulations. In the future, microfluid devices and novel stress models are expected to enhance the efficiency of screening for new anti-stress candidates.

Abstract:This study aims to investigate the effects of dietary gamma-aminobutyric acid (GABA) supplementation on the growth performance, intestinal immunity, intestinal GABAergic system, amino acid profiles and gut microflora of the weaned piglets. Totally sixteen healthy piglets were randomly assigned into two groups to be fed with the basal diet (Con group) or the basal diet with GABA (20 mg kg-1) supplementation. Body weights and feed intakes were monitored weekly. Piglets were sacrificed after 3 weeks of GABA supplementation to collect the blood, ileum, ileal mucosa and luminal content. Immune-associated factors, GABAergic system, amino acid profiles, and microbiota in the ileum and serum amino acid profiles were explored. The results showed that GABA supplementation improved the growth performance and modulated the intestinal immunity with inhibiting the gene expressions of IL-22, proinflammatory cytokines (IL-1 and IL-18), and Muc1, but promoted the expressions of anti-inflammatory cytokines (IFN-γ, IL-4, and IL-10), TLR6 and MyD88. GABA regulated a few components of the intestinal GABAergic system, increased the levels of most amino acids in the ileal mucosa but reduced the serum amino acid profiles. GABA regulated the population and diversity of intestinal microbiota, such as the abundances of the dominant microbial populations, the community richness, and diversity of the ileal microbiota. In conclusion, GABA supplementation modulated the intestinal functions, including intestinal immunity, intestinal amino acid profiles and gut microbiota, and the results can be helpful for understanding the functions of GABA in the intestine.
Effects of γ-aminobutyric acid on growth performance and antioxidant capacity in finishing pigs housed in thermal environment

Abstract:The experiment was conducted to study the effects of γ-aminobutyric acid on performance and metabolism of finishing pigs in thermal environment.According to sex and body weight,48 three-way crossbred Duroc ×(Landrace ×Yorkshire)finishing pigs(43 kg) were randomly divided into 2 groups and each group had 3 replicates of 8 pigs.Pigs of the control group were fed basal diets and the treatment group were fed basal diets supplemented with γ-aminobutyric acid 10 mg/kg.The results showed that the treatment group showed better growth performance and its feed efficiency than the control group(P < 0.01).The serum urea nitrogen (SUN) concentration of the treatment group was not significantly difference from that of the control group(P > 0.05),and its serum glucose (GLU) concentration was higher than that of the control group(P < 0.05),and its lactate dehydrogenase (LDH) was lower than that of the control group(P < 0.05),creatine kinase (CK) was higher than that of the control group(P < 0.01).The content of malondialdehyde decreased(P < 0.05)while glutathione peroxidase activities increased(P < 0.01),but superoxide dismutase was not significantly difference from that of the control group(P > 0.05).It was concluded that γ-aminobutyric acid had better effect on improving growth performance and metabolism response and improved the ability of resisting heat stress in finishing pigs in thermal environment.

Abstract:Gamma-aminobutyric acid (GABA) is widely distributed in nature and considered a potent bioactive compound with numerous and important physiological functions, such as anti-hypertensive and antidepressant activities. There is an ever-growing demand for GABA production in recent years. Lactic acid bacteria (LAB) are one of the most important GABA producers because of their food-grade nature and potential of producing GABA-rich functional foods directly. In this paper, the GABA-producing LAB species, the biosynthesis pathway of GABA by LAB, and the research progress of glutamate decarboxylase (GAD), the key enzyme of GABA biosynthesis, were reviewed. Furthermore, GABA production enhancement strategies are reviewed, from optimization of culture conditions and genetic engineering to physiology-oriented engineering approaches and co-culture methods. The advances in both the molecular mechanisms of GABA biosynthesis and the technologies of synthetic biology and genetic engineering will promote GABA production of LAB to meet people’s demand for GABA. The aim of the review is to provide an insight of microbial engineering for improved production of GABA by LAB in the future.

Abstract:Gamma-aminobutyric acid (GABA) is a four-carbon non-protein amino acid that is widely distributed among various organisms. Since GABA has several well-known physiological functions, such as mediating neurotransmission and hypotensive activity, as well as having tranquilizer effects, it is commonly used as a bioactive compound in the food, pharmaceutical and feed industries. The major pathway of GABA biosynthesis is the irreversible decarboxylation of l-glutamate catalyzed by glutamate decarboxylase (GAD), which develops a safe, sustainable and environmentally friendly alternative in comparison with traditional chemical synthesis methods. To date, several microorganisms have been successfully engineered for high-level GABA biosynthesis by overexpressing exogenous GADs. However, the activity of almost all reported microbial GADs sharply decreases at physiological near-neutral pH, which in turn provokes negative effects on the application of these GADs in the recombinant strains for GABA production. Therefore, ongoing efforts in the molecular evolution of GADs, in combination with high-throughput screening and metabolic engineering of particular producer strains, offer fascinating new prospects for effective, environmentally friendly and economically viable GABA biosynthesis. In this review, we briefly introduce the applications in which GABA is used, and summarize the most important methods associated with GABA production. The major achievements and present challenges in the biotechnological synthesis of GABA, focusing on screening and enzyme engineering of GADs, as well as metabolic engineering strategy for one-step GABA biosynthesis, will be extensively discussed.

Abstract:Inhibin INH is a heter-chinmeric glycoprotein hormone containing a common α-subunit and one of two β-subunit and are present in two form:inhibin A(α/B-A)and inhibin B(α/B-B),which is secreted by granular cells in ovarian and sertolic cells in testis. Inhibin selectively inhibits follicle stimulating hormone(FSH) synthesis and/or release. Immunoneutralization of inhibin specially stimulates the secretion of FSH by pituitary, facilitatea gamates production, and then raises animal reproduction. This article reviewed the research progress of inhibin and its effects on the reproduction of livestock.

Abstract:Gamma-aminobutyric acid (GABA) is a naturally occurring potential bioactive compound present in plants, microorganisms, animals, and humans. Especially, as a main inhibitory neurotransmitter in the central nervous system, GABA possesses a broad spectrum of promising bioactivities. Thus, functional foods enriched with GABA have been widely sought after by consumers. However, the GABA levels in natural foods are usually low, which cannot meet people's demand for health effects. With the increasing public awareness on the food securities and naturally occurring processes, using enrichment technologies to elevate the GABA contents in foods instead of exogenous addition can enhance the acceptability of health-conscious consumers. Herein, this review provides a comprehensive insight on the dietary sources, enrichment technologies, processing effects of GABA, and its applications in food industry. Furthermore, the various health benefits of GABA-enriched foods, mainly including neuroprotection, anti-insomnia, anti-depression, anti-hypertensive, anti-diabetes, and anti-inflammatory are also summarized. The main challenges for future research on GABA are related to exploring high GABA producing strains, enhancing the stability of GABA during storage, and developing emerging enrichment technologies without affecting food quality and other active ingredients. A better understanding of GABA may introduce new windows for its application in developing functional foods.

Abstract:The present experiment was conducted to investigate the effects of γ-aminobutyric acid (GABA) on the growth performance, immunity, antioxidant capacity, and intestinal microbiota of growing minks. One hundred minks were evenly allocated across five groups, with each group consisting of 10 males and 10 females. The minks in these groups were fed a basal diet supplemented with γ-aminobutyric acid (GABA) at 0 (control), 10, 20, 30, and 40 mg/kg of diet, respectively. The experiment lasted for eight weeks. The results showed that GABA significantly affected immunity, antioxidant capacity, and intestinal microbiota (p < 0.05). Compared to the control minks, minks in 20, 30, and 40 mg/kg GABA group had greater total protein quantitative (TP), immunoglobulin A (IgA), immunoglobulin M (IgM) content, total antioxidant capacity (T-AOC), and glutathione peroxidase (GSH-Px) activities in serum as well as interleukin-4 (IL-4) level in jejunal mucosa (p < 0.05), and had less serum blood urea nitrogen (BUN) content (p < 0.05). Furthermore, compared with the control, the supplementation of GABA at 30 mg/kg of diet improved average daily feed intake (ADFI) (p < 0.05), increased immunoglobulin G (IgG) content in serum, interleukin-10 (IL-10) and secreted immunoglobulin A (SIgA) levels in jejunal mucosa, and decreased jejunal mucosal interleukin-2 (IL-2), interleukin-12 (IL-12), and interferon-γ (IFN-γ) levels (p < 0.05). The weight and feed intake of males were higher than females, and the feed/gain ratio (F/G) was lower than females (p < 0.05). Males also had greater serum superoxide dismutase (SOD) and GSH-Px activities, and jejunal mucosa IL-2, IL-4, IL-12, SIgA, and IFN-γ levels (p < 0.05), and males had less serum IgA, IgM, and T-AOC contents, and jejunal mucosal tumor necrosis factor-α (TNF-α) level (p < 0.05). The results suggest that the supplementation of GABA at 30 mg/kg of diet can improve immune status and antioxidant capacity, and modulate the intestinal microbiota abundance of growing minks.
Gama-aminobutyric Acid (GABA) Alleviates Hepatic Inflammation Via GABA Receptors/tlr4/nf-Κb Pathways in Growing-Finishing Pigs Generated by Super-Multiparous Sows

Abstract:The offspring of super-multiparous sows face problems such as decreased growth performance, poor meat quality and even diseases in animal husbandry. Gama-aminobutyric acid (GABA) has long been known to promote growth and suppress inflammation, but little is known about the mechanisms. A total of 72 growing-finishing pigs from the 8th generation were randomly allotted to 2 groups with 6 replicates per treatment to receive a corn-soybean basal diet or the basal diet supplemented 20 mg/kg GABA for 60 d. After the animal-trial period, samples of serum and liver were collected for further analysis. Additionally, a lipopolysaccharide (LPS)-induced inflammatory model using HepG2 cells was established to explore the role of GABA on regulating hepatic inflammation. The results indicated that inflammatory cell infiltration occurs in the liver of progeny of super-multiparous sows, and dietary supplementation with GABA influenced liver morphology, increased activities of antioxidant enzymes and decreased the expression abundance of pro-inflammatory cytokines, including tumor necrosis factor-alpha (TNF alpha) and interleukin (IL)-1 beta, in the liver of growing-finishing pigs (P < 0.05). In addition, GABA supplementation increased mRNA expressions of peroxisome proliferator-activated receptor gamma (PPAR gamma) and GABA receptors (GABARs), and reduced the expression of toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-kappa B) signaling (P < 0.05). Additionally, an in vitro experiment demonstrated that GABA decreased the expressions of hepatic TLR4/NF-kappa B signaling via activating GABARs under LPS-stress (P < 0.05). In summary, liver injury may affect the growth performance of growing-finishing pigs by changing hepatic mitochondrial metabolism, the expression of pro-inflammatory cytokines and TLR4/NF-kappa B pathway and that GABA supplementation has a restorative effect by acting on GABARs.
