Optimization and Evaluation of Type 2 Diabetes Induced by High-Fat Diet and Low Dose of STZ in C57BL/6J Mouse Model

Abstract:Aim To optimize a rodent model of type 2 diabetes that would replicate the metabolic characteristics of the human syndrome and be suitable for pharmaceutical research.Methods By adopting the orthogonal experimental design method involving three factors and three levels,three main factors affecting the model including the frequency,dose and time of STZ injection were studied systematically.Different batches of mice were used and observed for more than 8 weeks to investigate the stability and reproducibility of the best combination.The suitability of the optimized Fat-fed/STZ mice model was determined for pharmaceutical testing,the response of this model to commonly used drugs for treatment of type 2 diabetes were tested.Results The result indicated that injection of streptozotocin(85mg/kg)twice every five days at the third week is the best for establishing rodent type 2 diabetes model.The optimized model was stable and reproducible.The blood glucose concentration was stable and could sustain until eight weeks after STZ injection.Biopsy also showed appropriate destruction of islet.Finally,Fat-fed/STZ mice were sensitive to metformin and pioglitazone.But Glimeperide still showed hypoglycemic effect at dose of up to 0.3mg/kg.Conclusion The rodent model of type 2 diabetes is successfully optimized,and there will be more convenient in evaluating and screening antidiabetic compounds.
