Abstract:Ovomucoid,ovalbumin,ovotransferrin and lysozyme are the major egg allergens,the allergenicity of which can be decreased by proper physical,chemical or biological process.The hypoallergenic egg products originated from physical process can be used as food and immunotherapy medicine for the population slightly allergic to egg,while the feasibility of industrial production of the hypoallergenic egg products originated from chemical or biological process still needs to be further investigated due to their potential safety risk.What's more,food processing can influence the structure of allergen protein,such as the disulfide bonds and high-level even primary structure,resulting in the change of its allergenicity.In short,the impact of processing on the structure and allergenicity of egg allergens is a scientific issue deserves in-depth exploration.Understanding this is very important for the development of hypoallergenic and non-allergenic egg products.

Abstract:Objective To investigate the immunogenicity of synthetic hen's egg protein components (Gal d1, Gal d2, Gal d3 and Gal d4) by using plasmid transduction, and the correlations among hen's egg protein components, specific IgE (f1) and the skin test results among hen's egg-allergic patients.Methods Hen's egg protein components were synthesized by using plasmid transduction, whereas the immunogenicity of the target proteins was analyzes based on Western blot.The clinical diagnostic values of each protein component were evaluated, based on the binding of protein components with specific IgE of hen's egg-allergic patients.Results Immunogenicity was shown successfully among synthetic hen's egg protein components (Gal d1, Gal d2, Gal d3 and Gal d4).The positive rate of skin prick test (SPT) of patients whose hen's egg specific IgE (f1)>10 kU/L was significantly higher than of patients whose specific IgE (f1)<10 kU/L (P=0.000 3).Ovomucin (Gal d1, D450 nm=0.81) and ovalbumin (Gal d2, D450 nm=0.567) are the major protein components of hen's egg allergen.Ovalbumin (Gal d2) specific IgE level was higher among patients whose hen's egg specific IgE (f1)>10 kU/L, whereas ovomucin (Gal d1) specific IgE level higher among patients whose hen's egg specific IgE (f1)>10 kU/L.Conclusion Immunogenicity was shown successfully among synthetic hen's egg protein components (Gal d1, Gal d2, Gal d3 and Gal d4), which should be of value for the purification of food allergens, and also of importance in the diagnosis of hen's egg allergy.

Abstract:Hen's egg is regarded as one of the eight major allergic foods by FAO,and the allergenicity can be reduced by proper processing,in which enzymatic hydrolysis is the most effective method.In this paper,four major allergens present in egg were described,and the impact of enzymatic hydrolysis on the structure and allergenicity of major allergens in egg were reviewed in detail.It offered the scientific basis for producing the hypoallergenic or non-allergenic egg products.

Abstract:BACKGROUND AND OBJECTIVE:Reducing the allergenicity of food allergens can suppress the clinical symptoms of food allergy. The objective of the present study was to investigate the effects of processing on the allergenic properties of hen's egg white proteins.METHODS:Eggs were processed by traditional Chinese cooking, including steaming, water boiling, frying, spicing and tea boiling. The contents of processed egg protein were assessed by sodium dodecyl sulfate polyacrylamide gel electrophoresis; the allergenicity was evaluated by Western blotting, enzyme-linked immunosorbent assay and enzyme allergosorbent test inhibition. Circular dichroism spectrum analysis of four major egg allergens from various egg products was performed as well. A mouse model of food allergy was developed to test the allergenicity of processed egg protein in vivo.RESULTS:Protein degradation was significant following tea boiling and spiced-tea boiling. The total allergenic potential of water-boiled egg and fried egg was relatively higher than that of steamed egg, spiced egg and tea-boiled egg. Challenge with proteins from raw egg, water-boiled egg and fried egg induced skewed T-helper 2 pattern responses (Th2 responses) in the intestine of mice sensitized to egg proteins; however, when the mice sensitized to egg proteins were challenged with proteins from steamed egg, spiced egg and tea-boiled egg, respectively, only weak Th2 responses were induced in their intestine.CONCLUSION:Processing by steaming, spicing, or tea boiling can weaken the allergenicity of egg proteins.
Preparation and Immunological Reactions of A Purified Egg Allergen Ovotransferrin

Abstract:Ovotransferrin is one of the major allergens in egg white. To define the properties of ovotransferrin, a simple protocol for ovotransferrin purification was developed and a broad physicochemical and immunological assay were used for the characterization. The prepared ovotransferrin revealed a single band on sodium dodecyl sulfate polyacrylamide gel electrophoresis, and had an estimated purity of more than 97%. The identity of the prepared protein identified by mass spectrometry analysis demonstrated that it was iron-free ovotransferrin with a molecular weight of 77,726 Da and pI value of 6.85, respectively, and its nature folded structure was kept well as defined by circular dichroism spectroscopy. Western blotting and enzyme-linked immunosorbent assays showed strong immune reactions between the prepared ovotransferrin and rabbit polyclonal antibody against ovotransferrin/egg-allergic patients' sera, suggesting that the prepared ovotransferrin bound to IgG and IgE strongly. In addition, 24 of the potential linear IgE binding epitopes on the prepared ovotransferrin were predicted using a web-based allergen database for food safety. The preparation method of ovotransferrin in the present work might provide scientific support to the authenticity of ovotransferrin for developing certified reference materials.
Differences in Protein Composition and Functional Properties of Egg Whites from Four Chicken Varieties

Abstract:Egg white protein (EWP) is widely used in the food industry for its technological properties, which are affected by diet-and breed-related changes in its composition. Therefore, the objective of this study was to analyse the relationship between hen variety and the protein composition, functional characteristics, and physicochemical properties of egg white (EW) obtained from Black Bone (BB), Gushi (GS), Hy-Line (HY-LINE), and Harbin White (HW) chickens. The protein composition of EW was analysed by high performance liquid chromatography (HPLC), while the secondary structure of EWP was assessed by Raman spectroscopy. BB and GS EWs had significantly higher ovalbumin content (183.38 and 168.56 mu g/mL, respectfully) than HY-LINE and HW EWs (148.15 and 147.44 mu g/mL, respectfully) (p < 0.05). In accordance with the higher ovalbumin content, BB and GS EWs possessed significantly better foaming and gel properties, along with greater sulfhydryl group and disulfide bond content in the EWPs, than HY-LINE and HW EWs (p < 0.05). Additionally, the ovotransferrin, ovomucin, and lysozyme contents also differed significantly among EWs from the four varieties (p < 0.05). Further, the EWPs possessed significant differences in amino acid (AA) content, including essential AAs, those contributing to egg flavour, and those associated with children's growth and preterm infant health. In conclusion, differences in protein composition likely led to dissimilar functional and physicochemical properties among EWs obtained from different hen varieties, which could provide a reference for food production and genetic breeding of laying hens.

Abstract:Ovalbumin (OVA), the most abundant protein in egg whites, has been widely used in various industries. Currently, the structure of OVA has been clearly established, and the extraction of high-purified OVA has become feasible. However, the allergenicity of OVA is still a serious problem because it can cause severe allergic reactions and may even be life-threatening. The structure and allergenicity of the OVA can be altered by many processing methods. In this article, a detailed description on the structure and a comprehensive overview on the extraction protocols and the allergenicity of OVA was documented. Additionally, the information on assembly and potential applications of OVA was summarized and discussed in detail. Physical treatment, chemical modification, and microbial processing can be applied to alter the IgE-binding capacity of OVA by changing its structure and linear/sequential epitopes. Furthermore, research indicated that OVA could assemble with itself or other biomolecules into various forms (particles, fibers, gels, and nanosheets), which expanded its application in the food field. OVA also shows excellent application prospects, including food preservation, functional food ingredients and nutrient delivery. Therefore, OVA demonstrates significant investigation value as a food grade ingredient.
Effect of Thermal Processing on Structure of Four Major Egg White Allergens

Abstract:In order to explore the impact of thermal processing on the structure of egg white allergens, four major allergens in hen's egg white were heated under different conditions of temperature and time. Circular dichroism, ultraviolet and fluorescence spectroscopy were used to detect the spatial structure of allergens. Results showed that ovotransferrin was unstable to heat, and its molecular structure was unfolded, with the aromatic residues and hydrophobic groups exposed on the molecular surface after heating. The secondary structure of ovalbumin became more ordered, ultraviolet absorbance value decreased, and surface hydrophobicity increased when it was heated slightly; however, with increasing temperature and heating time, ovalbumin became unordered and aggregated. The secondary structure of ovomucoid was relatively stable, and its molecules were unfolded after mild heating, leading to increased absorption and fluorescence intensities, and then aggregated partly after further heating. The secondary structure of lysozyme became disordered with the aromatic residues and hydrophobic groups gradually exposed after heating, but with increasing degree of heating, part of lysozyme became aggregated. The present study may provide a theoretical basis for studying the effect of heating on the allergenicity of egg allergens.

Abstract:A highly sensitive sandwich enzyme-linked immunosorbent assay (ELISA) kit was established for quantifying ovomucoid from hen's egg white, which has been considered as one of the major allergen in egg white. The detection limit reached 0.041 ng/mL, and linearity ranged from 0.1 to 6.25 ng/mL. Intra- and interassay coefficient variations were all lower than 5% at three concentrations (0.5, 2.5, and 5 ng/mL). No cross-reactivity was observed with bovine serum, horse serum, goat serum, human serum, duck egg white, goose egg white, quail egg white, and pigeon egg white, but a low level of cross-reactivity was found with chicken serum. The ELISA kit was established on the basis of two monoclonal antibodies (mAbs) recognizing different epitopes of ovomucoid. However, these mAbs Were generated using commercially purified ovalbumin as immunogen. Studies on the relative allergenicity and antigenicity of egg white protein have been performed by many researchers, but there were controversial opinions reported previously because of the impurity of each egg white protein used in various studies. In the present work we measured the degree of ovomucoid contamination in commercially purified ovalbumin sample, and the value was about 11%. We also determined the ovomucoid residue in influenza vaccine samples for the first time. These data-showed that the ELISA kit we established could serve as an effective method for precisely quantifying concentrations of ovomucoid in the egg industry and as a useful tool for the research of allergenicity and antigenicity of hen's egg proteins.
Identification of Egg White Proteins and Divergence in the Regulatory Region of the Ovalbumin Gene in Avians.

Abstract:Egg white proteins play an important role in avian reproductive systems and are an ideal resource for bioreactor construction. In this study, 1D electrophoresis and MALDI-TOF-MS were performed to analyze egg white proteins in four species. In total, 18, 11, 28, and 13 proteins were identified in the egg whites of the chicken, duck, goose, and pigeon, respectively. Egg white proteins in chickens have been studied previously; therefore, we focused on the proteins in goose and duck egg whites. Based on the amino acid sequence analysis and a comparison of the unique peptides, high similarity was observed between the goose and duck egg whites. Diversity in the regulatory region of the ovalbumin gene explained the higher ovalbumin expression in the duck and goose than in the chicken. These data clarify the evolutionary processes underlying to the unique peptides contributing to the differential expression of ovalbumin in avians.
Comparative Studies On the Allergenicity of Native Ovalbumin and Ovomucoid in Biological and Serologic Assays

Abstract:This study aimed to assess quantitatively the ability of native egg white allergens, ovalbumin (OVA) and ovomucoid (OVM), to release mediators from a passively sensitized rat basophil leukemia (RBL-2H3) cells and compare these functional results with findings on IgE immunoblots assay. RBL-2H3 cells were passively sensitized with murine IgE raised against OVA or OVM, and stimulated by serial dilutions of purified allergens in a dose-related manner. The allergen-specific degranulation was quantified by measuring the release of β-hexosaminidase in the supernatant. The intensity of the degranulation was expressed as EC50, corresponding to the dose of allergen that induced 50% of the maximum release. In addition, immunoreactivity was assessed by IgE immunoblotting using murine sera with IgE raised against egg protein extract. In the biological assay, the medians of EC50 obtained for OVA and OVM were 6 and 78 ng/ml, respectively, which indicated a higher degranulation potential for OVA compared with OVM (P < 0.05). Consistent with findings on the biological assay, IgE immunoblots with purified OVA and OVM showed substantially lighter binding of IgE to OVM compared with OVA. In the native egg white allergens, OVA is more potent than OVM for pooled murine IgE using an mediator release assay in combination with IgE immunoblots assay. Since eggs are heated to a certain degree prior to consumption, further studies are needed to compare the allergenicity of heated ovalbumin and ovomucoid based on the experiments reported here.
Specific Antibodies Detection in Egg-Allergic Children

Abstract:Objective To detect the serum levels of sIgE and IgG4 in egg-allergic children, and explore the influences of thermal processing on allergenicity of five different eggs. Methods The antigen were extracted from raw and denatured eggs. The eggs had been boiled 10 minutes for denaturazation. Serum sIgE and IgG4 were determined with ELISA in 30 egg-allergic children. Results For raw hen eggs, duck eggs, goose eggs, common quail eggs and pigeon eggs, the sIgE positive rates were 73.33% , 70.00% , 53.33% , 43.33% , 40.00% respectively; the IgG4 positive rates were 96.67%, 90.00%, 93.33%, 83.33%, 53.33% respectively. The sIgE positive rate of hen eggs and duck eggs were significantly higher than those of goose eggs, quail eggs and pigeon eggs (Pearson Chi-square χ2 = 11.201, P = 0.024). The IgG4 positive rate of hen eggs and goose eggs was significantly higher than those of duck eggs, quail eggs and pigeon eggs (Pearson Chi-square χ2 = 26.400, P = 0.000). For denatured hen eggs, duck eggs, goose eggs, common quail eggs and pigeon eggs, the sIgE positive rates were 70.00%, 63.33%, 46.67%, 43.33%, 33.33% respectively; IgG4 positive rates were 93.33%, 90.00%, 83.33%, 80.00%, 46.67% respectively. The sIgE positive rate was highest in hen eggs and lowest in pigeon eggs (Pearson Chi-square χ2 = 10.834, P = 0.028). The IgG4 positive rates of hen eggs and goose eggs were higher than those of quail eggs and pigeon eggs. Pearson Chi-square χ2 = 24.868, P = 0.000. There's no statistically significant difference in sIgE and IgG4 between the two groups. Conclusions Thermal processing has effects on immunogenicity of eggs and tend to decrease the immunogenicity. However, the immunogenicity has been partly kept and is still able to stimulate sIgE and IgG4. In these five different eggs, the hen eggs and duck eggs maybe have a higher allergenicity than pigeon eggs. However it needs more explorations. To avoid egg allergy, the egg-allergic children should avoid anything containing eggs.
Variation in the Allergenicity of Scrambled, Boiled, Short-Baked and Long-Baked Egg White Proteins

Abstract:BACKGROUND: Hen's egg white (HEW) is the most common cause of food allergy in children which induces mild to fatal reactions. The consultation for a proper restriction is important in HEW allergy. We aimed to identify the changes in HEW allergenicity using diverse cooking methods commonly used in Korean dishes.METHODS: Crude extract of raw and 4 types of cooked HEW extracts were produced and used for sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE), enzyme-linked immunosorbent assay (ELISA), and ELISA inhibition assays using 45 serum samples from HEW allergic and tolerant children. Extracts were prepared; scrambled without oil for 20-30 seconds in frying pan without oil, boiled at 100°C for 15 minutes, short-baked at 180°C for 20 minutes, and long-baked at 45°C for 12 hours with a gradual increase in temperature up to 110°C for additional 12 hours, respectively.RESULTS: In SDS-PAGE, the intensity of bands of 50-54 kDa decreased by boiling and baking. All bands almost disappeared in long-baked eggs. The intensity of the ovalbumin (OVA) immunoglobulin E (IgE) bands did not change after scrambling; however, an evident decrease was observed in boiled egg white (EW). In contrast, ovomucoid (OM) IgE bands were darker and wider after scrambling and boiling. The IgE binding reactivity to all EW allergens were weakened in short-baked EW and considerably diminished in long-baked EW. In individual ELISA analysis using OVA+OM+ serum samples, the median of specific IgE optical density values was 0.435 in raw EW, 0.476 in scrambled EW, and 0.487 in boiled EW. Conversely, it was significantly decreased in short-baked (0.406) and long-baked EW (0.012). Significant inhibition was observed by four inhibitors such as raw, scrambled, boiled and short-baked HEW, but there was no significant inhibition by long-baked HEW (IC50 > 100 mg/mL).CONCLUSION: We identified minimally reduced allergenicity in scrambled EW and extensively decreased allergenicity in long-baked EW comparing to boiled and short-baked EW as well as raw EW. By applying the results of this study, we would be able to provide safer dietary guidence with higher quality to egg allergic children.

Abstract:Chicken egg whites contain hundreds of proteins, and are widely used in the food, biological and pharmaceutical industries. It is highly significant to study the separation and purification of egg white proteins. This review first describes the structures and functional properties of several major active proteins in egg whites, including ovalbumin, ovotransferrin, ovomucoid, lysozyme, ovomucin, ovomacroglobulin and avidin. Then, the common techniques (including precipitation, chromatography and membrane separation) and some novel approaches (including electrophoresis, membrane chromatography, aqueous two-phase system and molecular imprinting technology) for the separation and purification of egg white proteins broadly reported in the current research are introduced. In addition, several co-purification methods for simultaneous separation of multiple proteins from egg whites have been developed to improve raw material utilization and reduce costs. In this paper, the reported techniques in the last decade for the separation and purification of chicken egg white proteins are reviewed, discussed and prospected, aiming to provide a reference for further research on egg proteins in the future.

Abstract:Antioxidant peptide is one of hotspots in the study of biological field,due to the features of high activity,small molecular weight and easy absorption. Egg is rich in protein,and its amino acid composition is close to the proportion of human essential amino acid,so it is a good source of preparation of antioxidant peptides. Antioxidant peptides derived from egg protein was reviewed,and its antioxidant activity and the relationship between structure and activity. In addition,this paper pointed out the problems existing in the research of antioxidant peptides derived from egg protein,and made outlooks about the research and application of antioxidant peptides derived from egg,these might offer some references for the related research.

Abstract:Currently,the egg production in China is the largest all over the world,but there are few high added-value products.It is very important to develop egg products with high value.Egg white protein contains essential amino acids for humans,and the amino acid composition is close to human demand.However,egg white protein has disadvantages such as high viscosity,gelling,foaming and allergy so that it is not suitable for use in the food industry.Hydrolysis can not only improve the functional properties of egg white protein,but also the hydrolysates have multiple biological activities such as antihypertensive activity,antioxidant property,anti-thrombin property,antimicrobial activity and immunomodulatory activity.The hydrolysates can be applied in the food industry widely.Therefore,in this paper,processing conditions of egg white protein as well as functional properties,bioactive activities and application in the food industry of egg white hydrolysates are described,which will provide new strategies for the utilization and development of egg white protein.

Abstract:In some food industries, the demand for egg yolk was large, and egg white was often discarded as waste, which not only causes environmental pollution, but also wastes this valuable protein resource. Therefore, the comprehensive utilization of egg white is urgent. The research at home and abroad is currently focused on lysozyme extraction methods, egg white cold sterilization technology, the functional properties and the biological activities of egg white hydrolysates, and recent progress on the 4 aspects were reviewed in this article.
