Abstract:D-tagatose,a novel low-calorie sugar-substituting sweetener,possesses multi-functionality applied in food and pharmaceuticals,such as anti-hyperglycemia and probiotics.In this article,the properties and functions,the anti-hyperglycemic mechanism,the applications in food and pharmaceuticals,and the development of production technology of tagatose were reviewed.Furthermore,the gene cloning and screening,the enzymatic engineering and the immobilization of L-arabinose isomerase,the tool enzyme for tagatose production,were elaborated.

Abstract:The functional sweetener xylooligosaccharides is one kind of new product developed for the patient, infant and pre-school children, aged people and subhealthy crowd. Nowadays, the sweetener produced and sold on the consumer market is low-grade. According to the balanced matching of result price of nutrition. Bagasse was used as raw materials by enzymatic methods to produce a new functional sweetener. The technology characteristic solves enzymes to raw materials, the uniqueness of bottom materials is strong, the product is high in purity. It is looked as milky white or yellowish powder. Xylooligosaccharides is apt to dissolve in water. The new functional sweetener tastes pure and delightful. It has a good acid- and heat-resisting stability. So there is a remarkable potential for xylooligosaccharides on practical utilization in many fields, including health products, food and drink, and with beneficial social and economy results.

Abstract:d-tagatose is a low-calorie alternative to sucrose natural monosaccharide that is nearly as sweet. As a ketohexose, d-tagatose has disease-relieving and health-promoting properties. Due to its scarcity in nature, d-tagatose is mainly produced through chemical and biological methods. Compared to traditional chemical methods, biological methods use whole cells and isolated enzymes as catalysts under mild reaction conditions with few by-products and no pollution. Nowadays, biological methods have become a very important topic in related fields due to their high efficiency and environmental friendliness. This paper introduces the functions and applications of d-tagatose and systematically reviews its production, especially by l-arabinose isomerase (L-AI), using biological methods. The molecular structures and catalytic mechanisms of L-AIs are also analyzed. In addition, the properties of L-AIs from different microbial sources are summarized. Finally, we overview strategies to improve the efficiency of d-tagatose production by engineering L-AIs and provide prospects for the future bioproduction of d-tagatose.

Abstract:Sweeteners are commonly used as food additives in our daily life, which, however, have been causing a number of undesirable diseases since the last century. Therefore, the detection and quantification of sweeteners are of great value for food safety. In this study, we used a taste biosensor to measure and analyze different sweeteners, both natural and artificial sweeteners included. Electrophysiological activities from taste epithelium were detected by the multi-channel biosensors and analyzed with spatiotemporal methods. The longtime signal result showed different temporal-frequency properties with stimulations of individual sweeteners such as glucose, sucrose, saccharin, and cyclamate, while the multi-channel results in our study revealed the spatial expression of taste epithelium to sweet stimuli. Furthermore, in the analysis of sweetener with different concentrations, the result showed obvious dose-dependent increases in signal responses of the taste epithelium, which indicated promising applications in sweetness evaluation. Besides, the mixture experiment of two natural sweeteners with a similar functional unit (glucose and sucrose) presented two signal patterns, which turned out to be similar with responses of each individual stimulus involved. The biosensor analysis of common sweeteners provided new approaches for both natural and artificial sweeteners evaluation.

Abstract:d-Tagatose is one of the several healthy sweeteners that can be a substitute for sucrose and fructose in our daily life. Whole cell-catalyzed phosphorylation and dephosphorylation previously reported by our group afford a thermodynamic-driven strategy to achieve tagatose production directly from starch with high product yields. Nonetheless, the poor structural stability of cells and difficulty in biocatalyst recycling restrict its practical application. Herein, an efficient and stable semiartificial cell factory (SACF) was developed by constructing an organosilica network (OSN) artificial shell on the cells bearing five thermophilic enzymes to produce tagatose. The OSN artificial shell, the thickness of which can be regulated by changing the tetraethyl silicate concentration, exhibited tunable permeability and superior mechanical strength. In contrast with cells, SACFs showed a relative activity of 99.5% and an extended half-life from 33.3 to 57.8 h. Over 50% of initial activity was retained after 20 reuses. The SACFs can catalyze seven consecutive reactions with tagatose yields of over 40.7% in field applications.

Abstract:Natural sweeteners generally refer to a sweet chemical component directly extracted from nature or obtained through appropriate modifications, mainly secondary metabolites of plants. Compared to the first-generation sweeteners represented by sucrose and the second-generation sweeteners represented by sodium cyclamate, natural sweeteners usually have high sweetness, low-calorie content, good solubility, high stability, and rarely toxic side effects. Historically, researchers mainly focus on the function of natural sweeteners as substitutes for sugars in the food industry. This paper reviews the bioactivities of several typical natural sweeteners, including anti-cancer, anti-inflammatory, antioxidant, anti-bacterial, and anti-hyperglycemic activities. In addition, we have summarized the extraction, physicochemical properties, and application of natural sweeteners. The article aimed to comprehensively collate vital information about natural sweeteners and review the potentiality of tapping bioactive compounds from natural products. Hopefully, this review provides insights into the further development of natural sweeteners as therapeutic agents and functional foods.

Abstract:Steviol glycosides (SGs) have been widely used as a natural non-calorie sweetener and a sugar substitute in food and beverage industry. Recent studies have demonstrated that SGs have notable interfacial activity and can be used as a new type of food-grade biosurfactants. The unique combination of high-intensity sweetness, biological activity and interfacial properties makes SGs have great potential as multifunctional ingredients for novel applications in food processing. Herein, we will first introduce the biological benefits of SGs and the strategies to improve their organoleptic properties. The micellar self-assembly of SGs in aqueous solutions and their interfacial behaviors are then discussed. The recent novel applications of SGs in active encapsulation and the stabilization of plant-based multiphase food systems are reported.

Abstract:Sugar is crucial as an essential nutrient for humans as well as for providing texture, sweetness and so on to food. But with the rise in people's pursuit of health, it is becoming increasingly clear that excessive consumption of sugar can locate a load on the body. It has been that excessive sugar is associated with many diseases, such as dental caries, obesity, diabetes, and coronary heart disease. Therefore, researchers and industries are trying to reduce or substitute sugar in food without affecting the sensory evaluation. Substituting sugar with sweeteners is alternatively becoming the most traditional way to minimize its use. So far, the sweeteners such as stevia and xylitol have been are commercially applied. Several studies have shown that technological innovation can partially compensate for the loss in sweetness as a result of sugar reduction, such as cross-modal interactions that stimulate sweetness with aroma, nanofiltration that filters disaccharides and above, enzyme-catalyzed sugar hydrolysis, and microbial fermentation that turns sugar into sugar alcohol. This review summarizes these studies to enhance the safety and quality of sugar-reduced products, and will provide some theoretical frameworks for the food industry to reduce sugar in foods, meet consumers' needs, and promote human health. Abstract:Due to healthy eating trends, the needs of diabetics, and economic advantages, the production of sweeteners, their range, and the range of beverages containing them are constantly expanding. At the same time, there are difficulties associated with the necessary selection of sweetening agents to solve specific problems. The data of modern scientific literature on the technological aspects of the use of sweeteners in the production of soft drinks are summarized and their choice for obtaining drinks of the corresponding functional direction is substantiated. All sweeteners can be divided into six groups: sugars, sweeteners (sweet alcohols), synthetic sweeteners, natural sweeteners, natural sugary products, and sugary products derived from starch or sugar. The main properties of various sweeteners are compared: sweetness factor, caloric content, glycemic index. The potential adverse effects that may occur when consuming beverages prepared with their use are analyzed. Natural sugary products are generally less sweet than sugar, while artificial sweeteners have a higher sweetness than sucrose. However, the risks and disadvantages of synthetic sweeteners outweigh the benefits of their use. In this regard, it is advisable to reduce or completely eliminate the consumption of food products containing sweeteners from the human diet. On the other hand, natural sources of sweeteners should be used to eliminate the negative effects associated with sugar consumption. When choosing sweeteners for functional beverages, many criteria are taken into account: sweetness factor, caloric content, effect on blood insulin levels (glycemic index), effect on the human digestive system, effect on dental health, and allowable daily intake. For the widespread introduction of natural sweeteners into beverage formulations, detailed research into how they are processed and used on an industrial scale is required.

Abstract:Improved human material living standards have resulted in a continuous increase in the rate of obesity caused by excessive sugar intake. Consequently, the number of diabetic patients has skyrocketed, not only resulting in a global health problem but also causing huge medical pressure on the government. Limiting sugar intake is a serious problem in many countries worldwide. To this end, the market for sugar substitute products, such as artificial sweeteners and natural sugar substitutes (NSS), has begun to rapidly grow. In contrast to controversial artificial sweeteners, NSS, which are linked to health concepts, have received particular attention. This review focuses on the extraction technology and biomedical function of NSS, with a view of generating insights to improve extraction for its large-scale application. Further, we highlight research progress in the use of NSS as food for special medical purpose (FSMP) for patients.

Abstract:In recent years, the worldwide increase in lifestyle diseases and metabolic disorders has been ascribed to the excessive consumption of sucrose and added sugars. For this reason, many approaches have been developed in order to replace sucrose in food and beverage formulations with alternative sweetening compounds. The raising awareness concerning the synthetic sweeteners due to their negative impact on health, triggered the need to search for alternative substances. Natural sweeteners may be classified in: (i) non-nutritive (e.g., neohesperidine dihydrochalcone, thaumatin, glycyrrhizin mogroside and stevia) and (ii) bulk sweeteners, including both polyols (e.g., maltitol, mannitol, erythritol) and rare sugars (e.g., tagatose and allulose). In this review we discuss the most popular natural sweeteners and their application in the main food sectors (e.g., bakery, dairy, confectionary and beverage), providing a full understanding of their impact on the textural and sensory properties in comparison to sucrose. Furthermore, we analyze the use of natural sweeteners in blends, which in addition to enabling an effective replacement of sugar, in order to complement the merits and limits of individual compounds. Finally, microencapsulation technology is presented as an alternative strategy to solving some issues such as aftertaste, bitterness, unpleasant flavors, but also to enhance their stability and ease of use.

Abstract:This multidisciplinary study details the biosynthesis of novel non-digestible oligosaccharides derived from rare sugars, achieved through transfructosylation of D-tagatose and L-sorbose by levansucrase from Bacillus subtilis CECT 39 (SacB). The characterization of these carbohydrates using NMR and molecular docking was instrumental in elucidating the catalytic mechanism and substrate preference of SacB. Tagatose-based oligosaccharides were higher in abundance than L-sorbose-based oligosaccharides, with the most representative structures being: β-D-Fru-(2→6)-β-D-Fru-(2→1)-D-Tag and β-D-Fru-(2→1)-D-Tag. In vitro studies demonstrated the resistance of tagatose-based oligosaccharides to intestinal digestion and their prebiotic properties, providing insights into their structure-function relationship. β-D-Fru-(2→1)-D-Tag was the most resistant structure to small-intestinal digestion after three hours (99.8% remained unaltered). This disaccharide and the commercial FOS clustered in similar branches, indicating comparable modulatory properties on human fecal microbiota, and exerted a higher bifidogenic effect than unmodified tagatose. The bioconversion of selected rare sugars into β-fructosylated species with a higher degree of polymerization emerges as an efficient strategy to enhance the bioavailability of these carbohydrates and promote their interaction with the gut microbiota. These findings open up new opportunities for tailoring natural rare sugars, like D-tagatose and L-sorbose, to produce novel biosynthesized carbohydrates with functional and structural properties desirable for use as emerging prebiotics and low-calorie sweeteners.

Abstract:Carbohydrates have a dietary role, but excessive consumption of high-calorie sugars can contribute to an increased incidence of metabolic diseases and dental caries. Recently, carbohydrates with sweetening properties and low caloric value, such as D-tagatose, have been investigated as alternative sugars. D-tagatose is a rare sugar that has nutritional and functional properties of great interest for health. This literature review presents an approach to the biological effects of D-tagatose, emphasizing its benefits for oral health. Studies report that D-tagatose has antioxidant and prebiotic effects, low digestibility, reduced glycemic and insulinemic responses, and the potential to improve the lipid profile, constituting an alternative for diabetes mellitus and obesity. It can also be observed that D-tagatose has an antioxidant action, favoring the elimination of free radicals and, consequently, causing a reduction in cellular oxidative stress. Furthermore, it also has antibacterial potential against oral species. Regarding oral health, studies have shown that D-tagatose efficiently reversed bacterial coaggregations, including periodontopathogenic species, and impaired the activity and growth of cariogenic bacteria, such as S. mutans. D-tagatose significantly inhibited biofilm formation, pH decrease and insoluble glucan synthesis in S. mutans cultures. Salivary S. mutans counts were also significantly reduced by the consumption of chewing gum containing D-tagatose and xylitol. In addition, there is evidence that tagatose is effective as an air-polishing powder for biofilm decontamination. The literature indicates that D-tagatose can contribute to the prevention of systemic diseases, also constituting a promising agent to improve oral health.

Abstract:d-tagatose is a ketohexose that can be used as a novel functional sweetener in foods, beverages, and dietary supplements. This study was aimed at developing a high-yielding d-tagatose production process using alginate immobilized Lactobacillus fermentum CGMCC2921 cells. For the isomerization from d-galactose into d-tagatose, the immobilized cells showed optimum temperature and pH at 65 °C and 6.5, respectively. The alginate beads exhibited a good stability after glutaraldehyde treatment and retained 90% of the enzyme activity after eight cycles (192 h at 65 °C) of batch conversion. The addition of borate with a molar ratio of 1.0 to d-galactose led to a significant enhancement in the d-tagatose yield. Using commercial β-galactosidase and immobilized L. fermentum cells, d-tagatose was successfully obtained from lactose after a two-step biotransformation. The relatively high conversion rate and productivity from d-galactose to d-tagatose of 60% and 11.1 g l−1 h−1 were achieved in a packed-bed bioreactor. Moreover, lactobacilli have been approved as generally recognized as safe organisms, which makes this L. fermentum strain an attracting substitute for recombinant Escherichia coli cells among d-tagatose production progresses.
Uncovering the Role of Impurity Sugars on the Crystallization of D-Tagatose Crystal: Experiments and Molecular Dynamics Simulations

Abstract:Impurity sugars produced in upstream process of functional sugars are significantly impacting the product quality. In this work, the effect of congeners (d-maltose (d-MAL), d-fructose (d-FRU), d-glucose (d-GLU)) on primary and secondary nucleation of d-tagatose (d-TAG) crystals was investigated. The impurity sugars showed an inhibition on primary nucleation of d-TAG crystals, while a promotion on the secondary nucleation of d-TAG. Interestingly, the impact of impurity sugars on d-TAG crystal growth was similar to that on primary nucleation. The diffusion ability, hydrogen bonding forming ability, interaction energy of d-TAG crystal surfaces and impurity sugars were evaluated by molecular dynamics (MD) simulations to reveal the nucleation and growth behavior. Based on the above findings, we designed the d-TAG crystallization experiments, and obtained d-TAG crystals with uniform particle size distribution and regular morphology. This study helps to understand the influence of impurity sugars on crystallization, guiding the industrial manufacturing of functional sugars.
