Study of neuregulin 1 protein levels of first-episode positive subtype schizophrenic patients.

Abstract:Objective To establish a method of detecting NRG1 protein and to detect the NRG1 protein levels in serum from first-episode schizophrenic patients.Methods Double antibody sandwich principle(labeled antibody: Monoclonal Anti-human NRG1/HRG1 Antibody;coated antibody: purified rabbit polyclonal antibody,and labled with 125I) was applied to establish a radioimmunoassay of detecting serum NRG1 level,and then the serum NRG1 level were measured in 33 normal controls and 21 first-episode schizophrenic patients.All patients took risperidone 4~6 mg/d four weeks.Results Compared with controls,patients exhibited lower level of NRG1 before and after 1,2 and 3 weeks of treatment.(P value is <0.001、<0.001、<0.001、0.006,respectively.) but not after 4 weeks of treatmentrence(t=1.38,P=0.17).Among treatment groups,the serum level of NRG1 was significantly higher in 4 weeks of treatment than in 1,2,or 3 weeks of treatment(P<0.05).Conclusions Double antibody sandwich method can be used to detect serum NRG1protein indirectly.Additionally,the serum level of NRG1 protein decrease in first-episodee schizophrenic patients and treatment with rispeeridone can attenuate the decrease in NRG1 protein.
