Abstract:Ligustrazine is a main active fraction of the traditional medicine known as Ligusticum chuanxiong hort, which has been used as clinical medication for cerebral thrombosis, coronary heart disease and stenocardia recently. The rapid metabolism and short half-life of ligustrazine seriously limits its application in clinical practice. Therefore, derivatives of ligustrazine are designed and synthesized in our and other labs, including piperazine, cinnamic acid, styrene, acylguanidine, amides, curcumin and triterpenes derivatives of ligustrazine. Most of these compounds present better pharmacodynamics activities and more favorable pharmacokinetic properties compared to the parent compound. Besides, some new biological activities of these compounds are discovered. Hence, this review continues the previous review of our group as well as aims to highlight recent prominent advances in this field in the past ten years.

Abstract:Recent advances in the structural modification of ligustrazine and studies on the bioactivities of ligustrazine derivatives are reviewed in this article. Due to its rapid metabolism and short half-life, ligustrazine is not suitable for application in clinical practice. Derivatives of ligustrazine were therefore designed and synthesized, including ether, amine, amide, hydrocarbon and deuterated derivatives of ligustrazine and ester and alcohol derivatives of 2-hydroxymethyl-3,5,6-trimethylpyrazine, an active metabolite of ligustrazine. Abstract:Objective:To investigate the pharmacological action of active ingredients of ligusticum wallichii based on bibliometrics in recent ten years.Methods:The literatures of pharmacological action of Ligustrazine, Ligustilide and Ferulic acid indexed in the CNKI and PubMed databases from 2005 to 2015 were analyzed by bibliometric methods.The multidimensional analysis and visualization of the literatures were conducted with bibliographic item co-occurrence matrix builder (BICOM) software and social network analysis and visualization tool (UCINET) software.Results:A total of 74 valid documents were screened out,and more researching attentions were given to Ligustrazine.Twenty high-frequency key words of Ligustrazine (≥ 4) and 17 high-frequency key words of Ferulic acid (≥ 2) were found.Taking these key words into co-occurrence analysis;Ligustrazine and Ferulic acid research hotspots were clustered.Conclusion:The pharmacological effects of Ligustrazine are mainly focused in the fields of ischemia reperfusion injury and apoptosis;and ferulic acid are focused in Alzheimer′s disease;apoptosis and oxidative stress.

Ligustrazine Exerts Cardioprotection in Animal Models of Myocardial Ischemia/Reperfusion Injury: Preclinical Evidence and Possible Mechanisms

Abstract:Ligustrazine (Lig) is one of the main effective components of Ligusticum Chuanxiong Hort, which possesses a variety of biological activities in the cardiovascular system. Here, we conducted a preclinical systematic review to investigate the efficacy of Lig for animal models of myocardial ischemia/reperfusion injury and its possible mechanisms. Twenty-five studies involving 556 animals were identified by searching 6 databases from inception to August 2017. The methodological quality was assessed by using Collaborative Approach to Meta-Analysis and Review of Animal Data from Experimental Studies (CAMARADES) 10-item checklist. All the data were analyzed using Rev-Man 5.3 software. As a result, the score of study quality ranged from 2 to 6 points. Meta-analyses showed Lig can significantly decrease the myocardial infarct size, cardiac enzymes and troponin compared with control (P < 0.01). The possible mechanisms of Lig for myocardial infarction are antioxidant, anti-inflammatory, anti-apoptosis activities and improving coronary blood flow and myocardial metabolism. In conclusion, the findings indicated that Lig exerts cardio protection through multiple signaling pathways in myocardial ischemia/reperfusion injury.

Abstract:The article reviewed the research progress of ligustilide in recent years and elaborated its pharmacological functions and mechanisms in detail, especially in ischemic brain injury. Its mechanism includes reducing cerebral infarct volumes and improving neurobehavioral deficits, anti-oxidant and anti-apoptosis, antithrombotic activity, calcium channel blockers function, and effect on erythropoietin. Other pharmacological effects of ligustilide including inhibiting vascular smooth muscle cell proliferation, anti-inflammatory and analgesic effects, effects on LPS-induced endotoxic shock, inhibiting constriction effect, suppression of the central nervous system, and ameliorating the memory impairment induced by scopolamine and so on, are also introduced. Ligustilide has potential pharmacological value, which provides a reference for its further research and development.
The Relation Between the Effect of Ligustrazine on Treating Pulmonary Hypertension and Endothelin

Abstract:Aim: To study the relation between the effect of ligustrazine on treating pulmonary hypertension and endothelin.Methods: Pulmonary arterial pressure was measued with right heart catherization.The levels of pulmonary arterial pressure and plasma endothelin were observed before ligustrazine was injected intravenously after 20 minutes and 45 minutes.Results: Before ligustrazine was injected intravenously,after 20 minutes and 45 minutes,the mPAP was(3.6±0.3)kPa,(2.7±0.19)kPa,(2.9±0.1)kPa respectively.The levels of plasma endothelin were(60.7±13.5)pg/mL,(25.1±12.2) pg/mL,(28.3±12.8)pg/mL respectively,the mPAP and ET after 20 minutes,45minutes compared with before ligustrazine was injected intravenously had significant difference(P0.01).Conclusion:Preventing the synthesis and secretion of endothelin may be the mechanism of ligustrazine on treating pulmonary hypertension.

Abstract:A series of novel Ligustrazinyl acylguanidines was designed, synthesized and evaluated for their protective effect on injured vascular endothelial cell (ECV-304). The preliminary results demonstrated that some compounds possessed more potent activities than that of Ligustrazine in stimulating replication of the injured endothelial cell. Among the active compounds, compounds 8b, 8f and 8l displayed remarkable antioxidative activity with low EC50 values of 0.097, 0.059 and 0.094 mM, respectively. Structure-activity relationships were briefly discussed.

Abstract:BACKGROUND: Migraine is one of the most common neurological diseases which has been treated by active substances from traditional Chinese medicine (TCM), such as ligustrazine, an extract of the Chinese herb Chuanxiong. However, the pathogenesis of migraine and the curative mechanisms of ligustrazine have remained unclear. The genes P2X3, TRPV1, ERK, and c-fos have been implicated to play a role. In this work, we attempted to elucidate the analgesic mechanism of ligustrazine using a classic migraine-representative rat model.METHODS: The migraine rat model was established by administration of nitroglycerin (NTG). Ligustrazine treatment was administered by intravenous injection. The animal's behavior was continuously recorded, and then trigeminal ganglions (TGs) were isolated. Total RNA was extracted from cells and total protein was extracted from TG. Quantitative real-time polymerase chain reaction (qRT-PCR) and western blot analyses were used to detect the levels of P2X3, TRPV1, c-Fos, and ERK in TG.RESULTS: Ligustrazine could reduce the neurological activities of NTG-induced migraine rats. The rats TG nerve showed varying degrees of expression of P2X3, TRPV1, c-Fos and ERK expression element. Ligustrazine could inhibit over-expression of P2X3, TRPV1, c-fos, and ERK in the TG nerve of NTG-induced migraine rats.CONCLUSIONS: Our results demonstrated that ligustrazine had potent activity against NTG-induced migraine rats through inhibition of the c-fos/ERK signaling pathway. This work may provide a comprehensive basis for a better understanding of the pathogenesis of migraine and the curative mechanisms of ligustrazine.

Ligustrazine improves atherosclerosis in rat via attenuation of oxidative stress.

Abstract:CONTEXT:Ligustrazine (Lig) is a compound isolated from the rhizome of Ligusticum chuanxiong Hort. (Umbelliferae) and has been reported to be effective for the treatment of a variety of vascular diseases. OBJECTIVE:The anti-atherosclerotic activities of Lig are evaluated in vivo for the first time in the present study. MATERIALS AND METHODS:We gave rats a single injection of vitamin D3 and then fed them with an atherogenic diet for 6 weeks to induce atherosclerosis. Lig was simultaneously given to rats by gavage at the dose of 20 or 80 mg/kg in the therapy groups. Multiple approaches including spectrophotometry, hematoxylin and eosin (H&E) staining, and quantitative RT-PCR were applied to investigate the effects of Lig on blood parameters, aorta and liver histology, and gene expression. In addition, the solely effects of Lig on food intake, body weight gain, and taste preference were also evaluated. RESULTS:We found that two doses of Lig treatment decreased the total cholesterol levels by 65.2 and 76.7%, respectively, in the plasma. Triglyceride (by 53.2 and 77.9%) and low-density lipoprotein (by 71.2 and 79.0%) levels were also decreased. However, high-density lipoprotein level was slightly increased. The circulating endothelial cells were decreased by 42.2 and 60.0% in Lig-treated rats, indicating the attenuation of endothelial injury. In contrast, Lig restored the total antioxidant capacity and superoxide dismutase 1 (SOD1) activity while decreasing the MDA generation. Furthermore, Lig improved liver dysfunction by decreasing ALT (by 13.0 and 49.7%) and AST (by 10.7 and 14.3%) levels. Histological examinations revealed that Lig suppressed atherosclerotic plaque progression in the thoracic aorta and lipid accumulation in the liver. At the transcriptional level, Lig inhibited the induction of antioxidant genes both in aorta and in liver. Lig also suppressed the mRNA expression of the genes involved in the hepatic fatty acid oxidation. Finally, Lig had a minimum effect on food intake, body weight gain, and taste preference. DISCUSSION AND CONCLUSION:Our results suggest that Lig suppresses the development of atherosclerosis and hepatic lipid accumulation via the alleviation of oxidative stress and the improvement of dyslipidemia.

Abstract:Cardio-cerebrovascular diseases (CVDs) are a serious threat to human health and account for 31% of global mortality. Ligusticum chuanxiong Hort. (CX) is derived from umbellifer plants. Its rhizome, leaves, and fibrous roots are similar in composition but have different contents. It has been used in Japanese, Korean, and other traditional medicine for over 2000 years. Currently, it is mostly cultivated and has high safety and low side effects. Due to the lack of a systematic summary of the efficacy of CX in the treatment of CVDs, this article describes the material basis, molecular mechanism, and clinical efficacy of CX, as well as its combined application in the treatment of CVDs, and has been summarized from the perspective of safety. In particular, the pharmacological effect of CX in the treatment of CVDs is highlighted from the point of view of its mechanism, and the complex mechanism network has been determined to improve the understanding of CX’s multi-link and multi-target therapeutic effects, including anti-inflammatory, antioxidant, and endothelial cells. This article offers a new and modern perspective on the impact of CX on CVDs.

Abstract:To study the molecular mechanism of action of ligustrazine, a low-density oligonucleotide microarray for cardiovascular disease-related genes, was constructed, and the preparation and hybridization protocols were optimized. Under the optimized conditions, the molecular mechanism of action of ligustrazine was analyzed with human umbilical vein endothelial cells. After 4 h of treatment with 100 microg/ml of ligustrazine, calcium-ATPase gene, sodium channel gene, P450c11 gene in human umbilical vein endothelial cells were up-regulated while apolipoprotein C-III gene was down-regulated. The results were shown to be reproducible. RT-PCR confirmed the results from microarray. These results suggest that ligustrazine may act on the function of human umbilical vein endothelial cells via modulating the expressions of cardiovascular disease-related genes. This also demonstrates the use of oligonucleotide microarray technology as an approach to studying targets of active components of Traditional Chinese Medicine.
Effect and Possible Mechanism of Ligustrazine on Expression of Vascular Cell Adhesion Molecule-1 and Monocyte Chemotactic Protein-1 in Cells of Wall Vascular

Abstract:Aim To observe the influence of ligustrazine on vascular cell adhesion molecule-1(VCAM-1),monocyte chemotactic protein-1(MCP-1) and nuclear factor-κB(NF-κB),expression by vascular endothelial cells and smooth muscle cells and to investigate the molecular mechanism of ligustrazine against atherosclerosis.Methods Human umbilical vein endothelial cells(hUVEC) and rat thorax aortic smooth muscle cells(SMC) were cultured and stimulated by ox-LDL,ox-VLDL,angiotensin Ⅱ and /or ligustrazine.Then,VCAM-1,MCP-1 and NF-κB expression by cultured cells were examed with immunocytochemistry staining or in situ hybridization.Adhesion test of monocytes was used to valuate the influence of ligustrazine on monocytes adhesion to endothelium. Results Comparing with contrast group(hUVEC:0.365±0.019,SMC: 0.416±0.013),ox-LDL,ox-VLDL and angiotensin Ⅱ could inhance VCAM-1 protein level both in hUVEC(0.552±0.008,0.460±0.006,0.486±0.025) and SMC(0.564±0.007,0.513±0.021,0.524±0.008)(P<0.01).As well,ox-LDL,ox-VLDL and angiotensin Ⅱ could inhance MCP-1 protein level both in hUVEC and SMC(P<0.01).By contrast,ox-LDL,ox-VLDL and angiotensin Ⅱ could inhance VCAM-1 and MCP-1 mRNA exprossionS both in hUVEC and SMC(P<0.01).And ligustrazine markedly reduced ox-LDL,ox-VLDL,angiotensin Ⅱ induced VCAM-1 and MCP-1 exprossion(P<0.01).By contrast,ox-LDL,ox-VLDL and angiotensin Ⅱpromoted NF-κB nuclear translocation both in hUVEC and SMC.Ligustrazine inhibited or blocked ox-LDL,ox-VLDL and angiotensin Ⅱ induced NF-κB nuclear translocation.Comparing with ox-LDL,and ox-VLDL group(2.047±0.011,1.936±0.014),ligustrazine reduced the amount of monocytes adhesion to endothelial cells(1.282±0.020,1.265±0.016)(P<0.01). Conclusion Ligustrazine resists atherosclerosis by inhibition of VCAM-1 and MCP-1 expression of vascular cells.This effect might be mediated by NF-κB pathway.
Effects of Ligustrazine on Myocardial Fibrosis in Rats with Pressure Overload

Abstract:Objective: To investigate the effects of ligustrazine (Li) on myocardial fibrosis in rats with pressure overload.Methods: Pressure overload rat models were established by constricting the abdominal aorta. Sixty-three SD rats were divided into 3 groups: Sham operated group (SOG, n = 21), operated group (OG,n = 21) and operated combined with ligustrazine group (OG+Li,n = 21). Each group was randomly assigned to seven time points: The 1st, 2nd, 4th, 7th, 14th, 21st and 30th day after operation. Three rats were included in each time point. Serial sections of cardiac tissues were examined and the morphological or morphometric analysis of the SOG, OG and OG+Li done by histopathological and computer image analyzer technique.Results: (1) It showed that there were reactive fibrosis (from the 4th day on after operation) and reparative fibrosis (from the 21st day on after operation) in the OG, while myocardial fibrosis in the OG+Li was alleviated. Though reactive fibrosis (from the 21st day on after operation) was shown, reparative fibrosis wasn’t seen. (2) Perivascular collagen area (PVCA) in the OG (2. 09 ± 0. 45) was significantly higher than SOG (0.83 ± 0.06) from the 1st day on after operation and then steadily increased, while in the OG+Li (1.16 ± 0.06), it was significantly lower than OG at the same time; collagen volume fraction (CVF) in the OG (3.08 ± 0. 56) significantly increased compared with the SOG (2. 78 ± 0. 64) from the 2nd day on after operation and showed a trend of rapid ascending from the 21st day on after operation; and in the OG + Li (4.69 ± 0.85), it was significantly decreased compared with the OG (7.56 ± 0.88) from the 21st day on after operation, with allP<0. 05.Conclusion: Ligustrazine could alleviate and postpone the accumulation of myocardial collagen and has protective effects on the heart.

Abstract:A series of novel Ligustrazinyl amides was designed, synthesized and evaluated for their protective effect on the injured vascular endothelial cells. Abstract:A series of novel Ligustrazine derivatives was designed, synthesized, and assayed for their protective effects on damaged ECV-304 cells and antiplatelet aggregation activities. The results showed that most Ligustrazine derivatives exhibited lower EC50 values for protective effects on the ECV-304 cells damaged by hydrogen peroxide in comparison with Ligustrazine. And some Ligustrazine derivatives presented better antiplatelet aggregation activities than Ligustrazine. The derivatives containing the bisphenylmethyl pharmacophore (7a-c) exhibited highest potency. Compound 7a displayed most potential protective effects on the ECV-304 cells damaged by hydrogen peroxide, and compound 7c was found to be the most active antiplatelet aggregation agent. Abstract:Aim: To assess the effectiveness and safety of ligustrazine for treatment of cerebral infarction. Methods: All of the randomized or quasi-randomized trials involving ligustrazine for cerebral infarction were reviewed systemically with the method used in the international Cochrane network. Results: Among 441 pertinent studies, seven trials with poor methodological quality were included. There were no studies on the long-term effect of ligustrazine in treatment of cerebral infarction. Four of the 7 trials reported adverse effect with low incidence, including intracranial hemorrhage and epistaxis. As to the neurologic impairment at the end of treatment period, ligustrazine was associated with a significant improvement(RR 0.34,95% CI 0.25-0.48). Conclusion: There is insufficient reliable evidence for treatment of cerebral infarction using ligustrazine due to the low quality of the trials and possible publication bias. Further randomized, double blind, placebo-controlled trials are needed.
