Comparative Study of Cryopreservation of Human Red Blood Cells with Different Cryoprotective Agents

Abstract:To explore ideal cryoprotective agent(CPA) which can be used as substitute for glycerol in the cryopreservation of human red blood cells(RBCs) and can simplify the washing process of rewarmed RBCs.According to the relationship between the concentration of hemoglobin and the absorbance of its solution at 412nm,the percentage of recovered cells could be determined.Cryoprotective effects of five CPAs were evaluated.The results indicate that 35.5%(w/v) glycerol is the best CPA with RBC recovery of 97.84%,and then is 25%(w/v) detran -40 with recovery of 85.08%,and after that is 1M trehalose(incubation at 37℃ for 17.5h) with recovery of 55.35% and 1M trehalose(without incubation) with recovery less than 35%,and at last is 10%(w/v) dimethylsulfoxide(Me 2SO) also with recovery less than 35%.In conclusion,the detran-40 may be used as substitute for glycerol in the cryopreservation of human red blood cells.
