Abstract:CF4 is one of the representative and environmentally hazardous perfluorinated compounds. Even though their decomposition via a catalytic thermal process is an effective process for dissociating C-F bonds by employing catalysts with Lewis acid sites, the related mechanisms are unclear. Herein, we report a theoretical mechanism for the hydrolysis of tetrafluoromethane (CF4) on an gamma-alumina surface via density functional theory calculations. The overall reaction mechanism was divided into two steps: CF2O formation and CO2 formation. After the dissociative adsorption of CF4 to the strongest Lewis acid site on the surface of the catalyst, several pathways of defluorination reaction were investigated considering the presence of water and the order of reaction steps. Regardless of the reaction pathway considered, the rate-determining step of the overall reaction was found to be the first dissociation of the C-F bond in CF4, which has the highest energy barrier of 42.31 kcal/mol. Interestingly, oxygen vacancies were readily formed on the catalyst surface during several reaction pathways following its interaction with the reaction intermediate, and these sites could be replaced by fluorine atoms detached from CF4. Our findings suggest that facile desorption of HF, in addition to the presence of strong acidic sites on the catalyst surface for C-F bond dissociation, is critical for improving the CF4 hydrolysis.

Abstract:Due to the uniqueness of fluorine atom and C—F bond,difluoromethylene has special properties.As a bioisostere of an oxygen or a carbonyl group,it plays an important role in medicines,pesticides and materials.Difluoromethyl heteroaryl sulfones,represented by 2-PySO2CF2H (Hu reagent),have been developed recently as difluoromethylation reagents,and widely recognized by synthetic chemists for their ease of preparation,good functional group tolerance and universal applicability to a wide range of carbonyl compounds.Through different types of reactions such as nucleophilic substitution,nucleophilic addition,Julia-Kocienski olefination reaction,and radical-mediated difunctionalization,they introduced difluoromethyl,difluoromethylene,gem-difluoroalkene and other fluorine-containing groups into aldehydes,ketones,and heterocyclic compounds.For the first time,the synthesis of fluorine-containing organic compounds involved in various difluoromethyl heteroaryl sulfones in the past decade is reviewed from the perspective of reaction types and their application studies.

Abstract:Abstract Diazocarbonyl compounds are useful substrates in di- and trifunctionalization reactions based on F/CF3/SCF3 introduction. In the presented reactions, various electrophilic F/CF3/SCF3-transfer reagents were used. The majority of the reactions were based on rhodium catalysis and the application of various oxygen nucleophiles, such as alcohols, cyclic/acyclic ethers, and carboxylic acids. The oxyfluorination reactions were further developed to provide a new fluorine-18 labeling method. Density functional theory (DFT) modeling studies were performed to get a deeper mechanistic understanding of these reactions. These DFT modeling studies indicated that the catalytic reactions proceed through formation of rhodium carbene and oxonium ylide intermediates. The oxonium ylides undergo tautomerization to enol ether type species that subsequently react with the electrophilic F/CF3/SCF3-transfer reagents. We also present an arylation–trifluoromethylthiolation reaction for simultaneous introduction of C–SCF3 and C–C bonds into diazocarbonyl compounds. Abstract:Hexafluoroisopropanol (HFIP)-catalyzed direct dehydroxydifluoro-alkylation of benzylic and allylic alcohols with difluoroenoxysilanes is developed. This procedure enables the synthesis of a broad range of alpha,alpha-difluoroketones, a class of highly valuable intermediates and building blocks in medicinal and organic chemistry. Here, we have demonstrated for the first time that HFIP could act as a powerful catalyst for fluorinated carbon-carbon bond formation. The application of this protocol in late-stage dehydroxydifluoroalkylation of potentially bioactive drugs and natural products has also been carried out.

PhSO2 SCF2 H is a powerful reagent for radical difluoromethylthiolation of aryl and alkyl boronic acids, decarboxylative difluoromethylthiolation of aliphatic acids, and a phenylsulfonyl-difluoromethylthio difunctionalization of alkenes under mild reaction conditions.

Abstract:Since the days of the “Manhattan Project” our knowledge of the chemistry of fluorine‐con‐taining compounds has increased tremendously. At first only considered out of curiosity, this area of research bordering between inorganic and organic chemistry has developed from modest beginnings to such a complexity of interesting ramifications that it would be very difficult to review all aspects in detail. The motivating force behind the developments in this field were, above all, the unusual properties the numerous compounds acquire on introduction of fluorine substituents. The spectrum of unusual modes of behavior extend from extreme stabilization in fluorine‐containing polymers and blood‐substitutes to drastic increases in reactivity in pharmacologically and phytomedicinally active substances and in dyes. Moreover, one physical peculiarity of the fluorine atom, its magnetic moment, opens a new route to the decyphering of metabolic processes.—Parallel to the ever increasing number of fluorinated compounds there has been a concomitant increase in the variety of methods of synthesis and reagents, which is frequently difficult to overview in entirety, even for the specialists. Older, simple methods such as halogen metathesis with metal fluorides and electrofluorination are being refined as far as higher selectivity is concerned, and completely new, very reactive substances such as hypofluorites and noble‐gas fluorides enable fascinating reactions on complex substrates, e. g. steroids and nucleobases. The present review is an attempted documentation and classification of the methods as well as a critical appraisal, not least from the standpoint of efficiency and economic aspects. The spectrum of changes the fluorine substituents can bring about in organic molecules extends from extreme stabilization (e. g. in polymers and blood‐substitutes) to drastic increases in reactivity (e. g. in Pharmaceuticals).—A selection of fluorinating agents is given below.

Abstract:Reaction of potassium fluoride or tetramethylammonium fluoride with thiophosgene leads to the formation of a nucleophilic source of trifluoromethanethiolate, suitable for the preparation of trifluoromethyl aryl sulfides from activated haloaromatics. Analysis of the by-products in the system demonstrates that complex molecules with up to C4 chains may be formed by the reaction of fluoride salts with thiophosgene.

Abstract:A number of novel tetra-arylfluorophosphoranes have been synthesised. The most stable form of these compounds is the ionic form PR4+F–(R = aryl) although various other forms have been shown to exist including a molecular monomer PR4F and a dimer PR4+ PR4F2–. The i.r. spectra of the ionic forms reveal strong P+⋯ F– interactions. The fluorides are prepared via the hydrogendiflurides PR4+HF2– which contain distorted HF2– ions.

Abstract:Fluorinated molecules have a wide range of applications and are used as medicines, agrochemicals and refrigerants and in smartphone liquid crystal displays, photovoltaic solar cells, Teflon tapes and the coatings of textiles and buildings. Fluorination and fluoroalkylation — incorporation of a trifluoromethyl, difluoromethyl or monofluoromethyl group — are the major strategies used for the construction of carbon–fluorine bonds and fluorinated carbon–carbon bonds, respectively. The past two decades have witnessed a rapid growth in fluorination and fluoroalkylation methods thanks to the development of new reagents and catalysts. This Primer aims to provide an overview of state-of-the-art strategies in fluorination, trifluoromethylation, difluoromethylation and monofluoromethylation, with an emphasis on using C–H functionalization, although other strategies for fluorination and fluoroalkylation are also discussed. Further landmark achievements are expected in the fields of fluorination and fluoroalkylation as organofluorine compounds are used increasingly in everyday applications.

Abstract:Although defluorinative functionalization of trifluoromethylarenes has been studied by several research groups, different kinds of catalysts are requested accordingly based on specific substrate families to obtain corresponding RCF2Ar compounds. Herein, we report a general photocatalytic approach for selective hydrodefluorination (HDF) and defluoroalkylation (DFA) of ArCF3. Using electronically-different arenethiolates as photosensitizer, 88 gem-difluoromethyl compounds were synthesized from a broad range of electronically-variable trifluoromethylarenes, including natural products and bioactive molecules. Our mechanistic studies uncovered photo-induced Electron Donor-Acceptor (EDA) complexation followed by the silane-assisted C-F bond activation.

Abstract:A series of cloro- and nitrophenyl trifluoromethyl sulphides and sulphones have been synthesised from the corresponding aryl halides [1]. The fluorination of these compounds by tetra-n-butyl ammonium fluoride and potassium fluoride has been investigated. Abstract:Organofluorine compounds have greatly benefited the pharmaceutical, agrochemical, and materials sectors. However, they are plagued by concerns associated with PFAS. Additionally, the widespread use of the trifluoromethyl group is facing imminent regulatory scrutiny. Defluorinative functionalization, which converts the trifluoromethyl to the difluoromethyl motifs, represents the most efficient synthetic strategy. However, general methods for robust C(sp3)–F bond transformations remain elusive due to challenges in selectivity and functional group tolerance. Here, we present a method for C(sp3)–F bond defluorinative functionalization of the trifluoromethyl group via difluoromethyl anion in flow. This new approach tames the reactive difluoromethyl anion, enabling diverse functional group transformations. Our methodology offers a versatile platform for drug and agrochemical discovery, overcoming the limitations associated with fluorinated motifs.

Abstract:Trifluoromethane (HCF3) is a byproduct of fluoropolymer production that has limited applications. It is often stored or destroyed at the point of production, but if released into the environment is a potent greenhouse gas with global warming potential of 14,600 times that of CO2. State-of-the-art chemical technologies for upgrading HCF3 typically occur with conservation of the CF3 group. These approaches will come under increased scrutiny as concern over the environmental impact of perfluoroalkyl substances (PFAS) continues to grow. A more sustainable approach involves synthetic transformations that repurpose the atomic content of HCF3 while also destroying the CF3 group. In this paper, we report a rare example of the transformation of HCF3 into a fluoroalkene functional group through defluoroalkylation. We rationalise product formation through DFT calculations, scale-up the synthesis through continuous flow methods, and show that a fluoroalkene reagent derived from HCF3 is a competent nucleophile for the fluoroethenylation of a range of aldehydes
