Abstract:SUMMARY Azithromycin (AZI) is a broad-spectrum antibacterial drug popular in the market, however its bitterness seriously limited its application. Coating is the most common method used to obstruct the bitter drugs contact with the taste buds. In this study, a novel polymeric coating composition composed of Eudragit RL 30D and Eudragit L30D-55 was used to coat AZI pellets for taste-masking. Due to the enteric properties of the coating film, no drug was released intraorally (pH 5.0) until the environment pH was lowered to 1.0 (gastric environment). The result indicated the combination of these two acrylic polymers provided an applicable approach to prepare AZI taste-masked dosage forms. INTRODUCTION Azithromycin(AZI) is a new semi-synthetic macrolide antibiotic derived from erythromycin. It is popular in the antibiotic product market due to several advantages, including broad-spectrum activity, long elimination half-life, chemical and biological stability, few adverse effects, and short therapeutic time. However, AZI is terribly bitter which reduces the patient compliance and limits its application. Therefore, it is necessary to develop AZI taste-masked dosage forms. The ideal taste-masked dosage form can specifically inhibit the bad taste (such as bitterness), but not affect other taste percepts (such as sweetness), as well as not change the drug bioavailability. By far, the taste-masking mechanisms can be summarized as following: (1) adding flavor agents to make taste buds produce illusion; (2) avoiding direct contact of bitter drugs with taste buds; (3) making the taste buds reversible and temporary paralysis; and (4) competing the bitter receptors to block bitter signals to the brain. In this study, coating technique was used to achieve the purpose of taste-masking. At present acrylic resins are mainly used for thin film coating. Among them, Eudragit RL 30D is insoluble but swellable in water and independent of pH, while Eudragit L 30D-55 is an enteric material dissolving in medium with pH above 5.5. Previous study showed when the two polymers were combined at certain ratio, the film property could be shifted from enteric to acid soluble. Taking advantage of this nature, a polymeric coating composition was novelly used for task-masking by simply adding Eudragit RL 30D into Eudragit L 30D-55. EXPERIMENTAL METHODS AZI and Polyvinylpyrrolidone K-90 (PVP K-90) were dissolved in 70% ethanol and sprayed on the microcrystalline cellulose (MCC) pellet cores (0.15-0.20 mm in size) until 20 wt% drug loading was achieved. The drug-layered pellets were dried in oven at 40°C for 4 h, and then sieved manually to collect pellets in size of 0.20-0.25 mm. The surface of pellets was examined by scanning electron microscopy (SEM). Subsequently the drug-loaded pellets were coated in a fluidized bed (bottom spraying) with the combination of Eudragit RL 30D and Eudragit L30D-55 pre-mixed at different ratios. Finally, the coated pellets were dried in oven at 40°C for 4 h, and sieved manually for the product in size of 0.20-0.25 mm. The taste-masking effect of the coated pellets was evaluated through in vitro drug release studies in pH 5.0 PBS and pH 1.0 acid media, which simulated the oral and gastric environment respectively. RESULTS AND DISCUSSION The SEM photographs of the drug-loaded pellets (Figure 1) illustrated the pellets were in uniform size and spheric or spheroidal shapes, which are appropriate for homogeneous coating.
