Abstract:Two new steroid glycosides, named linckosides A and B, were isolated from the Okinawan starfish Linckia laevigata, and their stereostructures were elucidated by spectroscopic methods and chemical derivatization. Each of them possesses two monosaccharide units at C-3 and C-29 of a polyhydroxylated steroid aglycon. These steroid glycosides showed not only notable neuritogenic activity against PC12 cells but also significant synergistic effects on the NGF-induced neuronal differentiation of PC12 cells. Linckoside B showed higher activities than those of linckoside A, though the structural difference is only the kind of a sugar.

Abstract:Five new steroid glycosides, linckosides M-Q (1-5), and two known related metabolites, mimics and/or enhancers of the neuritogenic activity of nerve growth factor (NGF), have been isolated from the Okinawan blue starfish Linckia laevigata. Their structures, including stereochemistry, were elucidated by spectroscopic methods and chemical derivatization. The most active metabolite linckoside P (4) induced neurite outgrowth in 55% of PC12 cells 6 days after treatment. Linckosides M-O (1-3) were less active, in the range 21 to 32%. The monoglycosides, linckoside Q (5) and the two known metabolites, 6 and 7, were much less active than the bisglycosides. All the metabolites enhanced the neuritogenic activity of a trace amount of NGF, from 11% to 60-99% three days after treatment. These structure-activity relationships suggest the importance of xylose on a side chain, especially at the C-24 carbon branch rather than at the terminal C-26, for NGF-mimic activity.

The key steroid aglycon was achieved by a dimethylaluminum chloride-mediated ‘ene’ reaction. A versatile strategy was employed for the construction of glycosidic bonds through Schmidt's procedure using a glycosyl trichloroacetimidate donor.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:Novel steroid glycosides, acanthasterosides A1, B1, and B3, have been isolated from the crown-of-thorns starfish Acanthaster planci. Acanthasterosides B1 and B3 having two separated xyloses induced neurite outgrowth as like as nerve growth factor (NGF) in the rat pheochromocytoma cell line PC12, whereas acanthasteroside A1, having one xylose, did not induce neurite outgrowth. The acanthasteroside B3 induced neuritogenesis via the significant activation of p38 mitogen-activated protein kinase after the activation of the small G-protein Cdc42 rather than via Ras-MEK-ERK pathway that is predominantly activated by NGF. Following subcutaneous administration, acanthasteroside B3 attenuated cognitive impairment of senescence-accelerated mice (SAMP8) in two different cognitive tests. Liquid chromatography-mass spectrometry-assisted quantitative analysis demonstrated that acanthasteroside B3 could be transported into the brain via the circulatory system in mice. Thus, acanthasteroside B3 (and possibly B1) are a novel class of potential drug candidates for neurodegenerative diseases.

Abstract:Two new neolignans, manneoinsigins A (1) and B (2), together with four known lignans, were isolated from the leaves and stems of Manglietia insignis. The new compounds were established on the basis of extensive spectroscopic analyses. All compounds except 2 were tested for their cytotoxic activity. Compound 3 showed weak cytotoxic activity against the HL-60 human tumor cell line with the IC50 value of 23.5 µM.

Abstract:Background: Neurotrophic factors such as nerve growth factor (NGF) play important roles in nervous system. NGF is a potential therapeutic drug for treatment of neurodegenerative diseases. However, because of physicochemical property, NGF cannot pass through the blood-brain barrier (BBB). Hence, small molecules which exhibit NGF-mimic activity and can pass through the BBB are considered to be promising drug candidates for treatment of such diseases.Purpose: The present study was designed to isolate NGF-mimic substance from extract of natural products, determine their structures and investigate mechanism of action of the active substance.Methods: Extract of Lindernia crustacean was partitioned between water and ethyl acetate to obtain water layer and ethyl acetate layer samples, respectively, and then evaluated their neuritogenic activity in PC12 cells. The active sample was separated by open columns, followed by HPLC purification to obtain active compound. Then, specific inhibitors were used to investigate signaling pathway of neurite outgrowth induced by the active compound. Finally, western blot analysis was performed to confirm the pathway proposed by inhibitor experiments.Results: The ethyl acetate layer sample of extract of Lindernia crustacea exhibited significant neuritogenic activity. Two new compounds, named as linderside A and lindersin B, were isolated; their structures were elucidated by spectroscopic and chemical derivatization methods. Linderside A is a cucurbitane glycoside, whereas lindersin B is a cucurbitane triterpenoid. Each compound has an unusual isopentene unit, namely, a double bond bound to an unmodified isopropyl group at the end of cucurbitane triterpenoid side chain. Among them, lindersin B induced significant neurite outgrowth in PC12 cells, while linderside A was inactive against PC12 cells. Western blotting analysis results showed that lindersin B-induced neuritogenic activity depended on the activation of the mitogen-activated protein kinase (MAPK)/extracellular signal-regulated kinase (ERK). Moreover, tyrosine kinase A (TrKA) and phosphatidylinositol 3 kinase (PI3K) were also involved in the signaling pathway.Conclusions: Two new cucurbitane triterpenoids, linderside A and lindersin B, were isolated from Lindernia crustacean. Abstract:A new steroidal saponin was isolated from Ophiopogon japonicus. This saponin possesses a modification by 2-hydroxy-3-methylvalerylation of the hydroxyl group at C-4' of the sugar, linked to C-1 of the aglycone. It exhibited significant neuritogenic activity for PC12 cells. The structure-activity relationship revealed the aglycone, rather than the sugar moieties and acylation, to be important for the neuritogenic activity.

Abstract:Chemical investigation of the marine sponge Dysidea avara, collected from the South China Sea, yielded 13 steroids, including nine new (1-9) and four known (10-13) ones. The new structures were elucidated as (3S,14R)-3,14-dihydroxycholesta-5,8-dien-7-one (1), (22E,24R)-7 alpha-ethoxy-5 alpha,6 alpha-epoxyergosta-8(14),22-dien-3 beta-ol (2), 3 beta-hydroxy-7 alpha-ethoxy-5 alpha,6 alpha-epoxy-8(14)-cholestene (3), 3 beta,5 alpha-dihydroxy-6 alpha-ethoxychofesta-7,9(11)-diene (4), 3 beta,5 alpha-dihydroxy-6 beta-ethoxycholest-7-ene (5), (22E,24R)-24-ethoxy-3 beta,5 alpha-dihydroxy-6 beta-ethoxyergosta-7,22-diene (6), (22E)-3 beta,5 alpha-dihydroxy-6 beta-ethoxycholesta-7,22-diene (7), 24-ethoxy-3 beta,5 alpha-dihydroxy-6 beta-ethoxycholest-7-ene (8 and 9), by extensive spectroscopic analyses, such as HR-ESI-MS, 1D and 2D NMR data. The absolute configuration of 1 was assigned by comparison the experimental ECD spectra with the calculated ones. Among the 13 metabolites, compounds 1, 4, 11, 12, and 13 showed NF-kappa B inhibitory activities in human HER-293 cells with IC50 values of 6.4, 18.7, 8.1, 9.6, and 7.5 mu M, respectively. Preliminary structure-activity relationship analysis unveiled that the conjugated ketones or unsaturated double bonds might be the functional groups for the five active steroids.

Abstract:Two new alkyl 2,3-dihydroxybenzoates, gentisides A and B, were isolated from the traditional Chinese medicine Gentiana rigescens Franch. They showed a significant neuritogenic activity at 30μM against PC12 cells that was comparable to that seen for the best nerve growth factor concentration of 40ng/mL.

Abstract:In the present study, nine new cembrane-type diterpenoids, namely ximaonanolobatins A–I (1–9), together with six related known analogs (10–15) and four known tetraprenylated alkaloids malonganenones D, E, H, and K (16–19) were isolated from the soft coral Sinularia nanolobata collected off the Ximao Island in the South China Sea. The structures of 1–9 were robustly established by a combination of detailed spectroscopic analyses, chemical reactions, quantum mechanical (QM)-NMR methods, biogenetic consideration, and the comparison with those literature data. The absolute configuration of 1 was confirmed by X-ray diffraction analysis, and the absolute configuration of 2 was determined by QM-NMR calculations and chemical transformation. In addition, the absolute configuration of 3 was determined using the modified Mosher's method. All these isolates were evaluated for their anti-inflammatory and anti-tumor activities. The results showed that only compound 15 exhibited potential anti-inflammatory effect against LPS-induced TNF-α release in RAW264.7 macrophages with an IC50 value of 12.6 μM.This article is protected by copyright. Abstract:Nigrograna sp. LY66, an endophytic fungus associated with the herbal medicinal plant Clematis shensiensis , produced four undescribed steroids, nigergostanes A−D ( 1 − 4 ), including an unusual ketal-containing nigergostane ( 1 ), and four undescribed sesquiterpenoids decorated with cyclohexanone motifs, nigbisabolanes A−D ( 7 − 10 ), along with three known compounds, 23 R -hydroxy-(20 Z ,24 R )-ergosta-4,6,8(14),20(22)-tetraen-3-one ( 5 ), ergosta-5,7,22-trien-3β-ol ( 6 ), and curculonone A ( 11 ). The structures and absolute configurations of these undescribed compounds were confirmed using spectroscopic data (NMR and HRESIMS), modified Mosher's method, and ECD experiments. Additionally, compounds 5 and 8 displayed significant inhibition of nitric oxide generation in lipopolysaccharide-induced BV-2 microglial cells with IC 50 values of 2.8 and 2.7 μM, respectively, and is thus more potent than that of the positive control, quercetin (IC 50 = 8.77 μM). A molecular docking study revealed that 23-OH of 5 binds to the Y347 residue of inducible nitric oxide synthase (iNOS), whereas the 2-OH and 9,10-diol moieties of 8 bind to R381 and W463 and haeme residues of iNOS, respectively, which has rarely been reported in previous studies. These findings provide a set of undescribed lead compounds that can be developed into anti-neuroinflammatory agents.

Abstract:Further chemical investigation of the Xisha soft coral Litophyton nigrum has resulted in the isolation of four new nardosinane-type sesquiterpenoids, namely linardosinenes D–G (1–4). The structures of new compounds were elucidated by extensive analyses of their spectroscopic data and by comparison with the reported data of known related ones. All compounds exhibited weak inhibitory effect against bromodomain-containing protein 4 (BRD4), a promising therapeutic target in various human diseases, at a concentration of 10 μM.
