Abstract:Objective:To optimize the processes of volatile oils extraction and β-cyclodextrin (CD)inclusion from angelica and safflower of Zhepujiuweirunzao Tang.Methods:An orthogonal design involving the extracting rate of the volatile oils was designed to evaluate the optimal process of the volatile oils extraction. The process of the inclusion complex of β-CD was optimized based on the transfer and recovery rate of the volatile oils in the complex.Results:The optimal extraction outcome was as follows: the crude herbal materials were soaked into an aqueous solvent (the ratio of water to herbs 8∶1) for 1 hour. Following after the refluxing of extracting solution for 10 hours, the extraction rate of the volatile oils was 3.7 mg·g -1 ( n =6,RSD=10.2%). The optimal inclusion complex condition was as follows:the β-CD formed a complex with the volatile oils in a ratio of 6∶1 under the refluxing and stirring for 1 h at 60℃. The recovery rate of inclusion compounds and the transfer rate of the oils were 79.1% ( n =3,RSD=1.7%) and 89.9%( n =3,RSD=9.1%), respectively. A GC-MS study confirmed that the content of the volatile oils showed no significant difference post the inclusion complex compared with the one prior to the inclusion complex. Conclusion:This optimal processes to manufacture Zhepujiuweirunzao Tang are attainable.
