Abstract:The marriage of energy transfer with electrochemiluminescence has produced a new technology named electrochemiluminescence energy transfer (ECL-ET), which can realize effective and sensitive detection of biomolecules. To obtain optimal ECL-ET efficiency, perfect energy overlapped donor/acceptor pair is of great importance. Herein, we present a sensitive ECL-ET based immunosensor for the detection of tumor markers, using energy tunable CdSeTe/CdS/ZnS double shell quantum dots (QDs) and gold nanorods (GNRs) as the donor and acceptor, respectively. Firstly a facile microwave-assisted strategy for the synthesis of green- to near-infrared-emitting CdSeTe/CdS/ZnS QDs with time- and component-tunable photoluminescence was proposed. And, on the basis of the adjustable optical properties of both CdSeTe/CdS/ZnS QDs and GNRs, excellent overlap between donor emission and acceptor absorption can be obtained to ensure effective ECL-ET quenching, thus improving the sensing sensitivity. This method represents a novel approach for versatile detection of biomolecules at low concentrations.

Abstract:Improving the sensitivity of the fluorescence method is crucial in biosensor fields. In this study, an ultrasensitive and excellent selectivity method was developed for the detection of trace Cu2+ ions using 3-mercaptopropionic acid capped CdSSe/CdS/ZnS quantum dots (MPA-CdSSe/CdS/ZnS QDs) as a fluorescent probe. Through energy band engineering to optimize the interior inorganic structure and passivate the surface defects, the photoluminescence quantum yield of the MPA-CdSSe/CdS/ZnS QDs in aqueous solution had as high as 70 %. The X-ray diffraction (XRD) and Transmission Electron Microscope (TEM) analysis revealed the prepared CdSSe/CdS/ZnS QDs were cubic zinc blende structure with average diameter 7.2 nm. Further, the fluorescence of the MPA-CdSSe/CdS/ZnS QDs was significantly quenched by the Cu2+ with fast response time through an efficient photo-induced electron transfer (PET) progress. By optimizing the thickness of CdS shell, the MPA-CdSSe/CdS/ZnS QDs probe with appropriate CdS shell thickness exhibited excellent linear linearity and sensitivity for the detection of Cu2+ ions with limit of detection as low as 0.05 nM. Additionally, ions interference experiments demonstrated the good selectivity of the probe towards Cu2+. Furthermore, this sensing system could also be applied to the measurement of Cu2+ ions in real water samples with satisfactory recoveries. This work provided new insights to design and synthesize high-performance QDs-based fluorescence probes with controllable exciton emission behavior for their applications in biosensor and biological fields.
Efficient Enhancement of Electrochemiluminescence from Tin Disulfide Quantum Dots by Hollow Titanium Dioxide Spherical Shell for Highly Sensitive Detection of Chloramphenicol

Abstract:In this study, modified hollow titanium dioxide and SnS2 quantum dots (SnS2 QDs) were used to build a novel electrochemiluminescence immunoassay (ECLIA) for ultrasensitive detection of chloramphenicol (CAP). The titanium dioxide hollow spheres (THS) modified with polyethyleneimine (PEI) and gold nanoparticles (Au NPs) was synthesized as a probe material for the novel ECLIA, meanwhile it also could be a coreaction accelerator which could greatly improve the interaction efficiency between SnS2 QDs and S2O82-. Then the modified THS was combined with chloramphenicol antibody to form ECLIA probes, and colloidal gold synthesized as a platform for an ECLIA by ascorbic acid (AA) reduction. A competitive immunoassay strategy was used for the detection of CAP, where CAP in the sample would compete with the coating antigen for the limited antibodies. The proposed ECLIA for CAP detection exhibited high sensitivity with a wide linear range from 0.01 ng mL(-1) to 100 ng mL(-1) and a low detection limit at 3.1 pg mL(-1). Furthermore, it could be seen from electrochemical analysis that the effect of THS on the enhancement of ECL signal is about twice as high as that of ordinary titanium dioxide. Importantly, this work not only successfully applied THS to amplify electrochemical-luminescence signal in the ECLIA, but also successfully prepared a stable and highly selective sensor for ultrasensitive detection of CAP.
High-Efficient Energy Funneling Based on Electrochemiluminescence Resonance Energy Transfer in Graded-Gap Quantum Dots Bilayers for Immunoassay

Abstract:A surprising electrochemiluminescence (ECL) enhancement effect in graded-gap CdSeTe@ZnS-SiO2 quantum dot (QD) bilayers was observed and used to create an ultrasensitive immunoassay. CdSeTe@ZnS-SiO2 QDs of two different sizes were used as a donor-acceptor pair, owing to their tunable energy and low biotoxicity. The graded-gap CdSeTe@ZnS-SiO2 QD bilayers were fabricated by layer-by-layer assembly of differently sized CdSeTe@ZnS-SiO2 QDs on a glutaraldehyde-activated electrode. Benefiting from a short interlayer distance and perfect spectral overlap in the graded-gap QD bilayers, highly efficient ECL resonance energy transfer (ECLRET)-based energy funneling was observed, wherein excitons from trapped states could be effectively recycled. Consequently, the observed ECL enhancement for the bilayers was more than four times greater than that observed for reference samples. The graded-gap QD bilayers were utilized in an ECL biosensor for the detection of carcinoembryonic antigen (CEA). The proposed method featured a detection limit of 0.4 pg mL(-1) CEA with a linear calibration range from 1 pg mL(-1) to 200 ng mL(-1). This method represents a novel approach for versatile detection of biomolecules in research and clinical applications.
Anodic Electrochemiluminescence of Cdte Quantum Dots and Its Energy Transfer for Detection of Catechol Derivatives

Abstract:This work reported for the first time the anodic electrochemiluminescence (ECL) of CdTe quantum dots (QDs) in aqueous system and its analytical application based on the ECL energy transfer to analytes. The CdTe QDs were modified with mercaptopropionic acid to obtain water-soluble QDs and stable and intensive anodic ECL emission with a peak value at +1.17 V (vs Ag/AgCl) in pH 9.3 PBS at an indium tin oxide (ITO) electrode. The ECL emission was demonstrated to involve the participation of superoxide ion produced at the ITO surface, which could inject an electron into the 1Se quantum-confined orbital of CdTe to form QDs anions. The collision between these anions and the oxidation products of QDs led to the formation of the excited state of QDs and ECL emission. The ECL energy transfer from the excited CdTe QDs to quencher produced a novel methodology for detection of catechol derivatives. Using dopamine and L-adrenalin as model analytes, this ECL method showed wide linear ranges from 50 nM to 5 microM and 80 nM to 30 microM for these species. Both ascorbic acid and uric acid, which are common interferences, did not interfere with the detection of catechol derivatives in practical biological samples.

Abstract:A novel electrochemiluminescent (ECL) sensing system was constructed for low-potential detection of metal ion by immobilizing surface-unpassivated CdTe quantum dots (QDs) on a glassy carbon electrode. The surface-unpassivated CdTe QDs were prepared using meso-2,3-dimercaptosuccinic acid (DMSA) as a stabilizer to cap CdTe QDs and characterized with scanning electron micrograph and X-ray photoelectron spectroscopy. The immobilized QDs showed a strong cathodic ECL emission peak at -0.87 V with an onset potential at -0.64 V (vs Ag/AgCl/saturated KCl) in air-saturated, pH 9.0 HCl-Tris buffer. On the basis of the competition of metal ion to the stabilizer, the quenching effect of metal ion on ECL emission was observed, which led to a sensitive chemical sensing application. Using cupric cation as a model analyte, the sensor showed a linear range from 5.0 nM to 7.0 muM with a detection limit of 3.0 nM and had been successfully applied in the detection of copper in human hair. It could be extended to detect other metal ions with stronger metal-S interaction than with the Cd-S bond. As an example, the sensor could be used to detect Hg(2+) down to 1.4 nM. The bidentate chelate QD-based sensor exhibited a promising platform for rapid detection of cations with strong metal-S interaction and could be further applied for development of other low-potential electrochemical sensing systems.

Abstract:This work used sulfite as a coreactant to enhance the anodic electrochemiluminescence (ECL) of mercaptopropionic acid modified CdTe quantum dots (QDs). This strategy proposed the first coreactant anodic ECL of QDs and led to a sensitive ECL emission of QDs in aqueous solution at relatively low potential. In the presence of dissolved oxygen, the stable ECL emission resulted from the excited QDs. Thus, an ECL detection method was proposed at +0.90 V (vs Ag/AgCl) based on the quenching of excited QDs by the analyte. Using tyrosine as a model compound, whose electrooxidized product could quench the excited QDs and thus the ECL emission, an analytical method for detection of tyrosine in a wide concentration range was developed. Furthermore, by combining an enzymatic cycle of trace tyrosinase to produce the oxidized product with an energy-transfer process, an extremely sensitive method for ECL detection of tyrosine with a subpicomolar limit of detection was developed. The sulfite-enhanced anodic ECL emission provided an alternative for traditional ECL light emitters and a new methodology for extremely sensitive ECL detection of mono- and dihydroxybenzenes at relatively low anodic potential. This strategy could be easily realized and opened new avenues for the applications of QDs in ECL biosensing.

Abstract:Herein, an efficient electrochemiluminescence energy resonance transfer (ECL-RET) system was first applied in an immunosensor for the detection of chloramphenicol (CAP). SnS2 quantum dots (SnS2 QDs) has an extremely narrow electrochemical emission spectrum, which was used as donor. Compared with conventional quantum dots, toxic-element-free SnS2 QDs exhibit superior environmental friendliness and biocompatibility. Ultrathin Ag@Au nanosheets (Ag@Au NSs) was selected as acceptor due to its excellent structure and controllable UV-vis absorption range. Furthermore, ZnO nanoflowers (ZnO NFs) was employed to modify the electrode surface to provide stable luminescent environment and support more SnS(2 )QDs. Taking these advantages, the absorption spectra of Ag@Au NSs and the ECL emission spectra of SnS2 QDs are highly matched which ensures the occurrence of ECL-RET. When the CAP analyte were absent, Ag@Au NSs was immobilized on the electrode, and its ultrathin thickness effectively shortens the path of ECL-RET to produce the weak ECL intensity. Inversely, CAP analyte would compete with coating antigen for the certain amount of antibody to remove excess Ag@Au NSs and cause relatively high ECL intensity. As a result, the proposed immunosensor performed satisfactory sensitivity with a wide linear range from 0.005 to 1000 ng(-1) and a low detection limit (1.7 pg mL(-1)).

Abstract:A hemin functionalized graphene sheet was prepared via the noncovalent assembly of hemin on nitrogen-doped graphene. The graphene sheet could act as an oxygen reduction catalyst to produce sensitive electrochemiluminescent (ECL) quenching of quantum dots (QDs) due to the annihilation of dissolved oxygen, the ECL coreactant, by its electrocatalytic reduction. With the use of the catalyst with high loading of hemin as a signal tag of the secondary antibody, a novel ultrasensitive immunoassay method for biomarker detection was proposed. In an air-saturated pH 8.0 buffer, the immunosensor constructed by a stepwise immobilization of bidentate-chelated CdTe QDs and capture antibody showed an intensive cathodic ECL irradiation, which could be scavenged upon the formation of the catalyst-bound sandwich immunocomplex. With the use of the carcinoembryonic antigen as a model analyte, the immunoassay method showed a linear range from 0.1 pg mL(-1) to 10 ng mL(-1) and a detection limit of 24 fg mL(-1). The immunosensor exhibited good stability, acceptable fabrication reproducibility, and practicability. The electrocatalytic reduction-based ECL quenching strategy provided a powerful avenue for the design of the ultrasensitive detection method, showing great promise for clinical application.

Abstract:This work elucidated the detailed electrochemiluminescence (ECL) process of the thioglycolic acid-capped CdSe quantum dots (QDs) film/peroxide aqueous system. The QDs were first electrochemically reduced to form electronsinjected QDs similar to - 1. 1 V, which then reduced hydrogen peroxide to produce OH center dot radical. The intermediate OH center dot radical was a key species for producing holes-injected QDs. The ECL emission with a peak at -1.114 V was demonstrated to come from the 1S(e)-1S(h) transition emission. Using thiol compounds as the model molecules to annihilate the OH center dot radical, their quenching effects on ECL emission were studied. This effect led to a novel strategy for ECL sensing of the scavengers of hydroxyl radical. The detection results of thiol compounds showed high sensitivity, good precision, and acceptable accuracy, suggesting the promising application of the proposed method for quick detection of both scavengers and generators of hydroxyl radical in different fields.

Ultrasensitive and Selective Detection of Cu2+ in Aqueous Solution with Fluorescence Enhanced CdSe Quantum Dots

Abstract:Trace Cu2+ ions in aqueous solution were detected with ultra-sensitivity and excellent selectivity by using CdSe quantum dots (QDs) with enhanced fluorescence properties as fluorescent reporters. CdSe QDs were first synthesized by using aqueous route with 3-mercaptopropionic acid (3-MPA) as capping agent. By further thermal-annealing the as-prepared 3-MPA capped CdSe QDs in the presence of imidazole (IM) under nitrogen atmosphere at 70°C, the fluorescence quantum yields of CdSe QDs in aqueous solution could be dramatically enhanced to be ∼10.9%. These CdSe QDs with enhanced fluorescence exhibited fast fluorescence response (within 1min), ultra-sensitivity and excellent selectivity toward Cu2+ ions over other metal ions in aqueous solution. The Cu2+-induced aggregation of CdSe QDs led to the significant fluorescence quenching of the CdSe QDs. The limit of detection (LOD) calculated for Cu2+ could reach as low as 0.13–0.16ppb (2.0–2.5nM). The co-existence of other metal ions did not interfere with the sensing of trace Cu2+ ions in aqueous solution.
Quenching study of Cu2S-MPA/NGODs composites in electrochemiluminescence detection by modulating resonance energy transfer and adsorption process

Abstract:This study explores the principles of resonance energy transfer and adsorption modulation using composites of Cu2S-MPA/NGODs. These composites can efficiently control the quenching process of electrochemiluminescence (ECL). Mercaptopropionic acid (MPA) was added during the synthesis of Cu2S-MPA to enhance its attachment to nitrogen-doped graphene quantum dots (NGODs). The UV absorption peaks of NGODs coincided with the emission peaks of luminol ECL, enabling resonance energy transfer and enhancing the quenching capability of Cu2S-MPA. Meanwhile, there is another quenching strategy. When the readily reducible Cu+ ions underwent partial reduction to Cu when they were bound to NGODs. This weakened the electrocatalytic effect on reactive oxygen species (ROS) and had a detrimental impact on electron transfer. Under optimal conditions, the immunosensor ECL intensity decreased linearly with the logarithm of carcinoembryonic antigen (CEA) concentration in the range of 0.00001-40 ng/mL, with a detection limit of 0.269 fg/mL. The sensor was effectively utilized for the identification of CEA in actual serum samples.
Electrogenerated Chemiluminescence Resonance Energy Transfer Between Lucigenin and CdSe Quantum Dots in the Presence of Bromide and Its Sensing Application

Abstract:Electrogenerated chemiluminescence (ECL) of lucignein often suffered from weak signal at positive potential in neutral condition. In the present study, strong anodic ECL was obtained in neutral lucigenin solution at a CdSe quantum dots modified glassy carbon electrode. Electrochemical results suggested that CdSe quantum dots can catalyze the oxidation of lucigenin and bromide, which can generate the anodic ECL. The fluorescence and the ECL spectra revealed that ECL resonance energy transfer (ECL-RET) can occur between lucigenin and CdSe quantum dots. The oxidation product of bromide can promote ECL-RET and increase the anodic ECL signal significantly. The mechanisms of the anodic ECL based on the ECL-RET were proposed. Cytochrome C exhibited apparent inhibiting effect on the anodic ECL emission, based on which a sensitive ECL sensor for the detection of cytochrome C was established. (C) 2015 Elsevier B.V. Abstract:Bidentate chelation, meso-2,3-dimercaptosuccinic acid (DMSA), was used as a stabilizer for the synthesis of CdTe quantum dots (QDs). The bidentate chelate QDs, characterized with FT-IR, PL, and UV/Vis spectroscopy; element analysis; and high-resolution transmission electron microscope, exhibited surface traps due to the large surface/volume ratio of QD particle and the steric hindrance of the DMSA molecule. The unpassivated surface of the QDs produced a narrower band gap than the core and electrochemiluminescent (ECL) emission at relatively low cathodic potential. In air-saturated pH 7.0 buffer, the QDs immobilized on electrode surface showed an intense ECL emission peak at -0.85 V (vs. Ag/AgCl). H(2)O(2) produced from electrochemical reduction of dissolved oxygen was demonstrated to be the co-reactant, which avoided the need of strong oxidant as the co-reactant and produced a sensitive analytical method for peroxidase-related analytes. Using hydroquinone/horseradish peroxidase/H(2)O(2) as a model system, a new, reagentless, phenolic, ECL biosensor for hydroquinone was constructed, based on the quenching effect of ECL emission of QDs by consumption of co-reactant H(2)O(2). The biosensor showed a linear range of 0.2-10 μM with acceptable stability and reproducibility. This work opens new avenues in the search for new ECL emitters with excellent analytical performance and makes QDs a more attractive alternative in biosensing.

Abstract:In this work, oligonucleotide encapusulated silver nanoclusters were applied in the electrochemiluminescence (ECL) system of CdS nanocrystals (NCs)/ K2S2O8 based on dual ECL quenching effects. We found that the ECL emission of CdS NCs matched well with the absorption band of oligonucleotide encapsulated Ag nanoclusters, which could act as the energy acceptor of CdS NCs ECL so as to lead to an effective ECL resonance energy transfer (RET). On the other hand, the Ag nanoclusters could also catalyze electrochemical reduction of K2S2O8, resulting in increased consumption of ECL coreactant near the working electrode and decreased ECL intensity from CdS NCs. Based on the dual ECL quenching effects, a sensitive ECL biosensor for detection of microRNA was successfully achieved with a wide linear range from 10 fM to 100 pM.

Abstract:Outstanding photoluminescence (PL) and electro-luminescence properties of quantum dots (QDs) promise possibilities for them to meet challenging expectations of electrochemiluminescence (ECL), which at present relies on inefficient and spectral-irresolvable emitters based on transition-metal complexes (such as Ru(bpy)(3)(2+)). However, ECL is reported to be extremely sensitive to the surface traps on the QDs likely because of the spatially and temporally separated electrochemical charge injections. Results here reveal that, by engineering the interior inorganic structure (CdSe/CdS/ZnS core/shell/shell structure) and inorganic-organic interface using new synthetic methods, the trap-insensitive QDs with near-unity PL quantum yield and monoexponential PL decay dynamics in water generated narrow band-edge ECL with efficiencies about six orders of magnitude higher than that of the standard Ru(bpy)(3)(2+)). The band-edge and spectrally resolved ECL from CdSe/CdS/ZnS core/shell/shell QDs demonstrated a new readout scheme using electrochemical potential. Excellent ECL performance of QDs uncovered here offer opportunities to realize the full potential of ECL for biomedical detection and diagnosis.
