Abstract:Infrared and resonance Raman spectra of thianthrene (Th), perdeuteriated thianthrene ([2H8]Th) and their radical cation perchlorates (Th˙+ClO–4 and [2H8]Th˙+ClO–4, respectively) have been measured. Comparison of the frequency shifts caused by the perdeuteriation and the radical cation formation of thianthrene enables evaluation of the free energy change of 137 cal mol–1 for the electron-exchange equilibrium Th +[2H8]Th˙+⇌ Th˙++[2H8]Th.

Abstract:A simplified and appealing strategy via a chiral catalyst to facilitate the electrochemical process and provide asymmetric induction of radical reactions is particularly desirable and would have potential applications in electrosynthesis, organic chemistry, and drug discovery. Here, we introduced a novel electrolytic system that diverts the standard ionic reactivity to new catalytic functions, enabling mechanistically distinct single-electron transfer-based enantioselective routes to exhibit a brand-new pattern of reactivity-electricity-driven asymmetric catalysis as a privileged chiral platform for enantioselective radical allylation. The nickel-catalyzed activation of nucleophiles triggered a single-electron transfer process to provide a chiral catalyst-bound radical cation intermediate, which could be applied as an alternative strategy for the development of stereocontrolled radical reactions.

Abstract:Deuterium-incorporated compounds are of high interest owing to their importance in the pharmaceutical industry, organic synthesis and materials science. So far, the integration of deuterium into the inert, saturated magic methyl or methylene groups of covalent molecules remains challenging. Here, we present a 1,4-H delivery of allylic metallic species to provide a highly stereoselective and straightforward approach to 3-methyl-2( E )-enals or -enones from readily available 2,3-allenols and organoboronic acids. The reaction accommodates many synthetically versatile functional groups as well as multi-pharmacophores, and is not limited to the formation of 3-methyl derivatives. By applying 1,4-H or D delivery, deuterium atom(s) from differently deuterated allenols can be edited into the methyl or methylene groups of versatile organic skeletons, resulting in the efficient formation of 4-monodeuterated, 1,4- and 4,4-doubly deuterated, and 4,4,4-triply deuterated 2( E )-enals or -enones. These powerful platform molecules can provide straightforward paths to other deuterated compounds for different purposes.
Cesium Amide‐Catalyzed Selective Deuteration of Benzylic C‐H Bonds with D2 and Application for Tritiation of Pharmaceuticals

Abstract:AbstractHydrogen isotope exchange (HIE) represents one of the most attractive labeling methods to synthesize deuterium‐ and tritium‐labeled compounds. Catalytic HIE methods that enable site‐selective C−H bond activation and exchange labeling with gaseous isotopes D2 and T2 are of vital importance, in particular for high‐specific‐activity tritiation of pharmaceuticals. As part of our interest in exploring s‐block metals for catalytic transformations, we found CsN(SiMe3)2 to be an efficient catalyst for selective HIE of benzylic C−H bonds with D2 gas. The reaction proceeds through a kinetic deprotonative equilibrium that establishes an exchange pathway between C−H bonds and D2 gas. By virtue of multiple C−H bonds activation and high activity (isotope enrichment up to 99 %), the simple cesium amide catalyst provided a very powerful and practically convenient labeling protocol for synthesis of highly deuterated compounds and high‐specific‐activity tritiation of pharmaceuticals.

Abstract:Deuterium labeling compounds play a crucial role in organic and pharmaceutical chemistry. The synthesis of such compounds typically involves deuterated building blocks, allowing for the incorporation of deuterium atoms and functional groups into a target molecule in a single step. Unfortunately, the limited availability of synthetic approaches to deuterated synthons has impeded progress in this field. Here, we present an approach utilizing alkyl-substituted thianthrenium salts that efficiently and selectively introduce deuterium at the α position of alkyl chains through a pH-dependent HIE process, using D2O as the deuterium source. The resulting α-deuterated alkyl thianthrenium salts, which bear two deuterium atoms, exhibit excellent selectivity and deuterium incorporation in electrophilic substitution reactions. Through in situ formation of isotopically labelled alkyl halides, these thianthrenium salts demonstrate excellent compatibility in a series of metallaphotoredox cross-electrophile coupling with (hetero)aryl, alkenyl, alkyl bromides, and other alkyl thianthrenium salts. Our technique allows for a wide range of substrates, high deuterium incorporation, and precise control over the site of deuterium insertion within a molecule such as the benzyl position, allylic position, or any alkyl chain in between, as well as neighboring heteroatoms. This makes it invaluable for synthesizing various deuterium-labeled compounds, especially those with pharmaceutical significance.
Lithium isotope separation by precipitation method coupled with solvent extraction with 1-Hydroxy-4-(p-toluidino) anthraquinone

Abstract:This paper reported a novel chelating agent and a green precipitation-extraction separation strategy for lithium isotope enrichment. 1-Hydroxy-4-(p-toluidino) anthraquinone and anisole were used as chelating agent and diluent, respectively. Three phases including organic phase, aqueous phase and precipitation could be preparated. The experiment indicated that excess sodium hydroxide was helpful to promote the single-stage precipitation rate and equilibrium time. The isotope result indicated that 6 Li was concentrated in the aqueous phase and 7 Li was concentrated in precipitation phase. The maximum single-stage lithium isotope speration factor reached to 1.013 ± 0.001. By doing thermodynamic analysis of the reaction, the free energy change (ΔG), enthalpy change (ΔH) and entropy change(ΔS) of the separation process were −8.97 J mol −1 , −21.37 J mol −1 and −0.0438 J mol −1 K −1 at 283 K. The results of DFT simulation and wave function analysis indicated that a lithium ion combined with a chelating agent anion formed a stable six-member ring between two oxygen, which was more stable than sodium ions combined with chelating agent anions (the Gibbs free energy of the lithium ions reaction was more negative). As interaction analysis result showed, the Li-O bond in complex interaction was significantly stronger than the Li-O bond interaction in aqueous phase, which was the theroy that led to the enrichment of 7 Li in organic phase. In the activated carbon treatment experiment, it was found that part of lithium chelator solved in aqueous phase, which resulted in that the lithium abundance of aqueous phase hardly changed. In order to increase the abundance of 6 Li in aqueous phase, it is necessary to add adjuvant to reduce the water solubility of lithium chelate. Overall, an economical and environmental-friendly precipitation method with a certain application prospect for lithium isotope separation was proposed, and the direction of improving the separation performance was pointed out in this paper.

Biocatalytic Generation of Trifluoromethyl Radicals by Nonheme Iron Enzymes for Enantioselective Alkene Difunctionalization

Abstract:The trifluoromethyl (–CF3) group represents a highly prevalent functionality in pharmaceuticals. Over the past few decades, significant advances have been made in the development of synthetic methods for trifluoromethylation. In contrast, there are currently no metal-loenzymes known to catalyze the formation of C(sp3)‒CF3 bonds. In this work, we demonstrate that a nonheme iron enzyme, hydroxy-mandelate synthase from Amycolatopsis orientalis (AoHMS), is capable of generating CF3 radicals from hypervalent iodine(III) reagents and directing them for enantioselective alkene trifluoromethyl azidation. A high-throughput screening (HTS) platform based on Staudinger ligation was established, enabling the rapid evaluation of AoHMS variants for this abiological transformation. The final opti-mized variant accepts a range of alkene substrates, producing the trifluoromethyl azidation products in up to 73% yield and 96:4 enanti-omeric ratio (e.r.). The biocatalytic platform can be further extended to alkene pentafluoroethyl azidation and diazidation by altering the iodine(III) reagent. In addition, anion competition experiments provide insights into the radical rebound process for this abiological transformation. This study not only expands the catalytic repertoire of metalloenzymes for radical transformations but also creates a new enzymatic space for organofluorine synthesis.

Abstract:A practical and scalable ortho-selective deuteration of aromatic aldehydes was accomplished by Pd-catalyzed hydrogen isotope exchange with deuterium oxide as an inexpensive deuterium source. The use of tert-leucine as a transient directing group facilitates the exchange, affording a wide range of ortho-deuterated aromatic aldehydes with deuterium incorporation up to 97%. The control experiments suggest that the addition of silver trifluoroacetate resists the unexpected reduction of Pd(II), while the theoretical study indicates a rapid reversible concerted metalation-deprotonation process.

Abstract:Enzymes facilitate chemical conversionsthrough the integratedactivity of multicomponents, but the aggregation of multiactive siteswith molecular containers to emulate enzymatic conversion remainschallenging. Herein, we report a new Ce-III-based octahedralcage with chloride ligands that boosts C(sp (3))-H photoactivation by marrying a Ce-IV -Cl photocatalyst and photoredox catalyst tointegrate photoinduced electron transfer (PET), ligand-to-metal chargetransfer (LMCT), and hydrogen-atom transfer (HAT) events through multiphotonexcitation. This molecular cage binds 2-chloro-9H-thioxanthen-9-one (TXO) for boosting PET via a pseudointramolecularpathway to oxidize Ce-III -Cl to Ce-IV -Cl and form TXO & BULL;-. The LMCT excitation of Ce-IV -Cl istriggered to form Cl-& BULL; that activates C(sp (3))-H via HAT, resultingin the regeneration of Ce-III -Cl andthe formation of an alkyl radical. TXO & BULL;- reducesthe reaction intermediate, which transforms C(sp (3))-H and facilitates recovery of TXOfor subsequent recycling. This approach offers a feasible strategyfor creating highly electrophilic radicals resembling enzymes anddemonstrates their potential for C(sp (3))-H functionalization.
Precise Introduction of the -Chnx3-n (X = F, Cl, Br, I) Moiety to Target Molecules by a Radical Strategy: A Theoretical and Experimental Study

Abstract:Addition of halomethyl radicals to form bioactive molecules has recently become an efficient strategy. The reaction has a bottleneck, however, which is the effective and selective generation of the proper halomethyl •CHnX3-n radical by combining CHnX4-n with a carbon radical. Understanding the reactivity and selectivity of carbon radicals in the hydrogen atom transfer (HAT) and halogen atom transfer (XAT) reactions of CHnX4-n is key to the development of such an attractive method. With the help of the emerging data-driven strategy, DFT calculations were used to explore various correlations. For selectivity, the relative energy barriers between HAT and XAT reactions (ΔG⧧H - ΔG⧧X) correlate reasonably well with the three parameters ΔGH, ΔGX, and IP, and the correlation studies reveal that the calculated IPinver and the experimental ΔBDE can be used to conveniently predict the selectivity. Predicted selectivities are consistent with experimental determinations. This work not only provides a possibility for selecting carbon radicals with the known or easily obtained physicochemical data but also demonstrates that the informatic workflow such as generating data and identifying correlations has potential applications in mining reaction rules.

Abstract:Deuterium- and tritium-labeled compounds play a crucial role in various studies throughout the drug discovery and development process. Although numerous hydrogen isotope exchange (HIE) labeling methods have been developed for deuteration and tritiation of organic molecules, most of them relays heavily on transition metal catalysts. It still remains highly desirable to develop complementary and operationally simple methods to enable HIE of a wide range of C–H bonds while using readily accessible catalysts and isotope sources of deuterium and tritium. To this end, we report the deuteration and tritiation of arenes and heteroarenes, including pharmaceutical molecules, by alkali metal amide catalysts MN(SiMe3)2 (M = K, Rb, Cs). The reactions proceed through a kinetic deprotonative equilibrium that establishes an exchange pathway between C–H bonds and D2 or T2 gas. The use of alkali metal amides in combination with D2/T2 promoted HIE of both benzylic and aromatic C–H bonds of various arenes, and allowed high regioselectivity and high isotope enrichment (up to 99%) for a large scope of organic compounds and pharmaceuticals. In view of the abundance, low cost, and low toxicity of alkali metals and the broad availability of D2 and T2 gas, the present combination of alkali metal amide catalysts and D2/T2 provided a practical and sustainable protocol for HIE labeling method.

Abstract:Recently, hydrogen isotope exchange (HIE) reactions have experienced impressive development due to the growing importance of isotope containing compounds in various fields including materials and life sciences, in addition to their classical use for mechanistic studies in chemistry and biology. Tritium-labeled compounds are also of crucial interest to study the in vivo fate of a bioactive substance or in radioligand binding assays. Over the past few years, deuterium-labeled drugs have been extensively studied for the improvement of ADME (absorption, distribution, metabolism, excretion) properties of existing bioactive molecules as a consequence of the primary kinetic isotope effect. Furthermore, in the emergent "omic" fields, the need for new stable isotopically labeled internal standards (SILS) for quantitative GC- or LC-MS analyses is increasing. Because of their numerous applications, the development of powerful synthetic methods to access deuterated and tritiated molecules with either high isotope incorporation and/or selectivities is of paramount importance.HIE reactions allow a late-stage incorporation of hydrogen isotopes in a single synthetic step, thus representing an advantageous alternative to conventional multistep synthesis approaches which are time- and resource-consuming. Moreover, HIE reactions can be considered as the most fundamental C-H functionalization processes and are therefore of great interest for the chemists' community. Depending on the purpose, HIE reactions must either be highly regioselective or allow a maximal incorporation of hydrogen isotopes, sometimes both. In this context, metal-catalyzed HIE reactions are generally performed using either homogeneous or heterogeneous catalysis which may have considerable drawbacks including an insufficient isotope incorporation and a lack of chemo- and/or regioselectivity, respectively.Over the past 6 years, we have shown that nanocatalysis can be considered as a powerful tool to access complex labeled molecules (e.g., pharmaceuticals, peptides and oligonucleotides) via regio- and chemoselective or even enantiospecific labeling processes occurring at the surface of metallic nanoclusters (Ru or Ir). Numerous heterocyclic (both saturated and unsaturated) and acyclic scaffolds have been labeled with an impressive functional group tolerance, and highly deuterated compounds or high molar activity tritiated drugs have been obtained. An insight into mechanisms has also been provided by theoretical calculations to explain the regioselectivities of the isotope incorporation. Our studies have suggested that undisclosed key intermediates, including 4- and 5-membered dimetallacycles, account for the particular regioselectivities observed during the process, in contrast to the 5- or 6-membered metallacycle key intermediates usually encountered in homogeneous catalysis. These findings together with the important number of available coordination sites explain the compelling reactivity of metal nanoparticles, in between homogeneous and heterogeneous catalysis. They represent innovative tools combining the advantages of both methods for the isotopic labeling and activation of C-H bonds of complex molecules.

Abstract:The synthesis of isotopically labeled organic molecules is vital for drug and agrochemical discovery and development. Carbon isotope exchange is emerging as a leading method to generate carbon-labeled targets, which are sought due to their enhanced stability in biological systems. While many bioactive small molecules bear carbon-containing stereocenters, direct enantioselective carbon isotope exchange reactions have not been established. We describe the first example of an enantioselective carbon isotope exchange reaction, where (radio)labeled α-amino acids can be generated from their unlabeled precursors in a single step using a chiral aldehyde mediator with isotopically labeled CO2. Many proteinogenic and non-natural derivatives undergo enantioselective labeling, including the late-stage radiolabeling of complex drug targets.

Abstract:C-H functionalization provides unparalleled benefits for the late-stage modification of complex molecules. In recent years, photocatalysis has progressed significantly due to its mild conditions, sustainability and selectivities. Current mechanisms for photocatalytic C-H functionalization primarily involve direct single-electron oxidation and direct hydrogen atom transfer (d-HAT) by radicals, both of which are invalid for the functionalization of highly electron-deficient C(sp3)-H bonds. Here, we developed a cooperative photoenzymatic system consisting of a flavin-dependent ene-reductase (ER) and an exogenous photocatalyst fluorescein (FI) to achieve enantiodivergent functionalization of electron-deficient C(sp3)-H bonds. Mechanistic studies revealed a novel pathway for radical intermediate formation via excited-state FI*-induced single-electron oxidation of carbanions under alkaline conditions. The overall catalytic efficiency is enhanced by the electron transfer (ET) between FMNox and FI radical anion, while the stereoselectivity is controlled by ERs through enantioselective hydrogen atom transfer (HAT). This suggests that thermodynamically unfavorable single-electron oxidation of highly electron-deficient species can occur through ion-to-radical conversion. We anticipate that our novel mode of photoenzymatic catalysis will inspire new pathways for generating free radical intermediates and foster innovative strategies for achieving photoenzymatic new-to-nature reactions.

Abstract:A highly regioselective visible light photoredox-catalyzed hydrogen isotope exchange (HIE) of benzylic positions in both simple and complex molecules is reported. The process follows a dual catalytic approach using an acridinium photocatalyst in combination with a thiol-based hydrogen atom transfer catalyst, while the use of D2O as an isotope source ensures operational simplicity and cost-effectiveness. High reactivity has been achieved for electron-rich benzylic positions. Moreover, targeted radical formation enables unprecedented selective HIE on intramolecular competing benzylic and alpha to heteroatom positions with moderate to excellent deuterium incorporation. The utility of the reaction was demonstrated on the late-stage HIE of several natural compounds and drug derivatives. Experimental studies and density functional theory (DFT) calculations suggested a single electron transfer (SET) mechanism followed by deprotonation to generate the benzylic radical, and revealed the importance of halogenated solvents or additives. Upon a weak complexation of the halogenated species to the substrate, an oxidation potential lowering effect is induced, as well as a stabilization of the radical-cation species through spin delocalization.

Abstract:Site- and enantioselective incorporation of deuterium into organic compounds is of broad interest in organic synthesis, especially within the pharmaceutical industry. This metal-free platform enables four types of deuterofunctionalization reactions of exocyclic olefins and allows deuteration at non-benzylic positions with high levels of enantioselectivity and deuterium incorporation. Computational studies reveal that attractive non-covalent interactions are responsible for stereocontrol. We anticipate that our findings will open up new avenues for asymmetric deuteration.

Abstract:Ortho-selective aromatic C-H functionalization is frequently used in organic synthesis and chemical/pharmaceutical industries. However, this reaction relies heavily on the use of directing groups suffering from limited substrate scope and extra steps to put on and remove the directing/protecting groups. Herein we present the previously neglected concept that enables good to nearly complete selective ortho position. Proton transfer was utilized to tune the electron density on the aryl ring and determine the positional selectivity of electrophilic substitution. Consistently with deuteration experiments and DFT studies, this work demonstrates that acid-promoted proton transfer directs accelerated ortho-selective halogenation of NH/OH contained aromatic amines/phenols with excellent selectivity (>40 examples; up to 98:2 ortho/para selectivity). The application potential of this Fe-catalyzed method is demonstrated by the convenient synthesis of three alkaloids and tizanidine. This report raises the possibility that proton transfer could serve as the basis of developing new selective C-H functionalization reactions.
