Involvement of the Nogo-A/NgR Pathway in Neuroprotective Effect of Ischemic Preconditioning on Focal Ischemia in Rats

Abstract:Objective To explore involvement of the Nogo-A/NgR pathway in neuroprotective effect of ischemic preconditioning (IPC) on focal ischemia in rats. Methods Male SD rats were randomly assigned to 4 groups: control (C), IPC, Tat-NEP1-40 and Tat-β-Gal groups. Temporary middle cerebral artery occlusion (MCAO) for 10min was used for IPC. Transient MCAO (tMCAO) was established at 72h after IPC. The neurologic deficit scores (NDS) were assessed at 24h after reperfusion. The animals were then decapitated and brain infarct volumes were evaluated with 2% 2, 3, 5-triphenyltetrazolium chloride (TTC). The expression of Nogo-A and NgR were determined by confocal laser scanning microscope. Results The NDS in IPC and Tat-β-Gal groups were lower than that in controls (P<0.01 and P<0.05, respectively). The infarct volume in IPC and Tat-β-Gal groups were significantly smaller than that in controls at 24h after reperfusion (P<0.05). There was no difference between Tat-NEP1-40 and Control groups. Tat-NEP1-40 partially reversed the amelioration of NDS and the reduction in infarct volume that was induced by IPC after focal ischemia, where as the vehicle (Tat-β-Gal) did not affect the NDS and volume after ischemia. Few Nogo-A and NgR was found in non-ischemic brain tissue. Cerebral ischemia induced expression of Nogo-A and NgR, which was markedly repressed in rats treated with IPC. Conclusion IPC induces delay ischemic tolerance possibly via a partial involvement of the Nogo-A/NgR pathway in the brain.
