Molecular detection of Pseudomonas syringae pv.actinidiae on bacterial canker of kiwifruit

Abstract:Bacterial canker of kiwifruit is the main disease in kiwifruit production.In order to establish the rapid diagnostic technology,a 1 300 bp specific fragment was obtained and sequenced from pathogenic bacteria by RAPD analysis.Based on the sequences of the specific fragments,a pair of specific primers(F7/R7) was designed and synthesized.Then,the amplification conditions were optimized and the specificity and sensitivity of the specific primers were verified.After that the genomic DNA of 14 bacterial strains were amplified by the specific primer set.The results showed that a 950 bp band was only amplified from the strains of Pseudomonas syringae pv.actinidiae.In addition,detections for diseased kiwifruit trunk tissues collected from orchard and inoculated kiwifruit trunk indicated that the primers detected the existence of Pseudomonas syringae pv.actinidiae.The detection sensitivity of the primer set was 100 fg/μL in the fresh weight.Therefore,using the specific primer F7/R7 under the optimized PCR reaction conditions,combined with the simple method of reagent kit for DNA extraction,the molecular detection for the pathogen of kiwifruit canker can be completed in a short time.
