Effects of Different Levels of Daidzein and Genistein on Rumen Liquor of Dairy Cows by Using Short-Term Artificial Rumen Fermentation

Abstract:This study was carried out to investigate the effects of different levels of Daidzein and Genistein on rumen gas production and fermentation by using short-term artificial rumen fermentation,so as to determine the appropriate level of Daidzein and Genistein in diets.Rumen liquor was collected from 3 dairy cows before morning feed and divided into eleven groups,including the control group based on the basal diet,five Daidzein groups and five Genistein groups(with the level of 20,40,60,80 and 100 mg/kg respectively).The gas production was recorded at 2,4,8,12,24,36 and 48 h after fermentation.Fermentation liquor from all groups was sampled to determine pH,NH3-N,TVFA,MCP and protozo population 48 h after fermentation.The results showed that pH,propionate concentration,butyrate concentration,ratio of acetate to propionate ratio and protozo population were not influenced regularly by adding different levels of Daidzein or Genistein(P>0.05).Both Daidzein and Genistein with high level(100 mg/kg of Daidzein,80~100 mg/kg of Genistein) could significantly raise the NH3-N concentration(P<0.05) and reduce the acetic acid concentration(P<0.05),TVFA concentration(P<0.05) and MCP production(P<0.05).These results indicated that 20~40 mg/kg Daidzein and 20 mg/kg Genistein had promoting effects on rumen fermentation,but 100 mg/kg Daidzein and 80~100 mg/kg Genistein held back the rumen fermentation(P<0.05).
