The nature of stretching forces on osteoblast biology in vitro

Abstract:The culture of osteoblastic cells facilitates an in vitro system to clarify the complex integration of factors and mechanisms of the adaptive metabolism of bone to mechanical strains. Stretching force attracts more investigators ' attention because of its metabolic effects on osteoblast activity and function. Although the external forces applied directly on bones could be qualified, the state of the micro-mechano-environment around the osteoblasts in vivo is hardly to be estimated. Up to date, many loading models on osteoblastic cell culture have been developed to generate either uniaxial or biaxial stretches. Moreover, various loading parameters, such as the magnitude, waveform, frequency, cycle number and duration of loading were selected according to the investigators’ preferences. Although the results of those in vitro experiments were incomparable from one to another, some common mechanical induced characteristics of osteoblasts could be interpreted. In this paper,the properties of various mechanical loading devices and the nature of stretching forces on the proliferation and differentiation of osteoblasts were reviewed. Understanding of these properties could have important implications for the clinical management of bone regeneration and bone engineering.
