Abstract:Aim:To observe the safety and efficacy of donepezil alone and combined with nimodipine on the patients with moderate to severe Alzheimer's disease.Methods:32 patients with moderate to severe Alzheimer's disease were randomly divided into donepezil group and donepezil combined with nimodipine group.All the patients administered the drugs for 16 weeks.The efficacy was evalued by mini-mental state examination(MMSE),clinical dementia rating(CDR)and global deterioration scale(GDS).Results:The scores of MMSE and CDR were obviously improved in donepezil group and nimodipine combined treatment group. The patients in combined treated groups performed better in GDS than pretreatment.No patient dropped out because of the adverse effects.Condusion:Both donepezil and donepezil combined with nimodipine can siginificanfly improve the cognitive function of the patients with moderate to severe Alzheimer's disease.The effect of donepezil combined with nimodipine on GDS may be better than that of donepezil alone.The two drugs were well tolerated and safe.
