The aza-Diels–Alder reaction of Danishefsky’s diene with poly(ethylene glycol) (PEG)-bound imines derived from PEG-bound aldehydes, followed by cleavage from the support, afforded 2, 3-dihydro-4-pyridones. On the other hand, the Staudinger reaction of PEG-bound imines with ketenes generated in situ from acid chlorides, followed by cleavage from the PEG, gave cis-b-lactams. The products were obtained in good yields and purities.

Abstract:An efficient and general ionic liquid supported synthesis of cis-beta-lactam library via multistep reactions have been successfully carried out in a single ionic liquid batch. The method exhibited the advantages over soluble and insoluble polymeric support strategies, such as high loading capacity, avoiding of large excesses of reagents and easy purification. Abstract:Gamma-iminolactones 2, dihydroquinolin-2-ones 3, and gamma-lactames 4 have been synthesized starting from beta-hydroxymethylcyclopropanylamides 1, mediated by SnCl4/NaI/NEt3, BF3.OEt2, and TiCl4/NEt3. The corresponding products 2, 3, and 4 were produced, respectively, in high to excellent yields.

Abstract:4-Acyl-β-lactams are important synthetic intermediates in both pharmaceutical and organic chemistry. Cis - and trans -4-acyl-β-lactams were synthesized stereoselectively from vicinal diketones via the formation of bulky and less bulky diimines as key intermediates, respectively. The diimines reacted with acyl chloride in the presence of triethylamine to give rise to the corresponding 4-imino-β-lactams, which were further hydrolyzed to afford 4-acyl-β-lactams. The cis - and trans selectivity is depended on the steric hindrance of the imine N -substituents. A series of cis -4-acyl-β-lactams were synthesized from vicinal ketoaldehydes via the formation of their monoimines and diimines as intermediates. Pyruvic aldehyde produced cis -4-acetyl-β-lactams and cis -4-formyl-β-lactams, respectively, through the reactions of its monoimine and diimine with acyl chlorides. Phenylglyoxal generated cis -4-benzoyl-β-lactams via its monoaldimine.

Abstract:Photolysis of alpha-diazo-N-methoxy-N-methyl (Weinreb) beta-ketoamides derived from enantiomerically pure (EP) alpha-amino acids affords the corresponding EP beta-lactams via an intramolecular Wolff rearrangement. The photochemistry is promoted with either standard UV irradiation or through the use of a 100 W compact fluorescent light; the latter affords a safe and environmentally friendly alternative to standard photolysis conditions. A continuous-flow photochemical reactor made from inexpensive laboratory equipment reduced reaction times and was amenable to scale-up. The diastereoselectivity (cis or trans) of the product beta-lactams has been shown to vary from modest to nearly complete. An extremely facile, atom-economical method for the epimerization of the product mixture to the trans isomer, which is generally highly crystalline, has been developed. Evidence for C3 epimerization of Weinreb amide structures via a nonbasic, purely thermal route is presented. Subsequent transformations of both the Weinreb amide at C3 (beta-lactam numbering) and the amino acid side chain at C4 are well-tolerated, allowing for a versatile approach to diverse beta-lactam structures. The technology is showcased in the synthesis of a common intermediate used toward several carbapenem-derived structures starting from unfunctionalized aspartic acid.

Abstract:Catalyzed by Rhodococcus erythropolis AJ270, a nitrile hydratase and amidase containing microbial strain, under mild conditions, kinetic resolution of racemic amides provides an efficient and scalable route to highly enantioenriched R-4-carboxymethyl-β-lactams and S-4-carbamoylmethyl-β-lactams despite the substrates contain a stereogenic center that is β-positioned to amide functionality. Synthetic potential of the method was demonstrated by the constructions of novel β-lactam-fused heterocyclic compounds through convenient and practical chemical transformations.

Abstract:The photoinitiated deaminative [3 + 2] annulation reaction of N-aminopyridinium salts with alkenes for the synthesis of functionalized γ-lactams is described. This transformation shows good functional group tolerance as well as excellent diastereoselectivity. Preliminary studies suggest that the employed N-aminopyridinium salts generate the key amidyl radical intermediates through N–N bond cleavage via a photoinduced single-electron transfer (SET) process. The amidyl radical species would add to the double bond of alkenes, followed by a radical-mediated annulation process, to afford the desired γ-lactams.

Abstract:An N-heterocyclic carbene (NHC)-catalyzed highly diastereo- and enantioselective formal [3+2] reaction of o-hydroxy aromatic aldimines and cinnamaldehydes for the preparation of enantioenriched trans-γ-lactams was developed. An internal hydrogen bond between the o-hydroxy and the imine function was crucial for the reactivity and chemical selectivity. Trans-γ-lactam 3 d was easily oxidized to multifunctional 1,4-benzoquinone 8, which could further be transformed to biaryl 9 in the presence of a phosphoric acid. Finally, preliminary results for a kinetic resolution of (±)-trans-γ-lactam 3 d under asymmetric NHC catalysis are reported.

Abstract:γ-Lactam containing α,β-contiguous stereogenic centers stands out as a pivotal motif in various bioactive compounds, while its efficient synthesis still needs to be enhanced. Herein, an asymmetric C-H activation strategy for accessing α,β-stereospecific γ-lactams in good yields (≤79%) with high enantio- and diastereoselectivities (≤96% ee and >20:1 dr) was described, which serves as an effective supplement to the existing strategies.

Abstract:Solid-phase organic synthesis (SPOS) has become an effective synthetic tool for the preparation of combinatorial libraries of non-oligomeric small molecules. Owing to their high efficacy and extremely safe toxicological profile, beta-lactam antibiotics are the first choice for bacterial infectious diseases. Moreover, beta-lactam compounds have also showed other biological activities that include inhibition of prostate specific antigen, thrombin, human cytomegalovirus protein, human leukocyte elastase and cholesterol absorption. Thus, the application of combinatorial and related methodologies to the chemistry of the beta-lactam ring has been recognized as a very attractive challenge by different research groups around the world. This review covers the solid-phase and combinatorial chemistry related to mono-and multicyclic beta-lactam compounds that has been reported in the literature from 1999 to 2004.

Abstract:Lactam rings are found in many biologically active natural products and pharmaceuticals, including important classes of antibiotics. Methods for the asymmetric synthesis of these molecules are therefore highly desirable, particularly through the selective functionalization of unreactive aliphatic C–H bonds. Here we show the development of a strategy for the asymmetric synthesis of β-, γ- and δ-lactams via the haemoprotein-catalysed intramolecular C–H amidation of readily accessible dioxazolone reagents. Engineered myoglobin variants serve as excellent biocatalysts for this transformation, yielding the desired lactam products in high yields with high enantioselectivity and on a preparative scale. Mechanistic and computational studies were conducted to elucidate the nature of the C–H amidation and enantiodetermining steps and provide insights into the protein-mediated control of the regioselectivity and stereoselectivity. Additionally, an alkaloid natural product and a drug molecule were synthesized chemoenzymatically in fewer steps (7–8 versus 11–12) than previously reported, further demonstrating the power of biosynthetic strategies for the preparation of complex bioactive molecules.

Abstract:Optically pure lactams have a wide diversity of applications, one of which is serving as important intermediates in the synthesis of agricultural bioactive compounds or pharmaceuticals. Efficient stereoselective synthesis of chiral lactams with low-cost amine donors under mild conditions is desired for manufacturing of these important chemicals. Herein, a two-step chemo-enzymatic strategy has been designed to produce (S)-lactams by engineering a natural amine dehydrogenase from Thermoanaerobacter thermohydrosulfuricus (TtherAmDH). TtherAmDH was subjected to eight cycles of directed evolution to enhance its reductive amination activity toward keto acid substrates. The variants showed improved specific activity toward 6 tested substrates and gave high stereoselectivities of up to 99% in the asymmetric synthesis of gamma- and delta- lactams. In particular, TtherAmDH(V8) showed a 237-fold higher specific activity toward the model substrate levulinic acid and gave (S)-5-methyl-2-pyrrolidone in 99% ee with a space-time yield of 75.3 g L-1 d(-1). These results indicate that the engineered amine dehydrogenase TtherAmDH can serve as an efficient biocatalyst for the manufacture of highly valued chiral lactams.

Abstract:The reaction of vicinal diimines and acyl chlorides in the presence of triethylamine produces 3-imino-β-lactams and/or bis-β-lactams chemo-, regio-, and stereoselectively, which are important intermediates in pharmaceutical and organic synthesis. The selectivities in the reaction have been investigated. The results indicate that all diimines react with various ketenes generated from acyl chlorides in the presence of triethylamine to give rise to cis-4-imino-β-lactams (mono-cis-β-lactams) diastereoselectively due to the electron-withdrawing property of the imino group in the vicinal diimines. Bis-β-lactams were obtained from diimines via the mono-cis-β-lactams as intermediates. Only ketenes with strong electron-donating substituents can react with the mono-cis-β-lactams to yield bis-β-lactams, affording a pair of C2-symmetric cis-bis-β-lactams with symmetric diimines, two or four pairs of diastereomeric bis-β-lactams with ketoaldehyde-derived unsymmetric diimines depending on the steric hindrance of their N-substituents. The current investigation provides very important information for the selective preparation of mono- and bis-β-lactams from vicinal diimines.
